Relationship between metabolic cost, muscle moments and co-contraction during walking and running.

BACKGROUND: The metabolic cost of locomotion is a key factor in walking and running performance. It has been studied by analysing the activation and co-activation of the muscles of the lower limbs. However, these measures do not comprehensively address muscle mechanics, in contrast to approaches using muscle moments and co-contraction. RESEARCH QUESTION: What is the effect of speed and type of locomotion on muscle moments and co-contraction, and their relationship with metabolic cost during walking and running? METHODS: Eleven recreational athletes (60.5 ± 7.1 kg; 169.0 ± 6.6 cm; 23.6 ± 3.3

Model-based interspecies interpretation of botulinum neurotoxin type A on muscle-contraction inhibition.

Botulinum neurotoxins (BoNTs) are commonly used in therapeutic and cosmetic applications. One such neurotoxin, BoNT type A (BoNT/A), has been studied widely for its effects on muscle function and contraction. Despite the importance of BoNT/A products, determining the blood concentrations of these toxins can be challenging. To address this, researchers have focused on pharmacodynamic (PD) markers, including compound muscle action potential (CMAP) and digit abduction scoring (DAS). In this study, we aimed to develop a probabilistic kinetic-pharmacodynamic (K-PD) model to interpret CMAP and DAS d

Estimating Palatal and Pharyngeal Muscle Contraction in Hindi Syllable Pronunciation using Computational Modeling.

Introduction  Speech, one of the main functions affected by cleft palate, involves a complex orchestra of sound deformation by various organs including the larynx, pharynx, epiglottis, palate, tongue, lip, and other structures complementing them. Although the effects of palatoplasty are traditionally evaluated subjectively, objective parameters have seldom been described to compare the pre- and postrepair functions. The study tries to extract the palatal and pharyngeal muscles while uttering various Hindi syllables and tries to find the percentage contraction of these structures as an objectiv

Melatonin MT2 receptor is expressed and potentiates contraction in human airway smooth muscle.

Nocturnal asthma is characterized by heightened bronchial reactivity at night, and plasma melatonin concentrations are higher in patients with nocturnal asthma symptoms. Numerous physiological effects of melatonin are mediated via its specific G protein-coupled receptors (GPCRs) named the MT1 receptor, which couples to both Gq and Gi proteins, and the MT2 receptor, which couples to Gi. We investigated whether melatonin receptors are expressed on airway smooth muscle; whether they regulate intracellular cyclic AMP (cAMP) and calcium concentrations ([Ca2+]i), which modulate airway smooth muscle

Comparison between surface electrodes and ultrasound monitoring to measure TMS evoked muscle contraction.

INTRODUCTION: Transcranial magnetic stimulation (TMS) is widely used to explore cortical physiology in health and disease. Surface electromyography (sEMG) is appropriate for superficial muscles, but cannot be applied easily to less accessible muscles. Muscle ultrasound (mUS) may provide an elegant solution to this problem, but fundamental questions remain. We explore the relationship between TMS evoked muscle potentials and TMS evoked muscle contractions measured with mUS. METHODS: In 10 participants, we performed a TMS recruitment curve, simultaneously measuring motor evoked potentials (MEPs

Effects of type 1 diabetes on reflexive cardiovascular responses to intermittent muscle contraction.

Studies have shown that early-stage type 1 diabetes mellitus (T1DM) leads to an exaggerated reflex pressor response to both static muscle contraction and tendon stretch. However, whether similar responses are present during dynamic exercise (i.e., intermittent contraction) is not known. Therefore, the purpose of this study was to determine whether T1DM leads to an exaggerated reflex pressor response to intermittent muscle contraction. We measured the exercise pressor reflex in unanesthetized, decerebrated T1DM (50 mg/kg streptozotocin; STZ) and healthy control (CTL) Sprague-Dawley rats by inte

Greater Muscle Stiffness during Contraction at Menstruation as Measured by Shear-Wave Elastography.

It is important to measure mechanical properties of muscle, since muscle stiffness is an important component of stabilizing or controlling joint stability. The levels of sex hormones especially estrogen vary over the phase of the menstrual cycle and impact the mechanical properties of soft tissue such as muscle, tendon, and ligaments due to the presence of 17-β estradiol receptor in human connective tissues. Recently, shear-wave elastography (SWE), based on ultrasound imaging, has been used as an accurate technique for visualizing and assessing tissue stiffness. The purpose of this study was t

Anal sphincter imaging: better done at rest or on pelvic floor muscle contraction?

