Complexed Micelles System Based on Ceria Oxide Nanozyme for Bone Remodeling and ROS Scavenging in Periodontitis.

Periodontitis is a chronic inflammatory disease caused by dental plaque and characterized by excessive accumulation of reactive oxygen species (ROS). Current treatments often fail to eliminate persistent inflammation and regenerate damaged tissue. Here, an ROS-responsive multifunctional micelle system (HA-PMs@Ce) constructed by self-assembly of the block copolymer polyethylene glycol-polylysine/phenylboronic acid (PLLA-b-PLys/PBA) and tannic acid (TA) is proposed, encapsulating cerium oxide (CeOx) nanozyme and coated with hyaluronic acid (HA). The catechol groups in TA form ROS-sensitive borat

Tumor-targeted near-infrared/ultraviolet-triggered photothermal/gas therapy nanoplatform for effective cancer synergistic therapy.

The integration of photothermal therapy (PTT) and gas therapy (GT) on a nanoplatform shows great potential in cancer treatment. In this paper, a tumor-targeted near-infrared/ultraviolet (NIR/UV) triggered PTT/GT synergistic therapeutic nanoplatform, PB-CD-PLL(NF)-FA, was designed based on Prussian blue (PB) nanoparticles, 5-chloro-2-nitrobenzotrifluoro (NF)-grafted polylysine (PLL(NF)), and folic acid (FA). PB serves as a core to load PLL(NF) through host-guest interaction and can further modify FA. PB-CD-PLL(NF)-FA can be enriched in tumor tissues by passive targeting with enhanced permeabili

An Injectable Hydrogel with Ultrahigh Burst Pressure and Innate Antibacterial Activity for Emergency Hemostasis and Wound Repair.

Uncontrolled bleeding and wound infections following severe trauma pose significant challenges for existing tissue adhesives, primarily due to their weak wet adhesion, slow adhesion formation, cytotoxicity concerns, and lack of antibacterial properties. Herein, an injectable hydrogel (denoted as ES gel) with rapid, robust adhesive sealing and inherent antibacterial activity based on ε-polylysine and a poly(ethylene glycol) derivative is developed. The engineered hydrogel exhibits rapid gelation behavior, high mechanical strength, strong adhesion to various tissues, and can sustain an ultrahigh

Thermoresponsive gel for sustained release of BMP4 to inhibit corneal neovascularization.

This work aimed to seek a sustained drug release system based on poloxamer-based thermoresponsive gel for sustained release drugs to inhibit corneal neovascularization (CNV) after eye operations. Thus, we designed and prepared a thermoresponsive gel with a phase transition temperature from 22 °C to 25 °C. When the concentrations of poloxamer (P) was 18% (w/w) and ε-Polylysine (EPL) was 0.5 mg/mL (P-18-EPL-05) in the thermoresponsive gel solution, the obtained thermoresponsive gel showed a suitable viscosity and strength in physiological condition. The viscosity, storage modulus G' and loss mod

Microencapsulation of human parathyroid cells: an "in vitro" study.

BACKGROUND: Patients affected by hypoparathyroidism of variable etiology are currently treated with exogenously administered vitamin D and calcium. Human parathyroid transplantation has long been investigated as a possible mean of treating these patients to prevent long-term hypocalcemia. However, the main obstacle for this treatment is represented by tissue rejection. A reliable method to efficiently protect the transplanted tissue from rejection and to allow long-term survival of the graft is the encapsulation of tissues or cells in alginate-polylysine-alginate membranes, which were successf

The role of cell adhesion molecules in neurite outgrowth on Müller cells.

The roles of neural cell adhesion molecule (NCAM), L1, N-cadherin, and integrin in neurite outgrowth on various substrates were studied. Antibodies against these cell surface molecules were added to explants of chick retina and the neurites from retinal ganglion cells were examined for effects of the antibodies on neurite length and fasciculation. On laminin, an anti-integrin antibody completely inhibited neurite outgrowth. The same antibody did not inhibit neurite outgrowth on polylysine or Müller cells. Antibodies to NCAM, L1, and N-cadherin did not significantly inhibit neurite outgrowth on

Pronounced therapeutic potential of oligonucleotides fixed on inorganic nanoparticles against highly pathogenic H5N1 influenza A virus in vivo.

