The growth and differentiation of mast cells.

Mast cells (MCs) are local immune cells involved in host defense mechanisms and allergic response. They usually develop from MC committed progenitor cells which in turn are derived from uncommitted hemopoietic stem cells. MC precursors are supposed to develop in the bone marrow (bm) cavities as well as in extramedullary tissues. MC precursor cells also have the potential to circulate in the blood stream. After homing in the tissues they give rise to mature MCs. Recruitment and differentiation as well as terminal maturation of MCs is regulated by a complex network of factors. Two major arms of

Differential Receptor Binding and Regulatory Mechanisms for the Lymphangiogenic Growth Factors Vascular Endothelial Growth Factor (VEGF)-C and -D.

VEGF-C and VEGF-D are secreted glycoproteins that induce angiogenesis and lymphangiogenesis in cancer, thereby promoting tumor growth and spread. They exhibit structural homology and activate VEGFR-2 and VEGFR-3, receptors on endothelial cells that signal for growth of blood vessels and lymphatics. VEGF-C and VEGF-D were thought to exhibit similar bioactivities, yet recent studies indicated distinct signaling mechanisms (e.g. tumor-derived VEGF-C promoted expression of the prostaglandin biosynthetic enzyme COX-2 in lymphatics, a response thought to facilitate metastasis via the lymphatic vascu

Iron deficiency in astrocytes alters cellular status and impacts on oligodendrocyte differentiation.

Iron deficiency (ID) has been shown to affect central nervous system (CNS) development and induce hypomyelination. Previous work from our laboratory in a gestational ID model showed that both oligodendrocyte (OLG) and astrocyte (AST) maturation was impaired. To explore the contribution of AST iron to the myelination process, we generated an in vitro ID model by silencing divalent metal transporter 1 (DMT1) in AST (siDMT1 AST) or treating AST with Fe3+ chelator deferoxamine (DFX; DFX AST). siDMT1 AST showed no changes in proliferation but remained immature. Co-cultures of oligodendrocyte precur

Transforming growth factor-β superfamily ligand trap ACE-536 corrects anemia by promoting late-stage erythropoiesis.

Erythropoietin (EPO) stimulates proliferation of early-stage erythrocyte precursors and is widely used for the treatment of chronic anemia. However, several types of EPO-resistant anemia are characterized by defects in late-stage erythropoiesis, which is EPO independent. Here we investigated regulation of erythropoiesis using a ligand-trapping fusion protein (ACE-536) containing the extracellular domain of human activin receptor type IIB (ActRIIB) modified to reduce activin binding. ACE-536, or its mouse version RAP-536, produced rapid and robust increases in erythrocyte numbers in multiple sp

STAT3-mediated astrogliosis protects myelin development in neonatal brain injury.

OBJECTIVE: Pathological findings in neonatal brain injury associated with preterm birth include focal and/or diffuse white matter injury (WMI). Despite the heterogeneous nature of this condition, reactive astrogliosis and microgliosis are frequently observed. Thus, molecular mechanisms by which glia activation contribute to WMI were investigated. METHODS: Postmortem brains of neonatal brain injury were investigated to identify molecular features of reactive astrocytes. The contribution of astrogliosis to WMI was further tested in a mouse model in genetically engineered mice. RESULTS: Activat

Regulation of prohormone convertases in hypothalamic neurons: implications for prothyrotropin-releasing hormone and proopiomelanocortin.

Recent evidence demonstrated that posttranslational processing of neuropeptides is critical in the pathogenesis of obesity. Leptin or other physiological changes affects the biosynthesis and processing of many peptides hormones as well as the regulation of the family of prohormone convertases responsible for the maturation of these hormones. Regulation of energy balance by leptin involves regulation of several proneuropeptides such as proTRH and proopiomelanocortin. These proneuropeptide precursors require for their maturation proteolytic cleavage by the prohormone convertases 1 and 2 (PC1/3 a

Mycobacterium tuberculosis VII secretion system effector molecule Rv2347c blocks the maturation of phagosomes and activates the STING/TBK1 signaling pathway to inhibit cell autophagy.

