Presence of Malassezia Hyphae Is Correlated with Pathogenesis of Seborrheic Dermatitis.

Seborrheic dermatitis (SD) is a common, chronic, and relapsing skin disease. The roles of Malassezia spp. in the pathogenesis of SD are still not clear due to the lack of direct evidence for the existence of hyphae within affected skin tissues. We set out to elucidate if Malassezia mycelium contributes to the onset and development of SD and if Malassezia mycelium is correlated with the clinical severity of SD patients. We detected Malassezia hyphae in patients with SD using potassium hydroxide (KOH) and calcofluor white (CFW) staining. Fluorescent microscopy was performed for the analysis of f

Oligomycins inhibit Magnaporthe oryzae Triticum and suppress wheat blast disease.

Oligomycins are macrolide antibiotics, produced by Streptomyces spp. that show antagonistic effects against several microorganisms such as bacteria, fungi, nematodes and the oomycete Plasmopara viticola. Conidiogenesis, germination of conidia and formation of appressoria are determining factors pertaining to pathogenicity and successful diseases cycles of filamentous fungal phytopathogens. The goal of this research was to evaluate the in vitro suppressive effects of two oligomycins, oligomycin B and F along with a commercial fungicide Nativo® 75WG on hyphal growth, conidiogenesis, conidial ger

[Mechanism of thymol inhibiting Botrytis cinerea: PAO-H2O2 system].

Gray mold disease caused by Botrytis cinerea infection is one of the major crop diseases. The application of environmental-friendly fungicides to control gray mold disease has been drawing great attention. Thymol, a natural compound, showed strong antifungal activity against Botrytis cinerea. We investigated the role of polyamine oxidase (PAO)-dependent hydrogen peroxide (H2O2) production in thymol-inhibited B. cinerea growth by using physiological and biochemical approaches. The results showed that: 1) Thymol significantly inhibited the growth of B. cinerea, with remarkable increases in H2O2

Intermolecular interactions of the malate synthase of Paracoccidioides spp.

BACKGROUND: The fungus Paracoccidioides spp is the agent of paracoccidioidomycosis (PCM), a pulmonary mycosis acquired by the inhalation of fungal propagules. Paracoccidioides malate synthase (PbMLS) is important in the infectious process of Paracoccidioides spp because the transcript is up-regulated during the transition from mycelium to yeast and in yeast cells during phagocytosis by murine macrophages. In addition, PbMLS acts as an adhesin in Paracoccidioides spp. The evidence for the multifunctionality of PbMLS indicates that it could interact with other proteins from the fungus and host.

Metabolites from a Pleurotus tuber-regium collected in Madagascar.

Pleurotus tuber-regium was isolated from a dead trunk of Raphia farinifera (Arecaceae) in a lowland moist forest in Antsohihy, Madagascar, and the species was confirmed by molecular analysis and morphological observations. The main bioactive metabolites of the mycelium extracts were identified by mass spectrometry techniques. Five structural diverse metabolites, tryptophol, pyroglutamic acid, prolyldiketopiperazine B, sporol and RKS-1778, were characterised by LC-MS qTOF analysis of the hydro-alcoholic extract. GC-MS analysis of both chloroform and ethyl acetate extracts revealed the presence

Fungi anaesthesia.

Electrical activity of fungus Pleurotus ostreatus is characterised by slow (h) irregular waves of baseline potential drift and fast (min) action potential likes spikes of the electrical potential. An exposure of the myceliated substrate to a chloroform vapour lead to several fold decrease of the baseline potential waves and increase of their duration. The chloroform vapour also causes either complete cessation of spiking activity or substantial reduction of the spiking frequency. Removal of the chloroform vapour from the growth containers leads to a gradual restoration of the mycelium electric

An electron microscopy study of wall expansion during Candida albicans yeast and mycelial growth using concanavalin A-ferritin labelling of mannoproteins.

Depending upon growth temperature, Candida albicans can exhibit two different morphologies, a budding yeast or a mycelium. By studying the distribution of concanavalin A-ferritin particles on the cell wall surface during bud and germ tube formation, we have elucidated the way cell wall extension occurs. Both processes initially require the localized lysis of the wall in order to allow the incorporation of the newly synthesized material. Later on, the cell wall behaves as an elastic structure, allowing extension by an intussusception process and, as a consequence, cell growth.

Host-parasite relationships in paracoccidioidomycosis.

