pH-sensitive imaging by low-frequency EPR: a model study for biological applications.

The use of pH-sensitive nitroxides, in conjunction with low-frequency EPR, offers a unique opportunity for non-invasive assessment of pH values (in the range 0 to 14) in living animals. In the present study, we have investigated the potential use of pH-sensitive nitroxide free radicals in conjunction with EPR imaging techniques at low and very low frequencies (280 MHz-2.1 GHz). In particular, we have measured the hyperfine splitting (hfs) of a pH-sensitive probe at three different EPR frequencies: 280 MHz, 1.1 GHz and 2.1 GHz. We have also developed EPR imaging experiments with phantoms simula

The role of pallidum in the neural integrator model of cervical dystonia.

Dystonia is the third most common movement disorder affecting three million people worldwide. Cervical dystonia is the most common form of dystonia. Despite common prevalence the pathophysiology of cervical dystonia is unclear. Traditional view is that basal ganglia is involved in pathophysiology of cervical dystonia, while contemporary theories suggested the role of cerebellum and proprioception in the pathophysiology of cervical dystonia. It was recently proposed that the cervical dystonia is due to malfunctioning of the head neural integrator - the neuron network that normally converts head

Chronic cortical and electromyographic recordings from a fully implantable device: preclinical experience in a nonhuman primate.

OBJECTIVE: Analysis of intra- and perioperatively recorded cortical and basal ganglia local field potentials in human movement disorders has provided great insight into the pathophysiology of diseases such as Parkinson's, dystonia, and essential tremor. However, in order to better understand the network abnormalities and effects of chronic therapeutic stimulation in these disorders, long-term recording from a fully implantable data collection system is needed. APPROACH: A fully implantable investigational data collection system, the Activa® PC + S neurostimulator (Medtronic, Inc., Minneapolis

Raslan Circuit: von Economo neurons as biological dipole antenna elements in a proposed FI-ACC communication pathway for multimodal integration and conscious access.

Consciousness remains incompletely explained at the anatomical, biophysical, and functional levels. Although existing theories characterize key functional correlates of conscious experience-including global broadcasting, information integration, attentional selection, working-memory-based awareness, and electromagnetic field dynamics-none has yet identified a specific cellular mechanism embedded within a defined long-range cortical pathway that could support the transmission of integrated sensory, interoceptive, emotional, and cognitive information toward anterior access and control systems. Volumetric mapping of the functional neuroanatomy of the respiratory network in the perfused brainstem preparation of rats.

KEY POINTS: The functional neuroanatomy of the mammalian respiratory network is far from being understood since experimental tools that measure neural activity across this brainstem-wide circuit are lacking. Here, we use silicon multi-electrode arrays to record respiratory local field potentials (rLFPs) from 196-364 electrode sites within 8-10 mm3 of brainstem tissue in single arterially perfused brainstem preparations with respect to the ongoing respiratory motor pattern of inspiration (I), post-inspiration (PI) and late-expiration (E2). rLFPs peaked specifically at the three respiratory phas

Computational imaging of cardio-magnetic sources: reconstruction of time-varying dipoles and epicardial potentials from magnetocardiography.

The electrical activities in living tissues are caused by ionic movements along cell membranes, particularly in excitable tissues like neurons, cardiac cells, and skeletal muscles. The heart muscles produce currents that flow through the tissue in the body's volume conductor. These currents generate potentials and magnetic field that are measurable at and beyond the thorax surface using detectors. Researchers have been studying these electrical activities by modeling their electrophysiology. This research work discusses the different approaches to model electrical activities of the heart. One

Scanning Probe Microscopy of Graphene Sensors Before and After Nitrogen Exposure.

Developing high-performance, miniaturized 2D material platforms requires a deep, multiscale understanding of gas-surface interactions and localized electronic behavior. In this study, we demonstrate an integrated experimental methodology that combines in situ environmental scanning electron microscopy (ESEM) with ex situ scanning probe microscopy (SPM) to analyze monolayer graphene field-effect transistor (GFET) sensors before and after controlled nitrogen (N2) exposure. In situ electrical measurements performed inside the ESEM chamber show a continuous, monotonic increase in graphene channel

Globus pallidus externus (GPe) alpha band activity decreases after deep brain stimulation in clinically responsive obsessive-compulsive disorder patients.

