Overexpression of SlREV alters the development of the flower pedicel abscission zone and fruit formation in tomato.

Versatile roles of REVOLUTA (REV), a Class III homeodomain-leucine zipper (HD-ZIP III) transcription factor, have been depicted mainly in Arabidopsis and Populus. In this study, we investigated the functions of its tomato homolog, namely SlREV. Overexpression of a microRNA166-resistant version of SlREV (35S::REV(Ris)) not only resulted in vegetative abnormalities such as curly leaves and fasciated stems, but also caused dramatic reproductive alterations including continuous production of flowers at the pedicel abscission zone (AZ) and ectopic fruit formation on receptacles. Microscopic analysi

Increased thiol biosynthesis of transgenic poplar expressing a wheat O-acetylserine(thiol) lyase enhances resistance to hydrogen sulfide and sulfur dioxide toxicity.

O-acetylserine(thiol) lyase (OASTL), a key enzyme of the plant sulfur assimilatory pathway, catalyses the formation of cysteine from sulfide and O-acetylserine. Transgenic hybrid poplar (Populus sieboldi x P. grandidentata 'Y63') plants expressing cys1, encoding a wheat cytosolic OASTL, were developed in order to examine the role of this enzyme in thiol production following hydrogen sulfide or sulfur dioxide exposure and in the extent of damage induced in the plants by these pollutants. The transgenic cys1 plants accumulated up to several-fold higher cysteine and glutathione levels and were si

Gene Editing in Forest Tree Breeding for Stress Resistance: From Mechanisms to Future Prospects.

Forest ecosystems face escalating threats from climate change alongside a surging demand for sustainable bioproducts. While conventional tree breeding is inherently constrained by long generation cycles, high heterozygosity, and complex genomes, CRISPR-based genome editing provides a precision framework for targeted genetic improvement. This review synthesises the fundamental principles and limitations of multiple gene-editing technologies, with a particular emphasis on CRISPR systems (Cas9, Cas12, and Cas13), in the specific context of woody perennial biology. Recent applications in key fores

A study of metal distribution from lignite fuels using trees as biological monitors.

Concentrations of five metals (Cd, Cu, Mn, Fe, Zn) were determined in tree leaves collected from an area with large coal-fired plants in Ptolemais, Macedonia, Greece. The analyses were carried out with flame atomic absorption spectroscopy. Mean heavy metal content in the tree leaves is, in descending order, Fe>Mn>Zn>Cu>Cd. In Populus nigra and Salix babylonica the dense piled leaves and the widespread root system are the reasons for high heavy metal uptake. Conifer trees Pinus nigra and Juniperus arizona, which have a rough leaf surface, also had elevated concentrations. Leaves with a smooth s

Mechanistic insights into Helicobacter pylori urease inhibition by poplar propolis ethanol extract.

ETHNOPHARMACOLOGICAL RELEVANCE: Propolis, a resinous mixture from plant exudates, is widely used in traditional ethnomedicine for gastrointestinal infections, implying anti-Helicobacter pylori (H. pylori) potential. However, its molecular mechanisms targeting H. pylori urease (HPU), a key virulence factor, remain unclear. AIM OF THE STUDY: This study aimed to characterize the inhibitory activity and kinetic properties of poplar propolis ethanol extract (PPEE) against HPU and jack bean urease (JBU), and to elucidate the underlying molecular mechanisms. MATERIALS AND METHODS: UPLC-MS/MS-based

CHD-18 g-modulated Pseudomonas taxa support poplar salt tolerance.

Against the background of global climate change, soil salinization has emerged as a major abiotic stressor constraining agroforestry productivity worldwide. Root-recruited microbes enhance plant stress resilience, and host-microbe interactions depend on plant root metabolism. Stress-tolerant plant genotypes exhibit a marked capacity to enrich beneficial root-associated microbes through specialized metabolic responses, thereby facilitating phenotypic plasticity. However, the molecular mechanisms underlying these plant-microbe interactions remain unclear. In this study, we compared salt toleranc

Functional insights into 4-coumarate-CoA ligase and caffeic acid O-methyltransferase genes involved in lignin biosynthesis in Chinese fir (Cunninghamia lanceolata).

