Quantitation of spinal cord demyelination, remyelination, atrophy, and axonal loss in a model of progressive neurologic injury.

Spinal cord pathology, such as demyelination and axonal loss, is a common feature in multiple models of central nervous system (CNS) injury and disease. Development of methods to quantify spinal cord pathology objectively would aid studies designed to establish mechanisms of damage, correlate pathology with neurologic function, and assess therapeutic interventions. In this study, we describe sensitive methods to objectively quantify spinal cord demyelination, remyelination, atrophy, and axonal loss following the initiation of a progressive inflammatory demyelinating disease with Theiler's muri

Incorporation of Oligo(Styrene), Oligo(Butadiene), Oligo(Methyl Methacrylate), and Oligo(tert-Butyl Acrylate) Droplets in Giant Unilamellar Vesicles.

In the environment, microplastics fragment and degrade into plastic oligomers, posing potential risks to ecosystems and human health. In the present study, the interactions between giant unilamellar vesicles (GUVs) and liquid droplets of oligo(styrene), oligo(butadiene), oligo(methyl methacrylate), and oligo(tert-butyl acrylate) were investigated using fluorescence microscopy. GUVs were used in a homogeneous fluid state or in a phase-separated state consisting of either a liquid-disordered (Ld) phase and a solid-ordered (So) phase, or a Ld phase and a liquid-ordered (Lo) phase. Age- and position-dependent effects of monaural acoustic deprivation in nucleus magnocellularis of the chicken.

The effect of a moderately severe monaural conductive hearing loss on the development of neuronal size in the avian nucleus magnocellularis (NM) was investigated. NM is considered to be the homologue of the mammalian anteroventral cochlear nucleus and receives large calyceal synaptic endings from the cochlear nerve. Silicone plastic earplugs which produce a 40 dB broadband conductive hearing loss were placed in one external auditory canal of chick embryos on the 18th day of incubation. After hatching, all animals were housed in communal brooders and sacrificed at 4, 10, 25, and 60 days of age.

EZH2 Inhibition Remodels Cell States and Enhances EGFRi Sensitization in Bladder Cancer.

BACKGROUND: Bladder cancer (BCa) displays interpatient heterogeneity and epigenetic plasticity that may limit treatment efficacy. OBJECTIVE: To investigate whether inhibition of Enhancer of zeste homolog 2 (EZH2) remodels tumor cell states and creates therapeutic vulnerabilities in BCa. DESIGN, SETTING, AND PARTICIPANTS: We integrated single-nucleus ribonucleic acid sequencing (snRNA-seq) of formalin-fixed paraffin-embedded (FFPE) primary tumors with single-cell ribonucleic acid sequencing (scRNA-seq) of matched patient-derived organoids (PDOs) from four BCas spanning stages Ta-T3. A Defined Hydrogel-Based Method For Generating Three-Dimensional Human Breast Organoids That Recapitulate Mammary Morphogenesis.

The development of physiologically relevant human model systems that recapitulate tissue architecture and cell-state dynamics remains a major challenge in studying breast development and early events in carcinogenesis. Conventional two-dimensional cultures and many three-dimensional systems fail to capture the structural organization and microenvironmental cues that define the human mammary gland. Here, we describe a reproducible method for generating three-dimensional human breast organoids from primary epithelial cells embedded within a defined hydrogel matrix composed of type I collagen, la

Single-cell analysis of sterol-induced Ca2+ signaling in human astrocytes by dynamic mode decomposition.

Ca2+ signaling in astrocytes is a central mechanism of intercellular communication in the brain and plays a key role in regulating neuronal excitability, synaptic plasticity, and energy metabolism. Disruption of astrocytic Ca2+ dynamics is a characteristic of neurodegenerative diseases, as are deviations in cholesterol trafficking and metabolism, which are essential for maintaining membrane structure and function. Although recent studies have begun to explore links between Ca2+ signaling and sterol homeostasis in astrocytes, unbiased analytical workflows and mechanistic insight into how choles

A protoplast-based method to visualize early cell biological events in plant cellular reprogramming and regeneration.

