Transcriptomic analysis of longitudinal Burkholderia pseudomallei infecting the cystic fibrosis lung.

The melioidosis bacterium, Burkholderia pseudomallei, is increasingly being recognised as a pathogen in patients with cystic fibrosis (CF). We have recently catalogued genome-wide variation of paired, isogenic B. pseudomallei isolates from seven Australasian CF cases, which were collected between 4 and 55 months apart. Here, we extend this investigation by documenting the transcriptomic changes in B. pseudomallei in five cases. Following growth in an artificial CF sputum medium, four of the five paired isolates exhibited significant differential gene expression (DE) that affected between 32 an

Bacterial and host factors affecting Pseudomonas aeruginosa colonization versus bacteremia in granulocytopenic patients.

In order to evaluate bacterial factors which might predispose to P. aeruginosa colonization or bacteremia in the granulocytopenic patient, 132 isolates recovered from 44 oncology patients were evaluated for antigenic serotype, iron correctable sensitivity to pooled human serum, antibiotic susceptibility, production of lecithinase, elastase, protease, gelatinase, pyocyanin and pyoverdin. Similarly, potential host factors, primarily total iron binding capacity, were evaluated in a subpopulation of acute leukemia patients composed of 13 control patients without P. aeruginosa cultured during their

A novel non-thermal hurdle strategy: Cold stress potentiated cold plasma to control Pseudomonas aeruginosa via integrated oxidative and metabolic disruption.

This study developed a synergistic sterilization strategy combining cold plasma (CP) and cold stress (CS) to control Pseudomonas aeruginosa (P. aeruginosa), a major spoilage bacterium in refrigerated foods. The optimized sequence of CS pretreatment (4 °C, 24 h) followed by CP treatment (50 kV, 180 s) achieved 88.90 ± 3.85% sterilization efficiency and induced irreversible cellular damage. The synergistic effect (CSCP24) initiated with CS-induced physiological sensitization, leading to severe membrane disruption (97.92% PI-positive cells), biomolecule leakage, and ultrastructural collapse. Marine Bacillus safensis DJ1 metabolites disrupt P. aeruginosa virulence: an integrated experimental and network pharmacology and dynamics approach.

Marine bioactive compounds are natural products known for diverse biological activities, including strong anti-quorum sensing (QS) properties. These compounds offer potential for developing antivirulence agents against pathogenic bacteria. Eighty-six bacterial isolates from the coastal region of Gujarat were screened for their ability to inhibit QS and virulence factors in Chromobacterium violaceum MK and Pseudomonas aeruginosa PA14. Isolate DKGJ2E6 (E6) (Bacillus safensis DJ1, identified by 16S rRNA sequencing) showed the strongest QS inhibitory activity and was selected for further analysis.

α-Mangostin attenuates Pseudomonas aeruginosa virulence by targeting PqsR and inhibiting PQS-mediated quorum sensing.

Due to the increasing global antibiotic resistance spread, novel antibacterial strategies targeting virulence factors instead of bacterial growth are clinically crucial. One promising approach involves interfering with bacterial quorum-sensing systems, which regulate virulence and enhance adaptability. To this end, exploring quorum sensing inhibitors from Chinese medicine monomers have gained attention. The key quorum-sensing systems of Pseudomonas aeruginosa (las, rhl, and pqs) control multiple virulence factors and contribute to pathogenicity. This study screened a library of Chinese medicin

Gain-of-function mutation in the sensor kinase CpxS modulates the antimicrobial resistance and virulence of Pseudomonas aeruginosa.

The CpxRA two-component system is a key regulator of envelope stress responses in Gram-negative bacteria. In this study, a gain-of-function mutant in Pseudomonas aeruginosa was generated by truncating the periplasmic sensing domain (PSD) of the sensor kinase CpxS in PAO1 (PAO1::cpxSΔ32-147), leading to constitutive activation of its cognate response regulator CpxR. The mutant exhibited a 2- to 4-fold increase in antimicrobial resistance while showing significantly attenuated virulence traits: swimming and swarming motility decreased by 44% and 34%, respectively, and production of pyoverdine, r

Computational exploration and in vitro validation of anti-quorum sensing potential of phytochemicals from Coleus amboinicus.

Development of antimicrobial resistance in bacteria is a major concern. The study of quorum sensing (QS), the bacterial communication system that coordinates important events in an infection, opens a new avenue for treating infections. QS helps in biofilm formation which acts as a physical barrier and alters the microenvironment which traps the molecules or slows down the antibiotic diffusion making them less effective. Since the standard drugs are not very effective against bacteria anymore, demand for the discovery of other alternatives arises. Phytochemicals extracted from commonly used pla

HPLC-MS/MS-based detection of Pseudomonas aeruginosa secondary metabolites and their application in metabolomics.

An HPLC-MS/MS method was developed for determining Pseudomonas aeruginosa metabolites in culture media and cosmetics. The samples were dispersed in saturated sodium chloride, purified by C18-SPE after acetonitrile extraction, filtered through a PTFE membrane, and then analyzed. The mobile phase system used was 0.1 % formic acid-methanol. Positive and negative ions are scanned simultaneously, and multiple reaction monitoring (MRM) mode was used for detection. The results revealed that the linear correlation coefficients (r) of the 17 metabolic substances were all greater than 0.99 within the li

Multidrug-resistant Pseudomonas aeruginosa: Pathogenesis, resistance mechanisms, and novel therapeutic strategies.

