Experimental infection of Mystromys albicaudatus with Leishmania braziliensis: pathology.

Thirty-four Mystromys albicaudatus were injected intradermally with Leishmania braziliensis promastigotes and examined and killed during a 12-week period. All animals developed single cutaneous lesions at the sites of inoculation that began as a papular thickening in the dermis and progressed to a 2.0 cm crateriform ulcer. The histopathology of these lesions was characterized by granuloma formation and diffuse infiltrates of lymphocytes and plasma cells. Parasites were most frequently observed in vacuolated macrophages immediately underlying the necrotic debris of the ulcer. Histiocytes in whi

Control of phlebotomine sandflies in confined spaces using diffusible repellents and insecticides.

The control of phlebotomine sandflies (Diptera: Psychodidae), the vectors of leishmaniasis, is directed mostly against adults as larvae develop in unknown or inaccessible habitats. In the current study we tested geraniol, a natural plant-derived product, as a space repellent and the synthetic pyrethroid prallethrin as a diffusible insecticide. Geraniol was dispersed in the air using diffusers with an electric fan and prallethrin was evaporated using electrically heated evaporators. Both substances were tested in inhabited bedrooms and in tents. Geraniol failed to effect significant reductions

Lipsosomal amphotericin B: a review of its properties, function, and use for treatment of cutaneous leishmaniasis.

The genus Leishmania includes a number of protozoan parasites that cause a wide range of infections named leishmaniasis. Leishmaniasis may be appear in three clinical forms - cutaneous (CL), visceral, and mucocutaneous (MCL) - with variation in their presentation and severity: diffuse CL and post-kala-azar dermal leishmaniasis). The prevalent signs of CL are nonhealing ulcers on exposed skin, but infected patients may have other dermatologic symptoms. In the 1960s, amphotericin B deoxycholate was introduced as a second-line therapy for CL and MCL. However, widespread administration of the agen

Histological spectrum of cutaneous leishmaniasis due to Leishmania tropica.

Skin biopsies from 117 cases of acute and 23 cases of lupoid leishmaniasis were studied. Of the 117 acute cases, anergic macrophage response were seen in 44, diffuse necrosis in 9 and focalized necrosis in 11 biopsies; the remaining 53 biopsies showed scanty or no parasites with a few to abundant epithelioid cells probably representing post necrotic changes before complete healing of the lesions. The 23 biopsies of lupoid leishmaniasis showed rather well organized epithelioid granulomata surrounded by lymphocytes, inconspicuous plasma cells, no amastigotes and no necrosis. Lupoid cases showed

Antigenic profiling of Leishmania amazonensis microsomal fraction to support vaccine development.

Vaccination remains the most promising long-term strategy for controlling leishmaniasis, as individuals who recover from primary infection generally develop protective immunity. However, no vaccine is currently available for human use. In this study, we identified two immunogenic proteins that may account for the protective effect previously observed with the Leishmania amazonensis microsomal fraction (MicF), which confers protection in murine models. Both proteins were expressed in Escherichia coli and used to immunize mice, eliciting specific humoral responses and promoting elevated producti

[Histopathology of mucocutaneous leishmaniasis caused by Leishmania (Vianna) braziliensis].

A histopathological study of mucocutaneous leishmaniasis due to Leishmania (Viannia) braziliensis was carried out on 28 cutaneous and 114 mucosal biopsies, taken from Bolivian and Peruvian patients. This study showed similar histopathological findings in cutaneous and mucosal lesions. The cutaneous biopsies showed a strong epidermal hyperplasia occasionnally budding in the dermis. In the ulcerative area, the epidermis was totally necrosed and replaced by a fibrino-leucocytic edge. In the dermis, histio-lympho-plasmocytic infiltration was constantly found. The histiocytes often gathered in foll

Review of the current treatments for leishmaniases.

Leishmaniases are vector-borne zoonotic diseases that are prevalent in tropical and subtropical areas in the world, with two million new cases occurring yearly. Visceral and tegumentary forms of leishmaniasis are known. The latter form may present as localized cutaneous or mucosal forms, disseminated, diffuse forms, or leishmaniasis recidiva cutis. Visceral leishmaniasis is caused by parasites of the species Leishmania (Leishmania) donovani and L. (L.) infantum, and tegumentary leishmaniasis is caused by 15 other species, with distinct distributions in the Old and New World. The varied clinica

Genetic variability within the species Leishmania aethiopica does not correlate with clinical variations of cutaneous leishmaniasis.

Leishmania aethiopica infections in man result in a spectrum of diseases from LCL to DCL. These clinical manifestations have been attributed to genetic differences within the host or the parasites. In this study two different PCR-based methods were used to elucidate genetic variation within the species L. aethiopica. Inter- and intra-specific variations were detected in the ITS of the ribosomal operon in different strains and species of Leishmania, using a PCR-RFLP approach, and by a PCR fingerprinting technique that used single non-specific primers to amplify polymorphic regions of the genomi

The cutaneous lesion in American leishmaniasis: leukocyte subsets, cellular interaction and cytokine production.

