High numbers of tumor-infiltrating programmed cell death 1-positive regulatory lymphocytes are associated with improved overall survival in follicular lymphoma.

PURPOSE: Tumor microenvironment influences the behavior of follicular lymphoma (FL), although the specific cell subsets involved are not well known. The aim of this study was to determine the impact of programmed cell death 1 (PD-1) -positive inhibitory immunoregulatory lymphoid cells in the clinicobiologic features and outcome of patients with FL. PATIENTS AND METHODS: We examined samples from 100 patients (53 men and 47 women; median age, 54 years) at diagnosis, as well as in 32 patients at first relapse, with a recently generated monoclonal antibody against PD-1. The cells were quantified

CD44 variant isoform v10 is expressed on tumor-infiltrating lymphocytes and mediates hyaluronan-independent heterotypic cell-cell adhesion to melanoma cells.

OBJECTIVE: Phenomenology, assessment, longitudinal, and psychosocial findings from an ongoing, controlled, prospective study of 93 subjects with a prepubertal and early adolescent bipolar disorder phenotype (PEA-BP) will be reviewed. METHODS: Unlike adult-onset bipolar disorder, for which there were over 50 years of systematic investigations, there were a paucity of rigorous data and much controversy and skepticism about the existence and characteristics of prepubertal-onset mania. With this background, issues to address for investigation of child-onset mania included the following: (i) What

Immunotherapy for Stewart-Treves syndrome. Usefulness of intrapleural administration of tumor-infiltrating lymphocytes against massive pleural effusion caused by metastatic angiosarcoma.

We describe a 56-year-old woman with Stewart-Treves syndrome who had severe dyspnea from a pleural effusion caused by metastatic angiosarcoma in the right lung. Tumor-infiltrating lymphocytes (TIL) in the pleural effusion were cultured and expanded in vitro in the continuous presence of recombinant interleukin 2 with periodic stimulation by CD3 antibody. The expanded TIL were administered intrapleurally seven times at 1- to 4-week intervals in combination with intravenous infusion of recombinant interleukin 2. A panel of T-cell clones was also obtained from TIL. Immunotherapy dramatically impr

The Association Between Lymphocyte-Monocyte Ratio, Tumor-Infiltrating Lymphocytes, and Tumor-Associated Macrophages with 3-Year Progression-Free Survival in Advanced Local Stage Nasopharyngeal Cancer Patients.

Nasopharyngeal cancer (NPC) is a major health burden in Indonesia, and reliable prognostic biomarkers are needed to guide treatment decisions. This study examined associations between lymphocyte-monocyte ratio (LMR), tumor-infiltrating lymphocytes (TILs), tumor-associated macrophages (TAMs), and 3-year progression-free survival (PFS) in advanced-stage NPC. A retrospective cohort was assembled at a national referral hematology-oncology clinic, including patients diagnosed from January 2015 to 2020. TIL and TAM expression were assessed by immunohistochemistry, and cutoffs for LMR, CD8, and CD163

Severe Cryptosporidiosis Following Tumor-Infiltrating Lymphocyte Therapy in a Patient With Metastatic Mucosal Melanoma.

Cryptosporidium is a protozoan parasite that can cause respiratory and gastrointestinal illnesses with watery diarrhea. Severe disease may occur in immunocompromised patients, especially those with acquired immunodeficiency syndrome or metastatic mucosal melanoma on immunosuppressive therapy. We report a 72-year-old man with metastatic mucosal melanoma treated with surgery, radiation, and tumor-infiltrating lymphocytes therapy complicated by hemophagocytic lymphohistiocytosis and autoimmune colitis. He presented with an acute profuse diarrhea despite sulfasalazine and budesonide therapy for hi

Role of Metastasectomy for Tissue Acquisition for Tumor-Infiltrating Lymphocyte Harvest and Biomarker/Genomic Testing.

Tumor-infiltrating lymphocyte therapy is an emerging paradigm with promising early results for a patient population without many good alternatives. In some cases where the primary tumor or soft tissues metastases cannot be harvested, there may be a role for the thoracic surgeon to harvest tissue from pulmonary metastases. Typically, these can be limited resections with most cases being amenable to a wedge resection as only 1 cm of tissue is usually required. As a result, in appropriately selected patients, there has been shown to be a low risk of pulmonary, cardiovascular, infectious, and woun

Pulmonary resection for tissue harvest in adoptive tumor-infiltrating lymphocyte therapy: Safety and feasibility.

