Neutrophil gelatinase-associated lipocalin suppresses cyst growth by Pkd1 null cells in vitro and in vivo.

Cyst growth in patients with autosomal dominant polycystic kidney disease is thought to be due to increased tubular cell proliferation. One model to explain this altered proliferation suggests that the polycystin proteins PC1 and PC2 localize to apical cilia and serve as an integral part of the flow-sensing pathway thus modulating the proliferative response. We measured proliferation and apoptosis in proximal tubule derived cell lines lacking PC1. These cells showed increased rates of proliferation, a decreased rate of apoptosis, compared to control heterozygous cell lines, and spontaneously f

Neutrophil gelatinase-associated lipocalin (NGAL) as a biomarker for acute renal injury after cardiac surgery.

BACKGROUND: The scarcity of early biomarkers for acute renal failure has hindered our ability to launch preventive and therapeutic measures for this disorder in a timely manner. We tested the hypothesis that neutrophil gelatinase-associated lipocalin (NGAL) is an early biomarker for ischaemic renal injury after cardiopulmonary bypass. METHODS: We studied 71 children undergoing cardiopulmonary bypass. Serial urine and blood samples were analysed by western blots and ELISA for NGAL expression. The primary outcome measure was acute renal injury, defined as a 50% increase in serum creatinine from

Endocytic delivery of lipocalin-siderophore-iron complex rescues the kidney from ischemia-reperfusion injury.

Neutrophil gelatinase-associated lipocalin (Ngal), also known as siderocalin, forms a complex with iron-binding siderophores (Ngal:siderophore:Fe). This complex converts renal progenitors into epithelial tubules. In this study, we tested the hypothesis that Ngal:siderophore:Fe protects adult kidney epithelial cells or accelerates their recovery from damage. Using a mouse model of severe renal failure, ischemia-reperfusion injury, we show that a single dose of Ngal (10 microg), introduced during the initial phase of the disease, dramatically protects the kidney and mitigates azotemia. Ngal acti

Identification of neutrophil gelatinase-associated lipocalin as a novel early urinary biomarker for ischemic renal injury.

Acute renal failure (ARF) secondary to ischemic injury remains a common and potentially devastating problem. A transcriptome-wide interrogation strategy was used to identify renal genes that are induced very early after renal ischemia, whose protein products might serve as novel biomarkers for ARF. Seven genes that are upregulated >10-fold were identified, one of which (Cyr61) has recently been reported to be induced after renal ischemia. Unexpectedly, the induction of the other six transcripts was novel to the ARF field. In this study, one of these previously unrecognized genes was further ch

Immunological functional studies of a putative lipocalin member LcApoM_AGPRP in large yellow croaker (Larimichthys crocea).

Lipocalins play crucial roles in immune defense in vertebrates; however, their functional mechanisms in teleost fish remain poorly understood. In this study, the economically important marine fish large yellow croaker (Larimichthys crocea) was used as a model to investigate a novel lipocalin-like protein, LcApoM_AGPRP, and to elucidate its role in immune responses. A putative antibacterial protein, LcApoM_AGPRP, which lacks functional annotation in the genome, was identified in large yellow croaker. LcApoM_AGPRP was highly constitutively expressed in the liver, head kidney, ovary, gill and spe

Diagnostic performance of fecal eosinophil-derived neurotoxin and lipocalin-2 in pediatric inflammatory bowel disease.

BACKGROUND: Non-invasive biomarkers for inflammatory bowel disease (IBD) diagnosis in children are needed to reduce dependence on invasive procedures. This study examined fecal eosinophil-derived neurotoxin (fEDN), lipocalin-2 (LCN-2), and calprotectin (FC) as potential non-invasive supportive markers in pediatric IBD. METHODS: This case-control study included 90 participants: 30 IBD patients (12 Crohn's disease, 18 ulcerative colitis), 30 acute diarrhea patients, and 30 healthy controls. Fecal samples were analysed for fEDN, LCN-2, and FC levels (ng/mL) using ELISA. Biomarker levels were cor

Neutrophil Gelatinase-Associated Lipocalin for the Assessment of Persistent Renal Dysfunction after Acute Kidney Injury.

INTRODUCTION: Renal injury is a significant complication in ST-segment elevation myocardial infarction (STEMI) and is associated with poor outcomes. The recently introduced term acute kidney disease (AKD) describes persistent renal dysfunction lasting 7-90 days post-acute kidney injury (AKI). Neutrophil gelatinase-associated lipocalin (NGAL), a biomarker for tubular injury, has shown promise in predicting AKI. This study aimed to evaluate NGAL's diagnostic utility for predicting AKD in STEMI patients. METHODS: This prospective observational study included 312 patients admitted with STEMI. NGA

Lipocalin-2-mediated oligodendrocyte pyroptosis contributes to post-stroke secondary white matter injury.

