Neurons that migrate from the olfactory epithelium in the chick express luteinizing hormone-releasing hormone.

In several mammalian species, luteinizing hormone-releasing hormone (LHRH) neurons have been shown to migrate from nasal regions to the brain during early development. Using immunocytochemistry, we have identified LHRH containing neurons in developing chick embryos. In embryonic day 4 (E4) and E5 animals, a small group of LHRH immunoreactive (IR) neurons were found just ventral to the olfactory pit. LHRH-IR neurons were also found within the immunoreactive (IR) neurons were found just ventral to the olfactory pit. LHRH-IR neurons were also found within the nasal epithelium. In E6 and E7 animal

Simulation of chaotic EEG patterns with a dynamic model of the olfactory system.

The main parts of the central olfactory system are the bulb (OB), anterior nucleus (AON), and prepyriform cortex (PC). Each part consists of a mass of excitatory or inhibitory neurons that is modelled in its noninteractive state by a 2nd order ordinary differential equation (ODE) having a static nonlinearity. The model is called a KOe or a KOi set respectively; it is evaluated in the "open loop" state under deep anesthesia. Interactions in waking states are represented by coupled KO sets, respectively KIe (mutual excitation) and KIi (mutual inhibition). The coupled KIe and KIi sets form a KII

Differential expression of RET receptor isoforms in the olfactory system.

The glial cell line-derived neurotrophic factor (GDNF) family supports neurons by activating the tyrosine kinase receptor RET. The two main isoforms of RET, RET9 and RET51, differ in their carboxyl termini and have been implicated with distinct functions in the enteric and central nervous systems. Previously we reported the cellular localization of GDNF, neurturin and RET9 in the olfactory system [Maroldt H, Kaplinovsky T, Cunningham AM (2005) J Neurocytol 34:241-255]. In the current study, we examined immunohistochemical expression of RET9 and RET51 in neonatal and adult rat olfactory neuroep

Differential expression of N-CAM, vimentin and MAP1B during initial pathfinding of olfactory receptor neurons in the mouse embryo.

Olfactory receptor neurons extend their primary axons from the nasal epithelium to the olfactory bulb primordium via the frontonasal mesenchyme. In the present study, expression of neuronal markers (vimentin and MAP1B) and N-CAM was immunohistochemically investigated in the development of the olfactory system in mouse embryos. Expression of vimentin and MAP1B was first observed at early day 10 of gestation (D10) in the posterosuperior region of the medial nasal epithelium, while N-CAM was initially detected in the mesenchyme adjacent to the vimentin- and MAP1B-positive nasal epithelium. As dev

Anatomy and forebrain projections of the olfactory and vomeronasal organs in axolotls (Ambystoma mexicanum).

We examined the anatomy of the nasal cavity and forebrain in the axolotl (Ambystoma mexicanum) to determine whether the olfactory and vomeronasal systems are present in this neotenic aquatic salamander. The current study was motivated by two considerations: (a) little is known of the anatomy of the vomeronasal system in aquatic vertebrates, and (b) the presence of both olfactory and vomeronasal systems in larval amphibians has broad implications for the evaluation of these systems in vertebrates. From cresyl-violet-stained sections of snouts we determined that the nasal cavity of axolotls is m

Olfactory but not vomeronasal mediation of scent marking by male golden hamsters.

Although olfactory and vomeronasal projections to the olfactory bulb and to the rest of the brain are quite distinct, it is not clear how the functions of these two chemosensory systems differ. In these experiments we attempted to determine the roles of the main olfactory and vomeronasal sensory systems in mediating scent marking by male golden hamsters, a behavior known to be stimulated by odors of other hamsters. Lesions of the main olfactory mucosa by irrigation of the nasal cavity with a solution of zinc sulfate dramatically reduced the frequency of marking. The degree of reduction in mark

Afferent and efferent connections of the nucleus sphericus in the snake Thamnophis sirtalis: convergence of olfactory and vomeronasal information in the lateral cortex and the amygdala.

This paper is an account of the afferent and efferent projections of the nucleus sphericus (NS), which is the major secondary vomeronasal structure in the brain of the snake Thamnophis sirtalis. There are four major efferent pathways from the NS: 1) a bilateral projection that courses, surrounding the accessory olfactory tract, and innervates several amygdaloid nuclei (nucleus of the accessory olfactory tract, dorsolateral amygdala, external amygdala, and ventral anterior amygdala), the rostral parts of the dorsal and lateral cortices, and the accessory olfactory bulb; 2) a bilateral projectio

Lectin binding to olfactory system in a shark, Scyliorhinus canicula.

Lectin histochemical studies were performed on the olfactory system of Scyliorhinus canicula to identify specific glycoconjugates on the cell surface of primary olfactory neurons. The olfactory receptor cells, the olfactory nerve fibers and their terminals in the bulbs were labelled with SBA, BSA-I and BSA-I-B4. The lectin staining patterns indicate that the membranes of small-spotted catshark olfactory neurons had glycoproteins with alpha-galactose residues. This carbohydrate moiety could be related to modulation of the cell-cell interactions in the olfactory system.

Relative contribution of brain and peripheral connections to postnatal growth and cell accretion in the rat olfactory bulb.

