Minimally invasive biopsy technique predicting breast pathological complete response after neoadjuvant therapy for breast cancer.

BACKGROUND: Neoadjuvant therapy (NAT) is widely used in the treatment of breast cancer, and the pathological complete response (pCR) rate is increasing. However, currently, the prediction of pCR still lacks accuracy. This study aimed to investigate the accuracy of minimally invasive biopsy techniques in predicting breast pCR (bpCR) after NAT in breast cancer. METHODS: From October 2022 to October 2024, a prospective single-arm study was conducted on 132 patients with primary breast cancer who achieved breast radiologic complete response (brCR) or breast radiologic partial response (brPR) afte

Predictive factors of axillary pathological complete response in HER2-positive breast cancer patients treated with neoadjuvant chemotherapy.

OBJECTIVE: Axillary lymph node dissection (ALND) is associated with significant morbidity in breast cancer patients. In HER2-positive (HER2+) breast cancer, neoadjuvant chemotherapy (NAC) combined with anti-HER2 therapy can achieve high rates of pathological complete response (pCR), supporting the de-escalation of axillary surgery. Identifying reliable predictors of axillary pCR is essential for safely omitting ALND in selected patients. MATERIALS AND METHODS: This retrospective cohort study included 60 women with HER2+ breast cancer and axillary involvement treated with NAC followed by surge

The patterns and risk factors for relapse in oesophageal squamous cell cancers that achieve pathological complete response to neoadjuvant chemoradiotherapy.

OBJECTIVES: The goal of this study was to investigate the patterns and risk factors for recurrence in patients with oesophageal squamous cell carcinoma with a pathological complete response (pCR) after neoadjuvant chemoradiotherapy (nCRT). METHODS: Between January 2008 and December 2018, a total of 96 patients with pCR were enrolled in this study. Lymph nodes with a pCR [LN-ypCR response (+)] were defined as those lymph nodes without residual tumour but with the presence of treatment response to nCRT. Prognostic factors for recurrence-free survival (RFS) were analysed with Cox proportional ha

[Savolitinib Induced Pathological Complete Response in Non-small Cell Lung Cancer with MET Amplification: A Case Report].

Mesenchymal-epithelial transition factor (MET) gene mutation is a large class of mutations commonly seen in non-small cell lung cancer (NSCLC). MET mutation includes subtypes such as MET exon 14 skipping mutation (METex14m) and MET amplification (METamp). For advanced NSCLC with METex14m, Savolitinib has a high sensitivity as a member of tyrosine kinase inhibitors (TKIs). METamp is a relatively rare genetic mutation type which can serve as a driver gene to mediate primary and later acquired drug resistance of epidermal growth factor receptor (EGFR)-TKIs. For advanced NSCLC with secondary METam

Racial Disparities in Pathological Complete Response Among Patients Receiving Neoadjuvant Chemotherapy for Early-Stage Breast Cancer.

IMPORTANCE: Among patients with breast cancer, inconsistent findings have been published on racial disparities in achieving pathologic complete response (pCR) after neoadjuvant chemotherapy (NACT). OBJECTIVE: To investigate whether racial disparities exist in achieving pCR and what factors contribute to them. DESIGN, SETTING, AND PARTICIPANTS: Within the ongoing Chicago Multiethnic Epidemiologic Breast Cancer Cohort (ChiMEC), which consists of a prospectively ascertained cohort of patients with breast cancer, 690 patients with stage I to III breast cancer receiving NACT were identified for t

Predictive markers for pathological complete response (pCR) after neo-adjuvant chemotherapy in HER2-positive breast carcinoma.

OBJECTIVES: Patients with HER2-positive invasive breast cancer that is node-positive and/or larger than 3 cm are generally treated with neoadjuvant chemotherapy (NAC). We aimed to identify predictive markers for pathological complete response (pCR) after NAC in HER2-positive breast carcinoma. METHODS: Hematoxylin/eosin-stained slides of 43 HER2-positive breast carcinoma biopsies were histopathologically reviewed. Immunohistochemistry (IHC) was performed on pre-NAC biopsies, comprising HER2, estrogen receptor (ER), progesterone receptor (PR), Ki-67, epidermal growth factor receptor (EGFR), muc

A case of pathological complete response with liposomal irinotecan + 5-FU/LV for unresectable locally advanced pancreatic cancer.