INTRODUCTION AND HYPOTHESIS: Exo-anal ultrasound imaging of the anal sphincter is usually undertaken on pelvic floor muscle contraction (PFMC) as this seems to enhance tissue discrimination. Some women are unable to achieve a satisfactory PFMC, and in this situation, the sphincter is assessed at rest. We aimed to determine whether sphincter imaging at rest is inferior to imaging on PFMC. METHODS: We analysed 441 women in this retrospective study. All underwent a standardised interview, including St Mark's incontinence score, clinical examination and 4D trans-labial ultrasound (TLUS). On analy

Inhibition of RhoA/Rho kinase pathway and smooth muscle contraction by hydrogen sulfide.

Hydrogen sulfide (H2 S) plays an important role in smooth muscle relaxation. Here, we investigated the expression of enzymes in H2 S synthesis and the mechanism regulating colonic smooth muscle function by H2 S. Expression of cystathionine-γ-lyase (CSE), but not cystathionine-β-synthase (CBS), was identified in the colonic smooth muscle of rabbit, mouse, and human. Carbachol (CCh)-induced contraction in rabbit muscle strips and isolated muscle cells was inhibited by l-cysteine (substrate of CSE) and NaHS (an exogenous H2 S donor) in a concentration-dependent fashion. H2 S induced S-sulfhydrati

[DEPENDENCE OF FUNCTIONAL PARAMETERS OF RAT SKELETAL MUSCLE CONTRACTION FROM LEVEL CIRCULATING TRIIODOTHYRONINE].

In experiments on white rats with different thyroid status (thyroidectomy, euthyroidism, hyper- and a thyrotoxicosis) in conditions in situ the relationship between the severity of functional parameters of the tibialis anterior muscle and individual level of circulating free triiodothyronine was learnt. The latent period of muscle contraction, the average and maximum speed of its contraction were determined. A comparative quantitative analysis of the values of indicators of the contractile function of the tibialis anterior muscle in euthyroid and thyroidectomy rats allowed to evaluate the «thy

Capacity of rectified vestibular evoked myogenic potential in correcting asymmetric muscle contraction power.

OBJECTIVES: Rectified vestibular evoked myogenic potential (rVEMP) is new method that simultaneously measures the muscle contraction power during VEMP recordings. Although there are a few studies that have evaluated the effect of the rVEMP, there is no study that has evaluated the capacity of rVEMP during asymmetrical muscle contraction. METHODS: Thirty VEMP measurements were performed among 20 normal subjects (mean age, 28.2±2.1 years; male, 16). VEMP was measured in the supine position. The head was turned to the right side by 0°, 15°, 30°, and 45° and the VEMPs were recorded in each positi

Trans-spinal direct current stimulation modulates motor cortex-induced muscle contraction in mice.

The present study investigated the effect of trans-spinal direct current (tsDC) on the firing rate, pattern, and amplitude of spontaneous activity of the tibial nerve and on the magnitude of cortically elicited triceps surae (TS) muscle contractions. The effect of combined tsDC and repetitive cortical electrical stimulation (rCES) on the amplitude of cortically elicited TS twitches was also investigated. Stimulation was applied by two disk electrodes (0.79 cm(2)): one was located subcutaneously over the vertebral column (T(10)-L(1)) and was used to deliver anodal DC (a-tsDC) or cathodal DC (c-

Soleus muscle in glycosylation-deficient muscular dystrophy is protected from contraction-induced injury.

The glycosylation of dystroglycan is required for its function as a high-affinity laminin receptor, and loss of dystroglycan glycosylation results in congenital muscular dystrophy. The purpose of this study was to investigate the functional defects in slow- and fast-twitch muscles of glycosylation-deficient Large(myd) mice. While a partial alteration in glycosylation of dystroglycan in heterozygous Large(myd/+) mice was not sufficient to alter muscle function, homozygous Large(myd/myd) mice demonstrated a marked reduction in specific force in both soleus and extensor digitorum longus (EDL) mus

Galpha13 regulates methacholine-induced contraction of bronchial smooth muscle via phosphorylation of MLC20.

