BACKGROUND: Heart failure is driven by pathological remodeling in which inflammation and mitochondrial dysfunction reinforce each other, yet the molecules that coordinate this interplay remain poorly defined. Netrin-3 is a guidance cue with no known function in the heart. We asked whether Netrin-3 acts in cardiomyocytes to affect mitochondrial integrity under inflammatory stress.
METHODS: We used neonatal rat cardiomyocytes treated with nigericin to activate the NLRP3 inflammasome. Netrin-3 was overexpressed via adenovirus or knocked down by siRNA. Its subcellular distribution was tracked by

BACKGROUND: Epidemiological studies show periodontitis and type 2 diabetes share a bidirectional aggravating relationship, yet the molecular mechanisms linking systemic metabolic dysregulation to local periodontal destruction remain unclear, and effective therapies for this comorbidity are lacking. Certain G protein-coupled receptors (GPCRs) enhance insulin secretion and attenuate inflammation. We hypothesized that GPCR regulators may contribute to crosstalk between systemic metabolic abnormalities and periodontal immunopathology.
METHODS: Using db/db mice with ligature-induced periodontitis,

BACKGROUND AND PURPOSE: Myocardial dysfunction is a significant complication associated with sepsis. However, there are currently no specific and effective treatments available. Inhibiting gasdermin D (GSDMD)-mediated pyroptosis has shown promise in mitigating sepsis-induced myocardial dysfunction. The GSDMD inhibitor Y2 (GI-Y2) has been demonstrated to directly bind to GSDMD. Nonetheless, it remains uncertain whether GI-Y2 offers a cardioprotective effect in the context of sepsis-induced myocardial dysfunction.
EXPERIMENTAL APPROACH: A mouse model of sepsis was created using lipopolysacchari

BACKGROUND: Sepsis-associated acute lung injury (ALI) remains a critical clinical challenge with limited therapeutic options. The NLRP3 inflammasome drives pathological inflammation in ALI, yet clinical translation of existing inhibitors is hindered by toxicity. Natural products offer safer alternatives, but their mechanisms in targeting NLRP3 assembly are poorly defined.
METHODS: We screened a natural compound library for NLRP3 inhibitors using LPS/nigericin-stimulated murine peritoneal macrophages, measuring IL-1β release by ELISA. Mechanistic studies included immunoprecipitation (NEK7-NLRP

Low-density lipoprotein receptor, a classic cholesterol regulatory receptor, has been found to inhibit NLR family pyrin domain containing protein 3 (NLRP3) inflammasome activation in neurons following ischemic stroke and to suppress the activation of microglia and astrocytes in individuals with Alzheimer's disease. However, little is known about the effects of low-density lipoprotein receptor

BACKGROUND: Spinal cord injury (SCI), which reportedly induces severe motor dysfunction, imposes a significant social and financial burden on affected individuals, families, communities, and nations. Acupuncture combined with moxibustion (AM) therapy has been widely used for motor dysfunction treatment, but the underlying mechanisms remain unknown. In this work, we aimed to determine whether AM therapy could alleviate motor impairment post-SCI and, if so, the potential mechanism.
METHODS: A SCI model was established in mice through impact methods. AM treatment was performed in SCI model mice

The present study aimed to explore the role of oxytocin (OT) in myocardial injury induced by ischemia/reperfusion (I/R) and hyperglycemia and its underlying mechanisms. In this study, the isolated rat hearts underwent I/R in Langendorff perfusion model and H9c2 cells were subjected to hypoxia/reoxygenation (H/R) to establish an in vitro model. I/R injury was induced by exposing the rat hearts to 40 min of global ischemia followed by 60 min of reperfusion. H9c2 cells were cultured under the normoglycemic or hyperglycemic condition with or without pretreatment of OT, and then exposed to 4 h of h

