Failure of mebendazole in the treatment of humans with Trichinella spiralis infection at the stage of encapsulating larvae.

Trichinella spiralis larvae infective for laboratory mice were collected from muscle biopsies performed at different times (from 1 day to 16 months) following the end of treatment, indicating the failure of mebendazole to kill Trichinella parasites when they are encapsulating in muscles.

A comparative study on the efficacy of albendazole and mebendazole in the treatment of ascariasis, hookworm infection and trichuriasis.

A comparative study of the efficacy of albendazole and mebendazole was carried out in Pattani Province in the southern part of Thailand. One hundred and ninety-six patients with single or multiple infections with Ascaris, hookworm, and Trichuris were randomized into 4 groups for different treatments. Stool examination by Kato-Katz technique were done before and on day 14 after treatment. Results of the study showed that (1) a single dose of 300 mg mebendazole (locally produced) resulted in cure rates of 50%, 0% and 0% and egg reduction rates of 87.3%, -15.3%, and 28.3%, for ascariasis, hookwor

Brugia malayi in Mastomys natalensis: efficacy of mebendazole in combination with Freund's complete adjuvant.

The chemotherapeutic efficacy of mebendazole given in combination with Freund's complete adjuvant (FCA) against Brugia malayi in multimammate rat was evaluated. Animals treated with mebendazole, orally at 200 mg/kg x 5 consecutive days along with FCA given subcutaneously (s.c.) on day -10, day 0 and day +15 of the drug treatment killed 48.51% of the adult worms. This drug given alone at the same regimen and by the same route showed only 18.7% mortality rate on adults. Mebendazole given intraperitoneally along with FCA given s.c., however, was four times more efficacious as filaricide than mebe

Radiological changes under effect of mebendazole on univesicular hydatid cysts in the liver and lung.

Mebendazole was given to a patient who had developed four univesicular hydatid cysts in 16 months following initial infection (two in the liver and two in the lungs). The size of the cysts varied from five to ten centimeters in diameter. The therapeutic effect of the drug was assessed by ultrasound, computed tomography and chest x-ray examination. A completely split wall was only observed in the smallest hepatic cyst after a three-month period with a dose of 2.4 g per day (50 mg/kg/day). Peak serum level of mebendazole was 45.2 ng/ml. Thereafter, the intake was increased to 4.5 g per day (86.5

Hydatid disease: the alveolar variety in Australia. A case report with comment on the toxicity of mebendazole.

The alveolar form of hydatid disease, caused by the larval stage of the cestode Echinococcus multilocularis, is virtually confined geographically to the northern hemisphere. It produces clinically malignant multivesicular infiltrating lesions most often in the liver, and is quite different from the usual cystic variety caused by E. granulosus. We present a case report of a patient with alveolar hydatid disease, whose treatment with mebendazole was brief due to side effects of alopecia and granulocytopenia; and review the features of this disease which, with increasing international migration,

[Filariasis in Gabon: treatment with mebendazole of filariasis due to M. perstans and Loa loa].

Mebendazole (Vermox 500) in long duration treatment 2 g/day for 3 to 4 weeks, is sometimes indicated in therapy of Loa Loa filariasis but remains the only active therapy on M. perstans filariasis. The perfect product tolerance and efficacy allow large indication in treatment of this last filariasis, the most often found in Gabon (Central Africa).

Determination of parasite-specific immunoglobulins using the ELISA in patients with echinococcosis treated with mebendazole.

Parasite-specific immunoglobulin classes were determined by the ELISA in sera of 19 patients with cystic or alveolar echinococcosis before and after 2 years (+/- 4 months) of continuous treatment with mebendazole at daily doses of 35-50 mg/kg body weight. In the majority of cases IgG decreased and IgA and IgE decreased or disappeared. In contrast, four patients with progressive disease showed reverse tendencies with increasing IgG, IgA and IgE concentrations during the same period.

A comparative trial of oxantel-pyrantel and mebendazole in multiple helminth infection in school children.

In a pilot study in 17 children, oxantel-pyrantel as one single dose of 10 to 20mg/kg of each, gave good results in ascariasis but poor cure rates in both trichuriasis and necatoriasis. However in the latter groups the infection was extremely heavy. In a controlled trial in 51 children, using oxantel-pyrantel at the same dose, but given daily for 3 consecutive days, or mebendazole 100mg twice daily for 3 days, the drugs gave similar response rates. Both gave 100% cure rates, as assessed by the absence of ova by both Beaver and brine-flotation techniques, in Ascaris lumbricoides infection. The

Impact of periodic selective mebendazole treatment on soil-transmitted helminth infections in Cuban schoolchildren.

