Effects of ultraviolet laser radiation on Venezuelan equine encephalomyelitis virus.

The effects of usual low-intensity continuous (lambda = 254 nm, I = 10 W/m2) UV radiation and high-intensity laser nanosecond (lambda = 266 nm, tau p = 10 ns, I = 10(9) W/m2) or picosecond (lambda = 266 nm, tau p = 23 ps, I = 10(12) W/m2) UV radiation on Venezuelan equine encephalomyelitis virus (a member of the Togaviridae family) were compared. The quantum yields of infectivity inactivation, pyrimidine dimer formation and RNA-protein crosslinking were determined.

Sunlight, Vitamin D, and Xeroderma Pigmentosum.

Sunlight, in particular UV-B radiation, is an important factor for endogenous vitamin D production as 80-90% of the required vitamin D needs to be photosynthesized in the skin. The active form of vitamin D, vitamin D3 or calcitriol, binds to the ligand-activated transcription factor vitamin D receptor (VDR) for genomic and non-genomic effects. Recently, calcitriol and analogs have been shown to have antiproliferative effects in mouse and human BCC and SCC cell lines in vitro. As UV radiation plays a critical role in the photosynthesis of vitamin D, stringent sun protection, as recommended for

Structural insight into π-stacking interactions of non-aromatic oxazolo[5,4-d]pyrimidine derivatives and their neuroactivity in a Galleria mellonella model.

In this study, the physicochemical characteristics, quantum-chemical properties, and neurobiological activity of 6-N-benzyloxazolo[5,4-d]pyrimidin-7(6H)-imine derivatives 1-9 were investigated. Solid-state structural analysis using single-crystal X-ray diffraction (XRD) revealed intermolecular π-stacking interactions involving the formally non-aromatic oxazolo[5,4-d]pyrimidine scaffold. Energy decomposition analysis (EDA) showed that the tested crystallographic dimers are energetically stabilizing, with dispersion representing the largest attractive contribution throughout the series. Compleme

Low amounts of the DNA repair XPA protein are sufficient to recover UV-resistance.

DNA integrity is threatened by the damaging effects of physical and chemical agents that can affect its function. Nucleotide excision repair (NER) is one of the most known and flexible mechanisms of DNA repair. This mechanism can recognize and remove damages causing DNA double-helix distortion, including the cyclobutane pyrimidine dimers (CPDs) and the pyrimidine-pyrimidone (6-4) photoproducts, promoted by ultraviolet light (UV). The human syndrome xeroderma pigmentosum (XP) is clinically characterized chiefly by the early onset of severe photosensitivity of the exposed regions of the skin, a

Defective global genome repair in XPC mice is associated with skin cancer susceptibility but not with sensitivity to UVB induced erythema and edema.

It is generally presumed that xeroderma pigmentosum (XP) patients are extremely sensitive to developing UV erythema, and that they have a more than 1000-fold increased skin cancer risk. Recently established mouse models for XP can be employed to investigate the mechanism of these increased susceptibilities. In line with human data, both XPA and XPC knockout mice have been shown to have an increased susceptibility to UVB induced squamous cell carcinomas. In XPA knockouts, nucleotide excision repair of UV induced DNA photolesions is completely defective (i.e., both global genome repair and trans

Restricted repair of aflatoxin B1 induced damage in alpha DNA of monkey cells.

We have investigated the processing of adducts formed by covalent binding of aflatoxin B1 (AFB1) to DNA in confluent cultures of African green monkey cells. Repair synthesis elicited by AFB1 adducts was deficient in alpha DNA sequences compared to that in bulk DNA, although the initial levels of modification were the same for these DNAs. The removal of the primary initial adduct, AFB1-N7-Guanine, was deficient in alpha DNA and the kinetics of its loss resembled those previously reported for removal from total DNA in xeroderma pigmentosum cells of complementation group A. Spontaneous loss of th

Unscheduled DNA synthesis in xeroderma pigmentosum cells after microinjection of yeast photoreactivating enzyme.

