Hippeastrum papilio (Amaryllidaceae) is a promising new source of galanthamine - an alkaloid used for the cognitive treatment of Alzheimer's disease. The biosynthesis and accumulation of alkaloids are tissue - and organ-specific. In the present study, histochemical localization of alkaloids in H. papilio's plant organs with Dragendorff's reagent, revealed their presence in all studied samples. Alkaloids were observed in vascular bundles, vacuoles, and intracellular spaces, while in other plant tissues and structures depended on the plant organ. The leaf parenchyma and the vascular bundles were

BACKGROUND: Chaihu is a popular traditional Chinese medicine that has been used for centuries. It is traditionally used to treat cold fever and liver-related diseases. Saikosaponins (SSs) are one of the main active components of chaihu, in addition to essential oils, flavonoids, and polysaccharides. Considerable effort is needed to reveal the biosynthesis and regulation of SSs on the basis of current progress.
OBJECTIVE: The aim of this study is to provide a reference for further studies and arouse attention by summarizing the recent achievements of SS biosynthesis.
METHODS: All the data com

PIII-containing H-phosphonate (HPO32-) and its monoethyl ester (fosetyl), essential pesticides for control of oomycete and fungal pathogens, are among the few pesticides transported by both xylem and phloem, making application as folia spray, soil spray, and trunk injection equally effective. To understand bioavailability and efficacy within soils, knowledge of adsorption to soil minerals is important. FeOOH(goethite) is often selected as an archetypal mineral surface. In the present work, H-phosphonate (with pKa values of 1.5 and 6.78) adsorption onto FeOOH is nearly complete below pH 6 and d

Microautoradiography (MAR) is a conventional imaging method based on the daguerreotype. The technique is used to visualize the distribution of radionuclide-labeled compounds within a tissue section. However, application of the classical MAR method to plant tissue sections is associated with several difficulties. In this study, we report an MAR method applicable to fresh-frozen plant sections. Our method had two features: (i) the sample was kept frozen from plant tissue collection to radioisotope detection, making it possible to fix solutes without solvent exchange; and (ii) 1.2 µm thick polyph

Transport via xylem of trichloroethylene (TCE) from roots to shoots in seedlings of wheat, corn, and tomato was measured following a 24-h exposure of plant roots to hydroponic solutions containing TCE. Dewdrops on plant leaves were also collected to test the foliar uptake by plants and the volatilization of TCE from shoot to air. Results indicated that the TCE concentration in xylem sap of wheat and corn decreased significantly with increasing TCE concentration in external solutions, where the initial concentration was set at 10-30 mg l(-1). The translocation stream concentration factor (TSCF)

We have studied autumn leaf senescence in a free-growing aspen (Populus tremula) by following changes in pigment, metabolite and nutrient content, photosynthesis, and cell and organelle integrity. The senescence process started on September 11, 2003, apparently initiated solely by the photoperiod, and progressed steadily without any obvious influence of other environmental signals. For example, after this date, senescing leaves accumulated anthocyanins in response to conditions inducing photooxidative stress, but at the beginning of September the leaves did not. Degradation of leaf constituent

The osmotic pressure of the body fluids of aphids is lower than in their diet of plant phloem sap. It is hypothesised that aphids reduce the osmotic pressure of ingested food by sucrase-mediated hydrolysis of dietary sucrose to glucose and fructose, and the polymerisation of glucose into oligosaccharides of low osmotic pressure per hexose unit. To test this hypothesis, the impact of the alpha-glucosidase inhibitor acarbose on the sugar relations and osmoregulation of aphids was explored. Acarbose inhibited sucrase activity in gut homogenates and the production of monosaccharides and oligosacch

Pepper ( Capsicum annuum L.) SAR8.2 genes, designated CASAR82A, B and C, which are induced by all the biotic and abiotic stresses, were isolated from a pepper cDNA library constructed with the mRNAs from pepper plants infected with Xanthomonas campestris pv. vesicatoria. The pepper CASAR82A, B and C gene products, which are very similar to each other in amino acid sequences, have 43-50% homology with those of tobacco SAR8.2 genes. The CASAR8.2 genes were not constitutively expressed in any of the organs of healthy pepper plants. In contrast, the CASAR82A gene was locally or systemically induce

Hemipteran plant sap feeders acquire food by inserting their piercing-sucking mouthparts into the apoplast (xylem sap feeders) or symplast (phloem sap feeders). When feeding, these insects seal their stylets inside the plant with a salivary sheath, minimising embolisms. The main impact of the insect on the host or food plant consists of sucking plant sap, transmitting phytopathogenic agents, or triggering galls as hypertrophic and/or hyperplastic neoformations. This paper proposes a rapid method for locating and counting the salivary sheaths of Philaenus spumarius (the Meadow Spittlebug), the

