A cross-species assessment of behavioral flexibility in compulsive disorders.

Lack of behavioral flexibility has been proposed as one underlying cause of compulsions, defined as repetitive behaviors performed through rigid rituals. However, experimental evidence has proven inconsistent across human and animal models of compulsive-like behavior. In the present study, applying a similarly-designed reversal learning task in two different species, which share a common symptom of compulsivity (human OCD patients and Sapap3 KO mice), we found no consistent link between compulsive behaviors and lack of behavioral flexibility. However, we showed that a distinct subgroup of comp

Dopamine depletion effects on cognitive flexibility as modulated by tDCS of the dlPFC.

BACKGROUND: Recent evidence suggests that transcranial direct current stimulation (tDCS) may interact with the dopaminergic system to affect cognitive flexibility. Objective/hypotheses: We examined whether putative reduction of dopamine levels through the acute phenylalanine/tyrosine depletion (APTD) procedure and excitatory anodal tDCS of the dorsolateral prefrontal cortex (dlPFC) are causally related to cognitive flexibility as measured by task switching and reversal learning. METHOD: A double-blind, sham-controlled, randomised trial was conducted to test the effects of combining anodal tDC

Effects of chronic cocaine self-administration and N-acetylcysteine on learning, cognitive flexibility, and reinstatement in nonhuman primates.

RATIONALE: Cocaine use disorder (CUD) is associated with cognitive deficits that have been linked to poor treatment outcomes. An improved understanding of cocaine's deleterious effects on cognition may help optimize pharmacotherapies. Emerging evidence implicates abnormalities in glutamate neurotransmission in CUD and drugs that normalize glutamatergic homeostasis (e.g., N-acetylcysteine [NAC]) may attenuate CUD-related relapse behavior. OBJECTIVES: The present studies examined the impact of chronic cocaine exposure on touchscreen-based models of learning (repeated acquisition) and cognitive

Operant procedures for assessing behavioral flexibility in rats.

Executive functions consist of multiple high-level cognitive processes that drive rule generation and behavioral selection. An emergent property of these processes is the ability to adjust behavior in response to changes in one's environment (i.e., behavioral flexibility). These processes are essential to normal human behavior, and may be disrupted in diverse neuropsychiatric conditions, including schizophrenia, alcoholism, depression, stroke, and Alzheimer's disease. Understanding of the neurobiology of executive functions has been greatly advanced by the availability of animal tasks for asse

Stuttering Thoughts: Negative Self-Referent Thinking Is Less Sensitive to Aversive Outcomes in People with Higher Levels of Depressive Symptoms.

Learning theories of depression have proposed that depressive cognitions, such as negative thoughts with reference to oneself, can develop through a reinforcement learning mechanism. This negative self-reference is considered to be positively reinforced by rewarding experiences such as genuine support from others after negative self-disclosure, and negatively reinforced by avoidance of potential aversive situations. The learning account additionally predicts that negative self-reference would be maintained by an inability to adjust one's behavior when negative self-reference no longer leads to

Testing the disgust conditioning theory of food-avoidance in adolescents with recent onset anorexia nervosa.

Anorexia nervosa is characterized by chronic food avoidance that is resistant to change. Disgust conditioning offers one potential unexplored mechanism for explaining this behavioral disturbance because of its specific role in facilitating food avoidance in adaptive situations. A food based reversal learning paradigm was used to study response flexibility in 14 adolescent females with restricting subtype anorexia nervosa (AN-R) and 15 healthy control (HC) participants. Expectancy ratings were coded as a behavioral measure of flexibility and electromyography recordings from the levator labii (d

Cognitive deficits and CNS damage after a 4-day binge ethanol exposure in rats.

Impairments of learning and memory are common neuropsychological sequelae of chronic alcohol abuse. Alcoholics often have impairments of anterograde memory, including spatial memory dysfunction, and a tendency toward response perseveration. This study was designed to assess the effects of binge ethanol exposure on neurodegeneration and cognitive function. Rats were given ethanol three times daily for 4 days. Silver staining revealed neurodegeneration in the olfactory bulb, piriform cortex, perirhinal cortex, entorhinal cortex, and dentate gyrus. After withdrawal, behavioral testing in the Morr

Avoidance behavior in vitamin B-6 deficient rats.

Pyridoxine deficiency was induced during the postweaning period in infant and young adult male rats and the effects on active avoidance and escape learning were studied. Five weeks of pyridoxine deficiency was sufficient to produce a deficit in active avoidance learning in the postweanling animals, whereas up to 10 weeks of deficient diet ingestion produced no effect on young adult animals. Mild motor impairment was produced in the young pyridoxine-deficient rats but the avoidance learning deficit could not be explained away on this basis, because a deficit in passive avoidance was also produc

Impact of Short-term Rehabilitation on Chronic Stress-induced Brain Alterations and Associated Cognitive Deficits.

BACKGROUND: Persistent stress is frequently comorbid with major depressive disorder and anxiety, leading to significant psychological and behavioural impairments. However, the neurophysiological mechanisms underlying stress reversal remain poorly understood. Although several reversal strategies, including pharmacological antidepressants, brain stimulation therapies and nutritional interventions, have been explored, their outcomes remain inconsistent. PURPOSE: This study aimed to evaluate the effectiveness of rehabilitation as a non-pharmacological intervention for reversing stress-induced beh

A Study of the Frequency of Lead Reversal at a Tertiary Care Institution.

