Immunophenotyping Studies in Nonhuman Primate Models.

Nonhuman primate (NHP) models are important in biomedical research due to their close evolutionary relationship with humans, which provides a high degree of anatomical, genetic, and physiological similarity. This close resemblance is particularly valuable for studying complex biological processes and diseases that are difficult to model in other animals. NHPs have hematopoietic and immune systems that closely mirror those of humans, making them crucial for researching hematopoietic stem cell biology, immunology, infectious diseases, and vaccine development, offering insights that are more like

Evolutionary specializations in primate cortical development.

Primates exhibit distinctive cognitive and behavioral repertoires relative to many other mammals. These capabilities are not associated with the emergence of entirely new cortical structures, but rather with coordinated evolutionary modifications within the cerebral cortex. To elucidate the complexity of these processes, this review adopts an analytical framework that progresses from macroscopic to microscopic levels and from structural changes to underlying mechanisms. We synthesize recent advances with a particular focus on multilevel features revealed by emerging technologies such as single

Visual Exploration and the Primate Hippocampal Formation.

During the 1990s and early 2000s, research in humans and in the nonhuman primate model of human amnesia revealed that tasks involving free viewing of images provided an exceptionally sensitive measure of recognition memory. Performance on these tasks was sensitive to damage restricted to the hippocampus as well as to damage that included medial temporal lobe cortices. Early work in my laboratory used free-viewing tasks to assess the neurophysiological correlates of recognition memory, and the use of naturalistic visual exploration opened rich avenues to assess other aspects of the impact of ey

"Ontogenetic Scaling of the Primate Middle Ear".

The study of primate auditory morphology is a significant area of interest for comparative anatomists, given the phylogenetic relationships that link primate hearing and the morphology of these auditory structures. Extensive literature addresses the form-to-function relationship of the auditory system (outer, middle, and inner ear) in primates and, by extension, provides insight into the auditory system of extinct primates and even modern humans. We add to this literature by describing the ontogenetic trajectory of the middle ear cavity and ossicular chain (malleus, incus, and stapes) due to t

Functional diversity in the output of the primate retina.

The visual image transmitted by the retina to the brain has long been understood in terms of spatial filtering by the center-surround receptive fields of retinal ganglion cells (RGCs). Recently, this textbook view has been challenged by the stunning functional diversity and specificity observed in ∼40 distinct RGC types in the mouse retina. However, it is unclear whether the ∼20 morphologically and molecularly identified RGC types in primates exhibit similar functional diversity, or instead exhibit center-surround organization at different spatial scales. Here, we reveal striking and surprisin

Anatomical organization of forebrain circuits in the primate.

The primate forebrain is a complex structure. Thousands of connections have been identified between cortical areas, and between cortical and sub-cortical areas. Previous work, however, has suggested that a number of principles can be used to reduce this complexity. Here, we integrate four principles that have been put forth previously, including a nested model of neocortical connectivity, gradients of connectivity between frontal cortical areas and the striatum and thalamus, shared patterns of sub-cortical connectivity between connected posterior and frontal cortical areas, and topographic org

A primate perspective on oocytes and transgenerational PCOS.

'Androgenized' rodent models are widely used to explore the pathophysiology underlying human polycystic ovary syndrome (PCOS), including reproductive and metabolic dysfunction. Based on a recent study using a dihydrotestosterone (DHT)-treated murine model, it has been proposed that prenatal androgen excess alone can predispose to transgenerational transmission of PCOS. From RNA sequencing analysis of metaphase II (MII) oocytes of androgenized lineages, the authors speculated that oocyte factors, including up-regulation of cytotoxic granulosa-associated RNA binding protein-like 1 (TiaL1), are s

A Primate Model for Viral Hemorrhagic Fever.

Lymphocytic choriomeningitis virus strain WE (LCMV-WE), a Risk Group 3 virus, causes a disease in rhesus monkeys that closely resembles human infection with Lassa fever virus, a Risk Group 4 agent. Three stages of disease progression have been defined and profiled in this model: pre-viremic, viremic, and terminal. The earliest or pre-viremic stage reveals changes in the blood profile predictive of the later stages of disease. In order to identify whether specific changes are pathognomonic, it was necessary to perform a parallel infection with an attenuated virus (LCMV-Armstrong). Here we revie

Adaptations for social cognition in the primate brain.