Total weight and DNA (cell number) determination in whole olfactory bulbs (OB) revealed that unilateral transection of the olfactory peduncle in newborn rats causes 2 X more (40%) deficit in cell number in the operated OB than section of the olfactory nerve. The deficit due to combined operations ('isolation' of OB) were additive. Similar effects were observed on weight gain. It is inferred that both olfactory nerve and sources in the brain exert marked influences on postnatal cell accretion and growth in the OB and that the greater share of the brain is due to its direct contribution of new m

Current generators and properties of early components evoked in rat olfactory cortex.

Depth-profile, current-source-density (CSD) and impedance analysis were used to determine the current generators of secondary waves "a" and "b" in the response evoked in pyriform cortex (PC) of the urethane anesthetized rat following OB or LOT stimulation. Positive peaks (sinks) in the second-derivative curves of the "a" and "b" waves were localized at 50-75 and 225-250 microns deep, respectively. Cortical impedance was significantly (p less than 0.01) correlated with the cell packing density of PC layers, being maximal close to the zero dipole point of the gross evoked response; magnitude of

[Electrical characteristics of the granular neurons of the carp olfactory bulb].

Using the intracellular recording technique the electrical activity of the carp olfactory bulb granular cells and secondary neurons was studied. Early and late EPSP and IPSP were the synaptic responses of granular cells both to the olfactory nerve and olfactory tract electrical stimulation. The comparison of responses of the granular cells and secondary neurons led to the conclusion that the granular cells are interneurons producing post-synaptic inhibition of the secondary neurons. It was supposed that the granular neurons are excited by dendro-dendritic and reccurent-collateral pathways and

Olfactory-induced locomotion in lampreys.

The olfactory system allows animals to navigate in their environment to feed, mate, and escape predators. It is well established that odorant exposure or electrical stimulation of the olfactory system induces stereotyped motor responses in fishes. However, the neural circuitry responsible for the olfactomotor transformations is only beginning to be unraveled. A neural substrate eliciting motor responses to olfactory inputs was identified in the lamprey, a basal vertebrate used extensively to examine the neural mechanisms underlying sensorimotor transformations. Two pathways were discovered fro

Spatiotemporal patterns of expression of extracellular matrix molecules in the developing and adult rat olfactory system.

Using immunocytochemical methods, we have examined extensively the spatial and temporal patterns of expression of three extracellular matrix molecules-laminin, fibronectin, and type IV collagen-in the embryonic, postnatal (days 2 and 11) and adult rat olfactory system. The study started at embryonic day 14 when olfactory fibres and their associated migrating cells course through the nasal mesenchyme. From embryonic day 14 to the adult, a sheet-like pattern of labelling for laminin, fibronectin and type IV collagen was observed along the basal surface of the olfactory epithelium and around the

Nuclear imaging with radiolabeled odor molecules in patients with olfactory disorder.

Smell loss originates from peripheral disorders, like intranasal obstruction and olfactory cell injury, as well as central pathway diseases. Information derived from electrophysiological and psychophysical tests are useful for identifying loss of smell, but not for discriminating between central and peripheral deficits. This is because conventional imaging modalities are unable to deliver information about functional olfactory performance. Although functional imaging is able to show abnormal changes in central olfactory pathways, it seems that it is only possible to observe such abnormalities

Development of the olfactory system in a wallaby (Macropus eugenii).

We used carbocyanine dye tracing techniques in conjunction with hematoxylin and eosin staining, immunohistochemistry for GAP-43, and tritiated thymidine autoradiography to examine the development of the olfactory pathways in early pouch young tammar wallabies (Macropus eugenii). The overarching aim was to test the hypothesis that the olfactory system of newborn tammars is sufficiently mature at birth to contribute to the guidance of the pouch young to the nipple. Although GAP-43 immunoreactive fibers emerge from the olfactory epithelium and enter the olfactory bulb at birth, all other componen

Unilaterally disrupted structural and functional connectivity of the fronto-limbic system in idiopathic hypogonadotropic hypogonadism.

OBJECTIVE: Idiopathic hypogonadotropic hypogonadism (IHH) is rare and can either be associated with normal or defective olfactory sensation, classified as normosmic IHH (nIHH) or Kallmann syndrome (KS). We do not yet understand the central processing pathways in the olfactory system. We aimed to compare the resting-state structural and functional connectivity (FC) of olfactory neural pathways in patients with IHH. We hypotheses that alterations of structural connectivity and FC may exist in the olfactory cortex pathways in IHH patients. DESIGN: STRUCTURAL AND FUNCTIONAL CONNECTIVITY DATA RESU

Intrabulbar projecting external tufted cells mediate a timing-based mechanism that dynamically gates olfactory bulb output.

In the mammalian olfactory system, intrabulbar projections (IBPs) mediated by a class of external tufted cells (ET cells) specifically link isofunctional odor columns within the same olfactory bulb. To study the function of these ET cells within the glomerular network, we developed a "hemibulb" preparation that maintains IBPs intact enabling the select activation of ET cells associated with specific glomeruli. Using P2-GFP mice, a line in which the P2 glomeruli are labeled with green fluorescent protein, we recorded from P2 mitral cells (MT cells) while selectively stimulating P2 ET cells. Her