Plant developmental biology lacks cell-based experimental systems comparable to the organoids and live-imaging platforms that have transformed mechanistic discovery in animal research. To address this gap, we present a robust, trackable protoplast regeneration platform in Arabidopsis thaliana that enables high-resolution, time-resolved analysis from the single-cell stage through microcolony formation and early regenerative development. Protoplasts are embedded in thin alginate matrices containing fluorescent fiducial beads, maintaining physical separation and allowing repeated return to the sa

From natural theology to the extended synthesis: Historical milestones and conceptual expansions in evolutionary biology.

This article explores the historical development of evolutionary biology-from Natural Theology to the Modern Synthesis (MS)-and the ongoing debate around the Extended Evolutionary Synthesis (EES). Over the past 2,500 years, evolutionary thinking has emerged from the interplay between empirical discoveries and dominant philosophical paradigms. Beginning with Aristotle and Saint Augustine, we trace how Darwin and Wallace introduced a scientific framework grounded in natural mechanisms. In the early 20th century, the MS unified Mendelian genetics and Darwinian selection, forming a gene-centered m

Not the Silver Bullet: Uncovering the Unexpected Limited Impacts of Silver-Containing Showerheads on the Drinking Water Microbiome.

The incidence of waterborne disease outbreaks in the United States attributed to drinking water-associated pathogens that can cause infections in the immunocompromised DWPIs (e.g., Legionella pneumophila, nontuberculous mycobacteria (NTM), and Pseudomonas aeruginosa, among others) appears to be increasing. An emerging technology adopted to reduce DWPIs are point-of-use devices, such as showerheads that contain silver, a known antimicrobial material. In this study, we evaluate the effect of silver-containing showerheads on DWPI density and the broader microbiome in shower water under real-use c

A Novel Hydraulic Actuation System Utilizing Magnetorheological Fluids for Single-Port Laparoscopic Surgery Applications.

Single-port laparoscopic surgery (SLS), which utilizes one major incision, can deliver favorable cosmetic outcomes with fewer patient hospitalization stays and less postoperative pain. However, current SLS instruments, which are rigid and slender, have been suffering from several drawbacks, including their inability to provide the optimum articulation required to complete certain SLS tasks. This paper reports on the development of a lightweight smart hydraulic actuation system that is proposed to be embedded at selected joints along current SLS instruments, in order to enhance their adaptabili

Alligator osteoderms: mechanical behavior and hierarchical structure.

Osteoderms are bony scutes embedded underneath the dermal layers of the skin acting as a protection of the alligator (Archosauria: Crocodylia) internal organs and tissues. Additionally, these scutes function as an aid in temperature regulation. The scutes are inter-linked by fibrous connective tissue. They have properties similar to bone and thus have the necessary toughness to provide protection against predators. The scutes consist of hydroxyapatite and have a porosity of approximately 12%. They have a disc-like morphology with a ridge along the middle of the plate, called the keel; the oute

Close microtopographical relationships between sympathetic nerve terminals and bulbous process endings of pinealocytes in the pineal gland of the Mongolian gerbil.

Previous studies have shown that pinealocytes of the gerbil pineal gland exhibit processes that form terminal swellings filled with abundant electron-lucent microvesicles. The membrane of these presumptive secretory microvesicles is known to contain synaptophysin, a major integral glycoprotein of neuronal synaptic vesicles. The present study was conducted to evaluate the microtopographical relationships between the vesicle-rich process swellings and intra-pineal nerve terminals. For this purpose, both nerve terminals and pinealocyte process endings were visualized immunohistochemically in the

Selective binding of colloidal gold-protein conjugates to epidermal phosphorus-rich keratohyaline granules and cornified cells.

