Acetylation Regulating Protein Stability and DNA-Binding Ability of HilD, thus Modulating Salmonella Typhimurium Virulence.

HilD, a dominant regulator of Salmonella pathogenicity island 1, can be acetylated by protein acetyltransferase (Pat) in Salmonella Typhimurium, and the acetylation is beneficial to its stability. However, the underlying mechanism of HilD stability regulated by acetylation is not clear. We show here that lysine 297 (K297) located in the helix-turn-helix motif, can be acetylated by Pat. Acetylation of K297 increases HilD stability, but reduces its DNA-binding affinity. In turn, the deacetylated K297 enhances the DNA-binding ability but decreases HilD stability. Under the Salmonella pathogenicit

Genetic characterization and protein stability analysis of a Chinese family with Von Hippel-Lindau disease.

BACKGROUND: Von Hippel-Lindau disease (VHL), a heritable autosomal dominant disease characterized by neoplasia in multiple organ systems, has rarely been reported in Asia. We genetically investigated a unique Chinese family with VHL disease and performed an analysis of the VHL protein stability. METHODS: Genomic deoxyribonucleic acid (DNA) extracted from peripheral blood was amplified by polymerase chain reaction (PCR) to three exons of the VHL gene in 9 members of the Chinese family with VHL disease. PCR products were directly sequenced. We estimated the effects of VHL gene mutation on the s

Covalent attachment of bacteriorhodopsin monolayer to bromo-terminated solid supports: preparation, characterization, and protein stability.

The interfacing of functional proteins with solid supports and the study of related protein-adsorption behavior are promising and important for potential device applications. In this study, we describe the preparation of bacteriorhodopsin (bR) monolayers on Br-terminated solid supports through covalent attachment. The bonding, by chemical reaction of the exposed free amine groups of bR with the pendant Br group of the chemically modified solid surface, was confirmed both by negative AFM results obtained when acetylated bR (instead of native bR) was used as a control and by weak bands observed

[Diurnal fluctuations of protein contents and the pH dependence of beta2-microglobulin stability in urine].

Diurnal fluctuations of protein excretion into urine and the effect of urinary pH on the urinary protein concentrations were studied in patients with various kidney diseases. The diurnal kinetics of gamma-immunoglobulin, transferrin, albumin, alpha1-microglobulin, gamma-immunoglobulin light chains, and the retinol-binding protein proved to positively correlate with the diurnal fluctuations of proteinuria and to negatively correlate with urinary pH. Diurnal changes in urinary beta2-microglobulin content did not correlate with those of any other protein. Oral bicarbonate intake alkalinized the u

Conformational stability of human frataxin and effect of Friedreich's ataxia-related mutations on protein folding.

The neurodegenerative disorder FRDA (Friedreich's ataxia) results from a deficiency in frataxin, a putative iron chaperone, and is due to the presence of a high number of GAA repeats in the coding regions of both alleles of the frataxin gene, which impair protein expression. However, some FRDA patients are heterozygous for this triplet expansion and contain a deleterious point mutation on the other allele. In the present study, we investigated whether two particular FRDA-associated frataxin mutants, I154F and W155R, result in unfolded protein as a consequence of a severe structural modificatio

The F-box protein Dia2 overcomes replication impedance to promote genome stability in Saccharomyces cerevisiae.

The maintenance of DNA replication fork stability under conditions of DNA damage and at natural replication pause sites is essential for genome stability. Here, we describe a novel role for the F-box protein Dia2 in promoting genome stability in the budding yeast Saccharomyces cerevisiae. Like most other F-box proteins, Dia2 forms a Skp1-Cdc53/Cullin-F-box (SCF) E3 ubiquitin-ligase complex. Systematic analysis of genetic interactions between dia2Delta and approximately 4400 viable gene deletion mutants revealed synthetic lethal/synthetic sick interactions with a broad spectrum of DNA replicati

Rapid release and unusual stability of immunodominant peptide 45-89 from citrullinated myelin basic protein.

Myelin basic protein (MBP) exists in a population of isoforms and isomers. The 18.5 kDa MBP-C1, the main human adult isoform, has 170 residues and is relatively unmodified, whereas the same isoform can be citrullinated on six arginine residues to create the MBP-C8 (MBP Cit6) isomer. MBP Cit6 dominates in MS brain, accounting for 45% rather than 25% of the population of MBP isomers. In the fulminant form of MS, known as Marburg's Disease, 18 of the 19 arginines in MBP are citrullinated (MBP Cit18). Citrullination of MBP could lead to instability of myelin or limited remyelination. In this inves

Protein kinase C regulates the magnitude and stability of CFTR currents in pancreatic duct cells.

