Emodin Induces Death in Human Cervical Cancer Cells Through Mitotic Catastrophe.

BACKGROUND: Anthraquinones, including emodin, are compounds with numerous pharmacological properties, including anticancer properties. The aim of this study experiment was to examine the effect of emodin, a natural compound present in the roots and rhizomes of Rheum palmatum, on the induction of mitotic catastrophe in cervical cancer cells. MATERIAL AND METHODS: HeLa celIs were treated with different emodin concentrations for 48 h, and cell growth was measured with 3-(4-,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolyl. The cell-cycle distribution and the level of apoptosis were determined by m

The ATPase SRCAP is associated with the mitotic apparatus, uncovering novel molecular aspects of Floating-Harbor syndrome.

BACKGROUND: A variety of human genetic diseases is known to be caused by mutations in genes encoding chromatin factors and epigenetic regulators, such as DNA or histone modifying enzymes and members of ATP-dependent chromatin remodeling complexes. Floating-Harbor syndrome is a rare genetic disease affecting human development caused by dominant truncating mutations in the SRCAP gene, which encodes the ATPase SRCAP, the core catalytic subunit of the homonymous chromatin-remodeling complex. The main function of the SRCAP complex is to promote the exchange of histone H2A with the H2A.Z variant. Ac

The Arabidopsis mutant jason produces unreduced first division restitution male gametes through a parallel/fused spindle mechanism in meiosis II.

In plants, whole-genome doubling (polyploidization) is a widely occurring process largely contributing to plant evolution and diversification. The generation and fusion of diploid gametes is now considered the major route of plant polyploidization. The parallel arrangement or fusion of meiosis II MII spindles (ps) is one of the most frequently reported mechanisms generating triploid offspring. Through a forward genetics screen of an Arabidopsis (Arabidopsis thaliana) ethyl methanesulfonate population, we identified Arabidopsis thaliana Parallel Spindles1 (AtPS1), which was recently reported as

Spatial control of protein phosphatase 2A (de)methylation.

Reversible methylation of the protein phosphatase 2A catalytic subunit (PP2A(C)(1)) is an important regulatory mechanism playing a crucial role in the selective recruitment of regulatory B subunits. Here, we investigated the subcellular localization of leucine carboxyl methyltransferase (LCMT1) and protein phosphatase methylesterase (PME-1), the two enzymes catalyzing this process. The results show that PME-1 is predominantly localized in the nucleus and harbors a functional nuclear localization signal, whereas LCMT1 is underrepresented in the nucleus and mainly localizes to the cytoplasm, Gol

Protective Effects of Taurine Against Zearalenone-Induced Quality Impairment During Porcine Oocyte Maturation.

Zearalenone (ZEN) is a common non-steroidal estrogenic mycotoxin that exists widely in contaminated feeds, and its toxicities target multiple organs, especially the reproductive system, since ZEN is shown to affect oocyte quality. In the present study, we reported that taurine (TAU), a sulfur-containing amino acid that exhibits antioxidant properties, protected the maturation quality of ZEN-exposed porcine oocytes. We showed that TAU significantly reversed the cumulus cell expansion and polar body extrusion defects of porcine oocytes under ZEN treatment. The rescue effect of TAU was attributed

Copper pyrithione exposure-induced copper accumulation compromises mouse oocyte maturation.

Copper exhibited serious toxicity while its accumulation was beyond the physiological threshold. Copper pyrithione (CPT) consists of two PT molecules and copper, is a new generation booster anti-fouling biocides agent widespread globally. The potential harmful impacts of CPT whether via copper accumulation have raised concern among people. Research has identified copper exposure caused deleterious effects on reproduction systems. Nevertheless, environmental-related concentrations of CPT exposure effects on female reproduction, especially the oocyte maturation and quality are still unclear. Her

Instability of dicentric plasmids in yeast.

Dicentric plasmids containing either two copies of centromere 4 or one copy of centromere 4 and one copy of centromere 3 in the yeast plasmid vector YRp17 were constructed in vitro and introduced into yeast cells by DNA transformation. The resulting colonies were heterogeneous for a mixed population of rearranged plasmids. The rearrangements always involved deletion of one or both centromere sequences originally present on the plasmid. Heterogeneity was due to the continued production of deleted plasmids from a pool of unrearranged dicentric plasmids maintained within some of the yeast cells i

Deubiquitinase USP8 regulates the spindle assembly checkpoint in oocytes.

The spindle assembly checkpoint (SAC) is vital for preventing oocyte aneuploidy, a leading cause of female infertility, miscarriages, and trisomy syndromes. However, whether deubiquitination participates in SAC regulation remains unknown. Here, we reveal that the deubiquitinase USP8 acts as a SAC regulator to prevent aneuploid egg formation. Mechanistically, depletion of USP8 inactivates the SAC, accelerates meiotic progression, and causes abnormal spindle assembly and chromosome alignment, ultimately leading to aneuploidy. Intriguingly, we identify USP8 in oocytes as a previously unidentified

Nuclear Tracking During Early Embryonic Divisions.