This study describes the effective attack of oligonucleotides on the viral genome of highly pathogenic H5N1 influenza A virus (IAV) in vivo using for the first time the new delivery system consisting of biocompatible low-toxic titanium dioxide nanoparticles and immobilized polylysine-containing oligonucleotides with the native (ODN) and partially modified (ODNm) internucleotide bonds. Intraperitoneal injection of the TiO2•PL-ODN nanocomposite provided 65-70% survival of mice, while intraperitoneal or oral administration of TiO2•PL-ODNm was somewhat more efficient (~80% survival). The virus tit

Molecular basis for the pathological actions of Clostridium perfringens iota toxin.

Clostridium perfringens type E iota toxin is composed of two separate and independent polypeptide chains that act synergistically in mouse lethal assays. The light chain is an enzyme that mono(ADP-ribosyl)ates certain amino acids. The enzyme displays substantial activity when homopoly-L-arginine is used as a substrate, but it shows little activity when polyasparagine, polylysine or polyglutamic acid are used. In keeping with the properties of an ADP-ribosylating enzyme, the toxin possesses the following characteristics. It produces incorporation of radioactivity into polyarginine when adenine-

Mechanisms and applications of condensed RNA-based molecular crowding on enhancing RNA aptamer performance.

BACKGROUND: RNA aptamers represent promising recognition bioreceptors for biosensing, yet practical applications are severely hampered by limited binding affinity, poor thermal stability and high susceptibility to nuclease-mediated degradation. Intracellular molecular crowding environments driven by liquid-liquid phase separation can remodel RNA conformation and improve biomolecular function, inspiring the construction of artificial condensed RNA systems to address the performance bottlenecks of RNA aptamers for foodborne pathogen detection. In this work, we engineered a CUG-trinucleotide-repe

Designing an antimicrobial film for wound applications incorporating bacteriophages and ε-poly-l-lysine.

Alginate-based dressings have been shown to promote wound healing, leveraging the unique properties of alginate. This work aimed to develop and characterize flexible individual and bilayered films to deliver bacteriophages (phages) and ε-Poly-l-lysine (ε-PLL). Films varied in different properties. The moisture content, swelling and solubility increased with higher alginate concentrations. The water vapour permeability, crucial in biomedical films to balance moisture levels for effective wound healing, reached optimal levels in bilayer films, indicating these will be able to sustain an ideal mo

Influence of phenothiazine molecules on the interactions between positively charged poly-l-lysine and negatively charged DPPC/DPPG membranes.

Phenothiazines are very effective antipsychotic drugs, which also have anticancer and antimicrobial activities. Despite being used in human treatment, the molecular mechanism of the biological actions of these molecules is not yet understood in detail. The role of the interactions between phenothiazines and proteins or lipid membranes has been much discussed. Herein, fourier-transform infrared (FTIR) spectroscopic studies were used to investigate the effect of three phenothiazines: fluphenazine (FPh); chlorpromazine (ChP); and propionylpromazine (PP) on the structures of a positively charged p

Biocompatibility investigation of polyethylene glycol and alginate-poly-L-lysine for islet encapsulation.

The use of microencapsulation with alginate-poly-l-lysine (PLL) as the encapsulation material has been hampered by overgrowth of collagen around implanted capsules. Studies have shown that poly(ethylene glycol) (PEG) has higher biocompatibility than PLL. In this project, we examined the biocompatibility of PEG in comparison with PLL in the Lewis rat model. Capsules made from either PEG or PLL were implanted into Lewis rats in three anatomical sites: subcutaneous (SC), intramuscular (IM), and intra-epididymis (IE). After 2 or 4 weeks, capsules were retrieved, sectioned, and stained with Sirius

Systemic antiangiogenic activity of cationic poly-L-lysine dendrimer delays tumor growth.

