In vivo studies of the regulation of neuropeptide stores in structures of the rat brain.

In order to evaluate the regulation of neuropeptide stores in structures of the rat brain, the content of the specific messenger ribonucleic acid for the peptide precursor, of the precursor and of its biologically active peptide fragment, resulting from the precursor processing was measured. Enkephalin stores of the striatum, but not that of other structures, were upregulated by repeated daily doses of dopaminergic receptor antagonists of the D-1 type. Morphine tolerance failed to change stores of enkephalin in brain but produced a down-regulation of the synthesis of pro-opiomelanocortin in th

60 YEARS OF POMC: Purification and biological characterisation of melanotrophins and corticotrophins.

The remarkable conservation of the primary structures and anatomical location of dogfish α-melanocyte-stimulating hormone (MSH), corticotrophin-like intermediate lobe peptide (CLIP) and adrenocorticotrophic hormone (ACTH) compared with mammals reinforced the tissue-specific processing hypothesis of ACTH peptides in the pituitary gland. The cloning of dogfish pro-opiomelanocortin (POMC) led to the identification of δ-MSH and simultaneously revealed the high conservation of the γ-MSH sequence during evolution. These studies have also shown that β-MSH is much less conserved during evolution and i

Evaluation of the Relationship Between Orexin A, Peptide YY, AgRP, and POMC Levels and Sleep Disorders in Children with Malnutrition.

BACKGROUND: Malnutrition and sleep disturbances are common in childhood and are associated with neuropeptides that regulate appetite and circadian rhythms. Hypothalamic peptides such as orexin A, agouti-related protein (AgRP), proopiomelanocortin (POMC), and peptide YY (PYY) play important roles in energy balance and eating behavior; however, their specific functions in pediatric malnutrition remain unclear. This study aimed to determine the levels of these peptides in malnourished children and to examine their relationship with eating and sleep behaviors. METHODS: This case-control, cross-se

Regulation of Melanogenesis in Skin Epidermal Keratinocytes via Activation of α7 Nicotinic Acetylcholine Receptor on the Expression of α-Melanocyte-Stimulating Hormone.

UV irradiation stimulates the production of pro-opiomelanocortin (POMC) and its derived peptides, for example, α-melanocyte stimulating hormone (α-MSH), from keratinocytes that subsequently stimulate melanin production in melanocytes. The idea of "skin synapse" describes the interplay between keratinocytes and melanocytes being modulated by cholinergic signaling, in which acetylcholine (ACh) release is induced by UV irradiation, regulating the process of pigmentation. ObjectiveHere, the role of the cholinergic system in regulating α-MSH production in keratinocytes during UV-induced skin pigmen

A cellular and molecular basis of leptin resistance.

Similar to most humans with obesity, diet-induced obese (DIO) mice have high leptin levels and fail to respond to the exogenous hormone, suggesting that their obesity is caused by leptin resistance, the pathogenesis of which is unknown. We found that leptin treatment reduced plasma levels of leucine and methionine, mTOR-activating ligands, leading us to hypothesize that chronic mTOR activation might reduce leptin signaling. Rapamycin, an mTOR inhibitor, reduced fat mass and increased leptin sensitivity in DIO mice but not in mice with defects in leptin signaling. Rapamycin restored leptin's ac

Mineralocorticoid receptor activates postnatal adiposity in zebrafish lacking proopiomelanocortin.

The proopiomelanocortin (Pomc)-derived peptides, including adrenocorticotropic hormone and α-melanocyte stimulating hormone (α-Msh), play both a central and a peripheral role in modulating the stress response. The central role is predominantly associated with nutrient homeostasis, while peripherally they play an important role in the synthesis of glucocorticoids (GCs) in response to stress. Pomc mutations are a major risk factor in the development of early-onset childhood obesity in humans. This is attributed primarily to their central effects on melanocortin receptor dysfunction leading to hy

GPSM1 in POMC neurons impairs brown adipose tissue thermogenesis and provokes diet-induced obesity.

