Generators and temporal succession of giant somatosensory evoked potentials in cortical reflex myoclonus: epicortical recording from sensorimotor cortex.

OBJECTIVE: To clarify the generator mechanism of giant somatosensory evoked potentials (giant SEPs) and the hyperexcitability of primary somatosensory and motor cortices (SI and MI). METHODS: In a patient with intractable focal seizures manifesting cortical reflex myoclonus of the left foot, giant SEPs to left tibial nerve stimulation were epicortically recorded as a part of presurgical evaluation with subdural electrodes. RESULTS: In the single pulse SEPs, enlarged P1-N1 components were observed at the foot area of the SI and MI (86.5-258.8 microV, respectively), and the peak latencies were

High-affinity uptake of GABA and glutamate decarboxylase activity in rat primary somatosensory cortex after sciatic nerve injury.

We have studied the changes in the GABAergic system in the rat somatosensory cortex 1-14 d after sensory deprivation of the hind-limb representation area. Glutamate decarboxylase (GAD) activity was measured in the individual cortical layers using serial sections cut on a freezing microtome parallel to the cortical surface. Gamma-aminobutyric acid (GABA) high-affinity uptake was studied in cortical homogenates of the hind-limb representation area. There was a less than or equal to 13% decrease in GAD activity in layer II-IV in both cortical hemispheres 3 d after sciatic nerve injury. In contras

LRP4+ Astrocytes: A Unique Subpopulation Crucial for Blood Vessel Maintenance and Function in the Somatosensory Cortex of Normal and 5xFAD Mice.

Astrocytes exhibit significant heterogeneity in morphology, molecular profiles, and function. While these cells perform diverse roles, their functional specialization across distinct subgroups remains poorly characterized. Lipoprotein receptor-related protein 4 (LRP4), a transmembrane receptor critical for agrin signaling for neuromuscular junction (NMJ) formation and maintenance, is selectively expressed in a subset of cortical and hippocampal astrocytes. However, the properties and functions of Lrp4+ astrocytes in the brain remain largely unknown. In this study, we investigated the distribut

Effects of transcranial ultrasound stimulation of primary somatosensory cortex and ventral posterolateral nucleus of the thalamus on acute pain perception.

Noninvasive neuromodulatory techniques provide important means to study the role of brain regions activated by nociceptive stimuli in pain perception. This study investigates the role of the primary somatosensory cortex (S1) and the ventral posterolateral nucleus (VPL) of the thalamus in acute pain perception using transcranial ultrasound stimulation (TUS). Twenty-five healthy participants underwent a double-blind, sham-controlled, within-subject experimental design. Transcranial ultrasound stimulation was applied to the left S1 and left VPL in separate sessions, with quantitative sensory test

Cortical reorganization following dorsal spinal injuries in newborn monkeys reveals a critical period in the development of the somatosensory cortex.

Lesions of the dorsal columns of the spinal cord in adult macaque monkeys lead to the loss of hand inputs and large-scale expansion of the face inputs in the hand region of the somatosensory cortex. Inputs from alternate spinal pathways do not reactivate the deafferented regions of area 3b. Here, we determined how transections of the dorsal columns done within a few days after birth affect the developing somatosensory cortex. Dorsal columns were transected between the 3rd and 12th postnatal day (PND), and the somatosensory cortex was mapped when the macaques were over 3 y old. There were two d

Longitudinal evaluation of the functional connectivity changes in the secondary somatosensory cortex (S2) of the monkey brain during acute stroke.

BACKGROUND: Somatosensory deficits are frequently seen in acute stroke patients and may recover over time and affect functional outcome. However, the underlying mechanism of function recovery remains poorly understood. In the present study, progressive function alteration of the secondary somatosensory cortex (S2) and its relationship with regional perfusion and neurological outcome were examined using a monkey model of stroke. METHODS AND MATERIALS: Rhesus monkeys (n = 4) were induced with permanent middle cerebral artery occlusion (pMCAo). Resting-state functional MRI, dynamic susceptibilit

Primary somatosensory cortex and periaqueductal gray functional connectivity as a marker of the dysfunction of the descending pain modulatory system in fibromyalgia.