BACKGROUND: Deep brain stimulation (DBS) of the anterior limb of the internal capsule (ALIC) is an effective treatment for severe obsessive-compulsive disorder (OCD). Identifying brain readouts of positive response may guide further DBS optimization. METHODS: We measured local field potential (LFP) changes from bilateral DBS leads in 10 OCD patients implanted at a uniform tractographic network target derived from prior DBS responders. We consistently stimulated dorsal lead contacts in the ALIC white matter, while recording LFP from the ventral lead contacts in grey matter of the anterior glob

Selection on Aedes aegypti alters Wolbachia-mediated dengue virus blocking and fitness.

The dengue, Zika and chikungunya viruses are transmitted by the mosquito Aedes aegypti and pose a substantial threat to global public health. Current vaccines and mosquito control strategies have limited efficacy, so novel interventions are needed1,2. Wolbachia are bacteria that inhabit insect cells and have been found to reduce viral infection-a phenotype that is referred to as viral 'blocking'3. Although not naturally found in A. aegypti4, Wolbachia were stably introduced into this mosquito in 20114,5 and were shown to reduce the transmission potential of dengue, Zika and chikungunya6,7. Oscillatory Waveform Shape and Temporal Spike Correlations Differ across Bat Frontal and Auditory Cortex.

Neural oscillations are associated with diverse computations in the mammalian brain. The waveform shape of oscillatory activity measured in the cortex relates to local physiology and can be informative about aberrant or dynamically changing states. However, how waveform shape differs across distant yet functionally and anatomically related cortical regions is largely unknown. In this study, we capitalize on simultaneous recordings of local field potentials (LFPs) in the auditory and frontal cortices of awake, male Carollia perspicillata bats to examine, on a cycle-by-cycle basis, waveform shap

Production of hydrogen peroxide in an intra-meander hyporheic zone at East River, Colorado.

The traditionally held assumption that photo-dependent processes are the predominant source of H2O2 in natural waters has been recently questioned by an increrasing body of evidence showing the ubiquitiousness of H2O2 in dark water bodies and in groundwater. In this study, we conducted field measurement of H2O2 in an intra-meander hyporheic zone and in surface water at East River, CO. On-site detection using a sensitive chemiluminescence method suggests H2O2 concentrations in groundwater ranging from 6 nM (at the most reduced region) to ~ 80 nM (in a locally oxygen-rich area) along the intra-m

N,N-Dimethyltryptamine attenuates spreading depolarization and restrains neurodegeneration by sigma-1 receptor activation in the ischemic rat brain.

Dimethyltryptamine (DMT), an endogenous ligand of sigma-1 receptors (Sig-1Rs), acts against systemic hypoxia, but whether DMT may prevent cerebral ischemic injury is unexplored. Here global forebrain ischemia was created in anesthetized rats and aggravated with the induction of spreading depolarizations (SDs) and subsequent short hypoxia before reperfusion. Drugs (DMT, the selective Sig-1R agonist PRE-084, the Sig-1R antagonist NE-100, or the serotonin receptor antagonist asenapine) were administered intravenously alone or in combination while physiological variables and local field potential

3D Finite Element Pseudodynamic Analysis of Deficient RC Rectangular Columns Confined with Fiber Reinforced Polymers under Axial Compression.

This paper utilizes the advanced potential of pseudodynamic three-dimensional finite-element modeling to study the axial mechanical behavior of square and rectangular reinforced concrete columns, confined with fiber reinforced polymer (FRP) jackets and continuous composite ropes in seismic applications. The rigorous and versatile Riedel-Hiermaier-Thoma (RHT) material model for concrete is suitably calibrated/modified to reproduce the variable behavior of characteristic retrofitted columns with deficient internal steel reinforcement detailing, suffering nonuniform local concrete cracking and cr

Silica nanoparticles with thermally activated delayed fluorescence for live cell imaging.