Secondary white matter injury occurs in regions remote from the primary infarct after ischemic stroke, contributing to long-term neurological deficits. However, the mechanisms underlying oligodendrocyte loss in secondary white matter injury remain poorly understood. Using a distal middle cerebral artery occlusion (dMCAO) model in mice, we demonstrated that focal cortical ischemia induced significant oligodendrocyte pyroptosis in the contralateral corpus callosum. This pathological process was accompanied by increased interleukin-1β, interleukin-18, and canonical pyroptosis markers, including G

The role of inflammatory biomarkers RBP4, Lipocalin-2, and hsCRP in prediabetes and type 2 diabetes mellitus.

BACKGROUND: Adipocytokines, along with macrophages infiltrating adipose tissue, contribute to chronic low-grade inflammation, which leads to insulin resistance and type 2 diabetes. Understanding insulin resistance in non-diabetic individuals and its cellular mechanisms is key for developing effective treatments and improving current protocols. This study aimed to investigate the levels of Retinol Binding Protein 4 (RBP4), Lipocalin-2, and high-sensitivity C-reactive protein (hsCRP) in individuals with impaired fasting glucose (IFG), impaired glucose tolerance (IGT), and type 2 diabetes mellitu

Shared inflammation in takotsubo syndrome and sepsis: Lipocalin-2 mediates neutrophil infiltration and cardiac damage.

BACKGROUND: Takotsubo syndrome (TTS) and sepsis often co-occur with poor outcomes, yet their underlying molecular mechanisms remain to be elucidated. Transcriptomic analysis is employed to detect diagnostic biomarkers and reveal shared pathophysiological mechanisms in sepsis-associated TTS. METHODS: Myocardial gene expression data (TTS, sepsis, and controls) from GEO were analyzed to identify shared differentially expressed genes. WGCNA and PPI networks were used for candidate gene selection. Key genes were further refined using random forest and LASSO algorithms. Immune cell infiltration was

Clustered Mitochondrial Homolog Inhibition by Lipocalin-2 Orchestrates Mitochondrial Disruption and Contributes to Kidney Disease.

KEY POINTS: Lipocalin-2 inhibited clustered mitochondrial homolog (CLUH) activity and caused mitochondrial dysfunction. Loss of CLUH in kidneys triggered metabolic reprogramming and impaired mitochondrial structure. CLUH expression decreased with kidney injury and correlated with kidney function decline in patients. BACKGROUND: Lipocalin-2 (LCN2) is a secreted protein involved in transporting hydrophobic molecules, regulating antibacterial responses and iron homeostasis. LCN2 expression increases after kidney damage and participates in CKD onset. We previously identified LCN2 as a regulator o

Deficiency of neutrophil gelatinase-associated lipocalin elicits a hemophilia-like bleeding and clotting disorder in mice.

Coagulation is related to inflammation, but the key pathway, especially innate immune system and coagulation regulation, is not well understood and need to be further explored. Here, we demonstrated that neutrophil gelatinase-associated lipocalin (NGAL), an innate immune inflammatory mediator, is upregulated in patients with thrombosis. Furthermore, it contributes to the initiation and amplification of coagulation, hemostasis, and thrombosis. This occurs by enhancing tissue factor expression on the cell surface, potentiating various clotting factors such as thrombin, kallikrein, factor XIa (FX

Systemic Neutrophil Gelatinase-Associated Lipocalin Alterations in Chronic Pancreatitis: A Multicenter, Cross-Sectional Study.

INTRODUCTION: Chronic pancreatitis (CP) is a progressive fibroinflammatory disorder lacking therapies and biomarkers. Neutrophil gelatinase-associated lipocalin (NGAL) is a proinflammatory cytokine elevated during inflammation that binds fatty acids (FAs) such as linoleic acid. We hypothesized that systemic NGAL could serve as a biomarker for CP and, with FAs, provide insights into inflammatory and metabolic alterations. METHODS: NGAL was measured by immunoassay, and FA composition was measured by gas chromatography in plasma (n = 171) from a multicenter study, including controls (n = 50), ac

Crosstalk between lipocalin-2 and IL-6 in traumatic brain injury: Closely related biomarkers.

Clinical biomarkers are crucial for diagnosing and predicting outcomes in patients with traumatic brain injury (TBI). In this study, we performed an unbiased analysis of plasma proteins in acute TBI patients using bead-based multiplex assays and identified a strong positive correlation between LCN2 and IL-6 levels. Based on these findings, we hypothesized that LCN2 and IL-6 are closely related circulating biomarkers for TBI. Our previous and current studies demonstrate that the expression of LCN2, IL-6, and its receptors is upregulated in patients with chronic traumatic encephalopathy, in mous

Elevated perioptic lipocalin-type prostaglandin D synthase concentration in patients with idiopathic intracranial hypertension.

The pathophysiology of vision loss and loss of visual field in patients with idiopathic intracranial hypertension with papilloedema is not fully understood. Although elevated CSF pressure induces damage to the optic nerve due to stasis of axoplasmic flow, there is no clear relationship between the severity of papilloedema and CSF pressure. Furthermore, there are cases of purely unilateral papilloedema and cases without papilloedema despite significantly elevated intracranial pressure as well as papilloedema that can persist despite a successfully lowered intracranial pressure. We hypothesize t
