Induction of tcI 7, a gene encoding a beta-subunit of proteasome, in tobacco plants treated with elicitins, salicylic acid or hydrogen peroxide.

We previously isolated, by differential display and 5' RACE (rapid amplification of cDNA ends), cDNAs corresponding to genes activated following cryptogein treatment of tobacco cell suspensions, among them tcI 7 (tcI for tobacco cryptogein Induced), a gene encoding a beta-subunit of proteasome. Here, we report that tcl 7 was up-regulated in tobacco plants treated with elicitins (cryptogein and parasiticein) that have been shown to induce a systemic acquired resistance (SAR). Moreover, subsequent inoculation of tobacco with the pathogen Phytophthora parasitica var. nicotianae (Ppn) was shown to

Characterization of acquired resistance in lesion-mimic transgenic potato expressing bacterio-opsin.

The lesion-mimic mutants of certain plants display necrotic lesions resembling those of the hypersensitive response and activate local and systemic defense responses in the absence of pathogens. We have engineered a lesion-mimic phenotype in transgenic Russet Burbank potato plants through constitutive expression of a bacterio-opsin (bO) proton pump derived from Halobacterium halobium. Transgenic potato plants exhibiting a lesion-mimic phenotype had increased levels of salicylic acid and overexpressed several pathogenesis-related messenger RNAs, all hallmarks of systemic acquired resistance (SA

Early Pep-13-induced immune responses are SERK3A/B-dependent in potato.

Potato plants treated with the pathogen-associated molecular pattern Pep-13 mount salicylic acid- and jasmonic acid-dependent defense responses, leading to enhanced resistance against Phytophthora infestans, the causal agent of late blight disease. Recognition of Pep-13 is assumed to occur by binding to a yet unknown plasma membrane-localized receptor kinase. The potato genes annotated to encode the co-receptor BAK1, StSERK3A and StSERK3B, are activated in response to Pep-13 treatment. Transgenic RNAi-potato plants with reduced expression of both SERK3A and SERK3B were generated. In response t

Characterization of Siderophores Produced by Bacillus velezensis YL2021 and Its Application in Controlling Rice Sheath Blight and Rice Blast.

Bacillus velezensis YL2021 has extensive antimicrobial activities against phytopathogens, and its genome harbors a catechol-type siderophore biosynthesis gene cluster. Here, we describe the characterization of siderophores produced by strain YL2021 and its antimicrobial activity in vitro and in vivo. A few types of siderophores were detected by chrome azurol S plates coupled with Arnow's test, purified, and identified by reversed-phase high-performance liquid chromatography. We found that strain YL2021 can produce different antimicrobial compounds under low-iron M9 medium or iron-sufficient Lu

A germin-like protein gene (CchGLP) of Capsicum chinense Jacq. is induced during incompatible interactions and displays Mn-superoxide dismutase activity.

A germin-like gene (CchGLP) cloned from geminivirus-resistant pepper (Capsicum chinense Jacq. Line BG-3821) was characterized and the enzymatic activity of the expressed protein analyzed. The predicted protein consists of 203 amino acids, similar to other germin-like proteins. A highly conserved cupin domain and typical germin boxes, one of them containing three histidines and one glutamate, are also present in CchGLP. A signal peptide was predicted in the first 18 N-terminal amino acids, as well as one putative N-glycosylation site from residues 44-47. CchGLP was expressed in E. coli and the

A multiplex PCR-Cas12a-lateral flow platform for pooled screening of four major tobacco pathogen groups.

Tobacco production is increasingly threatened by pathogens such as Ralstonia, Phytophthora, Alternaria, and Pseudomonas, which cause bacterial wilt, black shank, brown spot, and wildfire, respectively. Conventional diagnostics typically target single pathogens, and reliable methods for simultaneous multi-pathogen detection remain limited. Here, we developed a multiplex PCR-Cas12a-LFD platform for the pooled multi-target screening of four major tobacco pathogens. Multiplex PCR amplifies pathogen-specific 16S rRNA and ITS regions, while Cas12a-crRNA complexes specifically recognize the amplicons

A Pyrazole-Amide-Azobenzene scaffold for the photocontrollable modulation of succinate dehydrogenase: synthesis, photophysical and biological properties.

Aiming to resolve the issues resulting from the widespread use of antifungal in agricultural production, a series of novel Pyrazole-Amides-Azobenzenes (PAAs) has been designed and synthesized by incorporating the photochromic azobenzene moiety into the pharmacophore, endowing them with both antifungal and photoswitching properties. The majority of compounds (especially target compounds A3 and A4) achieved visible light-induced transition from the trans-isomer to cis-isomer. In addition, the in vitro inhibitory activity of compound A3 against Phytophthora capsici increased from an initial 64.2%

Late Blight Effector Pi05910 Suppresses Plant Immunity by Promoting Destabilisation of Host Scaffold Protein RACK1.