Thermally activated delayed fluorescence (TADF) has revolutionized the field of organic light emitting diodes owing to the possibility of harvesting non-emissive triplet states and converting them in emissive singlet states. This mechanism generates a long-lived delayed fluorescence component which can also be used in sensing oxygen concentration, measuring local temperature, or on imaging. Despite this strong potential, only recently TADF has emerged as a powerful tool to develop metal-free long-lived luminescent probes for imaging and sensing. The application of TADF molecules in aqueous and

Behavioral Context Determines Network State and Variability Dynamics in Monkey Motor Cortex.

Variability of spiking activity is ubiquitous throughout the brain but little is known about its contextual dependance. Trial-to-trial spike count variability, estimated by the Fano Factor (FF), and within-trial spike time irregularity, quantified by the coefficient of variation (CV), reflect variability on long and short time scales, respectively. We co-analyzed FF and the local coefficient of variation (CV2) in monkey motor cortex comparing two behavioral contexts, movement preparation (wait) and execution (movement). We find that the FF significantly decreases from wait to movement, while t

Evaluating repellence properties of a catnip essential oil-based mosquito repellent using the human landing catch method in Eastern Uganda.

Mosquitoes (Diperta: Culidae) act as vectors for several diseases, including malaria, dengue fever and yellow fever. Mosquito repellents represent one of the primary measures used to reduce the risk of these diseases in humans by reducing mosquito landing and biting events. Nepetalactone, a natural insect repellent primarily found in the essential oil of catnip (Lamiaceae; Nepeta cataria), has established mosquito repellence properties, but has not been widely exploited as a mosquito repellent in malaria-endemic regions such as Sub-Saharan Africa. Here, we evaluated the potential of a locally

Automatic flow planning for fetal cardiovascular magnetic resonance imaging.

BACKGROUND: Widening access to fetal flow imaging by automating real-time planning of two-dimensional (2D) phase-contrast flow imaging (OWL). METHODS: Two subsequent deep learning networks for fetal body localization and cardiac landmark detection on a coronal whole-uterus scan were trained on 167 and 71 fetal datasets, respectively, and implemented for real-time automatic planning of phase-contrast sequences. OWL was evaluated retrospectively in ten datasets and prospectively in seven fetal subjects (36+3-39+3 gestational weeks), with qualitative and quantitative comparisons to manual planni

Inner Ear Pharmacotherapy for Residual Hearing Preservation in Cochlear Implant Surgery: A Systematic Review.

Cochlear implantation initiates an inflammatory cascade in which both acute insertion trauma and chronic foreign body reaction lead to intracochlear fibrosis and loss of residual hearing. Several strategies have been proposed to attenuate the local reactive process after implantation, including intracochlear drug delivery. The present study gives an overview of what is being investigated in the field of inner ear therapeutics and cochlear implant surgery. The aim is to evaluate its potential benefit in clinical practice. A systematic search was conducted in PubMed, Embase, and Cochrane Library

Bone mass measurement in identification of women at risk for osteoporosis.

It is now possible to measure bone mass with highly precise, safe and noninvasive technology. Dual energy X-ray absorptiometry (DXA) can detect bone loss well before it becomes evident by conventional X-rays or by fracture. Because measurement of bone density is the single most important predictor of fracture risk, it is a critically important tool to apply to the population at risk, which includes women who have definable risk factors for osteoporosis, such as the menopause, as well as those with a family history of osteoporosis, life-long low calcium intake, smoking, extreme thinness, anorex

Simultaneous In Vivo Electrophysiology, Two-Photon Imaging, and Optogenetics for Probing Neurovascular Coupling.

Neuronal activity and cerebral blood flow are tightly coupled to support the high metabolic demands of the brain. Disruption of neurovascular coupling is a defining feature of many neurodegenerative disorders such as Alzheimer's disease, stroke, small vessel disease, Parkinson's disease, and aging. Progress in understanding the mechanisms underlying neurovascular coupling requires experimental approaches that can simultaneously measure neuronal activity and vascular dynamics with high spatial and temporal resolution, while also enabling targeted perturbations of the system. Here, we present a