BACKGROUND: Resting-state functional connectivity (rs-FC) may aid in understanding the link between pain-modulating brain regions and the descending pain modulatory system (DPMS) in fibromyalgia (FM). This study investigated whether the differences in rs-FC of the primary somatosensory cortex in responders and non-responders to the conditioned pain modulation test (CPM-test) are related to pain, sleep quality, central sensitization, and the impact of FM on quality of life. METHODS: This cross-sectional study included 33 females with FM. rs-FC was assessed by functional magnetic resonance imag

Acute anti-allodynic action of gabapentin in dorsal horn and primary somatosensory cortex: Correlation of behavioural and physiological data.

Neuropathic pain is a debilitating consequence of neuronal injury or disease. Although first line treatments include the alpha-2-delta (α2δ)-ligands, pregabalin and gabapentin (GBP), the mechanism of their anti-allodynic action is poorly understood. One specific paradox is that GBP relieves signs of neuropathic pain in animal models within 30min of an intraperitoneal (IP) injection yet its actions in vitro on spinal dorsal horn or primary afferent neurons take hours to develop. We found, using confocal Ca2+ imaging, that substantia gelatinosa neurons obtained ex vivo from rats subjected to sci

Spatiotemporal Changes of Neuronal Responses in the Primary Somatosensory Cortex to Noxious Tail Stimulation in Awake and Pentobarbital-Anesthetized Rats.

Primary somatosensory cortex (SI) is a key area in the processing of nociceptor inputs to our consciousness. To clarify the columnar and laminar organization of SI for pain processing, we compared spatiotemporal changes in neuronal activities of the primary sensorimotor cortex (SmI) of the rat in response to noxious laser heat stimulation applied to the mid-tail. Longitudinal and vertical array microelectrodes were chronically implanted in the cerebral cortex. Evoked neuronal activities, including intracortical local field potentials (LFP) and ensemble single-unit activity (SU) around SmI were

Somatosensory and visual deprivation each decrease the density of parvalbumin neurons and their synapse terminals in the prefrontal cortex and hippocampus of mice.

In the phenomenon known as cross-modal plasticity, the loss of one sensory system is followed by improved functioning of other intact sensory systems. MRI and functional MRI studies suggested a role of the prefrontal cortex and the temporal lobe in cross-modal plasticity. We used a mouse model to examine the effects of sensory deprivation achieved by whisker trimming and visual deprivation achieved by dark rearing in neonatal mice on the appearance of parvalbumin (PV) neurons and the formation of glutamic acid decarboxylase 67 (GAD67)-positive puncta around pyramidal neurons in the prefrontal

Metabolic activity pattern in the motor and somatosensory cortex of monkeys performing a visually guided reaching task with one forelimb.

The [14C]deoxyglucose method was used to map the metabolic activity in the primary somatosensory and motor cortex in monkeys (Macaca nemestrina) performing a unimanual task. The task required visually guided reaching and target holding at a rate of about 10 movements per min. The entire dorsoventral extent of the cortical region lying between the posterior crown of the arcuate and the anterior crown of the intraparietal sulci was reconstructed on the sagittal plane, from horizontal sections aligned on the fundus of the central sulcus. The metabolic mapping of the control monkey demonstrated ho

Functional preservation of benzodiazepine receptors of the primary somatosensory cortex in Creutzfeldt-Jakob disease: a pharmacologic-evoked potential study.

In previous studies, we demonstrated that the benzodiazepine (BZP) receptors of the visual system are functionally preserved in Creutzfeldt-Jakob disease (CJD). We hypothesized that such a functional preservation is not confined to the visual system. In a 74-year-old woman suffering from CJD, three consecutive recording sessions of somatosensory cortical evoked potentials (SEPs) by right median nerve stimulation were carried out: (a) basal condition, without any pharmacologic treatment; (b) 1 min after i.v. administration of 10 mg diazepam (DZP); (c) 2.5 min after i.v. administration of 3 mg F

Altered responses to cutaneous stimuli in the second somatosensory cortex following lesions of the postcentral gyrus in infant and juvenile macaques.

Infant macaques recover tactile abilities better than older animals after somatosensory cortical lesions. To investigate the neural basis of this phenomenon, we ablated the hand representation in primary somatosensory cortex (SI) of infant and juvenile Macaca mulatta and recorded in ipsilateral second somatosensory cortex (SII) a year later. We also made tracer injections to verify the lesion boundaries and to study the connections of SII after the lesion of SI. Similar to the report of Pons et al. (Science 237:417-420, '87), we found that substantial portions of the SII hand area were unrespo

Practical scan-length considerations for mapping upper limb movements to the somatosensory/motor cortex at 7T: A pilot study.