Activation of protein kinase C (PKC) inhibits adenosine 3',5'-cyclic monophosphate (cAMP)-stimulated fluid secretion in rat pancreatic ducts (N. Ashton, R. L. Evans, and B. E. Argent. J. Physiol. Lond. 452: 99P, 1992). Using the patch-clamp technique, we have investigated whether this inhibition of fluid secretion results from an effect of PKC on cystic fibrosis transmembrane conductance regulator (CFTR) Cl channels. Exposure to 100 nM 4 beta-phorbol 12,13-dibutyrate (PDBu) had no effect on CFTR current density in unstimulated duct cells, but caused a 31% increase in the magnitude of CFTR curr

Influence of iron, calcium, and zinc on the oxidative stability of omega-3 fatty acids in pea protein-stabilized oil-in-water emulsions.

BACKGROUND: Plant-based emulsified foods fortified with omega-3 fatty acids and minerals are nutritionally attractive, but their formulation is challenging because alpha-linolenic acid (ALA) is highly susceptible to oxidation and some mineral salts may act as pro-oxidants. This study evaluated how iron, zinc, and calcium salts affect pea protein isolate (PPI) properties and ALA oxidative stability in PPI/kappa-carrageenan (KC) gelled emulsions. Treatments included PPI-mineral systems containing ferrous glycinate (FeGly), zinc gluconate (ZnG), calcium gluconate (CaG), calcium lactate (CaL), cal

High internal phase emulsions stabilized by polyphenol-mediated Moringa seed protein for β-carotene encapsulation: Enhanced stability and bioaccessibility.

In this study, Moringa oleifera seed protein (MOSP) served as the raw material, while three polyphenols-epicatechin (EC), epigallocatechin gallate (EGCG), and tannic acid (TA)-were incorporated for modification to produce a high internal phase emulsion (HIPE). For each of the three polyphenols, the optimal addition level was around 15 μg per 1 mg of MOSP. The inclusion of polyphenols resulted in the interfacial tension being at the oil-water interface was significantly diminished, decreasing from 10.22 mN/m to 5.43 mN/m, with the emulsifying activity index (EAI) of MOSP-TA reaching 72.50 m2/g.

Fabrication of a Polysaccharide-Protein/Protein Complex Stabilized Oral Nanoemulsion to Facilitate the Therapeutic Effects of 1,8-Cineole on Atherosclerosis.

Atherosclerosis (AS) is a systemic disease characterized by lipid deposition in the blood vessel wall that urgently requires effective and safe therapeutic drugs for long-term treatment. An essential oil monomer-1,8-cineole (CIN) with ameliorative effects on vascular injuries has considerable potential for preventing the progression of AS because of its antioxidant, anti-inflammation, and cholesterol regulatory effects. However, the high volatility and instability of CIN result in low oral bioavailability and a short half-life, thereby limiting its clinical application. We formulated a nanoemu

Stabilizing the subtilisin BPN' pro-domain by phage display selection: how restrictive is the amino acid code for maximum protein stability?

We have devised a procedure using monovalent phage display to select for stable mutants in the pro-domain of the serine protease, subtilisin BPN'. In complex with subtilisin, the pro-domain assumes a compact structure with a four-stranded antiparallel beta-sheet and two three-turn alpha-helices. When isolated, however, the pro-domain is 97% unfolded. These experiments use combinatorial mutagenesis to select for stabilizing amino acid combinations at a particular structural locus and determine how many combinations are close to the maximum protein stability. The selection for stability is based

Cryoprotection of Tremella polysaccharides in silver carp myofibrillar protein: stability improvement against freeze-thaw damage.

BACKGROUND: Myofibrillar proteins of silver carp are susceptible to freezing denaturation, significantly degrading surimi quality. Tremella polysaccharide (TP), a natural edible mushroom polysaccharide, offers potential cryoprotective value and can inhibit this deterioration. The aim of this study was to investigate the cryoprotective effect of TP on the myofibrillar protein of silver carp. RESULTS: Tremella polysaccharide effectively mitigated protein aggregation and freeze-induced denaturation. Specifically, 2.0% TP enhanced freeze-thawed protein solubility by 107.23%, reduced turbidity by

Hydrophobic Ion Pairing Improves Protein Thermal Stability and Lipid Nanocarrier Integration through Controlled Surface Modification.