BACKGROUND AND OBJECTIVES: Adoptive T-cell therapies (ACTs) using expansion of tumor-infiltrating lymphocyte (TIL) populations are of great interest for advanced malignancies, with promising response rates in trial settings. However, postoperative outcomes following pulmonary TIL harvest have not been widely documented, and surgeons may be hesitant to operate in the setting of widespread disease. METHODS: Patients who underwent pulmonary TIL harvest were identified, and postoperative outcomes were studied, including pulmonary, cardiovascular, infectious, and wound complications. RESULTS: 83

[Tumor-infiltrating lymphocyte preparations have been in the fight against cancer for 40 years].

This review article summarizes the development of tumor-infiltrating lymphocyte (TIL) based immunotherapy, from the pioneering work of Rosenberg, Spiess and Lafreniere to the 2024 regulatory approval of lifileucel, the first cell therapy authorized for the treatment of solid tumors. A key advantage of TIL therapy over the chimeric antigen receptor (CAR) T-cell technology is its polyclonal T-cell receptor repertoire, which enables more effective recognition of heterogeneous tumor cell populations. The therapeutic approach is based on the ex vivo expansion and activation of lymphocytes isolated

Tumor-infiltrating lymphocyte (TIL) therapy: Historical context, clinical applications, and future directions.

In the past 15 years, immunotherapy has become a major strategy for cancer therapy. Tumor-infiltrating lymphocyte (TIL) therapy has been under investigation for nearly four decades and received its first Food and Drug Administration approval in 2024, when lifileucel was approved for patients with previously treated metastatic melanoma. TIL therapy is a form of cell-based immunotherapy that uses autologous T cells isolated from the tumor microenvironment, which are expanded ex vivo and reinfused to mediate tumor regression after lymphodepleting chemotherapy and high-dose interleukin-2. Tumor-infiltrating lymphocyte therapy in metastatic vulvar melanoma: A case report and review of the literature.

BACKGROUND: Vulvar melanoma is a rare and aggressive malignancy comprising less than 1% of all melanomas in women. Despite advances in immune checkpoint blockade, durable responses remain limited, and treatment of refractory disease poses a major challenge. Tumor-infiltrating lymphocyte (TIL) therapy has emerged as a promising option for patients with metastatic mucosal melanoma resistant to standard immunotherapy. CASE: We report a 48-year-old woman diagnosed with multifocal, unresectable vulvar melanoma who initially received ipilimumab and nivolumab, but discontinued due to immune-related

Tumor-Infiltrating lymphocyte dynamics as biomarkers of neoadjuvant treatment response in luminal breast cancer.

BACKGROUND: Tumor-infiltrating lymphocytes (TILs) are established immune biomarkers in breast cancer; however, their clinical relevance in hormone receptor–positive/HER2-negative (HR+/HER2−) disease remains controversial. In particular, the predictive value of TIL dynamics for pathological response and their prognostic significance for survival outcomes are not clearly defined in this subtype. METHODS: This retrospective study included 87 patients with HR+/HER2 − breast cancer who received neoadjuvant chemotherapy between 2017 and 2025. Stromal TILs were assessed in pre-treatment core biopsy s

Advances in Tumor-Infiltrating Lymphocyte (TIL) as a Prognostic Factor and for Treating Invasive Cutaneous Melanoma.

Invasive cutaneous melanoma is responsible for about 5% of skin tumors yet is liable for nearly 70% of skin cancer-related deaths. Despite notable advancements over the past decade, including immunotherapies and targeted treatments, more than half of invasive melanoma patients ultimately succumb to the disease due to therapeutic resistance. To overcome this obstacle, strategies such as combining immunotherapies with targeted drugs or adding epigenetic therapies have been investigated. Tumor-infiltrating lymphocytes (TILs) therapy has emerged as a promising option for patients whose disease con

Exogenous human herpesvirus 6 reinfection after tumor-infiltrating T-lymphocyte therapy.