Reversible airway constriction is induced by an increase in airway smooth muscle contractility in response to methacholine likely as a bronchospastic stimulus. Despite the finding of Galpha12 and Galpha13 up-regulation in airway hyperresponsive animals, their functional role of contraction in airway smooth muscle has not been directly explored. This study investigated the differential regulatory role of Galpha12/Galpha13 in methacholine-induced contraction of trachea and bronchus in Galpha12 or Galpha13 gene knockout mice after ovalbumin sensitization and challenges. Contraction of cultured human uterine smooth muscle cells after stimulation with endothelin-1.

To our knowledge, the problem of how to maintain isolated smooth cells in a "contractile" phenotypic state without deviation after subculturing has yet to be resolved. The present study characterized the in vitro contractile response of human uterine smooth muscle cell to endothelin-1, which induces contractions in isolated uterine strips. Contractile effects were qualitatively investigated using silicone rubber substrata. Endothelin-1 was able to distort and reduce the wrinkles in the silicone surface. Contractions were also quantified by measuring the resulting change in the collagen lattice

Cooling-induced bladder contraction: studies on isolated detrusor muscle preparations in the rat.

OBJECTIVES: Detrusor muscle contraction and uninhibited micturition after intravesical instillation of ice water is interpreted as a sign of upper motor neuron lesions. The basic mechanism of cooling-induced contraction (CIC) at the level of smooth muscle, however, has not been satisfactorily explained. We therefore designed model experiments with cooling of rat detrusor muscle. METHODS: We recorded isometric tension from strips of rat urinary detrusor muscle in organ baths during stepwise cooling. CIC was tested before and after addition of various standard agents interfering with known neur

Muscle cytoskeletal disruption occurs within the first 15 min of cyclic eccentric contraction.

The time course of loss of the 55,000-Da intermediate filament protein desmin was measured in rabbit muscles subjected to cyclic eccentric contraction. Rabbit extensor digitorum longus (EDL) and tibialis anterior (TA) muscles were examined 5 or 15 min after eccentric exercise and 1 h or 1 day after 30 min of an eccentric exercise protocol (n = 16 rabbits). The earliest change noted was a significant loss of desmin labeling in 2.5 +/- 0.63% of the rabbit EDL muscle fibers (P < 0.005) 5 min after initiation of eccentric exercise. Some loss of TA fiber desmin was also apparent at this time period

Alterations in intramuscular connective tissue after limb casting affect contraction-induced muscle injury.

This study examined the effect of alterations in rat intramuscular connective tissue (CT), secondary to limb immobilization, on the muscle's susceptibility to contraction-induced injury. Hindlimbs were casted for 3 wk with the extensor digitorum longus muscle fixed in a shortened (IM-SP) or lengthened position (IM-LP). An age-matched control group remained uncasted. Extensor digitorum longus muscles were injured in vivo by using a motorized foot pedal that repeatedly flexed and extended the foot while the muscle was electrically stimulated during plantar flexion. Four hours postinjury, maximum

Anti-stomach serum induces contraction of isolated smooth muscle cells from gastric antrum.

In this work, we investigated the ability of anti-tissue sera to cause contraction of smooth muscle cells (SMC) in vitro. Therefore, we compared the effects of a horse immune serum raised against hog gastric tissue (SER 292), of its globulin fraction (SER 292 globulin), and of its IgG fraction (SER 292 IgG), on concentration of isolated SMC from the gastric antrum of the rabbit. Our results showed that SER 292 IgG induced a dose-dependent contraction of SMC with a higher potency than SER 292 globulin and SER 292. Preincubation of SER 292 globulin with anti-F(ab')2 but not with anti-Fc reduced

O2 occlusion and cyanide induced immediate relaxation and contraction of murine skeletal muscle.

The acute changes of muscle tone and membrane current upon occlusion of oxygenation (O2 occlusion) were studied in vitro in mouse diaphragms. O2 occlusion immediately produced a contraction and a relaxation, respectively, in ryanodine- and high K(+)-contracted muscles while a biphasic change (an initial decrease then a late increase) of muscle tone was produced in muscles contracted with caffeine. The O2 occlusion effects were reversed after reoxygenation. CN- produced similar acute changes of muscle tone and abolished O2 occlusion effects. The O2 occlusion-induced relaxation in high K+ medium