This study describes the previously unreported intrinsic capacity of poly-L-lysine (PLL) sixth generation (G(6)) dendrimer molecules to exhibit systemic antiangiogenic activity that could lead to solid tumor growth arrest. The PLL-dendrimer-inhibited tubule formation of SVEC4-10 murine endothelial cells and neovascularization in the chick embryo chick chorioallantoic membrane (CAM) assay. Intravenous administration of the PLL-dendrimer molecules into C57BL/6 mice inhibited vascularisation in Matrigel plugs implanted subcutaneously. Antiangiogenic activity was further evidenced using intravital

Differently charged polypeptides in the prevention of post-surgical peritoneal adhesions.

OBJECTIVE: Peritoneal adhesions develop after almost all surgical interventions in the abdomen. We have developed an efficient treatment against post-surgical adhesions consisting of a combination of positively charged poly-L-lysine and negatively charged poly-L-glutamate. The aim of the present study was to further develop the concept of applying oppositely charged polypeptides in the prevention of adhesion formation, by evaluating different doses of the peptides, alterations in the way of administration, and also testing alternative components. MATERIAL AND METHODS: Eighty-five NMRI mice we

Analysis of nonlinear quenching of terbium(III):dipicolinic acid complex fluorescence by chelators and chelate-conjugated macromolecules.

The terbium-dipicolinic acid complex (Tb(III):DPA) fluorescence quenching method of Ma, Hwang, and Lee [Pharm. Res. (1993) 10, 204-207] was used at a lower initial concentration of Tb(III):DPA (0. 2 microM compared to 1.0 microM) to minimize the concentration of chelate-conjugated macromolecules required for estimating concentrations of metal-binding sites. Fluorescence quenching by either EDTA, diethylenetriaminepentaacetic acid (DTPA), poly (L-lysine)-DTPA, or poly (L-lysine)-DTPA linked to mouse IgG (IgG-PL-DTPA) was nonlinear, suggesting initial binding of chelator results in minimal chang

Enhancement of colloid uptake by tumor cell surface electrical charge modification.

Colloidal [51Cr]chromic phosphate uptake is considerably increased by preincubation of P388 ascites leukemia cells with poly(DL-lysine). The uptake increase is in direct relationship with the concentration and the degree of polymerization of poly(DL-lysine). The probable implication of cell surface electrical charge modification in these phenomena is discussed.

Enhancing gluten modification for celiac patients: harnessing microbial transglutaminase, ε-poly-l-lysine, and prolyl endopeptidase.

The immunogenicity of gluten poses a significant challenge for individuals with celiac disease. This study aimed to reduce gluten immunogenicity using a dual treatment strategy: microbial transglutaminase and ε-poly-l-lysine (ε-PL) for structural modification, followed by enzymatic degradation with prolyl endopeptidase. Structural characterization of gluten proteins treated with transglutaminase and ε-PL revealed substantial modification of gliadins, with higher ε-PL concentrations showing more pronounced effects. Low-molecular-weight glutenins were less affected, and no deamidation or impact

Surface tension-driven persistence: How hydrogel interfacial properties regulate fibroblast directional migration.

The development of viscoelastic biomaterials with tunable mechanical properties is a key issue in a wide range of applications in mechanobiology. While numerous foregoing works have revealed the impact of bulk matrix stiffness on cellular and multicellular responses, few have examined the effect of interfacial mechanical properties, such as surface tension σ. Owing to the elastocapillarity phenomenon, σ of soft materials can dominate their bulk mechanical properties and thus regulate cellular response. To address this complex issue of mechanotransduction largely overlooked in the literature, t

Nanocomplexation of thrombin with cationic amylose derivative for improved stability and hemostatic efficacy.

As a topical hemostatic agent, thrombin has wide application for many surgical treatments. However, native thrombin always suffers from its physical and chemical instabilities. In this work, a nanocomplexation strategy was developed for modifying the stability and hemostatic efficacy of thrombin, in which a water-soluble cationic amylose derivative containing poly(l-lysine) dendrons was prepared by a click reaction and then used to complex thrombin in an aqueous system. For resultant thrombin nanocomplexes, their morphology and particle size distribution were investigated. Their stabilities we
