Development of a multipurpose diagnostic marker using PCR, real-time PCR, and LAMP assays for rapid detection of Rhizoctonia solani in rice plants and soil.

Rhizoctonia solani AG1-IA is a major soil-borne pathogen responsible for sheath blight in rice, causing significant yield losses globally. Early, rapid, and accurate detection is essential for effective disease management, as traditional methods are time-consuming and less sensitive. In this study, we developed and validated a highly specific molecular marker, BrD3 targeting the bromodomain-containing Major Facilitator Superfamily (MFS) gene. This marker was successfully employed in PCR, quantitative PCR (qPCR), and Loop-mediated Isothermal Amplification (LAMP) assays for the detection of the

Raman spectroscopic analysis of cyanogenic glucosides in plants: development of a flow injection surface-enhanced Raman scatter (FI-SERS) method for determination of cyanide.

Cyanogenic glucosides were studied using Raman spectroscopy. Spectra of the crystal forms of linamarin, linustatin, neolinustatin, amygdalin, sambunigrin, and dhurrin were obtained using a Raman spectrograph microscope equipped with a 532 nm laser. The position of the signal from the C identical with N triple bond of the cyanohydrin group was influenced by the nature of the side group and was above 2240 cm(-1) for the three cyanogenic glucosides that contain a neighboring aromatic ring, and below or partially below 2240 cm(-1) for the non-aromatic cyanoglucosides. Signals from the CN bond of l

Strategies for development of functionally equivalent promoters with minimum sequence homology for transgene expression in plants: cis-elements in a novel DNA context versus domain swapping.

The cauliflower mosaic virus 35S (35S) promoter has been extensively used for the constitutive expression of transgenes in dicotyledonous plants. The repetitive use of the same promoter is known to induce transgene inactivation due to promoter homology. As a way to circumvent this problem, we tested two different strategies for the development of synthetic promoters that are functionally equivalent but have a minimum sequence homology. Such promoters can be generated by (a) introducing known cis-elements in a novel or synthetic stretch of DNA or (b) "domain swapping," wherein domains of one pr

Unveiling the carbohydrates profile of anti-diabetic medicinal plants by a developed and validated high-performance liquid chromatography coupled with refractive index detector (HPLC-RID).

Naturally occurring carbohydrates play a crucial role in the food nutritional and medicinal properties of plants. However, measuring these sugars in common herbal products is still not well established, especially for those intended for diabetic patients. The sugar content in these products directly impacts their safety, effectiveness, and authenticity. This study presents a straightforward isocratic high -performance liquid chromatography (HPLC) method with a refractive index (RI) detector. It allows for the simultaneous identification and measurement of seven sugars: D-(-)-Ribose, D-(+)-Xylo

Development of near-infrared fluorescent probes with AIE properties and their application in the detection and imaging of CN- in environmental water samples and multiple species (zebrafish/cells/plants).

Cyanide ions (CN-) are highly toxic pollutants that are widely present in industrial wastewater and the environment, posing a serious threat to ecosystems and human health. The development of highly sensitive and selective CN- detection technologies is an urgent need in environmental monitoring and biomedical fields. In this study, we designed and synthesized a near-infrared fluorescent probe, OTB, based on the aggregation-induced emission (AIE) mechanism. It features a methoxy-modified triphenylamine as the fluorescent moiety and incorporates a benzothiazole structure, achieving an ultra-larg

[Application of artificial DNA-binding proteins and artificial nucleases to prevention of virus infection: development of virus-resistant plants and protein-based anti-viral drugs].

Various DNA viruses are known to cause severe infectious diseases in both plants and mammals, including humans. For many of these infectious diseases, we have yet to find an effective prevention or treatment. Therefore, new methodologies for the prevention of virus infections in both agricultural crops and humans have been vigorously sought for a long time. One attractive approach to the prevention is inhibition of virus replication. We first inhibited virus replication by blocking binding of a viral replication protein, which initiates virus replication, to its replication origin, with using

Effects of a Plant-Based Diet During the First Month of Feeding on Alevin Rainbow Trout (Oncorhynchus mykiss) in the Development of Tongue Sensory System Regulating Feeding Behavior.

Taste perception is essential for animals to detect nutrients, providing critical dietary information necessary for growth and survival. Since the early growth performance of alevin rainbow trout (Oncorhynchus mykiss) can be affected by food intake influenced by terrestrial ingredients without fish meal and fish oil, our study aimed to evaluate the role of taste receptors in nutrient detection and the associated signaling pathways leading to central nervous system activation in the regulation of feeding behavior. We conducted a nutritional experiment from the first feeding to 30 days, comparin

DiZF-C3H1, a zinc finger transcription factor from the dove tree (Davidia involucrata Baill.), plays a negative role in seed development and plant growth in Arabidopsis and tobacco.

