Regional differences in cholinergic innervation and drug sensitivity in the smooth muscles of pig stomach.

1 To evaluate the regional differences in cholinergic mechanisms in the stomach, innervation and receptor distribution were investigated by isometric tension recording, receptor binding assay and measurement of nerve-related enzyme activity in longitudinal and circular smooth muscle layers isolated from various regions of the pig stomach. 2 The response to transmural nerve stimulation (TMS) varied with the muscle layer and portion of stomach tested. Contraction was predominant in the smooth muscle from the pyloric antrum, relaxation in the corpus region. 3 The contraction to TMS was abolished

Adrenocorticotropic hormone (ACTH) and centrally-acting cholinomimetic drugs improve survival of rats with severe hemorrhagic shock through distinct central cholinergic mechanisms.

Pharmacological doses (40-160 micrograms/kg) of adrenocorticotropic hormone (ACTH) intravenously injected to urethane-anesthetized rats subjected to otherwise lethal hemorrhagic shock (mean arterial pressure stabilized at 20-25 mmHg) promptly restore blood pressure to about the pre-bleeding values, and prevent death (anti-shock effect). Hemicholinium-3 (i.c.v. injected) and atropine sulphate, but not atropine methylbromide, antagonize these ACTH effects. Moreover, since pirenzepine, injected i.v. or i.c.v., does not affect the anti-shock activity of ACTH, the central cholinergic mechanism part

Endothelium-dependent dilatation of the ex vivo tail artery perfused with blood from a conscious rat.

A comparison was made of the endothelium-dependent vasodilatation produced by acetylcholine (Ach) in rat-tail arteries perfused with Krebs solution or whole rat blood. Segments of the tail artery of the rat were cannulated at both ends mounted in an organ bath and perfused at a constant 3 ml/min. Blood perfused the tail artery via an extra corporeal circuit by being withdrawn from and returned to a conscious donor rat previously prepared with exteriorized indwelling carotid artery and jugular vein cannulae. Perfusion pressure was increased by constricting the tail artery with an infusion of no

Comparisons between discrete lever-press and shuttle avoidance responses in mice: acquisition processes and effects of psychoactive drugs.

Acquisition processes of discrete lever-press (L-type) and shuttle (S-type) avoidance responses as well as effects of psychoactive drugs thereon were investigated in dd strain mice. The mice showed a more rapid acquisition of S-type avoidance than L-type. However, the mean avoidance rates and occurrences of good-performing mice (showing an avoidance rate of higher than 75%) were almost the same in both types when the training was carried out for more than 15 sessions of 1 hr each. The response rate of L-type avoidance was 2.5-3 times as high as that of S-type avoidance. Methamphetamine and coc

Mary Broadfoot Walker: 83 years since a historical discovery.

Mary Broadfoot Walker was a Scottish physician who, in 1935, described in great detail the effect of an anticholinesterase drug (physostigmine) on the signs and symptoms of myasthenia gravis. An original five-minutes movie is available online and the skepticism of her contemporary British medical doctors is understandable when the drastic effect of the treatment is shown in this movie. What Mary Walker taught us, more than eight decades ago, about myasthenia gravis continues to be the basis of a pharmacological diagnostic test and treatment of this disease.

Cholinomimetic effects of aqueous extracts from Carum copticum seeds.

An aqueous extract from roasted seeds of Carum copticum (omum) has cholinomimetic effects. It shows muscarinic effects on rabbit duodenum, guinea-pig ileum and rat jejunum, and on the blood pressure of rat and cat. These effects are blocked by atropine. It also has a nicotinic action on the frog rectus preparation and atropinized cat blood pressure. Its effect is potentiated by physostigmine and antagonized by cholinesterase or alkalinization. Paper and gas chromatography have confirmed the presence of acetylcholine and choline in the roasted omum seed extract.

Sustaining high acetylcholine levels in the frontal cortex, but not retrosplenial cortex, recovers spatial memory performance in a rodent model of diencephalic amnesia.

Although the thalamus and/or mammillary bodies are the primary sites of neuropathology in cases of diencephalic amnesia such as Wernicke Korsakoff Syndrome (WKS), there is also functional deactivation of certain cortical regions that contribute to the cognitive dysfunction. Acetylcholine (ACh) is a key neurotransmitter that modulates neural processing within the cortex and between the thalamus and cortex. In the pyrithiamine-induced thiamine deficiency (PTD) rat model of WKS, there are significant reductions in cholinergic innervation and behaviorally stimulated ACh efflux in the frontal (FC)

Sympathetic regulation of fructose secretion in the seminal vesicle of the guinea-pig.

Fructose secretion of everted guinea-pig seminal vesicles was studied in vitro. Carbachol produced dose dependent increase in fructose secretion. The effect was blocked by scopolamine but not by hexamethonium, mecamylamine, tetrodotoxin or previous denervation. High concentrations of acetylcholine also increased fructose secretion. This response was not augmented by physostigmine. Methoxamine reduced secretion. Methoxamine, terbutaline, clonidine and vasoactive intestinal peptide counteracted carbachol. Field stimulation produced increased secretion that was not blocked by autonomic drugs, tet

Cholinergic blockade and response timing in rats.

The effects of central cholinergic blockade on the temporal regulation of behaviour were studied with a two-level DRL schedule. Five-month-old male Wistar rats had to press lever A and then wait for a minimum of 5 s before pressing lever B to obtain the reinforcer (sweetened milk). After a stable baseline performance, subjects were injected in random order with the general cholinergic blocker, scopolamine, 0.15 and 0.5 mg/kg, the peripheral cholinergic blocker, methylscopolamine, 0.15 and 0.5 mg/kg, and a combination of the cholinesterase inhibitor, physostigmine, 0.2 mg/kg, and scopolamine, 0

A history of glaucoma pharmacology.