Late blight, caused by oomycete pathogen Phytophthora infestans, is one of the most devastating diseases of potato and tomato. Plant resistance to pathogens relies on a sophisticated innate immune system, which is often targeted by pathogen-derived effectors. P. infestans secretes numerous RXLR effectors to suppress host immunity. In this study, we show that the P. infestans core effector Pi05910 functions as a virulence factor and targets the host scaffold protein Receptor for Activated C Kinase 1 (RACK1), which is identified to positively regulate potato resistance to late blight. Functional

Halichonine B Inspired Design and Divergent Optimization of Drimanyl Amine for Novel Antimicrobial Leads.

Structurally unique halichonine B is promising for the design of pharmaceutical leads, while function-oriented optimization is unknown in agrochemical science. Our recent practical synthesis offers a great chance for the discovery of antimicrobial leads. "Linker plus replaceable substituents" is exerted, in which up to 9 unique linkers together with diverse substituents from a wide chemical space are investigated for optimization of the readily available drimanyl amine. The diamine- or amino amide-related C2- or C3-linker is preferred for increasing the bioactive potency. N,N'-bis(substituted)

Effects of an invasive forest pathogen on abundance of ticks and their vertebrate hosts in a California Lyme disease focus.

Invasive species, including pathogens, can have important effects on local ecosystems, including indirect consequences on native species. This study focuses on the effects of an invasive plant pathogen on a vertebrate community and Ixodes pacificus, the vector of the Lyme disease pathogen (Borrelia burgdorferi) in California. Phytophthora ramorum, the causative agent of sudden oak death, is a non-native pathogen killing trees in California and Oregon. We conducted a multi-year study using a gradient of SOD-caused disturbance to assess the impact on the dusky-footed woodrat (Neotoma fuscipes) a

The role of H2S in low temperature-induced cucurbitacin C increases in cucumber.

In this study, we first linked the signal molecule H2S with cucurbitacin C, which can cause the bitter taste of cucumber leaves and fruit, and specifically discuss its molecular mechanism. Cucurbitacin C (CuC), a triterpenoid secondary metabolite, enhances the resistance of cucumber plants to pathogenic bacteria and insect herbivores, but results in bitter-tasting fruits. CuC can be induced in some varieties of cucumber on exposure to plant stressors. The gasotransmitter hydrogen sulfide (H2S) participates in multiple physiological processes relating to plant stress resistance. This study focu

Biofilm formation and regulation of salicylic acid-inducible genes expression in Arabidopsis by Algerian indigenous bacteria from wheat and potatoes rhizospheres in semi-arid Sétif region.

45 bacterial isolates from potatoes and wheat rhizospheres near Sétif (Algeria) pre-selected for their antagonistic activity against three fungal plant pathogens, two necrotrophic Fusarium solani var. coeruleum and Phytophtora infestans, and a systemic F. oxysporum f. sp. albedinis. Molecular typing of the isolates showed abundance of Bacillus compared to Pseudomonas. Some of the tested strains have shown very high biofilm formation. Among the 24 Gram-positive bacilli screened for four cyclic lipopeptides genes, some isolates harbor two or more genes, while others have a single gene or have no

StLecRK. IX and StLRPK1 form a complex with StBAK1 to positively regulate potato late blight resistance.

In plants, numerous receptor-like kinases (RLKs) function as potential pattern recognition receptors (PRRs) involved in perceiving ligands to trigger pattern-triggered immunity (PTI); however, the roles of most RLKs remain uncharacterized. A previous study showed that the potato receptor-like kinase StLRPK1 interacts with the co-receptor SERK3A/BAK1 to positively regulate resistance against Phytophthora infestans, yet the underlying mechanism remains unknown. Here, we identified StLecRK. IX, a potato RLK that interacts with StLRPK1. The expression of StLecRK. IX was up-regulated in response to

A Systematically Enhanced LAMP Chip for Rapid, Sensitive, and Contamination-Free Pathogen Detection.

Main techniques for pathogen detection include molecular or nucleic acid-based methods, such as polymerase chain reaction (PCR) and loop-mediated isothermal amplification (LAMP). Among these, chip-based LAMP provides a particularly promising platform for point-of-care diagnostics. To ensure accurate, reliable, and robust pathogen detection, systematic optimization of the LAMP-chip system is essential. Building on our previously developed LAMP-chip platform, we report a systematically advanced version for the rapid and sensitive detection of Phytophthora infestans. By systematically investigati