Pseudomonas aeruginosa is a highly adaptable Gram-negative opportunistic pathogen and a major contributor to nosocomial infections, particularly in immunocompromised and critically ill patients. Its pathogenicity is mediated through an array of virulence determinants, including lipopolysaccharide (LPS), outer membrane proteins (OMPs), flagella, pili, and exopolysaccharides (alginate, Psl, Pel), which facilitate adhesion, immune evasion, and strong biofilm formation. The bacterium deploys an arsenal of secreted effectors such as exotoxins (ExoS, ExoT, ExoU, ExoY), pyocyanin, and elastases via s

Synergy Between Pseudomonas aeruginosa Filtrates And Voriconazole Against Aspergillus fumigatus Biofilm Is Less for Mucoid Isolates From Persons With Cystic Fibrosis.

Persons with cystic fibrosis (CF) frequently suffer from Pseudomonas aeruginosa and Aspergillus fumigatus co-infections. There is evidence that co-infections with these interacting pathogens cause airway inflammation and aggravate deterioration of lung function. We recently showed that P. aeruginosa laboratory isolates synergistically interact with the anti-fungal azole voriconazole (VCZ), inhibiting biofilm metabolism of several A. fumigatus laboratory strains. Interaction was usually mediated via pyoverdine, but also via pyocyanin or pyochelin. Here we used planktonic filtrates of 7 mucoid a

A scorpion venom peptide derivative BmKn‒22 with potent antibiofilm activity against Pseudomonas aeruginosa.

Pseudomonas aeruginosa is a leading cause of nosocomial and serious life-threatening infections and infections caused by this bacterium continue to pose a major medical challenge worldwide. The ability of P. aeruginosa to produce multiple virulence factors and in particular to form biofilms makes this bacterium resistant to all known antibiotics. As a consequence, standard antibiotic therapy are increasingly become ineffective to clear such infections associated with biofilms. In search for novel effective agents to combat P. aeruginosa biofilm infections, a series of the BmKn‒2 scorpion venom

The icmF3 locus is involved in multiple adaptation- and virulence-related characteristics in Pseudomonas aeruginosa PAO1.

The type VI secretion system (T6SS) is widely distributed in Gram-negative bacteria. Three separate T6SSs called H1-, H2-, and H3-T6SS have been discovered in Pseudomonas aeruginosa PAO1. Recent studies suggest that, in contrast to the H1-T6SS that targets prokaryotic cells, H2- and H3-T6SS are involved in interactions with both prokaryotic and eukaryotic cells. However, the detailed functions of T6SS components are still uncharacterized. The intracellular multiplication factor (IcmF) protein is conserved in type VI secretion systems (T6SS) of all different bacterial pathogens. Bioinformatic a

The puzzling construction of the conidial outer layer of Aspergillus fumigatus.

If the mycelium of Aspergillus fumigatus is very short-lived in the laboratory, conidia can survive for years. This survival capacity and extreme resistance to environmental insults is a major biological characteristic of this fungal species. Moreover, conidia, which easily reach the host alveola, are the infective propagules. Earlier studies have shown the role of some molecules of the outer conidial layer in protecting the fungus against the host defense. The outer layer of the conidial cell wall, directly in contact with the host cells, consists of α-(1,3)-glucan, melanin, and proteinaceous

Response to photo-oxidative stress of Pseudomonas aeruginosa PAO1 mutants impaired in different functions.

Clinicians often have to deal with infections that are difficult to control because they are caused by superbugs resistant to many antibiotics. Alternatives to antibiotic treatment include antimicrobial photodynamic therapy (aPDT). The photodynamic process causes bacterial death, inducing oxidative stress through the photoactivation of photosensitizer molecules in the presence of oxygen. No PDT-resistant bacteria have been selected to date, thus the response to photo-oxidative stress in non-phototrophic bacteria needs further investigation. The opportunistic pathogen Pseudomonas aeruginosa, in

Biogenic chitosan-encapsulated Cu/ZnO nanocomposite surface coating inhibits MDR Pseudomonas aeruginosa biofilm establishment on orthopedic implants.

Orthopedic implant-associated infections (IAIs), predominantly caused by Pseudomonas aeruginosa, represent a major clinical challenge due to antibiotic resistance and biofilm formation, which together contribute to their chronic nature. This research presents a biogenic biofilm-resistant chitosan encapsulated Cu/ZnO nanocoating (CS-Cu/ZnO NC) synthesized using Euphorbia thymofolia metabolites and applied on orthopedic implant surface (Tubular bone locked stainless steel plates). The synthesized CS-Cu/ZnO NCs were characterized physicochemically (UV-Vis, FTIR, XRD, SEM) and biologically (antiba

Role of cell-associated and secretion-independent virulence factors of Pseudomonas aeruginosa in microbial keratitis and anti-virulence therapeutic strategies.

PURPOSE: Microbial keratitis (MK) caused by Pseudomonas aeruginosa is a rapidly progressive and vision-threatening corneal infection worldwide. Even short delays in diagnosis or inappropriate therapy can lead to irreversible corneal scarring and blindness. This opportunistic pathogen colonizes, invades, and damages ocular tissue by taking advantage of disruptions in the corneal epithelial barrier and using a variety of virulence factors. The aim of this study was to summarize the current understanding of P. aeruginosa’s cell-associated, and secretion system-independent virulence factors involv