Studies of the factors affecting reproductive success in group-living monkeys have traditionally focused on competitive traits, like the acquisition of high dominance rank. Recent research, however, indicates that the ability to form cooperative social bonds has an equally strong effect on fitness. Two implications follow. First, strong social bonds make individuals' fitness interdependent and the 'free-rider' problem disappears. Second, individuals must make adaptive choices that balance competition and cooperation-often with the same partners. The proximate mechanisms underlying these behavi

Functional morphology of the primate head and neck.

The vertebral column plays a key role in maintaining posture, locomotion, and transmitting loads between body components. Cervical vertebrae act as a bridge between the torso and head and play a crucial role in the maintenance of head position and the visual field. Despite its importance in positional behaviors, the functional morphology of the cervical region remains poorly understood, particularly in comparison to the thoracic and lumbar sections of the spinal column. This study tests whether morphological variation in the primate cervical vertebrae correlates with differences in postural be

Social consequences of disability in a nonhuman primate.

Debates about the likelihood of conspecific care for disabled individuals in ancestral hominins rely on evidence from extant primates, yet little is known about social treatment (positive, neutral or negative) of physically disabled individuals in nonhuman primates. A group of free-ranging Japanese macaques (Macaca fuscata) at the Awajishima Monkey Center (AMC) in Japan presents a unique opportunity to investigate the relationships between physical impairment and social behavior, in the context of congenital limb malformation in adult nonhuman primates. We collected behavioral data on 23 focal

[Experimental whooping cough of nonhuman primate].

Despite considerable success in study of Bordetella pertussis virulence factors, pathogenesis of whooping cough, duration of B. pertussis bacteria persistence, types and mechanisms of immune response are still keep underinvestigated. It can be explained by the absence ofadequate experimental animal model for pertussis study. Our study estimates clinical and laboratory parameters of whooping cough in non-human primates of the Old World in the process of intranasan infection by virulent B. pertussis bacteria. Also the duration of B. pertussis bacteria persistence in animals was investigated. 14

Evolutionary time-scale of primate bocaviruses.

Human bocavirus (HBoV) is associated with acute gastroenteritis in humans, occurring mostly in young children and elderly people. Four bocavirus genotypes (HBoV1-HBoV4) have been found so far. Since there were no data on the contribution of HBoV to gastroenteritis in Russia, 1781 fecal samples collected from infants hospitalized with acute gastroenteritis in Novosibirsk, Russia during one year were tested for the presence of nucleic acids from HBoV and three major gastrointestinal viruses (rotavirus A, norovirus II, and astrovirus). HBoV was detected only in 1.9% of the samples: HBoV1 was dete

Perinatal asphyxia in a nonhuman primate model.

Perinatal asphyxia is a leading cause of brain injury in neonates, occurring in 2-4 per 1,000 live births, and there are limited treatment options. Because of their similarity to humans, nonhuman primates are ideal for performing preclinical tests of safety and efficacy for neurotherapeutic interventions. We previously developed a primate model of acute perinatal asphyxia using 12-15 min of umbilical cord occlusion. Continuing this research, we have increased cord occlusion time from 15 to 18 min and extended neurodevelopmental follow-up to 9 months. The purpose of this report is to evaluate t

Ductal plate malformation in a nonhuman primate.

During necropsy of a 6-year-old, male African Green monkey (Cercopithecus aethiops), multifocal pale, white to tan areas, 1-2 cm in diameter, were noted in the right lateral lobe, medial lobe, and capsular surface of the liver. Light microscopic examination of the liver revealed numerous spindle cells dispersed within interconnecting, broad bands of collagen, up to 200 microm in width, which often spanned and connected portal areas. A diagnosis of hepatic ductal plate malformation, most closely resembling congenital hepatic fibrosis with von Meyenburg complexes, was made. Few cases of ductal p

Regulation of gene expression in primate polyomaviruses.

Polyomaviruses are a growing family of small DNA viruses with a narrow tropism for both the host species and the cell type in which they productively replicate. Species host range may be constrained by requirements for precise molecular interactions between the viral T antigen, host replication proteins, including DNA polymerase, and the viral origin of replication, which are required for viral DNA replication. Cell type specificity involves, at least in part, transcription factors that are necessary for viral gene expression and restricted in their tissue distribution. In the case of the huma

Parasites and the evolutionary diversification of primate clades.

Coevolutionary interactions such as those between hosts and parasites have been regarded as an underlying cause of evolutionary diversification, but evidence from natural populations is limited. Among primates and other mammalian groups, measures of host diversification rates vary widely among lineages, but comparative studies have not yet identified a reliable explanation for this variation. In this study, we used a comprehensive data set of disease-causing organisms from free-living primates to illustrate how phylogenetic comparative methods can be used to examine mammalian lineage diversity
