Novel nucleotide-packaging vaccine delivers antigen and poly(I:C) to dendritic cells and generate a potent antibody response in vivo.

An issue with many current vaccines is the dependency on broadly inflammatory adjuvants, such as aluminum hydroxide or aluminum salts that affect many immune- and non-immune cells. These adjuvants are not necessarily activating all antigen-presenting cells (APCs) that take up the antigen and most likely they also activate APCs with no antigen uptake, as well as many non-immune cells. Conjugation of antigen and adjuvant would enable the use of smaller amounts of adjuvant and avoid unnecessary tissue damage and activation of bystander cells. It would ensure that all APCs that take up the antigen

NIPAM-based Microgel Microenvironment Regulates the Therapeutic Function of Cardiac Stromal Cells.

To tune the chemical, physical, and mechanical microenvironment for cardiac stromal cells to treat acute myocardial infarction (MI), we prepared a series of thermally responsive microgels with different surface charges (positive, negative, and neutral) and different degrees of hydrophilicity, as well as functional groups (carboxyl, hydroxyl, amino, and methyl). These microgels were used as injectable hydrogels to create an optimized microenvironment for cardiac stromal cells (CSCs). Our results indicated that a hydrophilic and negatively charged microenvironment created from poly( N-isopropyla

Amyloid Beta Aggregation in the Presence of Temperature-Sensitive Polymers.

The formation of amyloid fibrils is considered to be one of the main causes for many neurodegenerative diseases, such as Alzheimer's, Parkinson's or Huntington's disease. Current knowledge suggests that amyloid-aggregation represents a nucleation-dependent aggregation process in vitro, where a sigmoidal growth phase follows an induction period. Here, we studied the fibrillation of amyloid β 1-40 (Aβ40) in the presence of thermoresponsive polymers, expected to alter the Aβ40 fibrillation kinetics due to their lower critical solution behavior. To probe the influence of molecular weight and the e

Comparative evaluation of the prophylactic effect of slow release devices containing homidium bromide and isometamidium on Trypanosoma congolense in rabbits.

Two consecutive experiments were carried out to evaluate the prophylactic effect of biodegradable slow release devices (SRD), containing either isometamidium or homidium bromide. Rabbits subcutaneously implanted with SRD, were challenged with different Trypanosoma congolense stocks at regular intervals between 1 and 6.5 months after treatment. In a first experiment the efficacy of two types of isometamidium-SRD (poly(D,L-lactide) and poly(D,L-lactide-co-glycolide)) was compared with the classical intramuscular (i.m.) injection of the drug. Since the former polymer gave an average protection pe

Adjuvant combination enhances immune response of equine influenza virus vaccine in horses.

BACKGROUND: Despite routine vaccination, equine influenza outbreaks occur due to disparities of virus strains between vaccine and field, and immunity gaps between vaccinations. OBJECTIVES: To evaluate whether a combination of adjuvants can enhance immune responses to equine influenza virus (EIV) vaccines. STUDY DESIGN: In vivo experimental study. METHODS: Eight mixed-breed horses were immunised intramuscularly with a recombinant canarypox-based EIV vaccine, either alone or in combination with monophosphoryl lipid A (MPL) and polyinosinic-polycytidylic acid (poly I:C). A single vaccine dose

The Chlamydia M278 Major Outer Membrane Peptide Encapsulated in the Poly(lactic acid)-Poly(ethylene glycol) Nanoparticulate Self-Adjuvanting Delivery System Protects Mice Against a Chlamydia muridarum Genital Tract Challenge by Stimulating Robust Systemic and Local Mucosal Immune Responses.

Recently, we reported that our PPM chlamydial nanovaccine [a biodegradable co-polymeric PLA-PEG (poly(lactic acid)-poly(ethylene glycol))-encapsulated M278 peptide (derived from the major outer membrane protein (MOMP) of Chlamydia)] exploits the caveolin-mediated endocytosis pathway for endosomal processing and MHC class II presentation to immune-potentiate Chlamydia-specific CD4+ T-cell immune effector responses. In the present study, we employed the Chlamydia muridarum mouse infection model to evaluate the protective efficacy of PPM against a genital tract challenge. Our results show that mi

Development of a refined experimental mouse model of myasthenia gravis with anti-acetylcholine receptor antibodies.

Myasthenia gravis (MG) is an autoimmune disorder primarily caused by autoantibodies that target the acetylcholine receptor (AChR) at the neuromuscular junction (NMJ). The classical experimental autoimmune myasthenia gravis (C-EAMG) mouse model has long been used by immunizing mice with acetylcholine receptor from Torpedo fish (T-AChR), combined with complete Freund's adjuvant (CFA). This mixture is administered via subcutaneous injections into the hind footpads and back, but CFA often causes strong inflammatory reactions, including lesions at the injection sites. Our objective was to develop a

Prostate cancer in situ autovaccination with the intratumoral viral mimic poly-ICLC: Modulating the cold tumor microenvironment.

