High-pressure washing treatments to remove obscure mealybug (Homoptera: Pseudococcidae) and lightbrown apple moth (Lepidoptera: Tortricidae) from harvested apples.

Mixed life stages of obscure mealybug, Pseudococcus viburni (Signoret), and late 1st-instar or early 2nd-instar lightbrown apple moth, Epiphyas postvittana (Walker), on 'Royal Gala' apples (Malus domestica Borkhausen) were exposed to standard packhouse processing with and without addition of high-pressure apple washer treatments. Insect removal and mortality were assessed. After standard packhouse processing approximately 60% of P. viburni remained on their host apples. The 2 high-pressure apple washer treatments (500 and 800 psi at 2.0 rods/s) were equally effective and significantly reduced

Synergistic Bioconversion of Date Palm Leaves Into Ruminant Feed by a Leopard Moth (Zeuzera pyrina L.) Gut Bacterial Consortium and Nutrient Stimulators.

Date palm leaves (DPL), a widely available lignocellulosic by-product, are used as ruminant feed but are limited by high lignin and low protein content. This study evaluated the enhancement of DPL's nutritional value using a lignocellulose-degrading bacterial consortium (Staphylococcus sp., Brevibacterium sp., and Enterobacter sp.) isolated from the leopard moth (Zeuzera pyrina L.) gut, supplemented with microbial growth stimulators. Six treatments were applied: untreated DPL (control), DPL with M9 medium (T1), DPL with bacterial inoculum (T2), T2 + 0.5% glucose (T3), T2 + 0.5% urea (T4), and

Effects of successful or unsuccessful parasitism by the solitary endoparasitoid wasp Cotesia vestalis (Hymenoptera: Braconidae) on the testis development in several host moth species.

Parasitism by a parasitoid wasp alters the physiology and behavior of its host insects. This study investigated the effects of successful and unsuccessful parasitization on individual host development and testis growth in several moth species. Although the solitary koinobiont endoparasitoid wasp, Cotesia vestalis (Hymenoptera: Braconidae), primarily parasitizes the diamondback moth (Plutella xylostella, Lepidoptera: Plutellidae), its host range is relatively broad. Previous research confirmed successful parasitization of P. xylostella, Mythimna separata (Noctuidae), and Helicoverpa armigera ar

Cardio-renal protection and mechanisms of the antihypertensive activity of Alcalase-derived moth bean hydrolysate in DOCA-salt induced hypertensive Wistar rats.

Hypertension remains a major global health concern, often leading to severe cardiovascular and renal complications. In this study, we investigated the antihypertensive and cardiorenal protective effects of AF4, a bioactive Alcalase derived peptide fraction (< 1 kDa) derived from moth bean protein hydrolysate, in a DOCA-salt hypertensive rat model. AF4 administration significantly reduced systolic blood pressure in a dose-dependent manner, with the minimum effective dose (75 mg/kg body weight/day) selected for further mechanistic evaluation. Treatment with AF4 improved various physiological par

Male larval experience of cues from adult rivals alters lifetime sperm investment patterns in a sperm heteromorphic moth, Ephestia kuehniella.

Male animals may adjust their resource allocations for reproduction and other fitness functions in response to cues from rivals. For instance, adult males increase their investment in sperm for a higher paternity share when they perceive sperm competition risk in their surroundings. In nature, both juveniles and adults may coexist spatially and temporally. Yet, it is not clear how juvenile males of different ages respond to cues from adult rivals and fine-tune their lifetime investment in sperm production and ejaculation in any insect. Here we used the Mediterranean flour moth, Ephestia kuehni

Host Plants Affect the Foraging Success of Two Parasitoids that Attack Light Brown Apple Moth Epiphyas postvittana (Walker) (Lepidoptera: Tortricidae).

The light brown apple moth, Epiphyas postvittana is a key pest of wine grapes in Australia. Two parasitoids, Dolichogenidea tasmanica and Therophilus unimaculatus, attack the larval stage of this pest. D. tasmanica is dominant in vineyards, whereas T. unimaculatus is mainly active in native vegetation. We sought to understand why they differ in their use of habitats. Plants are a major component of habitats of parasitoids, and herbivore-infested plants influence parasitoid foraging efficiency by their architecture and emission of volatile chemicals. We investigated how different plant species

Role of ocular albinism type 1 (OA1) GPCR in Asian gypsy moth development and transcriptional expression of heat-shock protein genes.