Interactions between immunocompetent cells require the participation of T cell antigen receptor (TCR) and the integrin lymphocyte function-associated molecule-1 (LFA-1, CD11a/CD18). These interactions are mediated by interlinking cytokines, which are important in determining the type of immune response. In the present study, we have shown that in American cutaneous leishmaniasis (ACL) lesions, most infiltrating T cells expressed the alpha beta TCR including those selectively migrating to the epidermis. In contrast, gamma delta T cells were abundant in localized (LCL) and scarce in muco-cutaneo

Sugar-based colloidal nanocarriers for topical meglumine antimoniate application to cutaneous leishmaniasis treatment: Ex vivo cutaneous retention and in vivo evaluation.

The leishmaniases are a group of diseases caused by protozoan parasites from Leishmania species. Effectiveness therapies for cutaneous leishmaniasis (CL), the most common form, are still needed to be developed since the available drugs such as meglumine antimoniate (MA) present severe adverse reactions. Here, we develop and characterize maltodextrin polymeric colloidal nanocarriers containing MA (PCN-MA) for topical CL treatment. PCN-MA is composed of 5 to 8% maltodextrin, 0.3% NaCl, 1% MA in 21% of water as aqueous-internal phase, containing or no 3% Kolliphor® P-188, and 10% SF1540 dispersed

Limonene exerts antileishmanial effects via mevalonate pathway in Leishmania amazonensis.

Limonene, a monoterpene abundant in essential oils of citrus plants, exhibits well-documented activity against both promastigote and amastigote forms of Leishmania in vitro and in vivo. Leishmania (L.) amazonensis is one of the etiological agents of localized and diffuse cutaneous leishmaniasis in the Americas, clinical forms frequently associated with limited therapeutic efficacy and recurrent treatment failure. Here, we show that limonene inhibits the biosynthesis of key isoprenoids in L. amazonensis promastigotes, including ubiquinone, dolichol, and ergosterol. Mechanistically, limonene dis

Lipophosphoglycans from Leishmania amazonensis Strains Display Immunomodulatory Properties via TLR4 and Do Not Affect Sand Fly Infection.

The immunomodulatory properties of lipophosphoglycans (LPG) from New World species of Leishmania have been assessed in Leishmania infantum and Leishmania braziliensis, the causative agents of visceral and cutaneous leishmaniasis, respectively. This glycoconjugate is highly polymorphic among species with variation in sugars that branch off the conserved Gal(β1,4)Man(α1)-PO4 backbone of repeat units. Here, the immunomodulatory activity of LPGs from Leishmania amazonensis, the causative agent of diffuse cutaneous leishmaniasis, was evaluated in two strains from Brazil. One strain (PH8) was origin

Successful treatment with miltefosine of disseminated cutaneous leishmaniasis in a severely immunocompromised patient infected with HIV-1.

We describe here a case of disseminated cutaneous leishmaniasis due to Leishmania major in a severely immunocompromised patient from Burkino Faso, Africa, who is infected with human immunodeficiency virus-1. The skin lesions failed to respond to full treatment courses of amphotericin B, sodium stibogluconate, and liposomal amphotericin B but were successfully treated with miltefosine, an alkylphosphocholine analogue.

The Extraordinary Case of a Woman with a 30-Year-Long Diffuse Leishmaniasis Cured with One Single Ampoule of Intranasal Pentavalent Antimoniate.

Infection with Leishmania amazonensis and L. mexicana may lead to diffuse cutaneous leishmaniasis. The cure is exceptional, especially for the strange case of this lady. Case report: The patient acquired the disease in childhood and remained with lesions for over 30 years, albeit several treatments. She worsened after a pregnancy, developing disseminated lesions. Miltefosine with amphotericin B and pentamidine resulted in remission. Lesions reappeared after one year, accompanied by intra-nasal infiltration of the disease. The nasal spraying of a single ampoule of pentavalent antimoniate result

Effect of Two Different Isolates of Leishmania mexicana in the Production of Cytokines and Phagocytosis by Murine Dendritic Cells.

Species of the genus Leishmania are the causal agents of leishmaniasis, a disease with diametrically different clinical manifestations that have been attributed to the species and host immune response. Some Leishmania species, including Leishmania mexicana, are capable of causing both localized cutaneous leishmaniasis (LCL) and diffuse cutaneous leishmaniasis (DCL). Therefore, it is possible that intraspecific differences may exist that contribute to the development of distinct clinical forms. Dendritic cells (DC) are important host cells of Leishmania spp. parasites, and cytokine production a

Photodynamic treatment of cutaneous leishmaniasis.

Leishmaniasis is a widespread arthropod-borne protozoan zoonosis caused by more than 21 Leishmania species. Vectors are sandflies of different genera. The disease is classified into "Old World" versus "New World" leishmaniasis and further subclassified in cutaneous, mucocutaneous and visceral forms. Most therapeutic approaches are not evidence-based. We report a patient with facial cutaneous Leishmania tropica infection which proved to be resistant to various therapeutic regimes. Excellent results were achieved with photodynamic therapy.

Potentiating action of rifampicin and isoniazid against Leishmania mexicana amazonensis.

The treatment of the complex of diseases known as leishmaniasis, caused by infection with protozoal parasites, is often unsatisfactory. The response to chemotherapy depends not only on the species of infecting Leishmania but also on the immune reactions of the host. This creates difficulties in evaluating any new drug in clinical trials. In a Brazilian patient with diffuse cutaneous leishmaniasis, which classical antileishmanial drugs had failed to cure, antituberculous therapy of an intercurrent mycobacterial infection produced a striking remission of the leishmanial skin lesions.
