OBJECTIVES: β-Caryophyllene (β-Cary) exhibits anti-inflammatory and antioxidant activities. It can be obtained from clove oil, which is rich in eugenol; following eugenol removal, β-Cary can be isolated as the major constituent. However, the high lipophilicity of β-Cary limits its oral bioavailability. The present study investigated the anti-arthritic effects of β-Cary formulated in a self-nanoemulsifying drug delivery system (β-SNEDDS) in Holtzman rats with complete Freund's adjuvant-induced arthritis, emphasizing articular inflammation and systemic oxidative stress.
METHODS: Healthy and adj

β-caryophyllene (BCP) has been shown to alleviate neurological deficits in rats with cerebral ischemia-reperfusion injury (CIRI) induced by middle cerebral artery occlusion (MCAO). However, its molecular targets remain unclear. In this study, transcriptome analysis was conducted to identify BCP-responsive genes and potential therapeutic pathways. RNA sequencing revealed that BCP downregulated genes upregulated by CIRI, particularly those involved in extracellular matrix organization, leukocyte migration, angiogenesis regulation, and reactive oxygen species metabolism. KEGG analysis indicated e

Ochratoxin A (OTA) is a pervasive foodborne mycotoxin linked to multisystem toxicity, including pancreatic toxicity. Increasing evidence indicates that oxidative stress and inflammation caused by OTA are pivotal behind pancreatic damage. β-Caryophyllene (BCP), a natural sesquiterpene found in consumable flora, demonstrates significant antioxidant and anti-inflammatory effects. The objective of this study was to examine the protective effect of BCP against OTA-induced pancreatic toxicity, focusing on the modulation of the Nrf2/HO-1 and NQO1 antioxidant pathway and the NF-κB-mediated activation

BACKGROUND: Type I hypersensitivity is an allergic reaction characterized by the overactivity of the immune system provoked by normally harmless substances. Glucocorticoids, anti-histamines, or mast cell stabilizers are the choices of treatment for type I hypersensitivity. Even though these drugs have the anti-allergic effect, they can have several side effects in prolong use. Cedrol is the main bioactive compound of Cedrus atlantica with anti-tumor, anti-oxidative, and platelet-activating factor inhibiting properties.
METHODS: In this study, the preparation and anti-anaphylactic effect of ce

Cholangiocarcinomas (CCAs) are cancers originated in the biliary tree, which are characterized by their high mortality and marked chemoresistance, partly due to the activity of ATP-binding cassette (ABC) export pumps, whose inhibition has been proposed as a strategy for enhancing the response to chemotherapy. We have previously shown that β-caryophyllene oxide (CRYO) acts as a chemosensitizer in hepatocellular carcinoma by inhibiting ABCB1, MRP1, and MRP2. Here, we have evaluated the usefulness of CRYO in inhibiting BCRP and improving the response of CCA to antitumor drugs. The TCGA-CHOL cohor

Triple-negative breast cancer (TNBC) remains highly aggressive and refractory to conventional treatments, underscoring the need for novel combination strategies. Here, we employed 2D and 3D in vitro models, transcriptomic profiling, and in vivo xenograft studies to evaluate the anticancer efficacy of cannabinoids combined with the terpene β-caryophyllene (BC) in resistant TNBC models. Among the tested cannabinoids, cannabichromene (CBC) exhibited the greatest potency, and its combination with BC at sub-toxic concentrations significantly reduced IC50 values, enhanced cytotoxicity in spheroids,

RNA splicing, transcription and the DNA damage response are intriguingly linked in mammals but the underlying mechanisms remain poorly understood. Using an in vivo biotinylation tagging approach in mice, we show that the splicing factor XAB2 interacts with the core spliceosome and that it binds to spliceosomal U4 and U6 snRNAs and pre-mRNAs in developing livers. XAB2 depletion leads to aberrant intron retention, R-loop formation and DNA damage in cells. Studies in illudin S-treated cells and Csbm/m developing livers reveal that transcription-blocking DNA lesions trigger the release of XAB2 fro