The relationship between motor cortex (M1) and upper limb movements has been investigated extensively using functional MRI (fMRI). While most research has focused on applications, very few studies have focused on practical aspects of developing the fMRI protocol. Thus, the effect of scan length on M1 activations during various upper limb movements remains unclear. Scan length constraints are important for conducting motor experiments within a 60- or 90-minute scan session. We targeted this gap in the literature in this pilot study by investigating 7T fMRI activations in a male participant whil

First activity and interactions in thalamus and cortex using raw single-trial EEG and MEG elicited by somatosensory stimulation.

INTRODUCTION: One of the primary motivations for studying the human brain is to comprehend how external sensory input is processed and ultimately perceived by the brain. A good understanding of these processes can promote the identification of biomarkers for the diagnosis of various neurological disorders; it can also provide ways of evaluating therapeutic techniques. In this work, we seek the minimal requirements for identifying key stages of activity in the brain elicited by median nerve stimulation. METHODS: We have used a priori knowledge and applied a simple, linear, spatial filter on th

The synaptic inputs and thalamic projections of two classes of layer 6 corticothalamic neurons in primary somatosensory cortex of the mouse.

Although corticothalamic neurons (CThNs) represent the largest source of synaptic input to thalamic neurons, their role in regulating thalamocortical interactions remains incompletely understood. CThNs in sensory cortex have historically been divided into two types, those with cell bodies in Layer 6 (L6) that project back to primary sensory thalamic nuclei and those with cell bodies in Layer 5 (L5) that project to higher-order thalamic nuclei and subcortical structures. Recently, diversity among L6 CThNs has increasingly been appreciated. In the rodent somatosensory cortex, two major classes o

In-Depth Characterization of Layer 5 Output Neurons of the Primary Somatosensory Cortex Innervating the Mouse Dorsal Spinal Cord.

Neuronal circuits of the spinal dorsal horn integrate sensory information from the periphery with inhibitory and facilitating input from higher central nervous system areas. Most previous work focused on projections descending from the hindbrain. Less is known about inputs descending from the cerebral cortex. Here, we identified cholecystokinin (CCK) positive layer 5 pyramidal neurons of the primary somatosensory cortex (CCK + S1-corticospinal tract [CST] neurons) as a major source of input to the spinal dorsal horn. We combined intersectional genetics and virus-mediated gene transfer to chara

Correlates of Residual Limb Pain: From Residual Limb Length and Usage to Metabolites and Activity in Secondary Somatosensory Cortex.

Most recent studies attribute residual limb pain to peripheral pathological changes of the stump. However, in this paper, we focus on its associations with the residual limb length, usage, as well as the metabolic and functional alterations of the brain. The secondary somatosensory cortex (S2), one important area involved in pain intensity discrimination, was selected as the region of interest. Twenty-two upper-limb amputees were recruited and divided into two groups, i.e., amputees with residual limb pain (9/22) and without residual limb pain (13/22). The residual limb length, usage, as well

Proprioceptive pathways to posterior parietal areas MIP and LIPv from the dorsal column nuclei and the postcentral somatosensory cortex.

The posterior parietal cortex (PPC) serves as an interface between sensory and motor cortices by integrating multisensory signals with motor-related information. Sensorimotor transformation of somatosensory signals is crucial for the generation and updating of body representations and movement plans. Using retrograde transneuronal transfer of rabies virus in combination with a conventional tracer, we identified direct and polysynaptic somatosensory pathways to two posterior parietal areas, the ventral lateral intraparietal area (LIPv) and the rostral part of the medial intraparietal area (MIP)

[The effect of barbiturates, sodium oxybutyrate and ketamine on the cycle of primary response recovery in the second somatosensory area of the cerebral cortex].

Barbiturates (ethaminal sodium and hexenal) injected to freely moving cats were shown to decrease the testing primary response in the second somatosensory cortical zone to the paired electrical stimulation of the thalamocortical fibers. Sodium hydroxybutyrate increased and ketamine failed to change the testing primary response in the second somatosensory cortical zone.