A viewpoint of host-parasite relationships in paracoccidioidomycosis is presented. The characteristics of the fungus which are important to the host-parasite interaction are discussed. Aspects of inhibition of mycelium-to-yeast transformation by estrogens acting at receptors on the fungal wall and in the cytoplasm, and the role of polysaccharide components of the cell wall in virulence are reviewed. The natural mechanisms of host defense are also examined, including phagocytosis, complement system, natural-killer cells and genetic control of resistance and susceptibility. Finally, a discussion

H2O2-dependent decolorization of Poly R-481 by particulate fractions from Phanerochaete chrysosporium.

A cell-free preparation from Phanerochaete chrysosporium culture medium decolorized the polymeric dye Poly R-481. The majority of this decolorization activity sedimented when centrifuged at 150,000 X g, indicating that it was associated with a particulate body. The activity was sensitive to heat, azide and cyanide, was stimulated by exogenously added H2O2, and was optimal around pH 4. Electron micrographs of the sedimented culture medium fraction showed the presence of numerous particulate structures. A similar dye decolorization activity from sonicated mycelium also sedimented at 150,000 X g.

Long-term cryopreservation of ectomycorrhizal basidiomycetes: A comparative evaluation of the vermiculite and modified perlite protocols.

This study evaluated the long-term cryopreservation efficacy of two previously developed protocols-the Modified Perlite Protocol (MPP) and the Vermiculite Protocol (VP)-using 65 ectomycorrhizal basidiomycete culture strains stored for five years in the vapor phase of a liquid nitrogen tank. Survival rates were assessed based on recovery ratios, with both MPP and VP showing high viability: 64 of 65 strains showed complete recovery under each protocol. Colony morphology remained stable across all methods, showing no visible phenotypic changes due to cryopreservation. Regarding post-thaw mycelial

Degradation of rifamycin from mycelial dreg by activated persulfate: Degradation efficiency and reaction kinetics.

Rifamycin mycelial dreg (RMD) is a biological waste, and its residual rifamycin (RIF) is potentially harmful to both the environment and human health. In this work, thermally activated persulfate (PDS) oxidative degradation of RIF in RMD was developed for the first time. The effects of reaction temperature, initial PDS concentration, and pH on RIF degradation in RMD were investigated, and the treatment conditions were optimized using response surface methodology (RSM). The results showed that 90 °C, 50 mg/g PDS, and pH = 5.3 were the optimal pretreatment conditions, and 100% degradation effici

Microbe Profile: Aspergillus fumigatus: a saprotrophic and opportunistic fungal pathogen.

Aspergillus fumigatus is a saprotrophic fungus that continuously disseminates spores (conidia) into the environment. It is also the most common and opportunistic aerial fungal pathogen, causing allergic and chronic lung pathologies including the fatal invasive aspergillosis in immunocompromised patients. The pathobiology of aspergillosis is complex and depends on the competence of the host immune system. Moreover, A. fumigatus has become a model to study unique features of fungi. This includes the fungal cell wall, which not only acts as a rigid skeleton for protection against hostile environm

Differential Expression of Extracellular Lipase and Protease Activities of Mycelial and Yeast Forms in Malassezia furfur.

Malassezia furfur is a dimorphic yeast that is part of the human skin microflora. This fungus is a pathogen of a certain skin diseases, such as pityriasis versicolor, and in rare cases causes systemic infection in neonates. However, the role of dimorphism in the pathogenicity remains unclear. A modified induction medium (IM) was successfully able to induce mycelial growth of M. furfur under both solid and liquid condition. Filamentous elements with branching hyphae were observed when cultured in the IM. Furthermore, addition of bovine fetus serum into the liquid IM did not promote hyphal forma

[Aspergillosis of the brain].

A case of aspergillosis of the brain in a 49-year old woman is described. In view of the fact that there were no inflammatory changes in other organs, the assumption was put forward that the fungus had penetrated into the brain by the otogenic way, or through the accessory nasal sinuses. The results of experimental infection of laboratory animals aimed at obtaining a model of meningoencephalitis are discussed. It was shown that spores of the Aspergillus fumigatus fungus in low concentrations introduced into the otic veins penetrated into the lungs where they caused an inflammatory process, and

Antifungal Activity and Possible Mechanism of Streptomyces nojiriensis 9-13 Against Mycogone sp., Causing Wet Bubble Disease on Agaricus bisporus.

Wet bubble disease (WBD) in Agaricus bisporus caused by Mycogone species imposes a substantial economic loss to mushroom production in China. Currently, fungicide application is the main method to control WBD. However, excessive use of fungicides is challenged by the appearance of resistance and food safety. Therefore, it is necessary to explore safe and efficient strategies to control WBD. Strain 9-13, isolated from the rhizosphere soil of Taxus chinensis, showed strong inhibitory activity against three Mycogone species.