BACKGROUND: Neoadjuvant therapies for high-risk PCa have shown promise but remain confined to clinical trials. Translating neoadjuvant approaches into routine care thus underscores the critical need for innovative early-phase neoadjuvant trials to evaluate safety and efficacy in localized disease, where tumors are more responsive to intervention. METHODS: In this open-label phase 1 trial (NCT03262103), 12 patients with clinically localized intermediate- to high-risk PCa scheduled for radical prostatectomy (RP) received sequential intratumoral and intramuscular injections of poly-ICLC (Hiltono

Poly(I:C)/alum mixed adjuvant priming enhances HBV subunit vaccine-induced immunity in mice when combined with recombinant adenoviral-based HBV vaccine boosting.

BACKGROUND: Virus-specific cellular immune responses play a critical role in virus clearance during acute or chronic HBV infection. Currently, the commercially available HBV vaccine is combined with alum adjuvant, which stimulates mainly Th2 immune responses. Therefore, development of new therapeutic HBV vaccine adjuvants and immune strategies that also promote Th1 and CTL responses is urgently needed. METHODOLOGY/PRINCIPAL FINDINGS: To improve the immunity induced by the novel HBSS1 HBV vaccine, we evaluated the ability of adjuvants, including alum, CpG and polyriboinosinic polyribocytidylic

PLDLA/PCL-T Scaffold for Meniscus Tissue Engineering.

The inability of the avascular region of the meniscus to regenerate has led to the use of tissue engineering to treat meniscal injuries. The aim of this study was to evaluate the ability of fibrochondrocytes preseeded on PLDLA/PCL-T [poly(L-co-D,L-lactic acid)/poly(caprolactone-triol)] scaffolds to stimulate regeneration of the whole meniscus. Porous PLDLA/PCL-T (90/10) scaffolds were obtained by solvent casting and particulate leaching. Compressive modulus of 9.5±1.0 MPa and maximum stress of 4.7±0.9 MPa were evaluated. Fibrochondrocytes from rabbit menisci were isolated, seeded directly on t

Toxoplasma gondii infection induces the formation of host's nuclear granules containing poly(A)-binding proteins.

To study the mechanism by which human host cells respond to an infection of Toxoplasma gondii, we monitored the level of poly(A)-binding protein (PABP), an indicator of translation. Here, we report an observation of the relocalization of PABPs in human host cells upon T. gondii infection. Notably, the aggregates of PABPs formed upon infection are mainly found in the nucleus, which is a different response from that found after exposure to heat shock. Pyrimethamine treatment of the infected monolayers inhibits the multiplicity of the parasite and reverses the relocalization of PABP aggregates. T

A Lung-Targeted γδ T Cell Nanovaccine Couples Pulmonary Immune Priming to Systemic Antitumor Immunity in Non-Small-Cell Lung Cancer.

Non-Small-Cell Lung Cancer (NSCLC) often responds poorly to immune checkpoint blockade due to its immunosuppressive, "cold" tumor microenvironment. Activating alternative immune effectors may overcome this limitation. Here we identified lung-enriched γδ T cells as a key compartment in NSCLC and developed a lung-targeted lipid nanovaccine to activate them in situ. Analysis of patient transcriptomic data sets reveals that γδ T cell and CD1d signatures are associated with improved patient survival in NSCLC. Using this insight, we engineered α-galactosylceramide (α-GalCer) and poly(I:C)-loaded lip

CD20 + T lymphocytes in isolated Hashimoto's thyroiditis and type 3 autoimmune polyendocrine syndrome: a pilot study.

BACKGROUND: CD20+ T cells represent up to 5% of circulating T lymphocytes. These cells have been shown to produce higher levels of IL-17A and IFN-γ than those of CD20- T lymphocytes. Some reports described the role of CD20+ T cells in autoimmune disorders such as multiple sclerosis and rheumatoid arthritis possibly due to their ability to produce these inflammatory cytokines. This study is aimed at describing the behavior of CD20+ T lymphocytes in the most frequent autoimmune disorder, i.e., Hashimoto's thyroiditis (HT), presenting isolated or associated to further autoaggressive disorders in

A novel peptide-based sensor platform for detection of anti-Toxoplasma gondii immunoglobulins.