The ocular albinism type 1 gene, named OA1, is a coding pigment cell-specific G protein-coupled receptor exclusively localized in intracellular organelles. However, the function of OA1 in insects remains generally unknown. In the present study, we explore for the first time the function of LdOA1 in the Asian gypsy moth, Lymantria dispar. To identify the function of LdOA1 gene in the development and growth of the Asian gypsy moth, the LdOA1 gene in third instar larvae was knocked down by RNAi. Compared with the controls, the knockdown of LdOA1 increased larval mortality but did not significantl

Methyl salicylate, identified as primary odorant of a specific receptor neuron type, inhibits oviposition by the moth Mamestra brassicae L. (Lepidoptera, noctuidae).

The cabbage moth, Mamestra brassicae L. (Lepidoptera, Noctuidae), is a polyphagous species that is often choosing plants of Brassica as hosts for oviposition. In the search for biologically relevant odorants used by these moths, gas chromatography linked to electrophysiological recordings from single receptor neurons (RNs) has been employed, resulting in classification of distinct types of neurons. This study presents specific olfactory RNs responding to methyl salicylate (MeS) as primary odorant and showing a weak response to methyl benzoate, the 2 aromatic compounds occurring together in sev

The antennal benzoic acid receptor cell of the female silk moth Bombyx mori L.: structure-activity relationship studies with halogen substitutes.

Studies on structure-activity relationships were carried out to characterize the response specificity of the benzoic acid cell of the female of the moth Bombyx mori by means of single sensillum electrophysiological recordings. We demonstrated that this cell type responds best to a natural key substance (benzoic acid) and has similar response profiles for less effective compounds, including various halogen substitutes of benzoic acid, benzaldehyde and other derivates of the key compound. Using different halogen substitutes (F, Cl, Br, I), we showed that the cellular response decreases with incr

Effect of UVC light on growth, incorporation of thymidine, and DNA chain elongation in cells derived from the Indian meal moth and the cabbage looper.

After exposure to 10 or 20 J/m2 UVC light, cells of the UMN-PIE-1181 line, an embryonic cell line derived from the Indian meal moth, Plodia interpunctella, exhibited a rapid and prolonged depression in the rate of incorporation of [3H]thymidine, whereas cells of the TN-368 line, an ovarian cell line derived from Trichoplusia ni, the cabbage looper, showed only a slight drop in incorporation and a rapid recovery after exposure to 10 or 40 J/m2 UVC light. The extent of this depression was not correlated to the amount of cell killing by UVC light in these cell lines or in IAL-PID2 cells. Blockage

Larval and Larval-Pupal Parasitoids Associated with Major Owlet Moth Pests of Soybean and Maize in the Brazilian Savanna: Measures to Preserve Them in Crop Succession.

Owlet moths (Lepidoptera: Noctuoidea) are among the most threatening pests of soybean and maize systems under intensive cropping in tropical and subtropical regions. Their high dispersal, larval polyphagia, and voracious feeding behavior often lead to severe grain production losses. Many parasitoids are associated with owlet moths; however, the ecological importance of larval parasitism in the soybean-maize crop rotation of the Brazilian savanna remains uncertain. We assessed larval parasitism in major lepidopteran pests of soybean and maize crops over three agricultural seasons in eight field

Response to a Synthetic Pheromone Source by OX4319L, a Self-Limiting Diamondback Moth (Lepidoptera: Plutellidae) Strain, and Field Dispersal Characteristics of its Progenitor Strain.

The diamondback moth, Plutella xylostella L. (Lepidoptera: Plutellidae), is a global pest that infests vegetable and field crops within the Brassica family. A genetically engineered strain of P. xylostella, OX4319L, carrying a 'self-limiting' gene, has shown potential for managing P. xylostella populations, using sustained releases of OX4319L male moths. In order for such a strain to provide control, the transgenic individuals must exhibit attraction to female P. xylostella sex pheromone and adequate dispersal in the field. In this study, we tested these key traits. First, we compared the resp

Piperolein B and piperchabamide D isolated from black pepper (Piper nigrum L.) as larvicidal compounds against the diamondback moth (Plutella xylostella).