Exogenous human herpes virus 6 (HHV-6) reinfection has never been reported in patients receiving tumor-infiltrating T lymphocytes therapy. We report an unusual case of HHV-6 infection following infusion of HHV-6 infected autologous T lymphocytes. HHV-6 infection could interfere with the tumor antigen immune recognition and the efficacy of immunotherapy.

Defective proximal TCR signaling inhibits CD8+ tumor-infiltrating lymphocyte lytic function.

CD8+ tumor-infiltrating lymphocytes (TIL) are severely deficient in cytolysis, a defect that may permit tumor escape from immune-mediated destruction. Because lytic function is dependent upon TCR signaling, we have tested the hypothesis that primary TIL have defective signaling by analysis of the localization and activation status of TIL proteins important in TCR-mediated signaling. Upon conjugate formation with cognate target cells in vitro, TIL do not recruit granzyme B+ granules, the microtubule-organizing center, F-actin, Wiskott-Aldrich syndrome protein, nor proline rich tyrosine kinase-2

Analysis of the Tumor Reactivity of Tumor-Infiltrating Lymphocytes in a Metastatic Melanoma Lesion that Lost Major Histocompatibility Complex Class I Expression after Anti-PD-1 Therapy.

Major histocompatibility complex class I loss due to the abnormality of β2-microglobulin gene is one of the mechanisms underlying delayed relapses in melanoma patients long after the initial positive responses to anti-PD-1 therapy. However, the tumor-specific reactivity of tumor-infiltrating lymphocytes in tumor lesions that lost major histocompatibility complex class I expression has not been well evaluated. We report the case of a 55-year-old woman with two metastatic melanoma lesions. After a 12-month period of successful tumor suppression by anti-PD-1 antibody therapy, one lesion started t

Association between cumulative lifetime exposure to reproductive hormones and tumor-infiltrating lymphocytes, CD4 +, and CD8 + T cells among women with breast cancer.

BACKGROUND: Reproductive hormones may influence immune responses to tumors. Tumor-infiltrating lymphocytes (TILs), CD4 +, and CD8 + T cell densities are markers of immune activity and breast cancer prognosis. We examined associations between lifetime reproductive hormone exposure, tumor immune infiltration, and survival among women with breast cancer. METHODS: The study included invasive breast cancer cases in the Women's Circle of Health Study and the Women's Circle of Health Follow-up Study. Lifetime endogenous hormone exposure (LHEendo) was defined as reproductive years plus pregnancy dura

An Open Source Automated Tumor Infiltrating Lymphocytes Algorithm for Prognosis in Primary Small Bowel Adenocarcinoma Using Routine Hematoxylin and Eosin Stained Sections.

OBJECTIVE: While tumor-infiltrating lymphocytes (TILs) are prognostic in various cancers, their role in small bowel adenocarcinoma (SBA) is unexplored. This study evaluates the prognostic significance of digitally quantified TILs in SBA. METHODS: Using digital pathology (QuPath) on hematoxylin and eosin (H&E)-stained slides from 62 SBA cases, we quantified tumor cells, TILs (lymphocytes & plasma cells), and stromal components. Derived variables (eTILs, etTILs, esTILs, eaTILs, sTILs, iTILs, easTILs, and tumor-stroma ratio [TSR]) were correlated with clinicopathological features and survival. T

Prognostic Impact of the Immunoscore Based on Whole-Slide Image Analysis of CD3+ Tumor-Infiltrating Lymphocytes in Diffuse Large B-Cell Lymphoma.

An Immunoscore based on tumor-infiltrating T-cell density was validated as a prognostic factor in patients with solid tumors. However, the potential utility of the Immunoscore in predicting the prognosis of patients with diffuse large B-cell lymphoma (DLBCL) is unclear. Here, the prognostic value of an Immunoscore based on tumor-infiltrating CD3+ T-cell density was evaluated in 104 patients with DLBCL who underwent R-CHOP (rituximab, cyclophosphamide, doxorubicin, vincristine, and prednisolone) therapy. Digitally scanned whole-slide images were analyzed using Aperio ImageScope software. CD3+ c
