Construction and utilization of Fe3O4@Al3 + immobilized laccase for enhancing organic diethylstilbestrol removal: A multi-spectroscopy and molecular docking investigation.

As one of the endocrine-disrupting compounds (EDCs), diethylstilbestrol (DES) poses significant risks to ecosystems and human health. This study reports the development of a magnetic nanocomposite (Fe₃O₄@Al³⁺@laccase) via glutaraldehyde (GA) crosslinking to enhance DES degradation. The single factor experiments revealed that the optimum immobilization conditions were: temperature 40℃, pH 5, enzyme concentration 1.0 mg/mL, and immobilization time 1.0 h. Under these conditions, Fe₃O₄@Al³ ⁺@laccase achieved 91.71 %±2.95 % DES degradation within 12 h, as validated by HPLC, retaining 31.29 %±1.99 %

Two novel laccase-like nanozymes based on azole ligands for constructing pH-dependent sensor array for recognizing halogenated phenolic pollutants.

Halogenated phenolic compounds are widely used in industrial processes such as paper manufacturing and pesticide production. They are priority pollutants that need to be controlled and are discharged with industrial wastewater. Halogenated phenolics have much greater environmental impacts than phenolics due to elevated toxicity and reduced biodegradability caused by halogen substitution. Since the type and location of halogens have great influence on their toxicity and existing differentiation methods rely on large-scale instruments, there is an urgent need to develop new and convenient sensin

Facile Fabrication of 1-Methylimidazole/Cu Nanozyme with Enhanced Laccase Activity for Fast Degradation and Sensitive Detection of Phenol Compounds.

Facile construction of functional nanomaterials with laccase-like activity is important in sustainable chemistry since laccase is featured as an efficient and promising catalyst especially for phenolic degradation but still has the challenges of high cost, low activity, poor stability and unsatisfied recyclability. In this paper, we report a simple method to synthesize nanozymes with enhanced laccase-like activity by the self-assembly of copper ions with various imidazole derivatives. In the case of 1-methylimidazole as the ligand, the as-synthesized nanozyme (denoted as Cu-MIM) has the highes

Enzymatic degradation of ginkgolic acids by laccase immobilized on core/shell Fe3O4/nylon composite nanoparticles using novel coaxial electrospraying process.

Enzyme immobilization can increase enzyme reusability to reduce cost of industrial production. Ginkgo biloba leaf extract is commonly used for medical purposes, but it contains ginkgolic acid, which has negative effects on human health. Here, we report a novel approach to solve the problem by degrading the ginkgolic acid with immobilized-laccase, where core/shell composite nanoparticles prepared by coaxial electrospraying might be first applied to enzyme immobilization. The core/shell Fe3O4/nylon 6,6 composite nanoparticles (FNCNs) were prepared using one-step coaxial electrospraying and can b

Cellulose / waste Cu2+-activated carbon composite: A sustainable and green material for boosting laccase activity and degradation of bisphenol A in wastewater.

Low enzyme activity is one of the disadvantages of immobilized laccase. In this study, waste Cu2+-loaded activated carbon (Cu-AC) was successfully used in preparing a novel composite support, cellulose / Cu2+-loaded activated carbon beads (C / Cu-AC), and effectively boosted immobilized laccase activity. To achieve optimum conditions for immobilization of laccase, the immobilization time, pH and laccase concentration were examined. The highest immobilized laccase activity (34.21 U/g) was achieved under optimum conditions (T = 4 h, pH = 4, C = 5 g/L), which was increased by 35.86 % compared to c

Bio-Inspired Laccase-Mimicking Cu3-MOF Nanozyme as Colorimetric Sensor Array for Rapid Degradation and Visual Sensing of Phenolic Compounds.

Trinuclear copper complexes that emulate the active sites of multicopper oxidases (MCOs) are of broad biological interest. Here, a monoatomic-node strategy combined with solvent reduction was used to construct a rare trinuclear copper MOF featuring a planar, equilateral-triangular CuI 0.6CuII 2.4 core. The CuI 0.6CuII 2.4-MOF nanozyme shows enzymatic activity 37.2 times than that of laccase and 13.6 times than that of a CuII 3-MOF analogue, retains high chemical stability from pH 4 to 12, and lowers production cost by 16.2-fold relative to natural laccase. DFT calculations attribute the superi

The challenges associated with the practical application of natural laccase, such as poor reusability and intrinsic fragility, have driven the exploration of alternative solutions. In particular, cerium-based nanozymes emerge as promising candidates due to their high O2 mobility and inherent redox reactivity. However, there is a need for more in-depth and systematic research to enhance their catalytic activity and expand their application areas. Herein, a mesoporous cerium-based MOF material (MPUiO-66(Ce)) was synthesized with the cooperative assembly of amphiphilic molecular template and fram

Copper-manganese bimetallic oxide with excellent laccase-like activity for colorimetric detection of formaldehyde via the specific aldimine condensation reaction.

