Tissue-dependent effects of cis-9,trans-11- and trans-10,cis-12-CLA isomers on glucose and lipid metabolism in adult male mice.

Mixtures of the two major conjugated linoleic acid (CLA) isomers trans-10,cis-12-CLA and cis-9,trans-11-CLA are used as over the counter supplements for weight loss. Because of the reported adverse effects of CLA on insulin sensitivity in some mouse studies, we sought to compare the impact of dietary t10c12-CLA and c9t11-CLA on liver, adipose tissue, and systemic metabolism of adult lean mice. We fed 8 week-old C57Bl/6J male mice with low fat diets (10.5% Kcal from fat) containing 0.8% t10c12-CLA or c9t11-CLA for 9 or 38 days. Diets containing c9t11-CLA had minimal impact on the endpoints stud

Convergence of biological nitration and nitrosation via symmetrical nitrous anhydride.

The current perspective holds that the generation of secondary signaling mediators from nitrite (NO2(-)) requires acidification to nitrous acid (HNO2) or metal catalysis. Herein, the use of stable isotope-labeled NO2(-) and LC-MS/MS analysis of products reveals that NO2(-) also participates in fatty acid nitration and thiol S-nitrosation at neutral pH. These reactions occur in the absence of metal centers and are stimulated by autoxidation of nitric oxide ((•)NO) via the formation of symmetrical dinitrogen trioxide (nitrous anhydride, symN2O3). Although theoretical models have predicted physio

Microbial Metabolic Messengers from Fermented Foods: Multi-Organ Regulation of Energy Homeostasis and Obesity.

Mechanistic evidence was systematically reviewed on how major metabolites produced during fermentation of foods modulate obesity through gut microbiota remodeling and multiorgan metabolic regulation, with emphasis on the gut-brain axis, immunometabolic pathways, and epigenetic mechanisms. Obesity remains a major global public health challenge, and conventional management strategies are frequently limited by poor long-term efficacy, low adherence, and undesirable side effects. Increasing evidence positions the gut microbiome as a central metabolic organ that profoundly influences host energy ho

Deciphering the Relationship Between Circulating Metabolites and Osteoarthritis: A Comprehensive Genetic Correlation and Mendelian Randomization Studies.

The causal impact of blood metabolites on OA has yet to be definitively established, further studies are needed to explore the specific roles of metabolites in OA. This is a genetic correlation and two-sample bidirectional mendelian randomization study. GWAS summary data of metabolites and OA were extracted from large-scale GWAS study based on Europeans and Asians. LDSC was conducted to estimate the genetic correlations between 233 circulating metabolites and 11 OA phenotypes, MR was then performed to explore the casual association. 41.20% of the metabolic traits showed genetic correlation wit

High CLA-producing Lactiplantibacillus plantarum MNN1 alleviates irritable bowel syndrome through activation of intestinal PPARα signaling pathway.

Irritable bowel syndrome (IBS) is a common gut-brain axis disorder characterized by visceral hypersensitivity, abdominal pain, and anxiety. Although probiotics have emerged as a promising strategy for IBS management, the specific metabolite-driven mechanisms and their role in gut-brain axis modulation remain largely unclear. In this study, 18 Lactiplantibacillus plantarum strains were screened for conjugated linoleic acid (CLA) production, and strain MNN1 was identified as a high-CLA producer, with a yield of 426 μmol/L, approximately threefold higher than that of the reference strain WCFS1. Effect of feeding increasing levels of whole cottonseed on milk production, milk components and milk fatty acid profile in lactating dairy goats.

We assessed the feeding behavior, intake, apparent total tract digestibility, blood parameters, milk yield, milk composition and milk fatty acid profile in dairy goats fed diets containing increasing levels of whole cottonseed (WCS), specifically a control diet with no WCS and diets containing 120, 150 and 180 g WCS/kg DM. Eight lactating dairy goats were distributed in a replicated 4 × 4 Latin square design with four 21-day periods. Rumination and total chewing time increased linearly with WCS inclusion levels. However, no effects on dry matter intake or digestibility were observed. Milk prod

Pasture v. standard dairy cream in high-fat diet-fed mice: improved metabolic outcomes and stronger intestinal barrier.

Dairy products derived from the milk of cows fed in pastures are characterised by higher amounts of conjugated linoleic acid and α-linolenic acid (ALA), and several studies have shown their ability to reduce cardiovascular risk. However, their specific metabolic effects compared with standard dairy in a high-fat diet (HFD) context remain largely unknown; this is what we determined in the present study with a focus on the metabolic and intestinal parameters. The experimental animals were fed for 12 weeks a HFD containing 20 % fat in the form of a pasture dairy cream (PDC) or a standard dairy cr

Suppression of endothelin-1-induced cardiac myocyte hypertrophy by PPAR agonists: role of diacylglycerol kinase zeta.