Colloidal gold solutions conjugated with staphylococcal protein A (SpA) are widely used in high-resolution immunocytochemical studies to visualize antibodies bound at antigenic sites. Here we report that colloidal gold solutions conjugated with SpA, bovine serum albumin (BSA), or gelatin bind selectively to structures in glutaraldehyde-fixed, plastic-embedded epidermis of rabbit, mouse, and human. Two types of keratohyaline granules are present in epidermis, a phosphorus-rich (PR) and a sulphur-rich (SR) type. The PR keratohyaline granules were strongly labeled with gold particles, whereas SR

A cross-species analysis of cell wall mechanosensors.

The cell wall (CW) protects fungal cells from various challenges, making its integrity essential for cell survival. CW integrity is monitored by transmembrane sensors that activate effectors to promote CW synthesis in response to injuries. Sensors of the WSC (Wall Surface Component) family are found in most fungi, and share a conserved architecture, with a cytoplasmic tail, a single transmembrane domain, and a long Serine Threonine Rich domain (STR) prolonged by a WSC domain, both embedded in the CW. These extracellular domains promote force detection in the CW, sensor clustering and cell surv

BfrR-mediated biofilm regulation in the seafood-borne pathogen Vibrio parahaemolyticus.

Vibrio parahaemolyticus is a major seafood-associated pathogen whose biofilm formation enhances environmental persistence, antibiotic resistance, and food safety risks. In this study, we identified a LysR-type regulator, BfrR (VPA1687), as a critical mediator of biofilm development in V. parahaemolyticus. Deletion of bfrR severely impaired biofilm formation, switching colony morphology from wrinkled to smooth and reducing extracellular matrix components (exopolysaccharides (EPS), proteins, and extracellular DNA). BfrR also modulated metabolic activity in biofilm-embedded cells and exhibited du

Dual-Responsive Multi-Functional Silica Nanoparticles With Repaired Mitochondrial Functions for Efficient Alleviation of Spinal Cord Injury.

Preserved/rescued mitochondrial functions have a significant effect on maintaining neurogenesis, axonal carriage, and synaptic plasticity following spinal cord injury (SCI). We fabricated an ingenious redox-responsive strategy for commanded liberation of NADH (reduced form of nicotinamide-adenine dinucleotide) by bioactive diselenide-containing biodegradable mesoporous silica nanoparticles (Se@NADH). The nanocarrier-embedded NADH can be liberated in a controlled pattern through the cleavage of diselenide bonds in the presence of reactive oxygen species (ROS) or glutathione (GSH). The NAD+ was

Cell Reprogramming, Transdifferentiation, and Dedifferentiation Approaches for Heart Repair.

Cardiovascular disease (CVD) remains the leading cause of death globally, with myocardial infarction (MI) being a major contributor. The current therapeutic approaches are limited in effectively regenerating damaged cardiac tissue. Up-to-date strategies for heart regeneration/reconstitution aim at cardiac remodeling through repairing the damaged tissue with an external cell source or by stimulating the existing cells to proliferate and repopulate the compromised area. Cell reprogramming is addressed to this challenge as a promising solution, converting fibroblasts and other cell types into fun

Photoinduced Gelatin-Methacrylate Scaffolds to Examine the Impact of Extracellular Environment on Trabecular Meshwork Cells.

Glaucoma is the leading cause of irreversible blindness in the world, currently impacting 80 million people. Patients suffering from primary open-angle glaucoma experience aqueous humor accumulation within the eye causing an increase in intraocular pressure (IOP). The main cause of this rise in IOP is due to poor outflow of aqueous humor through the trabecular meshwork (TM), a tissue composed of collagen and glycosaminoglycans (GAGs) embedded with TM cells. The behavior of TM cells is impacted by their microenvironment, and studies conducted on two-dimensional plastic substrates do not necessa

Changes in predator exposure, but not in diet, induce phenotypic plasticity in scorpion venom.

Animals embedded between trophic levels must simultaneously balance pressures to deter predators and acquire resources. Venomous animals may use venom toxins to mediate both pressures, and thus changes in this balance may alter the composition of venoms. Basic theory suggests that greater exposure to a predator should induce a larger proportion of defensive venom components relative to offensive venom components, while increases in arms races with prey will elicit the reverse. Alternatively, reducing the need for venom expenditure for food acquisition, for example because of an increase in sca