BACKGROUND: Electrocardiography (ECG) remains an important point-of-care investigative modality in the screening and diagnosis of cardiac diseases due to rapidity, cost-efficiency, and a gentle learning curve for most emergent patterns. Mechanistical problems in recording ECGs can lead to distorted graphical representations of cardiac electrophysiology, which can lead to incorrect or obstructed care for the patient. AIM: To identify the prevalence of ECG lead reversal in a tertiary care hospital over two months and to discuss the various patterns of ECG lead misplacement. METHODS: A total of

D2 receptors and cognitive flexibility in marmosets: tri-phasic dose-response effects of intra-striatal quinpirole on serial reversal performance.

Behavioral flexibility, which allows organisms to adapt their actions in response to environmental changes, is impaired in a number of neuropsychiatric conditions, including obsessive-compulsive disorder and addiction. Studies in human subjects and monkeys have reported correlations between individual differences in dopamine D2-type receptor (D2R) levels in the caudate nucleus and performance in a discrimination reversal task, in which established contingent relationships between abstract stimuli and rewards (or punishments) are reversed. Global genetic deletion of the D2R in mice disrupts rev

Stroke Induces a BDNF-Dependent Improvement in Cognitive Flexibility in Aged Mice.

Stroke remains a leading cause of disability worldwide. Recently, we have established an animal model of stroke that results in delayed impairment in spatial memory, allowing us to better investigate cognitive deficits. Young and aged brains show different recovery profiles after stroke; therefore, we assessed aged-related differences in poststroke cognition. As neurotrophic support diminishes with age, we also investigated the involvement of brain-derived neurotrophic factor (BDNF) in these differences. Young (3-6 months old) and aged (16-21 months old) mice were trained in operant touchscree

Cognitive flexibility, impulsivity, and compulsivity in problematic consumption behaviours.

Different studies have identified cognitive inflexibility, impulsivity and compulsivity in substance use and gambling disorder, but it is unclear whether these deficits are present in other possible addictive behaviours. In this study, we evaluated cognitive inflexibility, as measured by the Probabilistic Reversal Learning Task (PRLT), impulsivity across UPPS-P model and the compulsivity in regular gamblers, buyers, video gamers, consumers of pornography and control group. Multivariate analysis was performed to evaluate the five groups, revealing higher scores across several impulsivity and co

Compulsivity and Inhibitory Control Deficits in Abstinent Individuals With Heroin Addiction and Their Biological Siblings Compared With Unrelated Healthy Control Participants.

BACKGROUND: Compulsivity represents the performance of persistent and repetitive acts despite negative consequences and is considered one of the critical mechanisms for drug addiction. Although compulsivity-related neurocognitive impairments have been linked to addiction, it remains unclear whether these deficits might have predated drug abuse as potential familial susceptibilities. METHODS: A large sample of 213 adult participants were recruited, including 70 abstinent individuals addicted to heroin (HAs), 69 unaffected biological siblings of the HAs (siblings), and 74 unrelated healthy cont

The D2/D3-receptor antagonist tiapride impairs concurrent but not sequential taste aversion learning.

Taste aversion learning (TAL) is a learning modality in which the animals reject a gustatory stimulus associated with the administration of noxious visceral substances. This learning can be established by concurrent or sequential procedures that involve different anatomical and functional mechanisms and may constitute distinct learning modalities. The dopaminergic system has been related to various learning processes and goal-directed behaviours. The present study examined the effect of the administration of tiapride, a D(2)/D(3) dopaminergic antagonist, on concurrent and sequential TAL. Resul

An fMRI Pilot Study of Cognitive Flexibility in Trichotillomania.

Trichotillomania is a relatively common psychiatric condition, although its neurobiological basis is unknown. Abnormalities of flexible responding have been implicated in the pathophysiology of obsessive-compulsive disorder and thus may be relevant in trichotillomania. The purpose of this study was to probe reversal learning and attentional set-shifting in trichotillomania. Twelve adults with trichotillomania and 13 matched healthy control subjects undertook a functional MRI task of cognitive flexibility. Group-level activation maps for extradimensional and reversal switches were independently

Impaired consolidation of spatial memory during sleep in patients with leucine-rich glioma-inactivated 1-associated limbic encephalitis.

Sleep promotes the systems consolidation of hippocampus (HC)-dependent spatial memories by reprocessing of previously encoded hippocampal representations. Hippocampal reprocessing involves pattern separation and pattern completion as central hippocampal functions performed by the dentate gyrus (DG) and cornu ammonis region 3 (CA3), respectively. The leucine-rich, glioma inactivated 1 (LGI1)-associated limbic encephalitis (LE) is an autoimmune brain disorder particularly affecting the DG and CA3 regions, thereby impairing hippocampal function. We studied 15 LGI1 patients (and matched healthy co

Stress-related modulation of hippocampal long-term potentiation in rats: Involvement of adrenal steroid receptors.

Stress is usually correlated with an increased release of glucocorticoids from the adrenal glands. Within the hippocampus, a structure long known to be involved in spatial learning, two corticosterone-binding receptors are identified: the glucocorticoid receptor (GR) and the mineralocorticoid receptor (MR). Activation of these receptors impairs or facilitates hippocampal long-term potentiation (LTP), respectively. Stress elicited by behavioral manipulations may interfere with cognitive modulations of LTP during learning experiments. Here, we explore the influence of two stress-inducing procedu