UNLABELLED: The VII secretion system is the main channel for Mycobacterium tuberculosis (MTB) to secrete virulence proteins. The ESAT-like proteins EsxA/B and EsxW/V in the RD region of its genome have been used as targets for vaccine antigens. However, the function of EsxO/P has not been explored, although it was predicted to potentially induce Th1 cell responses as a vaccine development target. In this study, the VII secretion system effector molecule Rv2347c was heterologously expressed in Mycobacterium smegmatis and found to inhibit the expression of the early marker RAB5 of phagosomes, th

Trkb signaling in pericytes is required for cardiac microvessel stabilization.

Pericyte and vascular smooth muscle cell (SMC) recruitment to the developing vasculature is an important step in blood vessel maturation. Brain-derived neurotrophic factor (BDNF), expressed by endothelial cells, activates the receptor tyrosine kinase TrkB to stabilize the cardiac microvasculature in the perinatal period. However, the effects of the BDNF/TrkB signaling on pericytes/SMCs and the mechanisms downstream of TrkB that promote vessel maturation are unknown. To confirm the involvement of TrkB in vessel maturation, we evaluated TrkB deficient (trkb (-/-)) embryos and observed severe car

Lysine-Proline-Valine attenuates oxidized low-density lipoprotein-induced foam cell formation and inflammatory responses in RAW264.7 macrophages.

Atherosclerosis develops through oxidative stress and inflammatory responses induced by oxidized low-density lipoprotein (oxLDL), and excessive lipid accumulation in macrophages accelerates disease progression by facilitating foam cell formation. In this study, we investigated the protective effects of the α-melanocyte-stimulating hormone-derived endogenous tripeptide Lysine-Proline-Valine (KPV) against oxLDL-induced cellular injury in RAW264.7 macrophages. Exposure to oxLDL markedly increased lipid accumulation in RAW264.7 macrophages, concomitantly enhancing inflammatory signaling and promot

Phosphorylation-dependent control of transcription factor activity regulates temporal patterning of cortical cell fate specification.

The development of the cerebral cortex requires precise temporal control of transcription factor (TF) activity to coordinate neuronal and glial lineage transitions. However, the molecular mechanisms coupling signaling dynamics to TF function remain unclear. Here, we combine proteomic, phosphoproteomic, and multi-omic analyses of the developing mouse cortex to define phosphorylation as a key post translational mechanism governing cortical cell fate decisions. Across the neurogenic to gliogenic transition, we identify a global remodeling of the phosphoproteome, including extensive phosphorylatio

A novel Au-Se nanoplatform for co-delivery of cGAMP and acriflavine synergizes with photothermal therapy to enhance anti-tumor immunity via STING activation and HIF-1α inhibition.

Activating the stimulator of interferon genes (STING) pathway is a potent strategy for promoting tumor vascular normalization and enhancing cancer immunotherapy. However, the poor metabolic stability of classical STING agonists limits their clinical translation. Herein, we engineered a self-assembled Au-Se nanoplatform (AAC) for the co-delivery of the STING agonist cyclic 2',3'-GMP-AMP (cGAMP) and the hypoxia-inducible factor-1α (HIF-1α) inhibitor acriflavine (Acf). Upon cellular internalization, the weakly acidic tumor microenvironment (TME) triggers the synchronized release of cGAMP and Acf

PPARα: Linking Cardiac Metabolism to Therapeutic Opportunities in Cardiovascular Diseases.

Peroxisome proliferator-activated receptor alpha (PPARα) is a key transcriptional regulator of lipid metabolism, highly expressed in metabolically active organs such as the heart. In cardiomyocytes, where approximately 70% of energy is derived from fatty acid oxidation, PPARα plays a central role in maintaining metabolic homeostasis. Moreover, the transcription factor is implicated in postnatal maturation of the heart and immune modulation. Dysregulation of PPARα signaling has profound consequences for cardiac energy balance, particularly under stress conditions. Accordingly, its role has been

A mitochondria targeting aggregation-induced emission phototheranostic agent for photodynamic synergistic therapy of triple-negative breast cancer.

Triple-negative breast cancer (TNBC) is the most aggressive subtype of breast cancer, characterized by poor prognosis and high mortality. However, current treatment options remain inadequate, necessitating the urgent development of novel, more effective therapeutic strategies. Here, we utilize amphiphilic DSPE-PEG2000 as a scaffold to co-assemble an anionic π+ self-assembled emission-inducing emitter (photosensitizer, TMP) and an immunostimulant, CpG, via electrostatic adsorption, constructing an effective phototherapeutic diagnostic agent (termed DTC NPs). The roles of pleiotrophin in brain injuries: a narrative review of the literature.

