Efficiency of lornoxicam in lung and trachea injury caused by peroxynitrite.

Peroxynitrite is involved in the pathogenesis of pulmonary diseases such as asthma, occupational pulmonary diseases and acute respiratory distress syndrome (ARDS) due to excessive production of nitric oxide or superoxide or both. Lornoxicam, a new oxicam derivative, is a potent anti-inflammatory agent. In this study, we evaluated the role of lornoxicam in a peroxynitrite-induced pulmonary and tracheal injury model by measuring myeloperoxidase (MPO) activity, malondialdehyde (MDA) and 3-nitrotyrosine (3-NT) levels in lung tissue and bronco-alveolar lavage fluid. The study protocol was based on

Xanthine oxidase converts nitric oxide to nitroxyl that inactivates the enzyme.

Xanthine oxidase (XO) was found to convert nitric oxide (NO* ) released from spermine-NONOate to nitroxyl (HNO), the one-electron reduction product of NO*, in the presence of its substrate hypoxanthine under anaerobic conditions. Under these conditions, XO lost its activity. Upon aerobic incubation of XO with its substrate, neither conversion of NO* to HNO nor inactivation of the enzyme was observed. Angeli's salt (an HNO generator) or synthetic peroxynitrite inactivated XO at low concentrations, whereas high concentrations of diethylamine-NONOate (an NO* donor) and SIN-1 (which generates pero

The effect of peroxynitrite on the catalytic activity of soluble guanylyl cyclase.

Soluble guanylyl cyclase (sGC) is a key enzyme of the *NO/cGMP pathway. Many cardiovascular disorders are associated with reduced *NO-mediated effects, while vascular superoxide (O(2)*(-)) production is increased. Both radicals rapidly react to peroxynitrite. We investigated whether peroxynitrite affects the activity and protein expression of sGC in intact vascular preparations. Catalytic sGC activity and expression of the sGC-beta(1) subunit was measured by conversion of radiolabeled GTP and western blot, respectively, using cytosolic extracts from rat aorta that had been incubated for 4 h wi

Role of intracellular calcium mobilization and cell-density-dependent signaling in oxidative-stress-induced cytotoxicity in HaCaT keratinocytes.

Peroxynitrite is a nitric-oxide-derived cytotoxic mediator produced in a broad range of inflammatory conditions, ranging from sunburn erythema to contact hypersensitivity. Our previous work has shown that in HaCaT cells the cytotoxic activity of peroxynitrite involves both apoptotic and necrotic routes with poly(ADP-ribose) polymerase activation serving as a mol-ecular switch diverting the default apoptotic pathway toward necrosis. Nonetheless, keratinocytes are regarded as highly resistant toward environmental noxa including oxidative stress. We set out to investigate the possible role of two

Nitration in cancer signaling.

Oxidative stress arises from an imbalance between the production of reactive oxygen and nitrogen species (ROS and RNS, respectively) and the capacity of cellular antioxidant defenses to neutralize them. In cancer, this imbalance drives pathological remodeling of signaling networks that promote tumor initiation and progression. Among RNS, nitric oxide (·NO) and its highly reactive derivative peroxynitrite (ONOO-) are central mediators of redox dysregulation within the tumor microenvironment. These species induce site-specific post-translational modifications (PTMs), most notably protein tyrosin

Reactive nitrogen species-driven oxidative and nitrative damage underlying cadmium/mercury co-exposure: A multimodal fluorescent visualization study.

Heavy metal contamination severely threatens ecological environment and human health. Few studies have established an integrated mechanistic framework linking heavy metal co-exposure, reactive nitrogen species (RNS) burst, and progressive cellular injury, especially for multi-metal combined pollution that better reflects real environmental scenarios. As downstream substance of nitric oxide (NO) and representative RNS, peroxynitrite (ONOO-) possess stronger oxidation/nitration capacity compared with hydrogen peroxide (H2O2). The oxidative/nitrifying stress ignited by RNS outburst under cadmium

Photoresponsive Nitric Oxide Release from Aggregation-Induced Emission-Based Ruthenium-Nitrosyl Complexes and Their Anticancer Activity through In Situ Formation of Peroxynitrite.

