IL-1R/TLR signaling plays a significant role in sensing harmful foreign pathogens and mounting effective innate and adaptive immune responses. However, the precise mechanism by which Leishmania donovani, an obligate intramacrophagic pathogen, breaches IL-1R/TLR signaling and host-protective immunity remains obscure. In this study, we report the novel biphasic role of Toll-interacting protein (Tollip), a negative regulator of the IL-1R/TLR pathway, in the disease progression of experimental visceral leishmaniasis. We observed that during early hours of infection, L. donovani induced phosphoryla

Transdifferentiation of quiescent dermal fibroblasts to secretory myofibroblasts has a central role in wound healing and pathological scar formation. This myofibroblast transdifferentiation process involves TGFβ-induced de novo synthesis of alpha smooth muscle cell actin (αSMA)+ fibers that enhance contractility as well as increased expression of extracellular matrix (ECM) proteins, including collagen and fibronectin. These processes are mediated upstream by the reactive oxygen species (ROS)-producing enzyme Nox4, whose induction by TGFβ is incompletely understood. In this study, we demonstrat

Rheumatoid arthritis (RA) is an autoimmune disease characterized by synovial inflammation and progressive joint destruction. Existing evidence indicates that hypoxia potentially contributes to the pathology of RA, though the specific mechanism remains unidentified. In this study, we explored the molecular mechanism through which the hypoxia-inducible factor (HIF-1α) contributed to the pathological process of RA. Our preliminary results suggested that hypoxia stimulates the activation of fibroblast-like synoviocytes (FLS) by inducing mitochondrial damage to activate cGAS-STING signaling, which

SUMO-specific protease 3 (SENP3) was reported to be dysregulated in osteosarcoma and ovarian cancer. However, the role of SENP3 in carcinogenesis of papillary thyroid cancer (PTC) has not been defined. Herein, we found that the expression of SENP3 protein was upregulated in PTC tissues and cell lines. Then, a PTC cell line, K-1 cells were transfected with SENP3 overexpression plasmid (pcDNA-SENP3) or small interfering RNA targeting SENP3 (SENP3 siRNA). We found that SENP3 overexpression facilitated cell proliferation and invasion, induced cell cycle arrest at G0/G1 phase and inhibited apoptosi

Lung adenocarcinoma (LUAD) is the major histological subtype of non-small cell lung cancer (NSCLC). The mechanistic target of rapamycin complex 1 (mTORC1), a master regulator of anabolic growth and metabolism, has been implicated in unfavorable prognosis and therapeutic resistance in LUAD. Nevertheless, how this association is mechanistically established remains insufficiently understood. Here, by integrating TCGA/GEO datasets with our institutional LUAD single-cell RNA-seq, we identified the atypical E2F factor E2F8 as the member most closely associated with the mTORC1 pathway and found that

BACKGROUND: Valproic acid (VPA) is widely used to treat epilepsy and bipolar disorder and is also a potent teratogen, but its teratogenic mechanisms are unknown. We have attempted to describe a fundamental role of the Polycomb group (Pc-G) in VPA-induced transformations of the axial skeleton.
METHODS: Pregnant NMRI mice were given a single subcutaneous injection of vehicle or VPA (800 mg/kg) on gestation day (GD) 8. The expression of genes encoding Polycomb and trithorax groups was measured by quantitative real-time RT-PCR using total RNA isolated from the embryos exposed to vehicle or VPA fo

Flowering is a key ornamental trait in chrysanthemum and a major agronomic trait affecting rice yield. Strigolactones (SLs) are a class of phytohormones that regulate diverse aspects of plant growth and development, including flowering time. However, the molecular mechanisms underlying SL-mediated regulation of flowering remain poorly understood. Here, we show that SL promotes flowering in chrysanthemum under inductive short-day conditions. The SL-specific signaling complex CmMAX2-CmD14.1-CmD53 was first identified in chrysanthemum. Silencing CmD14.1 or CmMAX2 causes late-flowering and SL inse

Duchenne muscular dystrophy (DMD) is characterized by progressive muscle wasting following repeated muscle damage and inadequate regeneration. Impaired myogenesis and differentiation play a major role in DMD as well as intracellular calcium (Ca2+) mishandling. Ca2+ release from the sarcoplasmic reticulum is mostly mediated by the type 1 ryanodine receptor (RYR1) that is required for skeletal muscle differentiation in animals. The study objective was to determine whether altered RYR1-mediated Ca2+ release contributes to myogenic differentiation impairment in DMD patients. The comparison of prim

BACKGROUND AND AIMS: Ischemia-reperfusion injury (IRI) represents a risk factor in liver transplantation (LT). We have shown that overexpression of heme oxygenase-1 (HO-1) mitigates hepatic IRI in LT recipients. Here, we hypothesized that human antigen R (HuR), the stabilizer of adenylate-uridylate (AU)-rich mRNAs, is required for hepatoprotection in LT.
APPROACH AND RESULTS: In an experimental arm, HuR/HO-1 protein expression was correlated with hepatic IRI phenotype. In an in vitro inflammation mimic model of hepatic warm IRI, induction of HuR/HO-1 and cytoplasmic localization following cyt

Moritella viscosa is a bacterial pathogen causing winter-ulcer disease in Atlantic salmon. The lesions on affected fish lead to increased mortality, decreased fish welfare, and inferior meat quality in farmed salmon. MicroRNAs (miRNAs) are small non-coding RNAs involved in post-transcriptional regulation by guiding the miRNA-induced silencing complex to specific mRNA transcripts (target genes). The goal of this study was to identify miRNAs responding to Moritella viscosa in salmon by investigating miRNA expression in the head-kidney and the muscle/skin from lesion sites caused by the pathogen.