BACKGROUND: Pancreatic cancer has one of the worst prognoses of any all cancers. 5-FU/leucovorin + irinotecan + oxaliplatin (FOLFIRINOX), gemcitabine (GEM) plus nab-paclitaxel regimens have been recognized as global-standard, first-line treatments for patients with advanced pancreatic cancer. The liposomal irinotecan (nal-IRI) + 5-FU/LV regimen is now included in treatment guidelines as a recommended and approved option for use in patients with metastatic pancreatic cancer that has progressed after GEM-based therapy and who have a suitable performance status and comorbidity profile. There is n

Impact of body mass index on pathological complete response and survival of breast cancer patients receiving neoadjuvant chemotherapy.

BACKGROUND: High body mass index (BMI) is regarded as a poor prognostic factor in breast cancer (BC). However, its association with pathological complete response (pCR) and survival after neoadjuvant chemotherapy (NAC) remains controversial. OBJECTIVE: We wanted to assess the prognostic impact of BMI in this setting. METHODS: Single-center, retrospective review of 314 BC patients undergoing NAC from 2010 to 2018. Patients were categorized as underweight/normal weight (UW/NW) (BMI < 25) or overweight/obese (OW/OB) (BMI ≥ 25). The relationship between BMI and other traditional clinical-patholo

Pathological Complete Response following Neoadjuvant Chemotherapy in Invasive Ductal Carcinoma with Mammary Paget's Disease: A Case Report.

Mammary Paget's disease is a rare malignancy. Mastectomy or breast-conserving surgery has been considered as the standard treatment, while there have been few reports of neoadjuvant chemotherapy (NAC). A 53-year-old woman with erythema and skin ulceration of the left breast was admitted to our institution. Breast examinations revealed left invasive ductal carcinoma (cT1bN0M0, cStage I), and a punch biopsy of the left mammary erythema indicated Paget's disease (cTisN0M0, cStage0). The patient received NAC because of multiple lesions. Consequently, the breast tumor clinically disappeared, and th

MRI Volume Changes of Axillary Lymph Nodes as Predictor of Pathologic Complete Responses to Neoadjuvant Chemotherapy in Breast Cancer.

INTRODUCTION: Longitudinal monitoring of breast tumor volume over the course of chemotherapy is informative of pathologic response. This study aims to determine whether axillary lymph node (aLN) volume by magnetic resonance imaging (MRI) could augment the prediction accuracy of treatment response to neoadjuvant chemotherapy (NAC). MATERIALS AND METHODS: Level-2a curated data from the I-SPY-1 TRIAL (2002-2006) were used. Patients had stage 2 or 3 breast cancer. MRI was acquired pre-, during, and post-NAC. A subset with visible aLNs on MRI was identified (N = 132). Prediction of pathologic comp

Impact of Genetics on Neoadjuvant Therapy with Complete Pathological Response in Metastatic Colorectal Cancer: Case Report and Review of the Literature.

Treatment of colorectal metastatic cancer is still challenging, despite recent improvements in chemotherapy. A genetic cancer profile, such as the KRAS (Kirsten rat sarcoma) gene status, plays a key role in individualized tailored therapy. Molecular targeted therapy added to neo-adjuvant chemotherapy can achieve a better pathological response and prolong survival. Pathological complete response of colorectal cancer stage IV is rare. A 47-year-old female patient presented with rectal adenocarcinoma and three liver metastases (cT3d/4, N2, Ml). After seven cycles of Bevacizumab and CAPOX in neoad

Identifying Predictors of Pathological Complete Response in Moroccan Patients With HER2-Positive Breast Cancer Treated With Neoadjuvant Therapy.

Background The prognosis of human epidermal growth factor receptor 2 (HER2)-positive breast cancer has significantly improved with the use of neoadjuvant chemotherapy combined with anti-HER2 therapy. Achieving pathological complete response (pCR) is commonly associated with favorable long-term outcomes. However, real-world data on predictive factors influencing pCR remain limited, particularly in Morocco and North Africa, where evidence is scarce. Generating region-specific evidence is therefore essential to complement findings from other populations. This study aimed to assess pCR rates and i

Evaluating the Surrogacy of Pathological Complete Response at Radical Cystectomy in Neoadjuvant-based Muscle-invasive Bladder Cancer Trials.

Overall survival (OS) remains the gold standard end point in neoadjuvant trials for muscle-invasive bladder cancer (MIBC), yet its assessment requires prolonged follow-up. Whether pathological complete response (pCR) at radical cystectomy (RC) can serve as a valid trial-level surrogate remains unclear. To test this hypothesis, we conducted a systematic review and meta-analytic surrogacy analysis (PROSPERO CRD420251050357) of prospective nonrandomized trials and randomized controlled trials (RCTs) evaluating neoadjuvant therapies followed by RC in nonmetastatic MIBC (2003-2026).