Nitric oxide (NO) is recognized as a gaseous chemical mediator that regulates various physiological functions. Two naphthalene-appended terpyridine-based mononuclear ruthenium-nitrosyl complexes with the molecular framework [1.Ru2+(L)NO](PF6)3 (L = 2,2'-bipyridine (bpy) or phenanthroline (phen) and 1 = 4'-(anthracene-9-yl)-2,2':6',2″-terpyridine), named 1.Ru(bpy)NO and 1.Ru(phen)NO, were reported for NO delivery. The introduction of ancillary ligands (bpy and phen) to the Ru-center led to the noticeable variations of photophysical properties and aggregation behavior in aqueous solution, which

Red-emitting fluorescent probe sequentially activated by peroxynitrite and monoamine oxidase for diagnosis and therapeutic evaluation of drug-induced liver injury.

Drug-induced liver injury (DILI) has emerged as a major public health issue due to the extensive use of prescription or non-prescription medications. Accordingly, the development of biomarker-based fluorescent probe which holds considerable potential for diagnosing DILI severity and evaluating the curative effect of putative hepatoprotectants is essential for improving therapeutic outcomes and expediting drug discovery. Considering that the dual-biomarkers-responsive probe offers superior specificity and higher signal-to-background ratios for liver pathologies surveillance compared with the si

Peroxynitrite-Triggered Carbon Monoxide Donor Improves Ischemic Stroke Outcome by Inhibiting Neuronal Apoptosis and Ferroptosis.

Cerebral ischemia-reperfusion injury produces excessive reactive oxygen and nitrogen species, including superoxide, nitric oxide, and peroxynitrite (ONOO-). We recently developed a new ONOO--triggered metal-free carbon monoxide donor (PCOD585), exhibiting a notable neuroprotective outcome on the rat middle cerebral artery occlusion model and rendering an exciting intervention opportunity toward ischemia-induced brain injuries. However, its therapeutic mechanism still needs to be addressed. In the pharmacological study, we found PCOD585 inhibited neuronal Bcl2/Bax/caspase-3 apoptosis pathway in

Electronic cigarette exposure causes vascular endothelial dysfunction due to NADPH oxidase activation and eNOS uncoupling.

We recently reported a mouse model of chronic electronic cigarette (e-cig) exposure-induced cardiovascular pathology, where long-term exposure to e-cig vape (ECV) induces cardiac abnormalities, impairment of endothelial function, and systemic hypertension. Here, we delineate the underlying mechanisms of ECV-induced vascular endothelial dysfunction (VED), a central trigger of cardiovascular disease. C57/BL6 male mice were exposed to ECV generated from e-cig liquid containing 0, 6, or 24 mg/mL nicotine for 16 and 60 wk. Time-dependent elevation in blood pressure and systemic vascular resistance

Dipyrone and aminopyrine are effective scavengers of reactive nitrogen species.

Reactive nitrogen species (RNS), namely nitric oxide (NO*) and peroxynitrite (ONOO-) are produced in the inflammatory sites and may contribute to the deleterious effects of inflammation. The aim of the present study was to evaluate the putative scavenging effect of a particular group of non-steroidal anti-inflammatory drugs (NSAIDs), the pyrazolone derivatives dipyrone, aminopyrine, isopropylantipyrine, and antipyrine against RNS, using in vitro non-cellular screening systems. The results obtained showed that dipyrone and aminopyrine were highly potent scavengers of NO* and ONOO- while antipyr

[How do blood vessels age? Mechanisms and clinical implications].

Ageing is one of the most important cardiovascular risk factors. Vascular ageing is determined by genetic, mechanic and hemodynamic factors. The latter is strengthened by the fact that age-associated changes in capillaries and veins, vessels which are less prone to changes in pulsatility and blood pressure, are less pronounced or even absent. Age-related morphologic changes in large resistance vessels include an intima-media thickening, increased deposition of matrix substances, thus ultimately leading to a reduced compliance. Vascular ageing is mainly characterized by an impaired endothelium-

The reaction of nitric oxide with 6-hydroxydopamine: implications for Parkinson's disease.

