BACKGROUND: Domoic acid (DA) is a neurotoxic compound causing amnesic shellfish poisoning, secreted by red algae and diatoms. As a glutamate analogue, DA accumulates in filter-feeding marine organisms, posing significant health risks to humans upon consumption. Detecting DA in marine environments remains challenging due to its low concentration and interference from complex matrices. Effective detection and removal require materials with high efficiency and selectivity, which traditional inorganic ionic materials lack due to their limited adsorption capacity and selectivity. Toxic species of the dinoflagellate genus Dinophysis can produce diarrheic toxins including okadaic acid (OA) and dinophysistoxins (DTXs), and the non-diarrheic pectenotoxins (PTXs). Okadaic acid and DTXs cause diarrheic shellfish poisoning (DSP) in human consumers, and also cause cytotoxic, immunotoxic and genotoxic effects in a variety of mollusks and fishes at different life stages in vitro. The possible effects of co-produced PTXs or live cells of Dinophysis to aquatic organisms, however, are less understood. Effects on an early life stage of sheepshead minnow (Cyprinodon variegatus), a co

Ciguatera food poisoning affects consumer health and fisheries' economies worldwide in tropical zones, and specifically in the Pacific area. The wide variety of ciguatoxins bio-accumulated in fish or shellfish responsible for this neurological illness are produced by marine dinoflagellates of the genus Gambierdiscus and bio-transformed through the food web. The evaluation of the contents of ciguatoxins in strains of Gambierdiscus relies on the availability of standards and on the development of sensitive and specific tools to detect them. There is a need for sensitive methods for the analysis

Domoic acid (DA) is a neuroexcitatory amino acid that is naturally produced by some marine diatom species of the genus Pseudo-nitzschia. Ingestion of DA-contaminated seafood by humans results in a severe neurotoxic disease known as amnesic shellfish poisoning (ASP). Clinical signs of ASP include seizures and neuronal damage from activation of ionotropic glutamate receptors. However, the impacts of DA exposure at levels below those known to induce outward signs of neurobehavioral exicitotoxicity have not been well characterized. To further understand the mechanisms of neurotoxic injury associat

Chronic and subchronic toxicity following exposure to the DSP (Diarrhetic shellfish poisoning) toxin okadaic acid (OA) is receiving increasing attention as a public human health biohazard. However information on ecological impacts induced by proliferation of the OA producing dinoflagellate Prorocentrum is scarce. In order to analyse the toxicity of these substances, in vivo experiments were conducted on medaka fish (Oryzias latipes) embryos used as an experimental model. The study was focused on two strains of benthic Prorocentrum species, P. arenarium and P. emarginatum, naturally found in th

Okadaic acid (OA) is the main marine toxin implicated in the diarrhetic shellfish poisoning (DSP) in humans after consumption of contaminated bivalve molluscs. We have previously shown that OA was an in vitro aneugenic compound that induced chromosome loss via micronuclei formation in CHO-K1 cells. The aims of this study were to investigate the chromosomal non-disjunction (ND) potential of OA in human lymphocytes and the pathways involved for aneuploidy in CHO-K1 cells. Firstly, we analysed the formation of micronuclei and the non-disjunction for chromosomes 1 and 17 in binucleated human lymph

Data on foodborne disease in Canada in 1977 were compared with data for 1976. A total of 777 incidents, comprising 660 outbreaks and 117 single cases, causing illness in 4810 persons was reported for 1977. The number of incidents and cases decreased by 9.7% and 10.4%, respectively, from 1976 to 1977. Unlike previous years, Salmonella spp. were responsible for more incidents (32) and cases (763) than any other agent. Other incidents were caused by Staphylococcus aureus (23), Clostridium perfringens (14), suspect mold and yeast (13), Bacillus spp. (11), Clostridium botulinum (5), Shigella sonnei

In 2021, the U.S. Food and Drug Administration (FDA), Centers for Disease Control and Prevention (CDC), and state partners investigated a multistate sample-initiated retrospective outbreak investigation (SIROI) consisting of a cluster of nine Salmonella Weltevreden illnesses associated with frozen, precooked shrimp imported from India. Import surveillance testing identified Salmonella Weltevreden recovered from a cooked shrimp sample from Supplier B. In total, nine patients with clinical isolates highly related via whole genome sequencing were reported in four states with illness onset dates b

Data on foodborne disease in Canada in 1983 are compared with those for 1982. A total of 963 incidents, comprising 752 outbreaks and 211 single cases, caused illnesses in 5,955 persons in 1983. These figures are slightly less than for 1982, although the numbers of incidents and cases are the second highest on record. Salmonella , Staphylococcus aureus , Clostridium perfringens and Bacillus cereus caused most of the illnesses. The main Salmonella serovars involved were S. typhimurium , S. hadar , and S. agona . The small outbreaks of hepatitis A were attributed to food served at a school cafete