The history of glaucoma pharmacology begins in 1862 with the isolation of physostigmine from the calabar bean. The discovery of epinephrine's intraocular pressure lowering capacity came along some 40 years later. During the 20th century, drug discovery and development accelerated, with the introduction of carbonic anhydrase inhibitors, beta blockers, and prostaglandin analogs. This survey of the history of glaucoma medications reviews some of the pivotal stories behind the development of the drugs that we use daily to manage our patients with glaucoma. In addition, some unmet needs that persis

Effects of direct and indirect acetylcholine receptor agonists on growth hormone secretion in humans.

Cholinergic pathways in the central nervous system positively influence growth hormone (GH) secretion. In fact pyridostigmine, a cholinesterase inhibitor, enhances both basal and GH-releasing hormone (GHRH)-induced GH secretion while, conversely, pirenzepine, an antagonist of muscarinic M1 receptors, inhibits the GH response to GHRH and to other physiological and pharmacological stimuli. The effect of the cholinergic system on GH secretion probably takes place via inhibition of the release of endogenous somatostatin. In this study in 36 normal adults (26 males and 10 females, age 22-35 years)

Argyll Robertson: 'twas better to be his pupil than to have his pupil.

The Scottish ophthalmologist Douglas Argyll Robertson (1837-1909) is best known for his description of the syphilitic pupil. He also made important contributions concerning the ocular effects of physostigmine and to filtration surgery for glaucoma. Outside of his medical practice, he was club champion at the Royal and Ancient Golf Club, St. Andrews, and the Honorable Company of Edinburgh Golfers many times. He was a champion archer of the Royal Company of Archers, the Queen's Bodyguard in Scotland. Robertson was honored as surgeon-oculist in Scotland to Queen Victoria and to King Edward VII.

Effects of the active constituents of Catha edulis on the neuromuscular junction.

(-)-Cathinone and d-norpseudoephedrine (DNE) in the dose range 0.2-1.2 mg/ml produced a reduction in contractions of skeletal muscle, evoked by direct and indirect electrical stimulation and antagonised the facilitatory action of physostigmine on the neuromuscular junction; but failed to antagonise a partial blockade induced by d-tubocurarine (dTb) as occurs with norepinephrine or epinephrine. The local anaesthetic actions of (-)-cathinone and DNE were found to be almost equivalent to that of lignocaine. These results indicate that (-)-cathinone and DNE may have a direct blocking action on the

Evidence for GABA-BZ receptor modulation in short-term memory passive avoidance task paradigm in mice.

The possible involvement of gamma-aminobutyric acid/benzodiazepine (GABA-BZ) receptor modulation in scopolamine-induced short-term memory deficit was investigated in mice. Passive avoidance step-down task behavior was observed. Latency of mice to reach shock-free zone (SFZ) and number of mistakes the animal made in 15 min were used as separate parameters for acquisition and memory retention, respectively. Scopolamine (0.3 mg/kg) caused a delay in reaching SFZ and an increased number of mistakes. Physostigmine reversed the scopolamine-induced increase in number of mistakes; however, it caused a

Narcolepsy in a Brahman bull.

Behavioral, electrophysiologic, and pharmacologic tests were performed on a 2 1/2-year-old Brahman bull suspected of having narcolepsy. Placement on a tilt table or electroejaculation induced apparent sleep and cataplexy. Electrophysiologic recordings during the episodes revealed the low-voltage, high-frequency electroencephalogram, reduced electromyogram amplitude, and rapid eye movements in the electrooculogram associated with rapid eye movements sleep. Physostigmine salicylate appeared to elicit and potentiate the episodes, whereas atropine sulfate and imipramine reduced or blocked them. Th

Central versus peripheral anticholinergic activity as assessed by two in vivo procedures in mice.

The activity of tertiary and quaternary anticholinergic drugs was compared in two different test procedures designed to measure cholinolytic activity in mice. The four drugs utilized were atropine sulfate, atropine methylnitrate, scopolamine hydrobromide, and scopolamine methylnitrate. The results led to the conclusion that one of these test procedures, the induction of mydriasis (increase in pupil size), primarily measures peripheral anticholinergic activity whereas the other procedure, inhibition of physostigmine lethality, primarily measures anticholinergic activity in the CNS. These two te

Antidepressant-like effects of guanfacine and sex-specific differences in effects on c-fos immunoreactivity and paired-pulse ratio in male and female mice.

RATIONALE: The a2A-noradrenergic agonist guanfacine can decreases stress-induced smoking in female, but not male, human smokers. It is not known whether these effects are due to effects on mood regulation and/or result from nicotinic-cholinergic interactions. OBJECTIVES: The objective of the study was to determine whether there are sex differences in the effect of guanfacine in tests of anxiolytic and antidepressant efficacy in mice at baseline and in a hypercholinergic model of depression induced by the acetylcholinesterase inhibitor physostigmine. METHODS: The effects of guanfacine were me

Towards an animal model of an antipsychotic drug-resistant cognitive impairment in schizophrenia: scopolamine induces abnormally persistent latent inhibition, which can be reversed by cognitive enhancers but not by antipsychotic drugs.

Schizophrenia symptoms segregate into positive, negative and cognitive, which exhibit differential sensitivity to drugs. Recent efforts to identify treatments targeting cognitive impairments in schizophrenia have directed attention to the cholinergic system for its well documented role in cognition. Relatedly, muscarinic antagonists (e.g. scopolamine) produce an 'antimuscarinic syndrome', characterized by psychosis and cognitive impairments. Latent inhibition (LI) is the poorer conditioning to a stimulus resulting from its non-reinforced pre-exposure. LI indexes the ability to ignore irrelevan
