This study investigated the ethnomedicine Celastrus rosthornianus Lose. for its chemical constituents and anti-nonalcoholic fatty liver disease (NAFLD) potential. Phytochemical analysis led to the isolation of six diterpenoids (1-6), including two new structures (1 and 2). In a HepG2 cell model, compounds 2 and 6 significantly inhibited intracellular lipid accumulation. Mechanistic studies revealed that these active diterpenoids exert their anti-NAFLD effects by interfering with the maturation of the lipogenic transcription factor SREBP1, thereby suppressing the de novo lipogenesis (DNL) pathw

The international Covid19-NMR consortium aims at the comprehensive spectroscopic characterization of SARS-CoV-2 RNA elements and proteins and will provide NMR chemical shift assignments of the molecular components of this virus. The SARS-CoV-2 genome encodes approximately 30 different proteins. Four of these proteins are involved in forming the viral envelope or in the packaging of the RNA genome and are therefore called structural proteins. The other proteins fulfill a variety of functions during the viral life cycle and comprise the so-called non-structural proteins (nsps). Here, we report t

The snake venom protein flavoridin, a polypeptide of 70 amino acid residues, is a potent inhibitor of blood platelet aggregation. It binds to cell-surface integrin receptors such as the fibrinogen receptor glycoprotein IIb/IIIa. The inhibitory properties of flavoridin have been attributed to the tripeptide segment Arg-Gly-Asp (residues 49 to 51). This paper describes the determination of the three-dimensional structure of flavoridin in aqueous solution based on two-dimensional nuclear magnetic resonance spectroscopy. A family of 18 conformers was selected to characterize the solution structure

Natural silk produced by silkworms and some arthropod spiders stands out as a traditional protein polymer. Fibroin and sericin, which are silk proteins obtained from cocoons, have been used in the medical field for centuries. This study is the first to evaluate the physicochemical properties and biological activities of these proteins, which were isolated from the boxwood moth (Cydalima perspectalis), as a biotechnological material. This followed this separation of the fibroin and sericin proteins obtained from the cocoons using a high-temperature and high-pressure method. The morphological, t

The mouse brachypodism locus encodes a bone morphogenetic protein (BMP)-like molecule called growth/differentiation factor 5 (GDF5). Here we show that Gdf5 transcripts are expressed in a striking pattern of transverse stripes within many skeletal precursors in the developing limb. The number, location and time of appearance of these stripes corresponds to the sites where joints will later form between skeletal elements. Null mutations in Gdf5 disrupt the formation of more than 30% of the synovial joints in the limb, leading to complete or partial fusions between particular skeletal elements, a

Insects have developed elaborate fibrillar (or hairy) attachment systems that allow them to attach reliably on a variety of different and unpredictable surfaces. These hairy adhesive pads consist of fine and long surface outgrowths (setae), terminated by thin plate-like tips of different shapes. Besides structural adaptations, recent work revealed material gradients along the length of the setae with spatula-shaped and pointed tip structures. It was shown that these setae have a rigid base and soft setal tips, which is assumed to enhance the adaptability to rough surfaces and prevent clusterin

Methicillin-resistant Staphylococcus aureus (MRSA) strains show strain-to-strain variation in resistance level, in genetic background, and also in the structure of the chromosomal cassette (SCCmec) that carries the resistance gene mecA. In contrast, strain-to-strain variation in the sequence of the mecA determinant was found to be much more limited among MRSA isolates examined so far. The first exception to this came with the recent identification of MRSA strain LGA251, which carries a new homolog of this gene together with regulatory elements mecI/mecR that also have novel, highly divergent s

The leucine-rich repeat kinase 2 (LRRK2) protein has been genetically and functionally linked to Parkinson's disease (PD), a disabling and progressive neurodegenerative disorder whose current therapies are limited in scope and efficacy. In this report, we describe a rigorous hit-to-lead optimization campaign supported by structural enablement, which culminated in the discovery of brain-penetrant, candidate-quality molecules as represented by compounds 22 and 24. These compounds exhibit remarkable selectivity against the kinome and offer good oral bioavailability and low projected human doses.

Nearly identical, repetitive elements in the genome contribute to the variability in genetic inheritance patterns, particularly in regions like the RCCX locus, where such repeats can lead to structural variations. In addition, during the formation of gametes as a result of meiosis, variants of loci with repetitive elements that do not code for the required proteins may occur. As a result, an individual with certain genetic rearrangements in this region may have an increased risk of developing a congenital disorder, particularly in cases where the non-functional allele is inherited dominantly.

