Transgenic pearl millet male fertility restorer line (ICMP451) and hybrid (ICMH451) expressing Brassica juncea Nonexpressor of pathogenesis related genes 1 (BjNPR1) exhibit resistance to downy mildew disease.

Brassica juncea Nonexpressor of pathogenesis-related genes 1 (BjNPR1) has been introduced into pearl millet male fertility restorer line ICMP451 by Agrobacterium tumefaciens-mediated genetic transformation. Transgenic pearl millet plants were regenerated from the phosphinothricin-resistant calli obtained after co-cultivation with A. tumefaciens strain LBA4404 harbouring Ti plasmid pSB111-bar-BjNPR1. Molecular analyses confirmed the stable integration and expression of BjNPR1 in transgenic pearl millet lines. Transgenes BjNPR1 and bar were stably inherited and disclosed co-segregation in subseq

Greatly Enhanced Removal of Volatile Organic Carcinogens by a Genetically Modified Houseplant, Pothos Ivy ( Epipremnum aureum) Expressing the Mammalian Cytochrome P450 2e1 Gene.

The indoor air in urban homes of developed countries is usually contaminated with significant levels of volatile organic carcinogens (VOCs), such as formaldehyde, benzene, and chloroform. There is a need for a practical, sustainable technology for the removal of VOCs in homes. Here we show that a detoxifying transgene, mammalian cytochrome P450 2e1 can be expressed in a houseplant, Epipremnum aureum, pothos ivy, and that the resulting genetically modified plant has sufficient detoxifying activity against benzene and chloroform to suggest that biofilters using transgenic plants could remove VOC

Utilization of Legume-Nodule Bacterial Symbiosis in Phytoremediation of Heavy Metal-Contaminated Soils.

With the increasing industrial activity of the growing human population, the accumulation of various contaminants in soil, including heavy metals, has increased rapidly. Heavy metals as non-biodegradable elements persist in the soil environment and may pollute crop plants, further accumulating in the human body causing serious conditions. Hence, phytoremediation of land contamination as an environmental restoration technology is desirable for both human health and broad-sense ecology. Legumes (Fabaceae), which play a special role in nitrogen cycling, are dominant plants in contaminated areas.

Edible Vaccines: A Patent-Driven Exploration of Immunization Technologies.

Vaccines are biological preparations that improve immunity to particular diseases. Particularly for poor developing nations, edible vaccines show significant potential as a financially advantageous, simple to administer, straightforward to store, fail-safe, and socially and culturally acceptable vaccine delivery system. A vaccine incorporates the gene-encoding bacterial or viral disease-causing agent in plants without losing its immunogenic property. Potatoes, tomatoes, rice, soybeans, and bananas are the primary plants for edible vaccines. It activates the systemic and mucosal immunity respon

CRISPR/Cas9-mediated multiplex gene editing of gamma and omega gliadins: paving the way for gliadin-free wheat.

Wheat is a staple cereal in the human diet. Despite its significance, an increasing percentage of the population suffers adverse reactions to wheat, which are triggered by wheat gluten, particularly the gliadin fractions. In this study, we employed CRISPR/Cas [clustered regularly interspaced palindromic repeats (CRISPR)/CRISPR-associated protein] multiplexing to introduce targeted mutations into γ- and ω-gliadin genes of wheat, to produce lines deficient in one or both immunogenic gliadin fractions simultaneously. For this study, eight single guide RNAs (sgRNAs) were designed and combined into

Production of transgenic Allium cepa by nanoparticles to resist Aspergillus niger infection.

BACKGROUND: Transgenic plants are becoming a more powerful tool in modern biotechnology. Genetic engineering was used in biotech-derived products to create genetically modified (GM) plants resistant to diseases. The onion (Allium cepa, L.) is a common, important perennial vegetable crop grown in Egypt for food and economic value. Onions are susceptible to a variety of fungal infections and diseases. Aspergillus niger is a common onion phytopathogen that causes diseases such as black mould (or black rot), which is a major issue, particularly when exporting onions. A. niger grows between the bul

Distinct defensive activity of phenolics and phenylpropanoid pathway genes in different cotton varieties toward chewing pests.