OBJECTIVE: G-protein-signaling modulator 1 (GPSM1) has been proved the potential role in brain tissues, however, whether GPSM1 in hypothalamic nuclei, especially in POMC neurons is essential for the proper regulation of whole-body energy balance remains unknown. The aim of our current study was to explore the role of GPSM1 in POMC neurons in metabolic homeostasis. METHODS: We generated POMC neuron specific GPSM1 deficiency mice and subjected them to a High Fat Diet to monitor metabolic phenotypes in vivo. By using various molecular, biochemical, immunofluorescent, immunohistochemical analyses

Mapping key neuropeptides involved in the melanocortin system in Atlantic salmon (Salmo salar) brain.

The melanocortin system is a key regulator of appetite and food intake in vertebrates. This system includes the neuropeptides neuropeptide y (NPY), agouti-related peptide (AGRP), cocaine- and amphetamine-regulated transcript (CART), and pro-opiomelanocortin (POMC). An important center for appetite control in mammals is the hypothalamic arcuate nucleus, with neurons that coexpress either the orexigenic NPY/AGRP or the anorexigenic CART/POMC neuropeptides. In ray-finned fishes, such a center is less characterized. The Atlantic salmon (Salmo salar) has multiple genes of these neuropeptides due to

Sex differences in the peripubertal response to a short-term, high-fat diet intake.

Obesity is one of the most important health problems facing developed countries because being overweight is associated with a higher incidence of type 2 diabetes, cardiovascular disease and cancer, as well as other comorbidities. Although increased weight gain results from a combination of poor dietary habits and decreased energy expenditure, not all individuals have equal propensities to gain weight or to develop secondary complications of obesity. This is partially a result not only of genetics, including sex, but also the time during which an individual is exposed to an obesogenic environme

Proopiomelanocortin Deficiency Treated with a Melanocortin-4 Receptor Agonist.

Patients with rare defects in the gene encoding proopiomelanocortin (POMC) have extreme early-onset obesity, hyperphagia, hypopigmentation, and hypocortisolism, resulting from the lack of the proopiomelanocortin-derived peptides melanocyte-stimulating hormone and corticotropin. In such patients, adrenal insufficiency must be treated with hydrocortisone early in life. No effective pharmacologic treatments have been available for the hyperphagia and obesity that characterize the condition. In this investigator-initiated, open-label study, two patients with proopiomelanocortin deficiency were tre

Adult pituitary cell maintenance: lineage-specific contribution of self-duplication.

The identification of a stable pool of progenitor/stem cells in the adult pituitary has renewed the interest of identifying mechanisms for maintenance of pituitary cells throughout life. Whereas developmental studies have shown that progenitor expansion is the major source of new differentiated cells during pituitary organogenesis, the contribution of these progenitors for maintenance of the adult tissue is not clear although progenitors were clearly involved in cell expansion following end-organ ablation, notably after adrenalectomy and/or gonadectomy. We have used a genetic trick that elimin

Anti-neuropeptide Y plasma immunoglobulins in relation to mood and appetite in depressive disorder.

Depression and eating disorders are frequently associated, but the molecular pathways responsible for co-occurrence of altered mood, appetite and body weight are not yet fully understood. Neuropeptide Y (NPY) has potent antidepressant and orexigenic properties and low central NPY levels have been reported in major depression. In the present study, we hypothesized that in patients with major depression alteration of mood, appetite and body weight may be related to NPY-reactive autoantibodies (autoAbs). To test this hypothesis, we compared plasma levels and affinities of NPY-reactive autoAbs bet

Nicotine excites hypothalamic arcuate anorexigenic proopiomelanocortin neurons and orexigenic neuropeptide Y neurons: similarities and differences.

Two of the biggest health problems facing us today are addiction to nicotine and the increased prevalence of obesity. Interestingly, nicotine attenuates obesity, but the underlying mechanism is not clear. Here we address the hypothesis that if weight-reducing actions of nicotine are mediated by anorexigenic proopiomelanocortin (POMC) neurons of the hypothalamic arcuate nucleus, nicotine should excite these cells. Nicotine at concentrations similar to those found in smokers, 100-1,000 nM, excited POMC cells by mechanisms based on increased spike frequency, depolarization of membrane potential,

Relative number and distribution of murine hypothalamic proopiomelanocortin neurons innervating distinct target sites.