Laccase, known as the "green catalyst", holds significant promise for applications in the textile industry and pollutant detection. However, the use of natural laccase is constrained by challenges associated with complex preparation and inherent instability. The emergence of nanozymes provides a pathway for developing laccase-like mimics. This study focuses on the synthesis of copper-manganese oxide (Cu-doped Mn3O4) nanoparticles, which demonstrate significant laccase-like activity through the oxidation coupling reaction between 2,4-dichlorophenol (2,4-DCP) and 4-aminoantipyrine (4-AAP) with o

Optimization and characterization of laccase (LccH) produced by Hexagonia hirta MSF2 in solid-state fermentation using coir pith wastes (CPW).

The accumulation of coir pith waste, a byproduct of coconut husk processing, poses environmental and logistical challenges. An innovative and sustainable solution involves using coir pith as a substrate for solid-state fermentation (SSF). In SSF, coir pith can be converted into valuable products, such as enzymes, organic acids, and bioactive compounds. The present study aimed to evaluate laccase production by Hexagonia hirta MSF2 through SSF using the coir pith waste as substrate. Physico-chemical parameters like moisture, pH, temperature, C source, N source, and CuSO4 concentrations were pre-

Molecular Docking of Lac_CB10: Highlighting the Great Potential for Bioremediation of Recalcitrant Chemical Compounds by One Predicted Bacteroidetes CopA-Laccase.

Laccases are multicopper oxidases (MCOs) with a broad application spectrum, particularly in second-generation ethanol biotechnology and the bioremediation of xenobiotics and other highly recalcitrant compounds. Synthetic pesticides are xenobiotics with long environmental persistence, and the search for their effective bioremediation has mobilized the scientific community. Antibiotics, in turn, can pose severe risks for the emergence of multidrug-resistant microorganisms, as their frequent use for medical and veterinary purposes can generate constant selective pressure on the microbiota of urba

Exploring the potential of Halalkalibacterium halodurans laccase for endosulfan and chlorophacinone degradation: insights from molecular docking and molecular dynamics simulations.

Pesticides are widely used in agriculture but at the same time, a majority of them are known to cause serious harm to health and the environment. In the recent past, laccases have been reported as key enzymes having the ability to degrade pollutants by converting them into less toxic forms. In this investigation, laccase from polyextremophilic bacterium Halalkalibacterium halodurans C-125 was analyzed for its structural, physicochemical, and functional characterization using in silico approaches. The 3D model of the said enzyme is unknown; therefore, the model was generated by template-indepen

Comprehensive Analysis of Rice Laccase Gene (OsLAC) Family and Ectopic Expression of OsLAC10 Enhances Tolerance to Copper Stress in Arabidopsis.

Laccases are encoded by a multigene family and widely distributed in plant genomes where they play roles oxidizing monolignols to produce higher-order lignin involved in plant development and stress responses. We identified 30 laccase genes (OsLACs) from rice, which can be divided into five subfamilies, mostly expressed during early development of the endosperm, growing roots, and stems. OsLACs can be induced by hormones, salt, drought, and heavy metals stresses. The expression level of OsLAC10 increased 1200-fold after treatment with 20 μM Cu for 12 h. The laccase activities of OsLAC10 were c

A Self-Unlocking Biocatalytic Cascade Enabled by a Laccase-Entrapping Metal-Organic Framework for Point-of-Care Detection of Alkaline Phosphatase.

The point-of-care testing (POCT) for alkaline phosphatase (ALP), a critical biomarker for hepatobiliary diseases, bone disorders, and specific malignancies, remains analytically challenging due to signal interference from complex biological matrices. Herein, we report the development of a laccase-entrapping zeolitic imidazolate framework-8 nanoprobe (LAC@ZIF-8) that enables a self-unlocking biocatalytic cascade (SUBC) for the selective colorimetric detection of ALP. The SUBC mechanism proceeds via three discrete steps: (1) ALP-catalyzed hydrolysis of disodium phenyl phosphate yields phenol and

Wet-chemical synthesis of AgCo@CQAS nanozyme: a novel high-activity copper-free laccase mimic for sensitive hydrogel-based pesticide detection.

Beyond peroxidase- and oxidase-like activities, the laccase-like activity of nanozymes has gradually gained significant attention. However, the catalytic performance of current copper-containing or non-copper laccase mimics remains relatively low, limiting their broader application. Inspired by the structure and properties of natural laccase, this study employed silver (Ag) and cobalt (Co) as catalytic active center, Prussian blue analog K3Co(CN)6 as ligand, and chitosan quaternary ammonium salt (CQAS) as electron transmitter, developed a novel copper-free laccase mimic, AgCo@CQAS, via a wet-c