AIMS: Ligand activation of peroxisome proliferator-activated receptors (PPARs) prevents cardiomyocyte hypertrophy, but the underlying signalling mechanisms remain unknown. We previously reported that the anti-hypertrophic effect of the dietary polyunsaturated fatty acid, conjugated linoleic acid (CLA), was associated with the upregulation of diacylglycerol (DAG) kinase (DGK). DGK catalyses phosphorylative conversion/attenuation of DAG, thereby modulating protein kinase C (PKC) and G-protein signalling. As the anti-hypertrophic effects of CLA were attenuated by inhibitors of PPARs, the present

Investigation of the formation mechanism of the oxidized soybean protein/linoleic acid aggregates: Effect of heating treatment on the soybean protein emulsion structure, antioxidant properties and accessibility of vitamin D3.

The aim of this study was to investigate the effects of heating-induced conjugation between soybean protein isolate (SPI) and linoleic acid (LA) on the structural modifications of the protein, the antioxidant activity of the oxidized protein, and the bioaccessibility of vitamin D3. The results revealed that the LA facilitated the formation of oxidized soy protein aggregates, which demonstrated that the protein oxidation degree increased. The simultaneous rheology and fourier transform infrared spectroscopy measurements made an analysis of the structural changes of proteins caused by oxidation,

CLA reduces inflammatory mediators from A427 human lung cancer cells and A427 conditioned medium promotes differentiation of C2C12 murine muscle cells.

Conjugated linoleic acid (CLA) is thought to have anti-proliferative and anti-inflammatory properties, but its effect on cancer cachexia is unknown. Two effects were here investigated: that of CLA on inflammatory mediator production in human lung cancer cells, and that of reduced mediators on the myogenic differentiation of murine muscle C2C12 cells. The latter cells were grown in medium conditioned by human lung cancer A427 cells, with or without CLA, to mimic only the effect of molecules released from the tumor "in vivo", excluding the effect of host-produced cachectic factors. The results o

Transient reductions in milk fat synthesis and their association with the ruminal and metabolic profile in dairy cows fed high-starch, low-fat diets.

Sub-acute ruminal acidosis (SARA) is sometimes observed along with reduced milk fat synthesis. Inconsistent responses may be explained by dietary fat levels. Twelve ruminally cannulated cows were used in a Latin square design investigating the timing of metabolic and milk fat changes during Induction and Recovery from SARA by altering starch levels in low-fat diets. Treatments were (1) SARA Induction, (2) Recovery and (3) Control. Sub-acute ruminal acidosis was induced by feeding a diet containing 29.4% starch, 24.0% NDF and 2.8% fatty acids (FAs), whereas the Recovery and Control diets contai

Ruminal cellulolytic bacteria abundance leads to the variation in fatty acids in the rumen digesta and meat of fattening lambs.

Ruminal cellulolytic bacteria could be a diagnostic tool for determining the subacute rumen acidosis (SARA) risk in individual ruminants; however, a limited number of studies have investigated the effects of the abundance of ruminal cellulolytic bacteria on the fatty acid (FA) composition of the rumen digesta and the muscle of sheep. Thus, the objective of this study was to evaluate the effect of the variation of rumen cellulolytic bacteria on the rumen fermentation, rumen digesta, and muscle FA composition of fattening lambs fed an identical diet. Forty-eight lambs were reared in individual u

Major scientific advances with dairy foods in nutrition and health.

A large body of scientific evidence collected in recent decades demonstrates that an adequate intake of calcium and other nutrients from dairy foods reduces the risk of osteoporosis by increasing bone acquisition during growth, slowing age-related bone loss, and reducing osteoporotic fractures. These results have culminated in the new (2005) Dietary Guidelines for Americans that now recommend 3 servings of milk products per day to reduce the risk of low bone mass and contribute important amounts of many nutrients that may have additional health attributes beyond bone health. A number of animal

Trans-11 vaccenic acid dietary supplementation induces hypolipidemic effects in JCR:LA-cp rats.

Trans-11 vaccenic acid [VA; 18:1(n-9)] is a positional and geometric isomer of oleic acid and is the precursor to conjugated linoleic acid (CLA) in humans. Despite VA being the predominant trans monoene in ruminant-derived lipids, very little is known about its nutritional bioactivity, particularly in conditions of chronic metabolic disorders, including obesity, insulin resistance, and/or dyslipidemia. The aim of this study was to assess the potential of VA to improve dyslipidemia, insulin sensitivity, or inflammatory status in obese and insulin-resistant JCR:LA-cp rats. The obese rats and age

Failure to withstand oxidative stress induced by phospholipid hydroperoxides as a possible cause of the lens opacities in systemic diseases and ageing.

Lipid peroxidation (LPO) is a causative factor of cataract. The increased concentrations of primary molecular LPO products (diene conjugates, lipid hydroperoxides) and end fluorescent LPO products, were detected in the lipid moieties of the aqueous humor samples obtained from patients with senile and complicated cataracts as compared to normal donors. The degrees of lens clouding were assessed quantitatively by measuring the optical density indices and areas of equidensities using digital image analysis. Human cataractous lenses showed decreased activity of glutathione peroxidase (GPX, catalyz