Oxidation of catecholamines is suggested to contribute to oxidative stress in Parkinson's disease. Nitric oxide (*NO) is able to oxidize cyclic compounds like ubiquinol; moreover, recent lines of evidence proposed a direct role of *NO and its by-product peroxynitrite in the pathophysiology of Parkinson's disease. The aim of this study was to analyze the potential reaction between 6-hydroxydopamine, a classic inducer of Parkinson's disease, and *NO. The results showed that *NO reacts with the deprotonated form of 6-hydroxydopamine at pH 7 and 37 degrees C with a second-order rate constant of 1.

Antifibrotic role of inducible nitric oxide synthase.

Long-term treatment in rats with l-NAME, an isoform-non-specific inhibitor of nitric oxide synthase (NOS), leads to fibrosis of the heart and kidney, suggesting that nitric oxide (NO) may play a role in preventing tissue fibrosis. In this process, a likely target of NO is the quenching of reactive oxygen species (ROS) through peroxynitrite formation, and one possible source for this NO is inducible NOS (iNOS). Using Peyronie's disease (PD) tissue from both human specimens and from a rat model of PD as the source of fibrotic tissue, we investigated if NO derived from iNOS could act as such an a

Increased activity and expression of iNOS in human duodenal enterocytes from patients with celiac disease.

The activity of nitric oxide synthase (NOS) was assayed in enterocytes isolated from human duodenal biopsies to determine its role in celiac disease. Patients were categorized into groups with irritable bowel syndrome, iron-deficiency anemia, B(12)/folate deficiency, and treated and untreated celiac disease. Enterocytes isolated from all groups showed 1400W-inhibitable Ca2+-independent NOS activity with a pH level and temperature optimum of 9.4 and 37 degrees C, respectively. Western blotting showed that enterocytes expressed the inducible NOS protein and proteins with nitrated tyrosine residu

An endoplasmic reticulum-targeted fluorescent probe for peroxynitrite visualization in drug-induced liver injury and hepatic ischemia-reperfusion injury.

Drug-induced liver injury (DILI) and hepatic ischemia-reperfusion injury (HIRI) are closely associated with aberrant accumulation of peroxynitrite (ONOO-) in the endoplasmic reticulum (ER). In this work, we designed and synthesized ER-ND, an ER-targeted turn-on fluorescent probe for the detection of ONOO-. This probe is composed of three key functional components: a 1,8-naphthalimide-based fluorophore, a 3,5-bis(trifluoromethyl)benzyl group serving as an ER-anchoring moiety, and a 4-(dimethylamino)phenoxy group acting as a responsive unit. ER-ND demonstrates a rapid response to ONOO-, along wi

Dual-channel fluorescent probe for simultaneously visualizing ONOO- and viscosity in epilepsy, non-alcoholic fatty liver and tumoral ferroptosis models.

Intracellular peroxynitrite anion (ONOO-) and viscosity play a major part in sustaining redox homeostasis, modulating substance transport and signal transduction. Abnormalities in these factors are closely associated with multiple physiological and pathological processes. Nevertheless, due to the absence of appropriate multifunctional fluorescent sensors, concurrent identification of ONOO- and viscosity has not been achieved in many diseases, such as epilepsy and tumoral ferroptosis models. Herein, a new near-infrared (NIR) fluorescent probe (QX-DP) was rationally conceived for concurrent dete

Imatinib binding to human serum albumin modulates heme association and reactivity.

Imatinib, an inhibitor of the Bcr-Abl tyrosine kinase, is approximately 95% bound to plasma proteins, α1-acid glycoprotein (AGP) being the primary carrier. However, human serum albumin (HSA) may represent the secondary carrier of imatinib in pathological states characterized by low AGP levels, such as pancreatic cancer, hepatic cirrhosis, hepatitis, hyperthyroidism, nephrotic syndrome, malnutrition, and cachexia. Here, thermodynamics of imatinib binding to full-length HSA and its recombinant Asp1-Glu382 truncated form (containing only the FA1, FA2, FA6, and FA7 binding sites; trHSA), in the ab