BACKGROUND: There is growing demand for the development of alternative pest control agents that are effective as well as non-toxic to human health and the environment. Plant protection products derived from plant extracts are an eco-friendly alternative to synthetic pesticides. The aim of this study was to identify larvicidal compounds isolated from a natural source against Plutella xylostella L. In a larvicidal activity assay, several solvent fractions from the methanol extract of Piper nigrum L. fruit showed larvicidal effects against P. xylostella. RESULTS: Screening results indicated that

Structural characterization and biological implications of sulfated N-glycans in a serine protease from the neotropical moth Hylesia metabus (Cramer [1775]) (Lepidoptera: Saturniidae).

Contact with the urticating setae from the abdomen of adult females of the neo-tropical moth Hylesia metabus gives rise to an urticating dermatitis, characterized by intense pruritus, generalized malaise and occasionally ocular lesions (lepidopterism). The setae contain a pro-inflammatory glycosylated protease homologous to other S1A serine proteases of insects. Deglycosylation with PNGase F in the presence of a buffer prepared with 40% H2 (18)O allowed the assignment of an N-glycosylation site. Five main paucimannosidic N-glycans were identified, three of which were exclusively α(1-6)-fucosyl

Low and high-density polyethylene and expanded polystyrene biodegradation by the greater wax moth Galleria mellonella L reveals a key role of the gut microbiome.

Polyethylene (PE) is a ubiquitous pollutant that takes long time to degrade naturally. PE breaks down into persistent micro- or nano-plastics that are even more dangerous for the environment and human health. Here we investigated the ability of the wax worm Galleria mellonella (Lepidoptera, Pyralidae) to degrade two different types of PE and expanded polystyrene (EPS). Results showed a reduction of up to 69 %, 73 %, and 50 % in the weight of LDPE, HDPE, and EPS, respectively. Antibiotic treatments indicated that PE consumption is highly dependent upon an intact population of at least eight dif

Transcriptome-Wide Identification of Cytochrome P450s in Tea Black Tussock Moth (Dasychira baibarana) and Candidate Genes Involved in Type-II Sex Pheromone Biosynthesis.

The tea black tussock moth (Dasychira baibarana), a devastating pest in Chinese tea plantations, uses a ternary Type-II pheromone blend containing (3Z,6Z)-cis-9,10-epoxyhenicosa-3,6-diene (Z3,Z6,epo9-21:H), (3Z,6Z,11E)-cis-9,10-epoxyhenicosa-3,6,11-triene (Z3,Z6,epo9,E11-21:H), and (3Z,6Z)-henicosa-3,6-dien-11-one (Z3,Z6-21:11-one) for mate communication. To elucidate the P450 candidates associated with the biosynthesis of these sex pheromone components, we sequenced the female D. baibarana pheromone gland and the abdomen excluding the pheromone gland. A total of 75 DbP450s were identified. Fu

Comparative transcriptome analysis of Arabidopsis thaliana infested by diamond back moth (Plutella xylostella) larvae reveals signatures of stress response, secondary metabolism, and signalling.

BACKGROUND: Plants are exposed to attack from a large variety of herbivores. Feeding insects can induce substantial changes of the host plant transcriptome. Arabidopsis thaliana has been established as a relevant system for the discovery of genes associated with response to herbivory, including genes for specialized (i.e. secondary) metabolism as well as genes involved in plant-insect defence signalling. RESULTS: Using a 70-mer oligonucleotide microarray covering 26,090 gene-specific elements, we monitored changes of the Arabidopsis leaf transcriptome in response to feeding by diamond back mo

Projections of male-specific receptor neurons in the antennal lobe of the Oriental tobacco budworm moth, Helicoverpa assulta: a unique glomerular organization among related species.

The macroglomerular complex in the primary olfactory center of male moths receives information from numerous pheromone-detecting receptor neurons housed in specific sensilla located on the antennae. We investigated the functional organization of the three glomeruli constituting this complex in Helicoverpa assulta, a unique species among heliothine moths as concerns the composition of the pheromone blend. By tip recordings from the male-specific receptor neurons combined with cobalt-lysine stainings, the axon terminals in the brain were traced and subsequently reconstructed by camera lucida dra