Lignin biosynthesis is vital for plant growth and development. In model plants, 4-coumarate: coenzyme A ligase (4CL) and caffeic acid O-methyltransferase (COMT) are key regulators of lignin content and composition. However, their functions in gymnosperms, especially conifers, remain largely unexplored. Here, we cloned and functionally characterized the orthologs of these genes in Chinese fir (Cunninghamia lanceolata), the major timber species in China. Enzymatic assays confirmed that Cl4CL catalyzes the conversion of caffeic and p-coumaric acids into CoA esters, while ClCOMT methylates caffeic

Promoted metabolic remolding by overexpression of AspAT9 ameliorates cadmium toxicity in Arabidopsis.

Cadmium (Cd) is a toxic heavy metal that poses a serious threat to crop safety and human health. Aspartate aminotransferase (AspAT) is a prime enzyme engaged in amino acid metabolism, contributing essential metabolic substances for plant growth and acclimatization to various stresses. In this study, we identified a novel AspAT9 gene with high responsiveness to Cd stress from poplar 'Nanlin895' and subsequently transformed it into Arabidopsis. The resulting transgenic AspAT9-1 plants (designated A9) and wild-type (WT) Arabidopsis were subjected to Cd treatment (T) or maintained under control co

Influence of aquifer properties on phytoremediation effectiveness.

Recent research has shown that planting deep-rooted trees, such as poplar, can take up and degrade important ground water pollutants such as trichloroethylene (TCE) as they transpire water from the capillary fringe of shallow contaminated aquifers. The effect of hydrogeologic factors on the minimum plantation area needed to prevent downgradient migration of contaminated ground water is not well known. Accordingly, the objective of this research was to identify the hydrogeologic parameters that control phytoremediation effectiveness. We used a numerical ground water flow model to evaluate the e

Effect of Alumina Nano-Particles on Physical and Mechanical Properties of Medium Density Fiberboard.

This research aims to explore the effects of nanoparticles such as alumina (Al2O3) on the physical and mechanical properties of medium density fiberboards (MDF). The nanoparticles are added in urea-formaldehyde (UF) resin with different concentration levels e.g., 1.5%, 3%, and 4.5% by weight. A combination of forest fibers such as Populus Deltuidess (Poplar) and Euamericana (Ghaz) are used as a composite reinforcement due to their exceptional abrasion confrontation as well as their affordability and economic value with Al2O3-UF as a matrix or nanofillers for making the desired nanocomposite sp

Growth-regulating factor 5 (GRF5)-mediated gene regulatory network promotes leaf growth and expansion in poplar.

Although polyploid plants have larger leaves than their diploid counterparts, the molecular mechanisms underlying this difference (or trait) remain elusive. Differentially expressed genes (DEGs) between triploid and full-sib diploid poplar trees were identified from two transcriptomic data sets followed by a gene association study among DEGs to identify key leaf growth regulators. Yeast one-hybrid system, electrophoretic mobility shift assay, and dual-luciferase assay were employed to substantiate that PpnGRF5-1 directly regulated PpnCKX1. The interactions between PpnGRF5-1 and growth-regulati

The mechanism of chelator improved the tolerance and accumulation of poplar to Cd explored through differential expression protein based on iTRAQ.

Appropriate chelator may increase plant tolerance and accumulation for Cd in soil, but its molecular mechanism is unclear. In this experiment, the technology of isobaric tags for relative and absolute quantitation (iTRAQ) was used to compare the differential expression proteins (DEPs) and differential expression genes (DEGs) characteristics of poplar accumulating Cd combined with EDTA and/or EGTA. The results showed that the Cd concentrations, biomasses and activities of antioxidant enzymes of poplar were significantly increased in the treatments of chelator addition compared to the TCd. A genetic basis for the manipulation of sink-source relationships by the galling aphid Pemphigus batae.

We examined how the galling aphid Pemphigus batae manipulates resource translocation patterns of resistant and susceptible narrowleaf cottonwood Populus angustifolia. Using carbon-14 ((14)C)-labeling experiments in common garden trials, five patterns emerged. First, although aphid galls on resistant and susceptible genotypes did not differ in their capacity to intercept assimilates exported from the leaf they occupied, aphids sequestered 5.8-fold more assimilates from surrounding leaves on susceptible tree genotypes compared to resistant genotypes. Second, gall sinks on the same side of a shoo

Cytoskeletal Regulation of Wood Formation and Stress Adaptation in Woody Plants.

