Secretome Profiling of Young Multipotent Stem Cells Reveals Angiogenic and Immunomodulatory Mechanisms Supporting Aged Neuromuscular Health.

Aging is the primary risk factor for many neuromuscular (NM) diseases that impair motor and cognitive function. Transplantation of young muscle-derived stem/progenitor cells (MDSPCs) has shown remarkable therapeutic potential across a range of age-related diseases, primarily through paracrine mechanisms. In this study, secretome profiling of young MDSPCs revealed a unique enrichment of pro-angiogenic and immunomodulatory proteins compared to their aged counterparts. Our systemic transplantation experiments also demonstrate that young MDSPCs activate biological pathways linked to these secreted

ADGRD1 promotes bladder cancer progression and angiogenesis via the PI3K/AKT/mTOR-mediated pro-angiogenic secretome.

BACKGROUND: Bladder cancer (BLCA) remains a major cause of cancer-related mortality, particularly in advanced and metastatic stages. Identifying molecular drivers of tumor aggressiveness is critical for developing targeted therapies. Adhesion G protein-coupled receptor D1 (ADGRD1, also known as GPR133) is an orphan aGPCR recently implicated in tumorigenesis. However, its role in bladder cancer and underlying signaling mechanisms remain unknown. METHODS: The clinical significance of ADGRD1 was analyzed using GEPIA3 datasets, focusing on clinical stage and prognosis. Stable ADGRD1-knockdown and

Therapy-induced senescence rewires the melanoma secretome to promote antitumor immune response and augment cell therapies.

BACKGROUND: Therapy-induced senescence (TIS) is a common outcome of diverse anticancer treatments, including chemotherapy, radiation, and small-molecule inhibitors. Senescence is characterized by stable growth arrest and the senescence-associated secretory phenotype (SASP), which includes various immune mediators. As the role of the immune system in controlling cancer becomes increasingly appreciated, understanding the impact of TIS on the tumor immune microenvironment (TIME) is critically important. Here, we investigated how senescence can be leveraged to enhance antitumor immune responses.

Stable, long-acting mesenchymal stromal cell secretome delivery via injectable starPEG-heparin hydrogels for sustained immunomodulation.

The secretome of mesenchymal stromal cells (MSCs) has emerged as a promising cell-free therapeutic strategy for regenerative medicine and immune modulation. However, its clinical translation remains limited by restricted stability and the short in vivo half-life of its bioactive factors, often requiring repeated administrations. Here, we describe an injectable star-shaped poly(ethylene glycol)-heparin (starPEG-heparin) hydrogel that enables the stable loading and sustained release (up to 14 days) of a lyophilized immunomodulatory MSC secretome (LS). Controlled delivery resulted from the interp

Severe hypoxia drives loss of ST6GAL1-mediated α2,6-sialylation in the epicardial secretome impairing angiogenic activity.

The epicardium contributes to cardiac development and regeneration, primarily through paracrine signalling. However, the impact of severe hypoxic stress, such as occurs immediately following myocardial infarction, on the activity of the epicardial secretome remains poorly defined. Here we investigated the angiogenic potential of the epicardial secretome under normoxic and hypoxic conditions. In contrast to expectation, the results demonstrate that the angiogenic effect of epicardial cells on vascular endothelium is hampered under severe hypoxia. Mechanistically, we found that hypoxia-mediated

Mesenchymal stem cell-derived secretome in traumatic brain injury: Stage-specific paracrine mechanisms and translational challenges.

Traumatic brain injury (TBI) is a stage-dependent disorder that evolves from acute neuroinflammation, blood-brain barrier (BBB) disruption, oxidative stress, excitotoxicity, and apoptosis to subacute neurovascular remodeling and chronic impairment of neuroregeneration and circuit repair. Current clinical management improves survival by stabilizing physiological status and limiting secondary injury, but it rarely rebuilds damaged neural networks or restores long-term neurological function. Mesenchymal stem cells (MSCs) have therefore attracted attention as regenerative candidates, with increasi

Adipose-Derived Mesenchymal Stem Cell Secretome for Post-Steroid Hypopigmentation and Skin Atrophy: A Case Report.

Skin hypopigmentation and atrophy are the most commonly reported local adverse effects following corticosteroid application. While these changes are typically reversible, the recovery may be delayed or incomplete in older adults due to diminished physiological reserves and age-related reductions in melanocyte function and dermal regenerative capacity. Adipose-derived mesenchymal stem cells (ADMSCs) secretome may offer substantial benefits in reversing the alterations. In this report, we present the clinical evaluation of an elderly patient with steroid-induced cutaneous manifestations receivin

Senescence and Hypoxia Regulate Colon Cancer Cell Transcriptome and Secretome: Insights into Cancer Cell Senescence Pathophysiology.

We investigated the effects of senescence and hypoxia on the transcriptome and secretome of the colon cancer cell, HCT-116, in an in vitro model. Senescence was confirmed using SA-β Gal staining and the expression of p53 and p21 proteins, and hypoxia using HIF-1α protein. Control (CN) and senescent (SN) cells were exposed to normoxia or hypoxia, control hypoxia (CH), and senescent hypoxia (SH). Senescence (SN, SH) enhanced the expression of kallikrein-related peptidases, TPp53, p21, optineurin, lipocalin, ADH-1, and stratifin by several folds. Stratifin, with tumor suppressive functions, was u

Targeted delivery of engineered adipose-derived stem cell secretome to promote cardiac repair after myocardial infarction.