Identifying the maximum level of inherent defense against harmful insects in natural variation among wild lineages of crop plants may result in high yield tolerant varieties and reducing use of chemical insecticides. However, knowledge of natural cotton genotypes with high insect-resistance is still indistinguishable at the biochemical or molecular level. In the present study, different cultivated Gossypium hirsutum varieties were evaluated for their inherent insect-tolerance against two major cottons chewing pests. The insect bio-assay identified two tolerant and one susceptible cotton variet

Caffeine and related purine alkaloids: biosynthesis, catabolism, function and genetic engineering.

Details of the recently elucidated biosynthetic pathways of caffeine and related purine alkaloids are reviewed. The main caffeine biosynthetic pathway is a sequence consisting of xanthosine-->7-methylxanthosine-->7-methylxanthine-->theobromine-->caffeine. Genes encoding N-methyltransferases involved in three of these four reactions have been isolated and the molecular structure of N-methyltransferases investigated. Pathways for the catabolism of caffeine have also been studied, although there are currently no reports of enzymatic and genetic studies having been successfully carried out. Metabo

Multiple phytochromes are involved in red-light-induced enhancement of first-positive phototropism in Arabidopsis thaliana.

The amplitude of phototropic curvature to blue light is enhanced by a prior exposure of seedlings to red light. This enhancement is mediated by phytochrome. Fluence-response relationships have been constructed for red-light-induced enhancement in the phytochrome A (phyA) null mutant, the phytochrome B- (phyB) deficient mutant, and in two transgenic lines of Rabidopsis thaliana that overexpress either phyA or phyB. These fluence-response relationships demonstrate the existence of two response in enhancement, a response in the very-low-to-low-fluence range, and a response in the high-fluence ran

Overexpression of EXO70E2 in Arabidopsis thaliana Disrupts Normal Development and Enhances Susceptibility to the Necrotrophic Fungus Botrytis cinerea.

Background: The exocyst complex, a conserved hetero-octameric complex including the EXO70 subunit, is a pivotal regulator of various cellular and developmental processes in plants. Among the diverse EXO70 isoforms, the specific function of EXO70E2-a primary organizer of exocyst-positive organelles (EXPOs)-remains to be fully elucidated. Methods: Here, we investigated the functional impact of constitutive EXO70E2 overexpression in Arabidopsis thaliana. Results: We observe that EXO70E2 overexpression leads to severe etiolation and dwarfism, accompanied by dose-dependent inhibition of primary roo

The miR166a-MpATHB8 module regulates anthocyanin accumulation to increase rust resistance in Malus "Profusion" leaves.

Apple rust, caused by the fungal pathogen Gymnosporangium yamadae, leads to substantial yield losses and significant economic damage. In the rust-resistant cultivar Malus "Profusion", rust infection triggers anthocyanin synthesis at infection sites as a defense mechanism to restrict fungal proliferation. Although small noncoding RNAs (miRNAs) play important roles in regulating anthocyanin biosynthesis, their specific functions under rust stress remain poorly characterized. In this study, small RNA sequencing revealed that miR166a is a key rust-responsive regulator. Its direct targeting and neg

Glycine max DREB1D transcription factor regulates plant height by reducing gibberellin levels and response.

Overexpression of GmDREB1D reduces plant height of soybean. Overexpression of GmDREB1D reduced active GA content and GmDREB1D can regulate GmGA2ox8. GmDREB1D may inhibit growth by reducing the levels of active GAs and response. Dehydration responsive element binding (DREB) proteins play an important role in growth and abiotic stress. Although the DREB1D transcription factor contributes to drought tolerance, the molecular mechanisms underlying growth inhibition are unclear. In this study, we reveal the function of GmDREB1D as a transcriptional regulator in growth. Overexpression of GmDREB1D red

SPOTTED-LEAF7 targets the gene encoding β-galactosidase9, which functions in rice growth and stress responses.

β-Galactosidases (Bgals) remove terminal β-D-galactosyl residues from the nonreducing ends of β-D-galactosidases and oligosaccharides. Bgals are present in bacteria, fungi, animals, and plants and have various functions. Despite the many studies on the evolution of BGALs in plants, their functions remain obscure. Here, we identified rice (Oryza sativa) β-galactosidase9 (OsBGAL9) as a direct target of the heat stress-induced transcription factor SPOTTED-LEAF7 (OsSPL7), as demonstrated by protoplast transactivation analysis and yeast 1-hybrid and electrophoretic mobility shift assays. Secretion-based production of prolyl-hydroxylated human type III collagen in scalable Physcomitrella photobioreactors.