The annual transport patterns of mineral nutrients in conifers remain poorly understood. Calcium (Ca) and magnesium (Mg) deficiencies often occur in planted conifer stands and cannot be easily remedied by simply adding these nutrients to soil. This study examined translocation and remobilization of Ca and Mg in young Nordmann fir (Abies nordmanniana) trees. Our data revealed that Ca and Mg absorbed by the roots do not reach the needles during the current growth season, indicating that internal nutrient remobilization is crucial for the growth of new needles, particularly shortly after the budb

Opium poppy is a crop of great commercial value as a source of several opium alkaloids for the pharmaceutical industries including morphine, codeine, thebaine, noscapine, and papaverine. Most enzymes involved in benzylisoquinoline alkaloid (BIA) biosynthesis in opium poppy have been functionally characterized, and opium poppy currently serves as a model system to study BIA metabolism in plants. BIA biosynthesis in opium poppy involves two biosynthetic gene clusters associated respectively with the morphine and noscapine branches. Recent reports have shown that genes in the same cluster are co-

BACKGROUND: Citrus huanglongbing (HLB) is a devastating disease caused by Candidatus Liberibacter asiaticus (CLas) that affects the citrus industry. In nature, CLas relies primarily on Diaphorina citri Kuwayama as its vector for dissemination. After D. citri ingests CLas-infected citrus, the pathogen infiltrates the insect's body, where it thrives, reproduces, and exerts regulatory control over the growth and metabolism of D. citri. Previous studies have shown that CLas alters the composition of proteins in the saliva of D. citri, but the functions of these proteins remain largely unknown.

UNLABELLED: Nutritional enhancement has been reported to effectively relieve infected symptoms of Huanglongbing, one of the most destructive diseases of citrus. However, few studies focused on the role of plant nutrition in citrus plant-vector (Asian citrus psyllid; Diaphorina citri Kuwayama) interactions, which is regarded as an important part to develop an effective management strategy.
METHOD: In the present study, a hydroponic culture was carried out to evaluate the effects of boron deficiency on psyllid feeding process to decode the molecular/biochemical basis of host-psyllid interaction

The study used scattered literature to summarize the effects of excess Cd, As, and Pb from contaminated soils on plant secondary metabolites/bioactive compounds (non-nutrient organic substances). Hence, we provided a systematic overview involving the sources and forms of Cd, As, and Pb in soils, plant uptake, mechanisms governing the interaction of these risk elements during the formation of secondary metabolites, and subsequent effects. The biogeochemical characteristics of soils are directly responsible for the mobility and bioavailability of risk elements, which include pH, redox potential,

BACKGROUND: The Asian citrus psyllid, Diaphorina citri Kuwayama, is the primary vector of the phloem-inhabiting bacterium Candidatus Liberibacter asiaticus putatively responsible for citrus greening (huanglongbing), a devastating citrus disease. Infestations of D. citri frequently develop on Citrus and other genera within the Rutaceae subfamily Aurantioideae including Murraya and Bergera. The genotype Poncirus trifoliata is also a member of the Aurantioideae and readily hybridizes with Citrus spp., but colonization by D. citri is reduced on this genotype.
RESULTS: Working with young potted se

An insect-induced gall is a highly specialized structure resulting from atypical development of plant tissue induced by a reaction to the presence and activity of an insect. The insect induces a differentiation of tissues with features and functions of an ectopic organ, providing nutrition and protection to the galling insect from natural enemies and environmental stresses. In this anatomical and cytological study, we characterized how the gall-inducing aphid Pemphigus betae reshapes the leaf morphology of the narrow-leaf cottonwood Populus angustifolia to form a leaf fold gall. Young galls di

A salicylic acid-inducible WRKY gene, PtrWRKY73, from Populus trichocarpa , was isolated and characterized. Overexpression of PtrWRKY73 in Arabidopsis thaliana increased resistance to biotrophic pathogens but reduced resistance against necrotrophic pathogens. WRKY transcription factors are commonly involved in plant defense responses. However, limited information is available about the roles of the WRKY genes in poplar defense. In this study, we isolated a salicylic acid (SA)-inducible WRKY gene, PtrWRKY73, from Populus trichocarpa, belonging to group I family and containing two WRKY domains,

To characterize the dynamics of Cd²⁺ flux in the rhizosphere and to study cadmium (Cd) plant-internal partitioning in roots, wood, bark and leaves in relation to energy metabolism, reactive oxygen species (ROS) formation and antioxidants, Populus × canescens plantlets were exposed to either 0 or 50 µM CdSO₄ for up to 20 days in the nutrient solution. A strong net Cd²⁺ influx in root apex was observed after Cd exposure for 24 h, even if net Cd²⁺ influx decreased gradually in roots. A large amount of Cd was accumulated in roots. Cd ions were uploaded via the xylem to leaves and further transport