OBJECTIVE: To evaluate the impact of periodic selective treatment with 500 mg mebendazole on soil-transmitted helminth (STH) infections in Cuban schoolchildren. METHODS: We followed up a cohort of 268 STH-positive schoolchildren, aged 5-14 years at baseline, at six-month intervals for two years and a final follow-up after three years. Kato-Katz stool examination was used to detect infections with Ascaris lumbricoides, Trichuris trichiura and hookworm. Common risk factors related to STHs were assessed by parental questionnaire. RESULTS: A significant reduction in the number of STH infections

A randomized controlled trial comparing mebendazole and albendazole against Ascaris, Trichuris and hookworm infections.

The efficacies and side effects of single dose treatments with 500 mg mebendazole (Janssen Pharmaceutica) and 400 mg albendazole (SmithKline Beecham) against intestinal nematodes were compared in a single-blind, randomized controlled trial among 2294 children aged 6 to 12 years on Pemba Island, Zanzibar, among whom infections with Ascaris, hookworms and Trichuris were highly prevalent. Both drugs were highly effective against Ascaris, with cure rates of over 97%. The cure rates for Trichuris were low, but mebendazole was significantly better than albendazole and produced a greater reduction in

Evaluating the Efficacy of Mebendazole Repurposing for Ovarian Cancer Therapy Using Optical Coherence Tomography.

Ovarian cancer (OvCa) remains the leading cause of gynecological cancer mortality, with most patients developing chemoresistance. Drug repurposing offers promising alternatives, with mebendazole (MBZ) showing anticancer activity. This study evaluates MBZ efficacy using Spectral Domain Optical Coherence Tomography (SD-OCT). We conducted longitudinal imaging of 40 wild-type (WT) and cisplatin-resistant (CPR) OVCAR8 multicellular tumor spheroids over 11 days. Four analyses were performed: volume analysis, optical attenuation analysis, uniformity analysis, and texture feature analysis. Volume anal

Phase 1 study of mebendazole therapy for refractory/progressive or recurrent pediatric brain tumors.

BACKGROUND: Mebendazole (MBZ) is an anti-helminthic that has shown antitumor activity in mice with gliomas and subsequently in medulloblastoma models. Safety and tolerability have been demonstrated in adults with brain tumors but not explored in children as a monotherapy. We characterized the safety and maximum tolerated dose of oral MBZ in pediatric patients with refractory or progressive brain tumors and assessed progression-free survival (PFS) as a secondary objective. METHODS: Patients up to 21 years of age with refractory or progressive brain tumors were enrolled at 2 centers in a 3 + 3

Mebendazole Inhibits Histoplasma capsulatum In Vitro Growth and Decreases Mitochondrion and Cytoskeleton Protein Levels.

Histoplasmosis is a frequent mycosis in people living with HIV/AIDS and other immunocompromised hosts. Histoplasmosis has high rates of mortality in these patients if treatment is unsuccessful. Itraconazole and amphotericin B are used to treat histoplasmosis; however, both antifungals have potentially severe pharmacokinetic drug interactions and toxicity. The present study determined the minimal inhibitory and fungicidal concentrations of mebendazole, a drug present in the NIH Clinical Collection, to establish whether it has fungicidal or fungistatic activity against Histoplasma capsulatum. Efficacy of single-dose 500 mg mebendazole in soil-transmitted helminth infections: a review.

Soil-transmitted helminthiasis (STH) is caused by Ascaris lumbricoides (roundworm), Trichuris trichiura (whipworm), and Ancylostoma duodenale and Necator americanus (hookworms). Mebendazole is one of the recommended preventive chemotherapy agents for STH. This review summarizes the efficacy data from 29 studies with single-dose 500 mg mebendazole in STH treatment and compares the results with those of a recently conducted phase 3 study of a 500 mg mebendazole chewable tablet against A. lumbricoides and T. trichiura infections. Studies that reported efficacy results against at least one STH inf

Recurrence rate after discontinuation of long-term mebendazole therapy in alveolar echinococcosis (preliminary results).

The recurrence rate was investigated in 19 patients with non-resectable alveolar echinococcosis after discontinuation of a long-term therapy with mebendazole (average treatment 4.3 years). A control group consisted of 14 patients who underwent radical surgery and finished a course of prophylactic postoperative mebendazole treatment of 2 years. In the controls, no recurrence was observed after a post-therapy period averaging 3.5 years. In contrast, recurrence occurred in 7/19 patients (37%) with non-resectable alveolar echinococcosis an average of 1.6 years after discontinuation of the long-ter