Proopiomelanocortin (POMC) neurons send projections widely throughout the brain consistent with their role in regulating numerous homeostatic processes and mediating analgesia and reward. Recent data suggest that POMC neurons located in the rostral and caudal extents of the arcuate nucleus of the hypothalamus may mediate selective actions, however it is not clear if POMC neurons in these regions of the arcuate nucleus innervate specific target sites. In the present study, fluorescent microspheres and cholera toxin B were used to retrogradely label POMC neurons in POMC-DsRed transgenic mice. Th

Pituitary hyperplasia.

Comprehensive article summarizing more than 25 years of experience with pituitary hyperplasia in surgical material. Morphologic forms of hyperplasia--diffuse and nodular--are defined and, for comparison, the normal morphology, frequency and intraglandular distribution of cell types are briefly reviewed. All cell types can give rise to hyperplasia, although their frequency, extent and clinical importance widely vary. Somatotroph hyperplasia is rare; it is limited to cases of GHRH overproduction by extrapituitary endocrine neoplasms and sporadic examples of gigantism. Prolactin cells display the

Diurnal pattern of proopiomelanocortin gene expression in the arcuate nucleus of proestrous, ovariectomized, and steroid-treated rats: a possible role in cyclic luteinizing hormone secretion.

Opiate peptides are thought to modulate the pattern of LH release in female rats. We tested the hypothesis that changes in proopiomelanocortin (POMC) gene expression occur in proestrous (PRO) and ovariectomized (OVX) steroid-treated rats which may explain their unique patterns of LH secretion. Using in situ hybridization, we examined whether diurnal changes in POMC gene expression occur in the arcuate nucleus. Four groups of rats were used in this study. 1) PRO rats were used after exhibiting at least two consecutive 4-day estrous cycles; 2) OVX rats were killed 9 days after ovariectomy; 3) es

Prevalence of POMC allele associated with obesity in feline population in Vietnam.

Feline obesity is an increasingly important health problem influenced by both genetic predisposition and husbandry practices. This study investigated the prevalence and phenotypic relevance of the feline proopiomelanocortin (POMC) c.28G > C (p.Gly10Arg) variant in a Vietnamese cat population and evaluated environmental factors associated with obesity. A total of 63 clinically healthy cats were classified as normal weight (body condition score [BCS] 5-6/9; n = 40) or obese (BCS ≥7/9; n = 23). Genotyping was performed using a newly developed PCR-restriction fragment length polymorphism assay and

Hypothalamic orexigenic and anorexigenic neuropeptides in the rotenone model of Parkinson's disease.

Beyond depression and anxiety, non-motor symptoms of Parkinson's disease (PD) also include body weight changes with an unclear neurobiological background. Here, we aimed to characterize the functional-morphological changes of the main orexigenic and anorexigenic hypothalamic peptidergic systems in the rotenone model of PD in male rats. We also tested their response to benserazide/levodopa (B/L) anti-PD therapy alone or in combination with antidepressant (fluoxetine) medication. In the model, anhedonia and compromised motor coordination were observed in sucrose preference and rotarod tests resp

TRH can stimulate the release of two POMC-derived pituitary hormones, ACTH and MSH, in medaka.

Anterior pituitary hormone secretion is generally considered to be under the strong regulation of hypothalamic neuropeptides. In mammals, adrenocorticotropic hormone (ACTH), which plays a crucial role in the stress response, is secreted from corticotropes and is regulated primarily by corticotropin-releasing hormone (CRH). In teleosts, although the pharmacological effects of hypothalamic factors have been demonstrated, their relative importance in regulating ACTH release remains controversial. One reason for this is the lack of methods for evaluating ACTH release at cellular resolution. Using