Stem cell secretome offers a promising alternative to stem cell transplantation for treating myocardial infarction (MI). However, its clinical application faces two major challenges: how to enhance the levels of growth factors within the secretome to promote cardiac cell survival and vascularization, and how to efficiently deliver the secretome to the infarcted heart during the acute MI phase without risking rupture of the weakened myocardium. To address these challenges, we upregulated angiogenic growth factors in the secretome from adipose-derived stem cells (ADSC-secretome) by conditioning

Diverse potential of secretome from natural killer cells and monocyte-derived macrophages in activating stellate cells.

Chronic liver diseases, such as non-alcoholic steatohepatitis (NASH)-induced cirrhosis, are characterized by an increasing accumulation of stressed, damaged, or dying hepatocytes. Hepatocyte damage triggers the activation of resident immune cells, such as Kupffer cells (KC), as well as the recruitment of immune cells from the circulation toward areas of inflammation. After infiltration, monocytes differentiate into monocyte-derived macrophages (MoMF) which are functionally distinct from resident KC. We herein aim to compare the in vitro signatures of polarized macrophages and activated hepatic

Research progress and application prospect of adipose-derived stem cell secretome in diabetes foot ulcers healing.

Diabetic foot ulcers (DFUs) are chronic wounds and one of the most common complications of diabetes, imposing significant physical and mental burdens on patients due to their poor prognosis and treatment efficacy. Adipose-derived stem cells (ADSCs) have been proven to promote wound healing, with studies increasingly attributing these beneficial effects to their paracrine actions. Consequently, research on ADSC secretome as a novel and promising alternative for DFU treatment has been extensively conducted. This article provides a comprehensive review of the mechanisms underlying refractory DFU

Integration of the cancer cell secretome and transcriptome reveals potential noninvasive diagnostic markers for bladder cancer.

PURPOSE: Bladder cancer (BLCA) is a major cancer of the genitourinary system. Although cystoscopy is the standard protocol for diagnosing BLCA clinically, this procedure is invasive and expensive. Several urine-based markers for BLCA have been identified and investigated, but none has shown sufficient sensitivity and specificity. These observations underscore the importance of discovering novel BLCA biomarkers and developing a noninvasive method for detection of BLCA. Exploring the cancer secretome is a good starting point for the development of noninvasive biomarkers for cancer diagnosis. EX

Fibroblasts' secretome from calcified and non-calcified dermis in Pseudoxanthoma elasticum differently contributes to elastin calcification.

Pseudoxanthoma elasticum (PXE) is a rare disease characterized by ectopic calcification, however, despite the widely spread effect of pro/anti-calcifying systemic factors associated with this genetic metabolic condition, it is not known why elastic fibers in the same patient are mainly fragmented or highly mineralized in clinically unaffected (CUS) and affected (CAS) skin, respectively. Cellular morphology and secretome are investigated in vitro in CUS and CAS fibroblasts. Here we show that, compared to CUS, CAS fibroblasts exhibit: a) differently distributed and organized focal adhesions and

PAX8 modulates the tumor microenvironment of high grade serous ovarian cancer through changes in the secretome.

High grade serous ovarian cancer (HGSC) arises from the fimbriated end of the fallopian tube epithelium (FTE), and in some cases, the ovarian surface epithelium (OSE). PAX8 is a commonly used biomarker for HGSC and is expressed in ∼90% of HGSC. Although the OSE does not express PAX8, murine models of HGSC derived from the OSE acquire PAX8, suggesting that it is not only a marker of Müllerian origin, but also an essential part of cancer progression, potentially from both the OSE and FTE. Previously, we have shown that PAX8 loss in HGSC cells causes tumor cell death and reduces cell migration an

Therapeutic potential of mesenchymal stem/stromal cell-derived secretome and vesicles for lung injury and disease.

Introduction: The acute respiratory distress syndrome (ARDS) is a devastating clinical condition common in patients with respiratory failure. Based largely on numerous preclinical studies and recent Phase I/II clinical trials, administration of stem cells, specifically mesenchymal stem or stromal cells (MSC), as a therapeutic for acute lung injury (ALI) holds great promise. However, concern for the use of stem cells, specifically the risk of iatrogenic tumor formation, remains unresolved. Accumulating evidence now suggest that stem cell-derived conditioned medium (CM) and/or extracellular vesi

Quantitative Proteomics of the Endothelial Secretome Identifies RC0497 as Diagnostic of Acute Rickettsial Spotted Fever Infections.

Mediterranean spotted fever is a reemerging acute tick-borne infection produced by the α-proteobacterium, Rickettsia conorii. Rickettsia conorii infects vascular endothelial cells producing disseminated plasma leakage, manifesting as nonspecific fever, headache, and maculopapular rash. Because there are no available tests of early infection, Mediterranean spotted fever is often undiagnosed and untreated, resulting in significant mortality. To address this critical need, we have applied a quantitative proteomics pipeline for analyzing the secretome of primary human umbilical vein endothelial ce