Integrating therapeutic proteins into lipid-based nanocarriers remains challenging due to fundamental incompatibilities between hydrophilic protein surfaces and lipophilic carrier matrices. Here, we demonstrate that controlled hydrophobic ion pairing (HIP) using lysozyme-sodium dodecyl sulfate (SDS) at stoichiometric ratios simultaneously improves thermal stability during pharmaceutical processing and lipid compatibility for nanocarrier integration. Building on our previous molecular characterization of lysozyme-SDS complexes, we investigated their application in nanostructured lipid carrier (

Chitinase-3-like Protein 1 Reduces the Stability of Atherosclerotic Plaque via Impairing Macrophagic Efferocytosis.

CHI3L1 is strongly associated with atherosclerosis, but its role in macrophages remains unknown. In this study, we observed a significant up-regulation of CHI3L1 in both carotid plaques and serum of symptomatic patients, and demonstrated that CHI3L1 impairs the efferocytosis of macrophages by down-regulating crucial efferocytic mediator MFGE8 through inhibiting ATF2, which binds directly to the enhancer of MFGE8. In human plaques, we observed a negative correlation between CHI3L1 expression and both ATF2 and MFGE8 levels, further proved their involvement in plaque destabilization. Using Ldlr-/

High interstitial fluid pressure enhances USP1-dependent KIF11 protein stability to promote hepatocellular carcinoma progression.

BACKGROUND: HCC is characterized by a high interstitial fluid pressure (HIFP) environment, which appears to support cancer cell survival. However, the mechanisms behind this phenomenon are not fully understood. METHODS: This study investigates the role of kinesin family member 11 (KIF11) in HCC under HIFP conditions, using both in vivo and in vitro models. In vitro experiments replicated the HIFP environment to observe changes in HCC cell behavior and protein expression, while in vivo studies involved mice portal hypertension to create an orthotopic liver cancer model, allowing for the evalua

New Advances in Anti-HIV-1 Strategies Targeting the Assembly and Stability of Capsid Protein.

The HIV-1 capsid has emerged as a highly attractive drug target due to its highly conserved sequence and critical role in the viral life cycle. By disrupting interactions between capsid proteins and impairing the proper assembly or disassembly of the capsid, the inhibitors can effectively suppress HIV-1 replication and infection. Based on this mechanism, numerous small-molecule agents targeting the HIV-1 capsid protein have been developed to date. In this review, we report the latest advances in such inhibitors and delve into their molecular mechanisms of action. We find a focus on small molec

RNA/DNA-binding protein TDP43 regulates DNA mismatch repair genes with implications for genome stability.

TDP43 is an RNA/DNA-binding protein increasingly recognized for its role in neurodegenerative conditions, including amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). As characterized by its aberrant nuclear export and cytoplasmic aggregation, TDP43 proteinopathy is a hallmark feature in over 95% of ALS/FTD cases, leading to detrimental cytosolic aggregates and a reduction in nuclear functionality in neurons. Building on our prior work linking TDP43 proteinopathy to the accumulation of DNA double-strand breaks (DSBs) in neurons, the present investigation uncovers a novel re

Magnetic nanosystems modified with natural polyphenol for white wine stability via efficient haze protein removal.

Protein haze is a significant concern for the white wine industry, impacting sensory quality, stability, and consumer experience. The predominant treatment currently employed in production is bentonite, which leads to high costs and significant wine production losses, while also adsorbing certain aroma and flavor compounds. Therefore, there is a need for cost-effective and efficient methods that do not compromise the wine's flavor and aroma. In this study, spherical superparamagnetic iron oxide (SPIO) particles, with a diameter of 26.0 ± 4.1 nm, were synthesized via a microwave heating method,

A Novel SIL1 Variant (p.E342K) Associated with Marinesco-Sjögren Syndrome Impairs Protein Stability and Function.

Marinesco-Sjögren syndrome (MSS) is a rare autosomal recessive neuromuscular disorder marked by ataxia, muscle weakness, cataracts, and often intellectual and skeletal abnormalities. It is commonly caused by loss-of-function variants in the SIL1 gene, which impair binding immunoglobulin protein (BiP) function, leading to protein misfolding and activation of the unfolded protein response. In a 2-year-old patient with typical MSS symptoms, we identified a previously unreported c.1024G>A (p.E342K) variant in SIL1 via whole-exome sequencing. The pathogenicity of this Sil1 variant was supported by