Toxoplasma gondii is an intracellular protozoan parasite responsible for toxoplasmosis, which affects humans and animals. Serologic detection of anti-T. gondii immunoglobulins plays a crucial role in the clinical diagnosis of toxoplasmosis. In this work, a novel electrochemical immunosensor for detecting anti-T. gondii immunoglobulins is reported, based on immobilization of an in silico predicted peptide (PepB3), obtained from membrane protein of T. gondii, on the graphite electrode modified with poly(3-hydroxybenzoic acid). Indirect ELISA confirmed infection and binding specificity of peptide

Development of immune cell delivery system using biodegradable injectable polymers for cancer immunotherapy.

Immune cell delivery using injectable hydrogel attracts much attention for improving its therapeutic effect. Specifically, dendritic cells (DCs) are the trigger cells for immune responses, and DC vaccines are studied for improving cancer immunotherapy. Hydrogel-assisted cell delivery is expected to enhance the viability of the implanted cells. We recently reported temperature-responsive biodegradable injectable polymer (IP) formulation utilizing poly(ε-caprolactone-co-glycolide)-b-poly(ethylene glycol)(PEG)-b-poly(ε-caprolactone-co-glycolide) (tri-PCG). Tri-PCG-based IP was reported to exhibit

The synergistic proapoptotic effect of PARP-1 and HDAC inhibition in cutaneous T-cell lymphoma is mediated via Blimp-1.

The therapy of advanced mycosis fungoides (MF) presents a therapeutic challenge, and the search for new therapeutic targets is ongoing. Poly(ADP-ribose) polymerase 1 was shown to be upregulated in patients with advanced MF and could be druggable by a new class of chemotherapeutic agents, PARP-1 inhibitors, which are already in clinical trials for other malignancies; however, the role of PARP-1 inhibitors in MF has never been established. We examined the efficacy of talazoparib in the murine model of cutaneous T-cell lymphoma. The cytotoxic effect of talazoparib on Moloney MuLV-induced T-cell l

Toward sustainable carbon utilization: Integrated LCA-TEA assessment of carbon dioxide-derived polymers.

The polymers using carbon dioxide (CO2) offer a promising pathway toward CO2 utilization, but their environmental and economic viability remains uncertain. This study implements Life Cycle Assessment (LCA) and Techno-Economic Analysis (TEA) to evaluate three polymer processes that use CO2 as a precursor: poly(trimethylene carbonate) (PTMC), tunable polypropylene carbonate/poly(propylene oxide/polylactic acid) (PPP), and polyhydroxybutyrate (PHB). TRACI 2.1 was used to quantify environmental impacts, while TEA models assess capital investment, operating costs, and key financial indicators under

A Poly(lactic Acid)-Porphyrin-Cobalt Vaccine Adjuvant System Elicits Protein- or Peptide-Specific Humoral and Cellular Immunity.

There is growing interest in vaccine adjuvant systems that are scalable, adaptable, and capable of stimulating humoral and cellular immune responses with recombinant protein or synthetic peptide immunogens. In this study, we introduced a "plug-and-play" nanoadjuvant system consisting of poly(lactic acid)-porphyrin-cobalt ion co-formulated with immunostimulatory saponin QS-21 (PPCoQ). PPCoQ facilitated subunit vaccine assembly by coordinating antigens' polyhistidine tags with immobilized cobalt (Co2+) within a porphyrin-modified poly(lactic acid). A BCMA-mRNA vaccine is a promising therapeutic for multiple myeloma.

Cancer vaccines are emerging as promising therapies to not only prevent cancer but to treat cancer. Here, we developed a therapeutic vaccine for multiple myeloma (MM) using B-cell maturation antigen (BCMA) protein as a target. Given the remarkable efficacy of COVID-19 messenger RNA (mRNA) vaccines, we first packaged sequence- and base-optimized BCMA mRNA into lipid nanoparticles (LNPs) using next-generation ionizable lipid, enhancing their accumulation in the spleen. A toll-like receptor 3 agonist, polyinosinic:polycytidylic acid [poly(I:C)], was also encapsulated in LNPs to further elicit BCM

Dual Treatment of Chronic Chagasic Cardiomyopathy and Parasitic Burden via Combination Nanotherapy.

In chronic Chagas disease, the persistence of the protozoan Trypanosoma cruzi (T. cruzi) is associated with an extensive inflammatory response that impacts cardiac function. The standard treatment, oral benznidazole, effectively targets the parasitic burden but does not address the chronic inflammation nor prevent the progression of severe cardiomyopathies. This presents an inherent immunotherapeutic challenge, as implementing an anti-inflammatory approach can have the unwanted effect of inhibiting beneficial parasite-specific immunity. Here, we investigated a combination therapy approach usin
