Monitoring the degradation of atropine and scopolamine in soil after spiking with naturally contaminated organic millet.

The spread of Datura sp. in European countries influences crop management and implies continuous food safety issues because of tropane alkaloids, atropine and scopolamine, the most relevant natural toxic compounds of this weed. These alkaloids can contaminate cereals to such a level that hampers food/feed related use and diverts batches of contaminated raw materials towards ultimate disposal such as burning. As no unambiguous information has been available on the fate of tropane alkaloids in soils, our study focused on the quantification and follow-up of these toxic residues in a soil experime

The context preexposure facilitation effect in mice: a dose-response analysis of pretraining scopolamine administration.

The context preexposure facilitation effect (CPFE) is an elaboration of contextual fear conditioning and refers to enhanced contextual conditioning resulting from preexposure to the context prior to a separate, brief context-shock episode. A version of the CPFE developed by Rudy and colleagues in rats has demonstrated greater sensitivity to pre-training hippocampal insult relative to standard contextual fear conditioning preparations. Our aim was to adapt the Rudy CPFE procedures to mice. In Experiment 1 we compared performance of young adult male C57BL6/J mice on two versions of the CPFE. One

Locomotion and stereotypy induced by scopolamine: contributions of muscarinic receptors near the pedunculopontine tegmental nucleus.

In this study, we test whether blockade of muscarinic receptors near mesopontine cholinergic cell groups may contribute to locomotor activation induced by scopolamine. Unilateral or bilateral injections of scopolamine (10-150 micrograms) into the pedunculopontine tegmental nucleus (PPT) increased horizontal locomotion by 2-15 times in a dose-related way. Unilateral or bilateral injections of scopolamine into the PPT increased stereotypic behaviors (such as sniffing in one location or over large areas), self-biting and grooming. Carbachol (4 micrograms) injected into PPT reduced locomotion for

Premedication with promethazine and transdermal scopolamine reduces the incidence of nausea and vomiting after intrathecal morphine.

Intrathecal morphine provides effective postoperative pain relief in major orthopaedic surgery. In use, however, is associated with unpleasant side effects like nausea and vomiting. The effect of different premedications on postoperative emetic sequelae induced by intrathecal morphine was studied in a prospective, double blind study. Sixty patients scheduled for arthroplasty surgery of the lower extremity were anaesthetized with spinal anaesthesia with a combination of isobaric bupivacaine 20 mg and morphine 0.3 mg. For premedication the patients were randomised to three groups of equal size.

A decrease in brain catecholamines prevents oxiracetam antagonism of the effects of scopolamine on memory and brain acetylcholine.

The effect of oxiracetam on passive avoidance conditioned response and acetylcholine (ACh) levels in rats with selective lesions of the central monoaminergic pathways was investigated. The lesions were followed by a marked decrease in cortical serotonin (-88%), noradrenaline (-54%) and striatal dopamine (-57%) levels, while neither the performance of a passive avoidance conditioned response nor brain ACh levels were affected. Scopolamine (hyoscine) administration (0.63 mg/kg, s.c.) to lesioned rats exerted the expected amnesic effect, associated with a decrease in hippocampal, cortical and str

Chin rub CRs are elicited by flavors associated with apomorphine, scopolamine, methscopolamine, physostigmine and neostigmine.

Three experiments were conducted in order to determine the pattern of behavioral conditioned responses (CRs) elicited by flavors paired with each of various drugs which effectively establish avoidance of a flavored solution. Each of the drugs employed supported both chin rub CRs and avoidance of a flavored solution. Experiment 1 employed apomorphine, a classic emetic agent which pharmacologically acts as a dopaminergic agonist. Experiment 2 and 3 employed cholinergic agonists and antagonists which were either peripherally or both peripherally and centrally acting agents. The results suggest th

Drug treatment of motion sickness: scopolamine alone and combined with ephedrine in real and simulated situations.

In two placebo-controlled, double-blind, randomized trials scopolamine (0.3 mg) alone or combined with ephedrine (25 mg) was tested for its effectiveness in the prevention of seasickness during 24 h at sea and of motion sickness in rotating chair tests in a laboratory. Scopolamine was effective both alone and in combination with ephedrine, which supports the hypothesis on central cholinergic overactivity in the pathogenesis of motion sickness. Ephedrine did not markedly increase the effectiveness of scopolamine. Side-effects were slight and did not disturb the operating ability of the voluntee

L-alpha-glycerylphosphorylcholine antagonizes scopolamine-induced amnesia and enhances hippocampal cholinergic transmission in the rat.

The effects of L-alpha-glycerylphosphorylcholine (alpha-GPC) on scopolamine-induced memory impairment and on brain acetylcholine (ACh) synthesis and release were investigated in rats. Oral administration of alpha-GPC 3 h before the behavioural test prevented the learning impairment induced by scopolamine given 30 min before the acquisition of a passive avoidance response. Similarly, retrograde amnesia induced by scopolamine, given immediately after acquisition training, was also completely reversed by the drug. These effects were dose-dependent with a maximum at 300 mg/kg. The mechanism of act

Transdermal scopolamine: a review of its effects upon motion sickness, psychological performance, and physiological functioning.

Scopolamine is the most effective single drug for the prophylaxis and treatment of motion sickness. However, oral or injected scopolamine displays a comparatively short duration of action (5-6 hours), and leads to deleterious side effects on autonomic and central nervous system cholinergic functions. The transdermal scopolamine system was designed to reduce these problems, but while it does deliver scopolamine over a prolonged time period (72 h), deleterious side effects are also produced. Transdermal scopolamine provides significant motion sickness protection, similar in extent to that provid

Gamma transcranial alternating current stimulation prevents scopolamine-induced damage to EEG activity and spatial memory.

BackgroundDementia is a serious public health concern, primarily caused by Alzheimer's disease (AD). Transcranial alternating current stimulation (tACS) can improve cognitive function by modulating neural oscillations. While tACS shows therapeutic potential for dementia, its preventive effects remain unknown. Because tACS has long-lasting modulatory effects, we propose that these offline effects may help prevent dementia.ObjectiveThis study aims to determine whether pre-administered 40 Hz tACS prevents scopolamine-induced impairment of electroencephalogram (EEG) activity and spatial cognition.

Scopolamine exerted antidepressant-like behavioral effects via restoring the synaptic plasticity in CRS-treated mice.

Depression stands as a widespread psychiatric illness, yet existing therapeutic approaches are constrained by delayed effectiveness and adverse reactions. Scopolamine, as a M-AChRs antagonist, has shown significant antidepressant-like effects in preclinical studies, but its mechanisms remain unclear. The present study is designed to explore the molecular mechanisms that underpin the antidepressant-like actions of scopolamine. Four-week chronic restraint stress (CRS) was employed to induce depression-like behaviors, while behavioral assessments—including TST (the tail suspension test) and FST (