AIMS: Calcium phosphate (CaP) particle deposits are found in several inflammatory diseases including atherosclerosis and osteoarthritis. CaP, and other forms of crystals and particles, can promote inflammasome formation in macrophages leading to caspase-1 activation and secretion of mature interleukin-1β (IL-1β). Given the close association of small CaP particles with vascular smooth muscle cells (VSMCs) in atherosclerotic fibrous caps, we aimed to determine if CaP particles affected pro-inflammatory signalling in human VSMCs.
METHODS AND RESULTS: Using ELISA to measure IL-1β release from VSM

Rabbit corneal epithelial cells, loaded with the pH-sensitive fluorescent dye 2',7'-bis(2-carboxy-ethyl)-5(6)-carboxyfluorescein, show a profound acidification (pH 7.33 to 6.75) in HCO3(-)-free Ringer solution when exposed to the Na(+)-H+ exchange inhibitor amiloride. This indicates that the cells are under a constant acid load that is being countered by Na(+)-H+ exchange. Amiloride-induced acidification was affected neither by incubation in Cl(-)-free Ringer solution nor by hypoxia, indicating that the potential acid loaders, Cl(-)-HCO3- (or OH-) exchange or glycolytic metabolism, are not con

Malaria, caused by Plasmodium spp. parasites, presents treatment challenges due to the emergence of resistant strains to frontline antimalarials. In this study, we explored activities of 24 furoxan derivatives, originally designed for leishmaniasis treatment, against intraerythrocytic Plasmodium falciparum parasites. The studies were carried out with genetically-modified P. falciparum expressing GCaMP3, a cytosolic calcium indicator, and maintained in human red blood cell culture. The most potent compound Furoxan (1,2,5-oxidiazole-2N-oxide) 4m exhibited significant antimalarial activity (EC50

Acylcoenzyme A:cholesterol acyltransferase (ACAT) and/or cholesteryl esters have been implicated as important factors in the normal assembly of apolipoprotein (apoB)-containing lipoproteins. The predominant substrate for ACAT is believed to originate from cholesterol contained within the plasma membrane. To investigate a possible role of intestinal plasma membrane cholesterol in triacylglycerol-rich lipoprotein synthesis and secretion, CaCo-2 cells were incubated with agents that are known to interfere with cholesterol transport from the plasma membrane to the ER. Progesterone, verapamil, and

Lysosomal transport of monoiodotyrosine was characterized in countertransport experiments using rat FRTL-5 thyroid cell lysosomes. Monoiodotyrosine carrier activity was temperature-dependent (Ea = 11.65 kcal/mol) and had a pH optimum of 7.5. Carrier activity was minimally inhibited by KCl and NaCl, but unaffected by the presence of other ions or ATP. Monoiodotyrosine transport was unaffected by the presence of carbonyl cyanide m-chlorophenylhydrazone, nigericin, or ammonium chloride, indicating that a proton or K+ gradient is not necessary for monoiodotyrosine transport across the lysosomal me

BACKGROUND: Inflammatory bowel disease (IBD), a chronic immune disorder, has increasing global incidence and poor treatment outcome. Abnormal macrophage function is implicated in the pathophysiology of IBD. In this study, we investigated the mechanism by which human umbilical cord mesenchymal stem cell-derived exosomes (hucMSC-Ex) inhibit inflammation in IBD mouse and macrophage inflammation models.
METHODS: We established a dextran sodium sulfate (DSS)-induce BALB/c mice model of IBD and treated with hucMSC-Ex via tail vein to evaluate their repair effect on IBD mice. An in vitro macrophage

The NLRP3 inflammasome is a multiprotein complex that mediates caspase-1 activation and the release of proinflammatory cytokines, including interleukin (IL)-1β and IL-18. Gain-of-function variants in the gene encoding NLRP3 (also called cryopyrin) lead to constitutive inflammasome activation and excessive IL-1β production in cryopyrin-associated periodic syndromes (CAPS). Here we present functional screening and automated analysis of 534 NLRP3 variants from the international INFEVERS registry and the ClinVar database. This resource captures the effect of NLRP3 variants on ASC speck formation s