High-risk Human Papillomaviruses (HPV) has been found to be associated with carcinomas of the cervix, penis, vulva/vagina, anus, mouth and oro-pharynx. As the main tumorigenic effects of the HPV have been attributed to the expression of E6 and E7 genes, different gene therapy approaches have been directed to block their expression such as antisense oligonucleotides (ASO), ribozymes and small interfering RNAs. In order to develop a gene-specific therapy for HPV-related cancers, we investigated a potential therapeutic strategy of gene silencing activated under illumination. Our aim according to

Cytoreductive surgery is the treatment protocol for colorectal cancer. Nonetheless, a major medical challenge remains to fully eliminate malignant tumor cells, along with a number of complications such as peritoneal adhesion and tumor peritoneal metastasis. The occurrence of peritoneal adhesions compromises not only the ability to do subsequent surgery, but also the efficacy of adjunct chemotherapy. More and more evidence suggest that the process of mesothelial-mesenchymal transition (MMT) influenced by transforming growth factor-β1 (TGF-β1) has a role to play in these disturbances, therefore

Begomoviruses, the largest genus of the family Geminiviridiae, are ssDNA viruses transmitted by whiteflies. Betasatellite molecules are occasionally found to be associated with begomoviruses and greatly influence viral pathogenesis. During viral invasion, plants activate a well-coordinated antiviral immune response to inhibit the propagation of the pathogen. In this study, we have reported an RNA-binding glycine-rich protein (RBGA) getting differentially expressed during infection due to begomovirus-betasatellite complex in Nicotiana benthamiana. Over-expression of the protein successfully res

Chlamydia trachomatis persistent infection is the leading cause of male prostatitis and female genital tract diseases. Inhibition of host cell apoptosis is the key to maintaining Chlamydia survival in vivo, and long noncoding RNAs (lncRNAs) play important roles in its developmental cycle and pathogenesis. However, it is not clear how lncRNAs regulate persistent Chlamydia infection. Here, using a microarray method, we identified 1718 lncRNAs and 1741 mRNAs differentially expressed in IFN-γ-induced persistent C. trachomatis infection. Subsequently, 10 upregulated and 5 downregulated differential

Verticillium wilt, caused by Verticillium dahliae Kleb., is a serious threat to cotton (Gossypium spp.) crop production. To enhance our understanding of the plant's complex defensive mechanism, we examined colonization patterns and interactions between V. dahliae and two cotton species, the resistant G. barbadense and the susceptible G. hirsutum. Microscopic examinations and grafting experiments showed that the progression of infection was restricted within G. barbadense. At all pre- and postinoculation sampling times, levels of salicylic acid (SA) were also higher in that species than in G. h

Bruton's tyrosine kinase (BTK) is a critical mediator of survival in B-cell neoplasms. Although BTK inhibitors have transformed therapy in chronic lymphocytic leukemia (CLL), patients with high-risk genetics are at risk for relapse and have a poor prognosis. Identification of novel therapeutic strategies for this group of patients is an urgent unmet clinical need, and therapies that target BTK via alternative mechanisms may fill this niche. Herein, we identify a set of microRNAs (miRs) that target BTK in primary CLL cells and show that the histone deacetylase (HDAC) repressor complex is recrui

AIMS/HYPOTHESIS: The proline-rich Akt substrate of 40 kDa (PRAS40) is a component of the mammalian target of rapamycin complex 1 (mTORC1) and among the most prominent Akt substrates in skeletal muscle. Yet the cellular functions of PRAS40 are incompletely defined. This study assessed the function of PRAS40 in insulin action in primary human skeletal muscle cells (hSkMC).
METHODS: Insulin action was examined in hSkMC following small interfering RNA-mediated silencing of PRAS40 (also known as AKT1S1) under normal conditions and following chemokine-induced insulin resistance.
RESULTS: PRAS40 kn

Although doxorubicin (DOX) is a common chemotherapeutic drug, the serious nephrotoxicity caused by DOX-induced renal fibrosis remains a considerable clinical problem. Tanshinone IIA (Tan IIA), a compound extracted from Salvia miltiorrhiza, has been reported to have an anti-fibrotic effect. Therefore, this study investigated the molecular pathway whereby Tan IIA protects the kidneys from DOX administration. DOX (3 mg/kg body weight) was intraperitoneally administered every 3 d for a total of 7 injections (cumulative dose of 21 mg/kg) to induce nephrotoxicity. Then, Tan IIA (5 or 10 mg/kg/d) was

Sepsis is characterized by dysregulated inflammation and coagulation leading to organ dysfunction, yet the molecular mechanisms linking these processes remain elusive. Through transcriptomic profiling of leukocytes from septic and non-septic patients, we identified Kruppel-like factor 7 intronic transcript 1 (KLF7-IT1) as one of the most robustly upregulated long non-coding RNAs in sepsis. Elevated KLF7-IT1 expression correlated strongly with hypercoagulability, inflammatory cytokines, and multi-organ injury, suggesting a pathogenic role in septic coagulopathy. Functional studies revealed that

BACKGROUND: Traumatic spinal cord injury (SCI) includes the primary insult as well as a sequela of biochemical and cellular cascades that amplifies the initial injury. This degenerative process, known as secondary injury, is often mediated by both reactive oxygen and nitrogen species released from damaged cells. Previous data suggests that dysregulated production of nitric oxide via inducible nitric oxide synthase (iNOS) is detrimental to spinal cord recovery. M1 macrophages have been implicated to overexpress iNOS post-SCI. In this work, we propose to inhibit iNOS expression through small int