Low seed fertility seriously limits the survival and adaption of rare plant species. Here, we identified a seed-specific gene, DiZF-C3H1, from the dove tree and verified its function. Overexpression of DiZF-C3H1 caused retarded root development, delayed anthesis, abnormal floral organs, and deformed siliques in transgenic Arabidopsis lines. No offspring were obtained in transgenic Arabidopsis lines due to serious seed abortion. Therefore, we performed further verification in tobacco. Similarly, overexpression of DiZF-C3H1 retarded root development and reduced berry size and seed yield in trans

Development of a next generation DNA sequencing-based multi detection assay for detecting and identifying Leishmania parasites, blood sources, plant meals and intestinal microbiome in phlebotomine sand flies.

Leishmaniasis is a disease caused by Leishmania parasites transmitted by phlebotomine sand flies (Diptera: Psychodidae). Human infections with different Leishmania species cause characteristic clinical manifestations; cutaneous or visceral leishmaniasis. Here we describe the development and application of a Miseq Next GenerationSequencing (NGS)-based Multi Detection Assay (MDA) designed to characterize metagenomics parameters pertinent to the sand fly vectors which may affect their vectorial capacity for Leishmania. For this purpose, we developed a MDA by which, DNA fragments were amplified th

The cytokinin requirement for cell division in cultured Nicotiana plumbaginifolia cells can be satisfied by yeast Cdc25 protein tyrosine phosphatase: implications for mechanisms of cytokinin response and plant development.

Cultured cells of Nicotiana plumbaginifolia, when deprived of exogenous cytokinin, arrest in G2 phase prior to mitosis and then contain cyclin-dependent protein kinase (CDK) that is inactive because phosphorylated on tyrosine (Tyr). The action of cytokinin in stimulating the activation of CDK by removal of inhibitory phosphorylation from Tyr is not a secondary downstream consequence of other hormone actions but is the key primary effect of the hormone in its stimulation of cell proliferation, since cytokinin could be replaced by expression of cdc25, which encodes the main Cdc2 (CDK)-Tyr dephos

A review on the green synthesis of metal (Ag, Cu, and Au) and metal oxide (ZnO, MgO, Co3O4, and TiO2) nanoparticles using plant extracts for developing antimicrobial properties.

Green synthesis (GS) is a vital method for producing metal nanoparticles with antimicrobial properties. Unlike traditional methods, green synthesis utilizes natural substances, such as plant extracts, microorganisms, etc., to create nanoparticles. This eco-friendly approach results in non-toxic and biocompatible nanoparticles with superior antimicrobial activity. This paper reviews the prospects of green synthesis of metal nanoparticles of silver (Ag), copper (Cu), gold (Au) and metal oxide nanoparticles of zinc (ZnO), magnesium (MgO), cobalt (Co3O4), and titanium (TiO2) using plant extracts f

Development of Botanical Pesticides: Exploration on the Phenotype of Vestigial Wings of Insect Pests Induced by Plant Natural Products or Their Derivatives by Blocking Tyrosine Phosphorylation of Insulin Receptor 1.

Unlike faster-acting conventional insecticides, some botanical insecticides exhibit growth inhibitory activity against some insect pests. One of the distinguishing features of growth inhibitory activity appears to be in malformed moths with vestigial wings. However, the molecular mechanism underlying vestigial wings of insect pests induced by plant natural products or their derivatives is still elusive. In this work, based upon the phenotype of the vestigial wings of Mythimna separata Walker (as a model pest) induced by a podophyllotoxin derivative 2a (as a model compound), we found that compo

Ectopic expression of foxtail millet zip-like gene, SiPf40, in transgenic rice plants causes a pleiotropic phenotype affecting tillering, vascular distribution and root development.

Plant architecture determines grain production in rice (Oryza sativa) and is affected by important agronomic traits such as tillering, plant height, and panicle morphology. Many key genes involved in controlling the initiation and outgrowth of axillary buds, the elongation of stems, and the architecture of inflorescences have been isolated and analyzed. Previous studies have shown that SiPf40, which was identified from a foxtail millet (Setaria italica) immature seed cDNA library, causes extra branches and tillers in SiPf40-transgenic tobacco and foxtail millet, respectively. To reconfirm its

Method development for the determination of iron in milligram amounts of rice plants (Oryza sativa L.) from cultivation experiments using graphite furnace atomic absorption spectrometry.

The amount of sample that is available for analysis in laboratory plant cultivation experiments is usually very limited. Highly sensitive analytical techniques are therefore required, even for elements that are present in the plants at mg g(-1) concentrations, and graphite furnace atomic absorption spectrometry (GFAAS) was chosen in this work because of its micro-sampling capability, and its relatively simple operation. Four micro-methods were investigated for the determination of iron in roots and leaves of rice plants: i) a micro-digestion with nitric and hydrochloric acids, ii) a slurry pro