Positioning of the largest organelle, the nucleus, provides important cues for the geometry of cell divisions and cell fate determination. A primary function of nuclear positioning is to determine the spindle position and cleavage plane localization during cell division. Thus, centrally located nuclei lead to symmetrical divisions typical of mitosis, and off-center spindles culminate in symmetry-breaking as in mammalian oocytes. More recently, nuclear positioning has also been implicated in regulating mechanotransduction and gene expression in mouse oocytes. In human embryos, equal blastomere

Neurymenolide A, a Novel Mitotic Spindle Poison from the New Caledonian Rhodophyta Phacelocarpus neurymenioides.

The marine α-pyrone macrolide neurymenolide A was previously isolated from the Fijian red macroalga, Neurymenia fraxinifolia, and characterized as an antibacterial agent against antibiotic-resistant strains that also exhibited moderate cytotoxicity in vitro against cancer cell lines. This compound was also shown to exhibit allelopathic effects on Scleractinian corals. However, to date no mechanism of action has been described in the literature. The present study showed, for the first time, the isolation of neurymenolide A from the New Caledonian Rhodophyta, Phacelocarpus neurymenioides. We con

Spindle microtubule dysfunction and cancer predisposition.

Chromosome segregation and spindle microtubule dynamics are strictly coordinated during cell division in order to preserve genomic integrity. Alterations in the genome that affect microtubule stability and spindle assembly during mitosis may contribute to genomic instability and cancer predisposition, but directly testing this potential link poses a significant challenge. Germ-line mutations in tumor suppressor genes that predispose patients to cancer and alter spindle microtubule dynamics offer unique opportunities to investigate the relationship between spindle dysfunction and carcinogenesis

A soft cortex is essential for asymmetric spindle positioning in mouse oocytes.

At mitosis onset, cortical tension increases and cells round up, ensuring correct spindle morphogenesis and orientation. Thus, cortical tension sets up the geometric requirements of cell division. On the contrary, cortical tension decreases during meiotic divisions in mouse oocytes, a puzzling observation because oocytes are round cells, stable in shape, that actively position their spindles. We investigated the pathway leading to reduction in cortical tension and its significance for spindle positioning. We document a previously uncharacterized Arp2/3-dependent thickening of the cortical F-ac

Asymmetric inheritance of mother versus daughter centrosome in stem cell division.

Adult stem cells often divide asymmetrically to produce one self-renewed stem cell and one differentiating cell, thus maintaining both populations. The asymmetric outcome of stem cell divisions can be specified by an oriented spindle and local self-renewal signals from the stem cell niche. Here we show that developmentally programmed asymmetric behavior and inheritance of mother and daughter centrosomes underlies the stereotyped spindle orientation and asymmetric outcome of stem cell divisions in the Drosophila male germ line. The mother centrosome remains anchored near the niche while the dau

Podophyllotoxin Exposure Affects Organelle Distribution and Functions in Mouse Oocyte Meiosis.

Podophyllotoxin (POD) is one of the most characterized lignans that is commonly found in podophyllum, and its preparations and derivatives are widely used in clinical treatment due to strong antitumor and antivirus activities. POD has been reported for its neurotoxicity, liver toxicity, and potential reproductive toxicity. In the present study, we investigated the effects of POD on the organelles of mouse oocytes during meiosis. Our results showed that exposure to POD significantly reduced the developmental competence of mouse oocytes. Further analysis revealed that the endoplasmic reticulum (

Asteroid bodies: derivatives of the cytosphere. An electron microscopic contribution to the pathology of the cytocentre.

An electron microscopic study of asteroid bodies in granulomatous diseases of man, in particular in sarcoidosis and foreign body reactions, has led to the following conclusions. 1. Asteroid cytoplasmic inclusions are composed of organic proteineous structures. The body and arms of the stars consist mainly of longitudinally orientated, partially helically wound, collagen free, microfilaments with a diameter of approximately 50 A. Microtubules participate in their composition to a lesser degree. The filaments intertwine in various directions within the corpus of the stars. 2. Deeply osmiophilic

Mga2-mediated transcription supports mitotic nuclear expansion under lipid saturation conditions in stearoyl-CoA desaturase Ole1 mutant.

Membrane organelles are dynamic structures that depend on fluid membranes for their integrity and function, with the fluidity primarily derived from loosely packed unsaturated lipids. We investigated how cells respond to lipid saturation and its effect on nuclear dynamics in the budding yeast Saccharomyces cerevisiae. We found that the lipid desaturase mutant ole1-20 upregulates various genes, including OLE1, primarily through the lipid saturation-sensing transcription factor Mga2. The ole1-20 mutant displays prolonged anaphase and impaired nuclear membrane expansion, which can be rescued by t

Microcephaly protein Asp focuses the minus ends of spindle microtubules at the pole and within the spindle.

Depletion of Drosophila melanogaster Asp, an orthologue of microcephaly protein ASPM, causes spindle pole unfocusing during mitosis. However, it remains unclear how Asp contributes to pole focusing, a process that also requires the kinesin-14 motor Ncd. We show that Asp localizes to the minus ends of spindle microtubule (MT) bundles and focuses them to make the pole independent of Ncd. We identified a critical domain in Asp exhibiting MT cross-linking activity in vitro. Asp was also localized to, and focuses the minus ends of, intraspindle MTs that were nucleated in an augmin-dependent manner