Photoreactivating enzyme (PRE) from yeast causes a light-dependent reduction of UV-induced unscheduled DNA synthesis (UDS) when injected into the cytoplasm of repair-proficient human fibroblasts (Zwetsloot et al., 1985). This result indicates that the exogenous PRE monomerizes UV-induced dimers in these cells competing with the endogenous excision repair. In this paper we present the results of the injection of yeast PRE on (residual) UDS in fibroblasts from different excision-deficient XP-strains representing complementation groups A, C, D, E, F, H and I (all displaying more than 10% of the U

DNA repair in human/embryonic chick heterokaryons. Ability of each species to aid the other in the removal of ultraviolet-induced damage.

Cultured human and embryonic chick fibroblasts possess different enzyme-mediated processes to repair cyclobutyl pyrimidine dimers induced in their deoxyribonucleic acid (DNA) by ultraviolet (UV) radiation. While dimers are corrected in human cells by excision repair, a photoenzymatic repair process exists in embryonic chick cells for the removal of these potentially deleterious UV photoproducts. We have utilized a sensitive enzymatic assay to monitor the disappearance, i.e. repair, of dimer-containing sites in fused populations of human and chick cells primarily consisting of multinucleate hum

Stereodynamics of Hemiacetal Formation in Pyridine-3-Carbaldehyde and Pyrimidine-5-Carbaldehyde Derivatives: Implications for the Soai Reaction.

The Soai reaction is a unique example of self-amplifying asymmetric autocatalysis, in which a chiral zinc alkoxide not only promotes its own formation from pyridine-3-carbaldehydes and pyrimidine-5-carbaldehydes by alkylation with diisopropylzinc but also amplifies the enantiomeric excess of the added nonracemic alcohol. Both the asymmetric autocatalysis and the positive nonlinear effect have been attributed to equilibria involving the zinc alkoxide product, including dimerization and hemiacetalate formation with the substrate aldehyde. Hemiacetal formation is therefore of central importance f

CARM1/PRMT4 facilitates XPF-ERCC1 heterodimer assembly and maintains nucleotide excision repair activity.

The structure-specific endonuclease, XPF-ERCC1, plays a central role in DNA damage repair. This nuclease is known to be important for nucleotide excision repair, interstrand crosslink repair, and DNA double-strand repair. We found that the arginine methyltransferase, CARM1/PRMT4, is essential for XPF stabilization and maintenance of intracellular protein levels. Loss of CARM1 results in a decrease in XPF protein levels and a concomitant decrease in ERCC1 protein. A similar destabilization of XPF protein was observed in cells expressing a mutant in which XPF arginine 568 was replaced by lysine.

UV Protection in the Cornea: Failure and Rescue.

Ultraviolet (UV) irradiation induces DNA lesions in all directly exposed tissues. In the human body, two tissues are chronically exposed to UV: the skin and the cornea. The most frequent UV-induced DNA lesions are cyclobutane pyrimidine dimers (CPDs) that can lead to apoptosis or induce tumorigenesis. Lacking the protective pigmentation of the skin, the transparent cornea is particularly dependent on nucleotide excision repair (NER) to remove UV-induced DNA lesions. The DNA damage response also triggers intracellular autophagy mechanisms to remove damaged material in the cornea; these mechanis

Photoprotection of DNA (in vitro) by acyclothymidine dinucleosides.

Excessive exposure to sunlight is primarily implicated in ultraviolet (UV) induced skin cancers worldwide. Direct absorption of UV radiation by DNA leads to the formation of cyclobutane pyrimidine dimers (CPDs) resulting in DNA damage. The molecular mechanisms involved in the mutagenicity of CPDs are well established. Photoprotection of the skin from the detrimental effects of UV is essential in preventing skin damage. A variety of formulations, which essentially contain UV filters have been used as photoprotective agents of the skin. These comprise aromatic and inorganic molecules, whose mech

Defective transcription-coupled repair in Cockayne syndrome B mice is associated with skin cancer predisposition.

