Anterior and Posterior Tongue Regions and Taste Papillae: Distinct Roles and Regulatory Mechanisms with an Emphasis on Hedgehog Signaling and Antagonism.

Sensory receptors across the entire tongue are engaged during eating. However, the tongue has distinctive regions with taste (fungiform and circumvallate) and non-taste (filiform) organs that are composed of specialized epithelia, connective tissues, and innervation. The tissue regions and papillae are adapted in form and function for taste and somatosensation associated with eating. It follows that homeostasis and regeneration of distinctive papillae and taste buds with particular functional roles require tailored molecular pathways. Nonetheless, in the chemosensory field, generalizations are

Purinergic Signaling in Oral Tissues.

The role of the purinergic signal has been extensively investigated in many tissues and related organs, including the central and peripheral nervous systems as well as the gastrointestinal, cardiovascular, respiratory, renal, and immune systems. Less attention has been paid to the influence of purines in the oral cavity, which is the first part of the digestive apparatus and also acts as the body's first antimicrobial barrier. In this review, evidence is provided of the presence and possible physiological role of the purinergic system in the different structures forming the oral cavity includi

Surface architecture of the oropharyngeal cavity and the digestive tract of Mulloidichthys flavolineatus from the red sea, Egypt: A scanning electron microscope study.

Surface architecture of the buccal cavity and the surface organization of the luminal mucosa of the oesophagus, stomach, and intestine of the carnivorous fish M. flavolineatus from the Red Sea were studied by using SEM. The results revealed that M. flavolineatus has four kinds of teeth; curved-blunt, wedge-shaped, flattened crowns, molariform and papilliform. Three types of taste buds (type I, II and III) were recorded in the oropharyngeal cavity. It was observed that taste buds and teeth are co-located in the pharyngeal region. Characteristic patterns of microridges of the surface cells in th

Gustatory information of umami substances in three major taste nerves.

The chorda tympani (CT), glossopharyngeal (GL), and greater superficial petrosal (GSP) nerves, the three major branches of cranial nerves innervating taste buds, respond with considerable differences to various taste stimuli. To examine which nerve is responsible for transmitting umami taste in rats, we conducted electrophysiological and behavioral experiments. In the electrophysiological study, responses to umami substances were compared among these three nerves. The CT and GSP were more responsive to mixtures of monosodium L-glutamate (MSG) and 5'-inosine monophosphate (IMP) than the GL. Syn

Might Helicobacter pylori infection be associated with distortion on taste perception?

Helicobacter pylori (H. pylori) has been found in dental plaque, saliva and lingual sites. To date, taste or olfaction disorders related to H. pylori infections have never been reported. In a review of the literature we found two papers just referring to a sour taste sensation during H. pylori infection. Studies in animal models suggest that changes in taste perception may relate to infections which damage taste buds. We observed an interesting clinical case of a 24-year-old Ghanaian woman with documented H. pylori gastric infection, complaining of cacosmia and cacogeusia. Taste evaluation ind

Sensory functions beyond taste: multifaceted role of solitary chemosensory cells and taste receptors in mucosal immune defense and disease pathogenesis.

Taste receptors are most commonly associated with the experience of flavor. However, these receptors are also crucial participants in wider physiological mechanisms, particularly immunological defense. Solitary chemosensory cells (SCCs) express taste receptors (e.g., TAS2Rs and TAS1Rs) and are distributed throughout many organs, including the oral cavity, respiratory system and gastrointestinal tract. These cells function as sensors for dangerous bacteria and trigger immunological responses to preserve the integrity of mucosal tissues and prevent disease. Recent research has revealed that SCCs

Extragustatory bitter taste receptors in head and neck health and disease.

Taste receptors, first described for their gustatory functions within the oral cavity and oropharynx, are now known to be expressed in many organ systems. Even intraoral taste receptors regulate non-sensory pathways, and recent literature has connected bitter taste receptors to various states of health and disease. These extragustatory pathways involve previously unexplored, clinically relevant roles for taste signaling in areas including susceptibility to infection, antibiotic efficacy, and cancer outcomes. Among other physicians, otolaryngologists who manage head and neck diseases should be

The endocrinology of taste receptors.

Levels of obesity have reached epidemic proportions on a global scale, which has led to considerable increases in health problems and increased risk of several diseases, including cardiovascular and pulmonary diseases, cancer and diabetes mellitus. People with obesity consume more food than is needed to maintain an ideal body weight, despite the discrimination that accompanies being overweight and the wealth of available information that overconsumption is detrimental to health. The relationship between energy expenditure and energy intake throughout an individual's lifetime is far more compli

A Novel Mechanism for T1R-Independent Taste Responses to Concentrated Sugars.

Recent findings from our laboratory demonstrated that the rostral nucleus of the solitary tract (rNST) retains some responsiveness to sugars in double-knock-out mice lacking either the T1R1+T1R3 (KO1+3) or T1R2+T1R3 (KO2+3) taste receptor heterodimers. Here, we extended these findings in the parabrachial nucleus (PBN) of male and female KO1+3 mice using warm stimuli to optimize sugar responses and employing additional concentrations and pharmacological agents to probe mechanisms. PBN T1R-independent sugar responses, including those to concentrated glucose, were more evident than in rNST. Simil

Prion infection of oral and nasal mucosa.