The cytoskeleton regulates cell division, intracellular transport and cell-wall patterning, but its roles in secondary growth and long-term stress responses in woody plants remain poorly understood. We classify individual claims as direct functional evidence from woody species (Level I), correlative or functionally non-specific evidence from woody systems (Level II), mechanisms demonstrated only in non-woody models (Level III), or relationships untested in woody plants (Hypothesis). Direct functional evidence remains concentrated in Populus. In hybrid poplar, PagPCaP1a promotes salt-induced mi

Integrating Multivariate Ordination and Machine Learning to Disentangle the Environmental Drivers of Xylem Sap Redox Metabolism in Trees.

Xylem sap is increasingly recognized as a dynamic biological matrix reflecting whole-plant physiological status, but its biochemical variation under field conditions remains insufficiently characterized. We investigated oxidative stress markers, osmolytes, antioxidant enzymes, and redox-related enzymes in xylem sap from three focal tree individuals representing Fraxinus excelsior, Populus nigra, and Pinus sylvestris. Sap was collected by passive stem tapping using a custom-built apparatus, and biochemical patterns were evaluated using multivariate statistical and machine-learning approaches. T

Mechanisms of Isoprene Decoupling in Poplar: Precursor Dynamics and VOC Fluxes Under Acute Thermal Exposure and Elevated CO2.

Rising temperatures and atmospheric CO2 exert complex, interacting effects on plant carbon metabolism and volatile organic compound (VOC) emissions. This study investigated the physiological mechanisms underlying acute thermal tolerance in Populus nigra by integrating leaf gas exchange with high-resolution proton-transfer-reaction time-of-flight mass spectrometry (PTR-TOF-MS). We employed a factorial design (25-40 °C; 400 and 800 ppm CO2) to examine how metabolic regulation and pulse-induced signalling interact across thermal gradients. Our results identify a critical metabolic tipping point a

Evolutionary relationships of Pemphigus and allied genera (Hemiptera: Aphididae: Eriosomatinae) and their primary endosymbiont, Buchnera aphidicola.

Aphids harbor primary endosymbionts, Buchnera aphidicola, in specialized cells within their body cavities. Aphids and Buchnera have strict mutualistic relationships in nutrition exchange. This ancient association has received much attention from researchers who are interested in endosymbiotic evolution. Previous studies have found parallel phylogenetic relationships between non-galling aphids and Buchnera at lower taxonomic levels (genus, species). To understand whether relatively isolated habitats such as galls have effect on the parallel relationships between aphids and Buchnera, the present

The Virulence-Associated CAP/PR1-Like Proteins From Cytospora chrysosperma Target a Subunit of RNA Polymerase II to Suppress Plant Immunity.

Plant pathogenesis-related 1 (PR1) proteins are members of the CAP protein superfamily and are widely used immune markers. The precise function and mode of action of CAP proteins, however, remains poorly understood. In Cytospora chrysosperma, the causative agent of poplar canker disease, three CAP family members, CcCAP1-3, have been identified. CcCAP1 is required for fungal virulence and suppresses the plant immune response to facilitate fungal colonisation, in a nuclear-localisation dependent way. In this study, we functionally characterised the other two members of the C. chrysosperma CAP fa

Genome-wide identification of the oligopeptide transporter gene family in poplar and their responses to iron and cadmium.

BACKGROUNDS: The oligopeptide transporter (OPT) family belongs to a type of proton-coupled symporter and functions in the transport of metal ions, glutathione, phytochelatins (PCs), and metal-PC complexes, as well as in the regulation of metal homeostasis. Polplar, a fast-growing woody model species which has been resequenced multiple times, shows great potential in phytoremediation. However, the identification of OPT family genes and their responses to cadmium (Cd) and iron (Fe) remain poorly understood. RESULTS: This study identified 30 OPT genes (17 yellow stripe-like (YSL) subfamily membe

Ethylene response factor 113 regulates the growth and development of poplar through the tryptophan-dependent auxin metabolism.

ERF transcription factors play a pivotal role in orchestrating plant growth and development processes. In this study, PagERF113 was isolated from the 84 K poplar (Populus alba × P. glandulosa) and its function was detected using molecular and genetic methods. Subcellular localization and trans-activation assays indicated that PagERF113 lacks transcriptional activation capability, residing in both the cell membrane and nucleus. Phenotypic analyses revealed that transgenic poplar lines overexpressing PagERF113 (OE) exhibited weedy growth compared to wild-type (WT), characterized by small leaves,