A mouse model for the nucleotide excision repair disorder Cockayne syndrome (CS) was generated by mimicking a truncation in the CSB(ERCC6) gene of a CS-B patient. CSB-deficient mice exhibit all of the CS repair characteristics: ultraviolet (UV) sensitivity, inactivation of transcription-coupled repair, unaffected global genome repair, and inability to resume RNA synthesis after UV exposure. Other CS features thought to involve the functioning of basal transcription/repair factor TFIIH, such as growth failure and neurologic dysfunction, are present in mild form. In contrast to the human syndrom

In vitro complementation of xeroderma pigmentosum.

In vitro complementation is a powerful strategy for isolation and characterization of individual components of multienzyme biochemical pathways. Application of the method to elucidate DNA metabolic pathways in prokaryotes enabled the successful identification of pathways of DNA replication and repair. In practice the technique requires the availability of genetic mutants that display defective operation of a selected biochemical pathway, and an in vitro assay system that allows detection of effective operation of the pathway. Xeroderma pigmentosum is a human disease syndrome characterized by p

UV-B Inhibition of Phytochrome-Mediated Anthocyanin Formation in Sinapis alba L. Cotyledons : Action Spectrum and the Role of Photoreactivation.

An action spectrum was measured for ultraviolet (UV) radiation-induced damage to (inhibition of) phytochrome-induced anthocyanin formation in cotyledons of 40-hour-old Sinapis alba L. seedlings. The action spectrum showed maximum effectiveness in the 260 to 280 nanometer waveband with little effect above 295 nanometers. The damaging effect of UV could be photorepaired by subsequent exposure to sunlight or to long wavelength (360 nanometers) UV radiation. Because this form of damage is subject to photorepair (photoreactivation), it is probably due to the formation of pyrimidine dimers, and the

Differential repair of the two major UV-induced photolesions in trichothiodystrophy fibroblasts.

Defects in nucleotide excision repair have been shown to be associated with the photosensitive form of the disorder trichothiodystrophy (TTD). Most repair-deficient TTD patients are mutated in the XPD gene, a subunit of the transcription factor TFIIH. Knowledge of the kinetics and efficiency of repair of the two major UV-induced photolesions in TTD is critical to understand the role of unrepaired lesions in the process of carcinogenesis and explain the absence of enhanced skin cancer incidence in TTD patients contrarily to the xeroderma pigmentosum D patients. In this study, we used different

A physical sunscreen protects engineered human skin against artificial solar ultraviolet radiation-induced tissue and DNA damage.

Sunscreens are known to-protect against sunlight-induced erythema and sunburn, but their efficiency at protecting against skin cancer is still a matter of debate. Specifically, the capacity of physical sunscreens to prevent or reduce tissue and DNA damage has not been thoroughly investigated. Our objective was to assess the ability of a broad-spectrum sunscreen containing TiO2 to protect human skin against tissue and DNA damage following ultraviolet radiation. For this purpose, engineered human skin (EHS) was generated and either treated or not treated with an SPF 28 physical sunscreen and exp

Epidermal proliferation but not quantity of DNA photodamage is correlated with UV-induced mouse skin carcinogenesis.

The hairless SKH-1 mouse strain has a higher skin tumor incidence, shorter tumor latency, and higher tumor yield in response to ultraviolet (UV) irradiation than the SENCAR strain. In this study we assessed the differences in UV susceptibility of both strains by measuring DNA photodamage and epidermal proliferation after one UV treatment and after 1, 3, 6, and 9 wk of chronic UV irradiation. Induction rates for cyclobutane pyrimidine dimers (CPDs) and pyrimidine (6-4) pyrimidone photoproducts [(6-4) PDs] were significantly greater in the SKH-1 strain than the SENCAR strain, but no strain diffe

Diagnosis of Xeroderma pigmentosum variant in a young patient with two novel mutations in the POLH gene.

We describe the characterization of Xeroderma Pigmentosum variant (XPV) in a young Caucasian patient with phototype I, who exhibited a high sensitivity to sunburn and multiple cutaneous tumors at the age of 15 years. Two novel mutations in the POLH gene, which encodes the translesion DNA polymerase η, with loss of function due to two independent exon skippings, are reported to be associated as a compound heterozygous state in the patient. Western blot analysis performed on proteins from dermal fibroblasts derived from the patient and analysis of the mutation spectrum on immunoglobulin genes pr