Centrifugal spread of the prion agent to peripheral tissues is postulated to occur by axonal transport along nerve fibers. This study investigated the distribution of the pathological isoform of the protein (PrP(Sc)) in the tongues and nasal cavities of hamsters following intracerebral inoculation of the HY strain of the transmissible mink encephalopathy (TME) agent. We report that PrP(Sc) deposition was found in the lamina propria, taste buds, and stratified squamous epithelium of fungiform papillae in the tongue, as well as in skeletal muscle cells. Using laser scanning confocal microscopy,

The adaptation of the frog tongue to various taste solutions: the effect on gustatory neural responses to bitter stimuli.

1. After the frog tongue was adapted for 10 sec to various salts and sugars, the initial phasic component of gustatory neural responses to almost all of quinine hydrochloride (Q-HCl), quinine sulfate (Q-H2SO4). Brucine, caffeine and picric acid was suppressed. 2. Following 10 sec adaptation to acetic acid, the phasic responses to Q-HCl and Q-H2SO4 were unchanged, those to brucine and caffeine were enhanced, and that to picric acid was depressed slightly. 3. The response to any one of Q-HCl, Q-H2SO4. brucine and caffeine was suppressed after adaptation to the other three, while those to picric

Behavioral and neurophysiological taste responses to sweet and salt are diminished in a model of subclinical intestinal inflammation.

There is strong evidence for gut-taste bud interactions that influence taste function, behavior and feeding. However, the effect of gut inflammation on this axis is unknown despite reports of taste changes in gastrointestinal (GI) inflammatory conditions. Lipopolysaccharide (LPS), an inflammatory stimulus derived from gram-negative bacteria, is present in the normal GI tract and levels increase during high-fat feeding and gut infection and inflammation. Recordings from the chorda tympani nerve (CT), which transmits taste information from taste buds on the anterior tongue to the brain, previous

Sour ageusia in two individuals implicates ion channels of the ASIC and PKD families in human sour taste perception at the anterior tongue.

BACKGROUND: The perception of sour taste in humans is incompletely understood at the receptor cell level. We report here on two patients with an acquired sour ageusia. Each patient was unresponsive to sour stimuli, but both showed normal responses to bitter, sweet, and salty stimuli. METHODS AND FINDINGS: Lingual fungiform papillae, containing taste cells, were obtained by biopsy from the two patients, and from three sour-normal individuals, and analyzed by RT-PCR. The following transcripts were undetectable in the patients, even after 50 cycles of amplification, but readily detectable in the

Formulation of poorly water-soluble drugs in lipid nanocarriers: a suitable approach for taste masking or bitter-taste enhancing?

The taste of a drug formulation, especially of orodispersible and liquid dosage forms, is of crucial importance due to its direct contact with the human taste buds. In order to achieve high patient acceptance with these formulations, drugs with an unpleasant, predominantly bitter taste have to be masked. One strategy for masking taste particularly suitable for lipophilic drugs is to dissolve the drug molecules in lipid carrier systems, such as nanoemulsions. Although this strategy has often proven effective as a formulation strategy for improving bioavailability and masking an unpleasant taste

The tongue map and the spatial modulation of taste perception.

There is undoubtedly a spatial component to our experience of gustatory stimulus qualities such as sweet, bitter, salty, sour, and umami, however its importance is currently unknown. Taste thresholds have been shown to differ at different locations within the oral cavity where gustatory receptors are found. However, the relationship between the stimulation of particular taste receptors and the subjective spatially-localized experience of taste qualities is uncertain. Although the existence of the so-called 'tongue map' has long been discredited, the psychophysical evidence clearly demonstrates

The selective serotonin reuptake inhibitor paroxetine does not alter consummatory concentration-dependent licking of prototypical taste stimuli by rats.

Serotonin and the 5HT(1A) receptor are expressed in a subset of taste receptor cells, and the 5HT(3) receptor is expressed on afferent fibers innervating taste buds. Exogenous administration of the selective serotonin reuptake inhibitor, paroxetine, has been shown to increase taste sensitivity to stimuli described by humans as sweet and bitter. Serotonergic agonists also decrease food and fluid intake, and it is possible that modulations of serotonin may alter taste-based hedonic responsiveness; alternatively, or in combination, serotonin may interact with physiological state to impact ingesti

Expression of Calca gene-derived peptides in the murine taste system.

Taste cell regeneration and taste signaling are regulated by myriad growth factors and signaling molecules secreted by neurons and taste papillae-resident cells. The calcitonin-related polypeptide alpha (Calca) gene is a source of 4 biologically active peptides with varied physiological roles. Alternative splicing of the Calca messenger RNA generates either preprocalcitonin gene-related peptide (CGRP) or preprocalcitonin-encoding transcripts. Proteolytic processing of preprocalcitonin generates procalcitonin, calcitonin and katacalcin. Calcitonin is a ligand for the G protein coupled receptor

Impaired responses to sweet taste in vitamin A-deficient rats.

In brief-exposure, two-choice preference tests (sucrose solution v water), vitamin A-deficient (VAD) rats exhibited a decreased preference for sucrose relative to control rats. There was no difference in total fluid intake from both choices between the two groups, nor was any significant difference found in circumvallate taste papilla keratin size. It is concluded that the impaired preference for sucrose in VAD rats is due to a specific impairment in taste sensation rather than general malaise.