BACKGROUND: Pleiotrophin (PTN), a secreted multifunctional growth factor, is highly expressed in the developing brain. Recently, many studies have indicated that PTN participates in the development of brain and plays a neuroprotection after brain injury, especially promoting neuronal survival and neurite outgrowth, stimulating oligodendrocyte maturation and myelination, modulating neuroinflammation, and so on. OBJECTIVE: However, no reviews comprehensively summarize the roles of PTN in brain injuries. Considering this, this review focuses on the roles and related regulatory pathways of PTN in

Antibacterial chitosan/organic rectorite nanocomposite-conjugated gelatin/β-cyclodextrin hydrogels with improved hemostasis performance for wound repair.

Bacterial infections and severe bleeding continue to pose significant challenges in wound repair. There is an urgent need for innovative, nature-inspired hydrogel dressings with antibacterial and hemostatic properties. A Ge-β-CD-CS-OREC conjugate hydrogel was developed by grafting β-CD and CS-OREC nanocomposites into a Ge matrix using EDC/NHS crosslinking, as confirmed by FT-IR and EDX analyses. Compared to single Ge-β-CD cross-linked hydrogels, the addition of CS-OREC enhanced the hydrogel's properties, including increased pore size (60 ± 14 μm), improved wettability (WCA = 28.82 ± 0.6°), enh

Involvement of NK Cells in IL-28B-Mediated Immunity against Influenza Virus Infection.

IL-28B is a member of the newly discovered type III IFN family and exhibits unique antiviral properties compared with other family members. NK cells play a critical role in defending against viruses; however, little is known about the role of IL-28B in NK cell function. In a mouse model of influenza A virus (mouse adapted influenza A/PR/8/34 strain) infection, long-term overexpression of IL-28B induced by hepatocyte-specific gene delivery exerted a strong antiviral effect in the presence of NK cells. In IL-28B-overexpressing wild-type mice, the percentages and absolute numbers of NK cells in t

Oligodendrocyte precursor cells support blood-brain barrier integrity via TGF-β signaling.

Trophic coupling between cerebral endothelium and their neighboring cells is required for the development and maintenance of blood-brain barrier (BBB) function. Here we report that oligodendrocyte precursor cells (OPCs) secrete soluble factor TGF-β1 to support BBB integrity. Firstly, we prepared conditioned media from OPC cultures and added them to cerebral endothelial cultures. Our pharmacological experiments showed that OPC-conditioned media increased expressions of tight-junction proteins and decreased in vitro BBB permeability by activating TGB-β-receptor-MEK/ERK signaling pathway. Secondl

Notch signaling drives multiple myeloma induced osteoclastogenesis.

Multiple myeloma (MM) is closely associated with bone destruction. Once migrated to the bone marrow, MM cells unbalance bone formation and resorption via the recruitment and maturation of osteoclast precursors. The Notch pathway plays a key role in different types of cancer and drives several biological processes relevant in MM, including cell localization within the bone marrow, proliferation, survival and pharmacological resistance. Here we present evidences that MM can efficiently drive osteoclastogenesis by contemporaneously activating Notch signaling on tumor cells and osteoclasts through

Excess cholesterol induces mouse egg activation and may cause female infertility.

The HDL receptor scavenger receptor, class B type I (SR-BI) controls the structure and fate of plasma HDL. Female SR-BI KO mice are infertile, apparently because of their abnormal cholesterol-enriched HDL particles. We examined the growth and meiotic progression of SR-BI KO oocytes and found that they underwent normal germinal vesicle breakdown; however, SR-BI KO eggs, which had accumulated excess cholesterol in vivo, spontaneously activated, and they escaped metaphase II (MII) arrest and progressed to pronuclear, MIII, and anaphase/telophase III stages. Eggs from fertile WT mice were activate

Electrospun fibers with plasmid bFGF polyplex loadings promote skin wound healing in diabetic rats.

Deep or chronic skin wounds are difficult to heal spontaneously due to the lack of scaffold to guide cell growth and reduced levels and activities of endogenous growth factors. Emulsion electrospinning process integrated with DNA condensation techniques indicated potentials to gradually release DNA, but no attempt has been made to clarify the advantages in promoting tissue regeneration and wound recovery. In this study, polyplexes of basic fibroblast growth factor-encoding plasmid (pbFGF) with poly(ethylene imine) were incorporated into electrospun fibers with a core-sheath structure, and poly