Wolbachia are obligate intracellular bacteria that occur in insects and filarial worms. Strains that infect insects have genomes that encode mobile genetic elements, including diverse lambda-like prophages called Phage WO. Phage WO packages an approximately 65 kb viral genome that includes a unique eukaryotic association module, or EAM, that encodes unusually large proteins thought to mediate interactions between the bacterium, its virus, and the eukaryotic host cell. The Wolbachia supergroup B strain, wStri from the planthopper Laodelphax striatellus, produces phage-like particles that can be

At present, physical methods of chemical analysis are constantly improving providing large amount of data on elemental composition of organs and tissues. However, only few works describe the correlation (or the potential connection) between the general or local bioelemental imbalances and specific biochemical reactions that are involved in pathogenesis of certain diseases. This review describes primary open-angle glaucoma (POAG) - one of the most common ophthalmic diseases - in terms of elemental chemistry. The authors look into the impact that various subgroups of elements have on passive and

Heat shock protein 90 (Hsp90) is a central biological macromolecule and essential molecular chaperone in protozoan parasites, whose functional cycle relies on tightly regulated conformational dynamics and allosteric communication between domains. Despite its relevance as an antiparasitic target, the high conservation of Hsp90 across species complicates the identification of selective regulatory mechanisms. Here, we investigate the allosteric regulation of Leishmania braziliensis Hsp90 (LbHsp90) using a celastrol-derived modulator, named SS2, as a mechanistic probe. We identify a previously unr

Sjögren's disease (SjD) is marked by dysfunction of the salivary gland (SG) caused by epithelial cell death. However, the mechanism remains unclear. Here we discovered that NRIP1 was abnormally upregulated in SjD and formed a protein complex with estrogen receptor α (ERα) to inhibit saliva secretion and lead to epithelial cell death. NRIP1 interacted with ERα and altered the estradiol (E2)-ERα downstream signal in the SG epithelium. In the context of SjD, NRIP1-ERα suppressed aquaporin-5 (AQP5) expression and promoted MYC expression. The NRIP1-ERα complex bound to the estrogen response element

Alpha haemoglobin stabilising protein (AHSP) binds free alpha-globin chains and plays an important role in the protection of red cells, particularly during beta-thalassaemia. Murine ASHP was discovered as a GATA-1 target gene and human AHSP is directly regulated by GATA-1. More recently, AHSP was rediscovered as a highly erythroid Kruppel-like factor (EKLF) -dependent transcript. We have determined the organisation of the murine AHSP gene and compared it to orthologs. There are two CACC box elements in the proximal promoter. The proximal element is absolutely conserved, but does not bind EKLF

BACKGROUND: Infection with hepatitis B virus (HBV) is major public health concern. The limitations of available antiviral drugs require development of novel approaches to inhibit HBV replication. This study was conducted to identify functional elements and new siRNA target sites within the highly conserved regions of the 533 base post-transcriptional regulatory element (PRE) of HBV RNAs.
RESULTS: Computational analysis of the PRE sequence revealed several conserved regulatory elements that are predicted to form local secondary structures some of these within known regulatory regions. A deleti

Babesia divergens, a zoonotic tick-borne pathogen, causes bovine and human babesiosis in Europe. The Elongation Factor 1 alpha (EF-1α) protein is important in many cellular processes and has emerged as a possible target for subunit vaccine development against parasitic infections, and its intergenic region (IG) is an important tool for genetic manipulation of Babesia parasites. While the EF-1α locus of B. divergens has been described, structural variation between isolates was poorly defined. In order to fill this gap, we performed a comparative analysis of the EF-1α-IG in B. divergens human (R

The NUCLEAR FACTOR Y (NF-Y) transcription factor family plays a crucial regulatory role in various aspects of plant development, physiological responses, and light signaling pathways. Nevertheless, there are limited reports on the characteristics and functions of NF-Ys in orchids. This study identified the NF-Y gene family in four orchids (Gastrodia elata, Gastrodia menghaiensis, Platanthera guangdongensis, and Platanthera zijinensis) and analyzed their structural domains, physicochemical properties, phylogenetic relationships, gene collinearity, and the cis-elements in their promoter region.

Interruption of the renin-angiotensin-aldosterone system (RAAS) at different levels is target-organ protective in several disease states; however, complete blockade is unlikely to be achieved due to escape mechanisms whenever blockade is attempted, incomplete knowledge of the role of all elements of the RAAS, and lack of pharmacotherapy against some elements that have been shown to contribute to disease states. Aldosterone has been overlooked as a mediator of RAAS escape and a key factor in target-organ injury despite the use of available RAAS blockers. Aldosterone is thought to play a role in

Group B Streptococcus (GBS) is the leading cause of bacterial pneumonia, sepsis, and meningitis among neonates and an important cause of morbidity among pregnant women and immunocompromised adults. Invasive diseases due to GBS are attributed to the ability of the pathogen to translocate across human epithelial surfaces. The alpha C protein (ACP) has been identified as an invasin that plays a role in internalization and translocation of GBS across epithelial cells. The soluble N-terminal domain of ACP (NtACP) blocks the internalization of GBS. We determined the 1.86-A resolution crystal structu

UNLABELLED: Herpesviruses alternate between the latent and the lytic life cycle. Switching into the lytic life cycle is important for herpesviral replication and disease pathogenesis. Activation of a transcription factor replication and transcription activator (RTA) has been demonstrated to govern this switch in Kaposi's sarcoma-associated herpesvirus (KSHV). The protein encoded by open reading frame 49 from KSHV (ORF49KSHV) has been shown to upregulate lytic replication in KSHV by enhancing the activities of the RTA. We have solved the crystal structure of the ORF49KSHV protein to a resolutio