BACKGROUND: An assay that utilizes the relationship between the amount of toxin administered and the lethal time of mice is called a mouse bioassay (MBA) and has been used as an assay for marine neurotoxins such as paralytic shellfish poison toxins and pufferfish toxins. The MBA for tetrodotoxin (TTX), which is possessed by pufferfish, has been used for many years in Japan. In the Japanese reference method, equivalent to the official method, the toxicity of TTX is expressed in mouse units (M.U.), and 1 M.U. is defined as the amount of the toxins required to kill a 20 g mouse in 30 min and is s

Temporal lobe epilepsy with hippocampal sclerosis (TLE-HS) is one of the more common forms of chronic epilepsy. Its aetiology is unknown, though an early developmental insult is thought by some to be an important trigger. There is not a strong genetic predisposition; gene-environment interactions are more significant considerations. Environmental risk factors for TLE-HS are under-researched. Domoic acid (DA) is an environmental neurotoxin of algal origin that can contaminate marine food webs. DA can cross the placenta, is significantly more toxic to the developing brain compared to the adult b

Pinnatoxins and pteriatoxins are a group of cyclic imine toxins that have hitherto only been isolated from Japanese shellfish. As with other cyclic imine shellfish toxins, pinnatoxins cause rapid death in the mouse bioassay for lipophilic shellfish toxins, but there is no evidence directly linking these compounds to human illness. We have identified the known pinnatoxins A (1) and D (6), and the novel pinnatoxins E (7), F (8) and G (5), in a range of shellfish and environmental samples from Australia and New Zealand using LC-MS. After isolation from the digestive glands of Pacific oysters, the

The proliferation of harmful cyanobacterial algal blooms is of concern due to the associated release of toxins affecting ecosystems and human health. The paralytic shellfish poison saxitoxin (STX) is a small polar alkaloid that can occur in inland and marine aquatic environments. Here, we optimized a fast and sensitive analytical method for the determination of STX, neosaxitoxin (NeoSTX), and their decarbamoyl analogues in surface waters. The method involves a simple filtration, addition of isotope-labelled internal standard (ILIS), and analysis by on-line solid-phase extraction coupled to hyd

This review briefly describes the origin, chemistry, molecular mechanism of action, pharmacology, toxicology, and ecotoxicology of palytoxin and its analogues. Palytoxin and its analogues are produced by marine dinoflagellates. Palytoxin is also produced by Zoanthids (i.e. Palythoa), and Cyanobacteria (Trichodesmium). Palytoxin is a very large, non-proteinaceous molecule with a complex chemical structure having both lipophilic and hydrophilic moieties. Palytoxin is one of the most potent marine toxins with an LD50 of 150 ng/kg body weight in mice exposed intravenously. Pharmacological and elec

The highly toxic methyl-, ethyl- and phenylmercury species that may exist in the three main anatomical parts - the adductor muscle, the mantle and the visceral mass - of bay scallops (Argopecten irradias) were quantitatively released by cupric chloride, zinc acetate, sodium chloride and hydrochloric acid (HCl) under ultrasonic extraction. After centrifugation, the mercury species in the supernatant were concentrated by thio (SH)-bearing chelating resins, eluted with HClO4 and HCl and extracted with toluene. Separation was achieved by capillary GC equipped with programmed temperatures, a consta

Minamata disease is methylmercury poisoning from consuming fish and shellfish contaminated by industrial waste. The polluted seafood was widely consumed in the area around Minamata, but many individuals were never examined for or classified as having Minamata disease. Following the determination of the Supreme Court of Japan in October 2004 that the Japanese Government was responsible for spreading Minamata disease, over 13,000 residents came forward to be examined for Minamata disease. We studied 197 residents from the Minamata area who had a history of fish consumption during the polluted pe

PLTX (Palytoxin) is a fatty-chain polyether marine biotoxin originally produced by benthic dinoflagellates, particularly Ostreopsis ovata and Ostreopsis siamensis. Via the marine food web, PLTX bioaccumulates in numerous organisms such as corals and shellfish, and ranks among the most lethal non-proteinaceous natural compounds described to date. Thus far, its toxicological mechanism remains unclear, and no specific clinical antidote is available. In this study, we employed a limited proteolysis‑based chemical proteomics approach (Lip‑SMap) to analyze the protection of protease cleavage sites c

The dinoflagellate genus Alexandrium has been well known for causing paralytic shellfish poisoning (PSP) worldwide. Several non-PSP toxin-producing species, however, have shown to exhibit fish-killing toxicity. Here, we report the allelopathic activity of Alexandrium leei from Malaysia to other algal species, and its toxicity to finfish and zooplankton, via laboratory bioassays. Thirteen microalgal species that co-cultured with Al. leei revealed large variability in the allelopathic effects of Al. leei on the test algae, with the growth inhibition rates ranging from 0 to 100%. The negative all
