Development and evaluation of the neutralizing capacity of horse antivenom against the Brazilian spider Loxosceles intermedia.

Spider bites due to Loxosceles intermedia are currently a major public health problem in South Brazil. About 3000 cases are reported annually. Specific treatment for spider bites is provided by the polyvalent anti-arachnidic antiserum produced by Butantan Institute, São Paulo, Brazil by immunizing horses with mixtures of venoms from Tityus serrulatus and T. bahiensis scorpions, as well as Phoneutria nigriventer and L. gaucho spiders. Due to the large amounts of the anti-arachnidic antivenom required and since L. intermedia venom has some biochemical and pharmacological variations, we have prod

A Brief Overview of the Toxic Sphingomyelinase Ds of Brown Recluse Spider Venom and Other Organisms and Simple Methods To Detect Production of Its Signature Cyclic Ceramide Phosphate.

A special category of phospholipase D (PLD) in the venom of the brown recluse spider (Loxosceles reclusa) and several other sicariid spiders accounts for the dermonecrosis and many of the other clinical symptoms of envenomation. Related proteins are produced by other organisms, including fungi and bacteria. These PLDs are often referred to as sphingomyelinase Ds (SMase Ds) because they cleave sphingomyelin (SM) to choline and "ceramide phosphate." The lipid product has actually been found to be a novel sphingolipid: ceramide 1,3-cyclic phosphate (Cer1,3P). Since there are no effective treatmen

Innovative Immunization Strategies for Antivenom Development.

Snakes, scorpions, and spiders are venomous animals that pose a threat to human health, and severe envenomings from the bites or stings of these animals must be treated with antivenom. Current antivenoms are based on plasma-derived immunoglobulins or immunoglobulin fragments from hyper-immunized animals. Although these medicines have been life-saving for more than 120 years, opportunities to improve envenoming therapy exist. In the later decades, new biotechnological tools have been applied with the aim of improving the efficacy, safety, and affordability of antivenoms. Within the avenues expl

Upper eyelid necrosis and reconstruction after spider byte: case report and review of the literature.

Spider bites are not very common, especially in the Mediterranean area, and those affecting the ocular-palpebral region involving reconstructive surgery are particularly rare. In May 2010, the case of a Caucasian 24-year-old female patient was brought to the attention of the Dermatology Department, University of Cagliari, Italy. The patient reported she woke up feeling an intense pain with itching and that also she had noticed a spider of an unknown species on her bed. The dermatosis had affected the right orbital region, where there was a considerable red and violet erythema and a hard edema,

Arthropod bites.

The phylum Arthropoda includes arachnids and insects. Although their bites typically cause only local reactions, some species are venomous or transmit disease. The two medically important spiders in the United States are widow spiders (Latrodectus), the bite of which causes intense muscle spasms, and the brown recluse (Loxosceles), which may cause skin necrosis. Widow bites usually respond to narcotics, benzodiazepines, or, when necessary, antivenom. Most recluse bites resolve uneventfully without aggressive therapy and require only wound care and minor debridement. Tick bites can transmit dis

A comparison of the toxicity of the venoms of twelve common Australian spider species on rodent vital organ systems.

1. Venom samples from twelve different Australian spider species were compared for their toxic effects on the heart and respiratory systems of anaesthetized rats. 2. The first and most serious effect of all venoms was on breathing, cardiotoxicity mostly being secondary to respiratory failure. 3. Only Atrax infensus and Selenocosmia stirlingi venoms proved highly toxic to rats, and therefore potentially toxic to humans, when single doses were used. 4. The low potency of most of the other venoms tested showed that several simultaneous spider bites would be necessary to cause significant toxicity

A Nocardia brasiliensis Infection Secondary to a Loxosceles reclusa Bite: A Spinal Cord Stimulation Case Report.

BACKGROUND: Nocardia brasiliensis, an uncommon human pathogen, is rarely seen in infections secondary to insect bites or stings. However, Nocardia brasiliensis can infect an individual through an ulcerative wound, such as one mediated by the phospholipase D toxin immune response following a Loxosceles reclusa (i.e., brown recluse spider) bite. CASE REPORT: This report presents the case of a patient who underwent a spinal cord stimulator (SCS) trial with an unknown brown recluse spider bite on her left foot. We describe the Nocardia brasiliensis cutaneous infection following a brown recluse bi

The health problem of arachnidism.

The authors give data on arachnidism (the systemic poisoning caused by the bite of the spider Latrodectus) in general, on its distribution in the world and in Yugoslavia in particular, on the Latrodectus spider, and on the characteristic clinical syndrome of arachnidism. The bite of the Latrodectus causes great pain, disables the bitten person for a certain time, and may even cause death in a few instances. The results of the authors' own observations in the clinic and on experimental animals are given. Of all forms of therapy tried by them, the simultaneous application of calcium, antivenom,

Loxoscelism: old obstacles, new directions.

Loxosceles spiders have a worldwide distribution and are considered one of the most medically important groups of spiders. Envenomation (loxoscelism) can result in dermonecrosis and, less commonly, a systemic illness that can be fatal. The mechanism of venom action is multifactorial and incompletely understood. The characteristic dermonecrotic lesion results from the direct effects of the venom on the cellular and basal membrane components, as well as the extracellular matrix. The initial interaction between the venom and tissues causes complement activation, migration of polymorphic neutrophi

Silently Suffering: A Pediatric Black Widow Spider Envenomation.

BACKGROUND: Black widow spiders are distributed worldwide and, although rarely fatal, account for significant morbidity. Diagnosis can be challenging, and children are at risk of increased morbidity due to their small size. CASE REPORT: We present a case of a 3-year-old boy who was brought to our emergency department because of sudden ear pain followed by labored breathing, abdominal pain, refusal or inability to speak, and grunting respirations. A black widow spider bite was suspected based on additional history obtained, and the spider was found in his helmet, confirming the diagnosis. The

Heat Tolerance of the Brown Recluse Spider (Araneae: Sicariidae): Potential for Pest Control.

The brown recluse spider, Loxosceles reclusa Gertsch & Mulaik, is a well-known venomous spider common in the south-central United States where infestations can reach thousands of individuals in a single structure. Bites from this spider pose a risk of dermonecrotic lesions (loxoscelism) or, rarely, more serious systemic effects. The heat tolerance of this spider is understudied but may offer an alternative pest control solution to pesticides or fumigation, both of which have their disadvantages. We subjected brown recluse spiders to increasing temperatures to establish the upper lethal tempera

[Severe loxoscelism with lethal outcome. Report of one case].

Loxoscelism, is caused by the bite of Loxosceles laeta spiders. It has two clinical forms: cutaneous loxoscelism (CL) and viscerocutaneous loxoscelism (VCL). VCL is characterized by hematuria, hemoglobinuria, jaundice, fever and sensorial involvement. In severe cases there is massive hemolysis and renal failure, with high lethality. We report a 71 year-old man, brought to the hospital five days after suffering a spider bite. The patient was admitted with a severe kidney failure, hemolysis, metabolic acidosis and clotting disorder. The patient was managed with adrenal steroids and dialisys but