Scalable moss bioreactors enable the production of high-quality recombinant prolyl-hydroxylated human collagen without heterologous P4H expression, offering a sustainable and vegan alternative to conventional collagens derived from animals. Collagens are structural proteins of the extracellular matrix essential for skin elasticity and integrity. They are widely used in dietary supplements and cosmetics. Conventional collagens of animal origin raise concerns regarding ethics, safety, and sustainability. As a vegan alternative, we report on the production of a 30 kDa prolyl-hydroxylated human co

Plant Biofactories for Vaccination: Progress in Edible Vaccine Technologies as a Sustainable Approach to Global Immunization.

Globally, the pathogen-driven enteric diseases remain a leading cause of morbidity and mortality, disproportionately affecting populations in low-income regions. While significant strides have been made in vaccine research, challenges such as high production costs, complex manufacturing processes, and inefficient distribution continue to hinder global vaccine accessibility. To overcome these challenges, plant-based edible vaccines have emerged as a promising and innovative alternative, offering advantages such as affordability, ease of administration, and reduced reliance on cold-chain infrast

The Targeted Regulation of BDUbc and BDSKL1 Enhances Resistance to Blight in Bambusa pervariabilis × Dendrocalamopsis grandis.

Arthrinium phaeospermum is the major pathogen responsible for the significant stem disease "blight" in B. pervariabilis × D. grandis. The interacting proteins of the key pathogenic factor ApCtf1β, BDUbc and BDSKL1, have previously been obtained by two-hybrid, BiFC, GST pull-down yeast assays. However, the functions of these interacting proteins remain unknown. This study successfully obtained transgenic plants overexpressing BDUbc, BDSKL1, and BDUbc + BDSKL1 via Agrobacterium-mediated gene overexpression. qRT-PCR analysis revealed significantly increased expression levels of BDUbc and BDSKL1 i

Plant-Made Vaccines Targeting Enteric Pathogens-Safe Alternatives for Vaccination in Developing Countries.

Enteric diseases by pathogenic organisms are one of the leading causes of death worldwide, particularly in low-income countries. Despite antibiotics, access to clean water and vaccination are the most economically affordable options to prevent those infections and their health consequences. Vaccines, such as those approved for rotavirus and cholera, have played a key role in preventing several enteric diseases. However, vaccines for other pathogens are still in clinical trials. Distribution and cost remain significant barriers to vaccine access in developing regions due to poor healthcare infr

Carotenoid Biofortification of Crops in the CRISPR Era.

Carotenoids are micronutrients important for human health. The continuous improvements in clustered regularly interspaced short palindromic repeats (CRISPR)-based genome-editing techniques make rapid, DNA/transgene-free and targeted multiplex genetic modification a reality, thus promising to accelerate the breeding and generation of 'golden' staple crops. We discuss here the progress and future prospects of CRISPR/Cas9 applications for carotenoid biofortification.

Africa's inevitable walk to genetically modified (GM) crops: opportunities and challenges for commercialization.

High relative poverty levels in Africa are attributed to the continent's under performing agriculture. Drought, low-yielding crop varieties, pests and diseases, poor soils, low fertilizer use, limited irrigation and lack of modern technologies are among the problems that plague African agriculture. Genetically modified (GM) crops may possess attributes that can help overcome some of these constraints, but have yet to be fully embraced in the mix of technology solutions for African agriculture. Cognizant of this, South Africa, Burkina Faso and Egypt are steadily growing GM crops on a commercial

Remodeling of the tuberization-controlling FLOWERING LOCUS T genes from non-tuberizing ancestor to potato.

Potato (Solanum section Petota) originated from a natural hybridization between tomato and Etuberosum ancestors, through which the trait of tuber formation evolved. Genetically, tuberization in potato is controlled by FLOWERING LOCUS T (FT) homologs. However, from non-tuberous ancestors to potato, what remodeling of these tuber formation determinants emerged remains unclear. Using homology-based discovery, expression profiling, CRISPR knockouts, and genetic complementation, we here show that tuberization-controlling FT-like genes underwent Petota-specific expansion, restructuring, and divergen
