Transformation of double-hit follicular lymphoma to plasmablastic lymphoma: a partial role of MYC gene rearrangement.

Follicular lymphoma (FL) is genetically characterized by BCL2/IGH translocation. Some FL cases histologically transform to high-grade lymphoma, and the majority of cases transform to diffuse large B-cell lymphoma. We report herein an unusual FL case that transformed to plasmablastic lymphoma (PBL) with MYC gene rearrangement as early as 12 months after FL diagnosis. IGH/MYC translocation, the most common cytogenetic abnormality seen in de novo PBL, was also detected in the transformed tumor (double-hit lymphoma). The patient became resistant to chemotherapy and died 4 months after transformati

Deep Infiltrating Endometriosis Malignant Invasion of Cervical Wall and Rectal Wall With Lynch Syndrome: A Rare Case Report and Review of Literature.

BACKGROUND: Malignant transformation of deep infiltrating endometriosis (DIE) invading the cervix and rectum is quite rare, especially in patients combined with Lynch syndrome (LS). We report a rare case of a 49-year-old perimenopausal woman with endometrioid carcinoma arising from the pouch of Douglas, invading the cervix and rectum 1 year after a unilateral salpingo-oophorectomy treatment for ovarian endometriosis. The genetic testing of the patient showed germline mutations in MSH2, which combined with the special family history of colorectal cancer of the patient, was also thought to be as

[An integrated system for transient expression and stable transformation in Nicotiana benthamiana suspension cells].

Nicotiana benthamiana suspension cells, exhibiting advantages such as short growth cycles, facile genetic manipulation, ease of product purification, and suitability for large-scale cultivation, are emerging as a novel industrial biosynthesis platform for plant-derived natural products. Transient expression and stable transformation are core technologies for validating gene function and constructing plant cell factories. However, current methods have been developed primarily for whole-plant systems, systematic investigations in plant suspension cells are lacking, let alone a technological fram

Efficient assembly of a large fragment of monkeypox virus genome as a qPCR template using dual-selection based transformation-associated recombination.

Transformation-associated recombination (TAR) has been widely used to assemble large DNA constructs. One of the significant obstacles hindering assembly efficiency is the presence of error-prone DNA repair pathways in yeast, which results in vector backbone recircularization or illegitimate recombination products. To increase TAR assembly efficiency, we prepared a dual-selective TAR vector, pGFCS, by adding a PADH1-URA3 cassette to a previously described yeast-bacteria shuttle vector, pGF, harboring a PHIS3-HIS3 cassette as a positive selection marker. This new cassette works as a negative sel

Epithelial transition zones: merging microenvironments, niches, and cellular transformation.

Transition zones (TZs) are regions in the body where two different types of epithelial tissue meet resulting in the appearance of a distinct abrupt transition. These TZs are found in numerous locations within the body, including the cornea-conjunctiva junction, esophagogastric junction, gastro-duodenal junction, endo-ectocervix junction, ileocecal junction, and anorectal junction. Several of these TZs are often associated with the development of cancer, in some cases due to viral transformation by the human papilloma virus (HPV). The underlying molecular and cellular basis for this tumor susce

Transformation by the human adenoviruses.

Oncogenic transformation by the human adenoviruses involves the concerted action of two genes, E1A and E1B. Over the last few years the products of these genes have been characterised in considerable detail using genetic, immunological and biochemical means. The E1A gene by itself can immortalise primary cells and can cooperate to effect full morphological transformation not only with E1B but also with other known oncogenes. The immortalisation and cooperation activities of E1A require multiple functions that are directed by structurally and functionally independent regions of the E1A protein.

Protoplast Transformation of Phytophthora spp.

At the core of assays to understand the role(s) of specific genes is the ability to stably transfer genes into Phytophthora through transformation. A key method for achieving this has been based on polyethylene glycol (PEG)/CaCl2 transformation of protoplasts, but efficiency has often been low. Improving transformation efficiency is necessary for many applications, such as gene knockouts. Here we describe improvements through successive rounds of "mock" transformation, leading to improved efficiency in Phytophthora infestans and other species.

Development of an Optimized in Planta Transformation System in Sugarcane and Its Application on Sh4CL13 in Chlorogenic Acid Biosynthesis.

Sugarcane (Saccharum spp. hybrid) is the most important sugar crop. However, its genetic improvement is severely constrained by genotype-dependent regeneration recalcitrance and lengthy tissue culture cycles. Here, we established a simple, efficient, and genotype-flexible in planta transformation system using two-leaf-stage plantlets. By systematic optimization of three key parameters: Agrobacterium cell density (OD600 = 0.5), dark incubation duration (2 weeks), and infection frequency (two rounds), we achieved a maximum transformation efficiency of 51.2%. The protocol was successfully applied

High-efficiency stable transformation of the model fern species Ceratopteris richardii via microparticle bombardment.

Ferns represent the most closely related extant lineage to seed plants. The aquatic fern Ceratopteris richardii has been subject to research for a considerable period of time, but analyses of the genetic programs underpinning developmental processes have been hampered by a large genome size, a lack of available mutants, and an inability to create stable transgenic lines. In this paper, we report a protocol for efficient stable genetic transformation of C. richardii and a closely related species Ceratopteris thalictroides using microparticle bombardment. Indeterminate callus was generated and m

Atypical Burkitt's lymphoma transforming from follicular lymphoma.

Amongst follicular lymphoma that transforms into a high-grade lymphoma, majority are diffuse large B cell lymphoma. Here we reported a rare atypical Burkitt's lymphoma transformation from an asymptomatic follicular lymphoma. Lymph node biopsy showed a composite lymphoma with infiltration of the inter-follicular areas by high grade small non-cleaved lymphoma cells amongst neoplastic follicles. Moreover, FISH and molecular genetic study confirmed concomitant MYC translocations and t(14;18) in the high-grade component, thereby suggesting the transformation of atypical Burkitt's lymphoma from an u

Large DNA fragment ISEc9-mediated transposition during natural transformation allows interspecies dissemination of antimicrobial resistance genes.

PURPOSE: Antimicrobial resistance poses a significant global health challenge, contributing to a lack of effective therapeutic agents, especially against Gram-negative bacteria. Resistance dissemination is accelerated by horizontal gene transfer (HGT) mechanisms. The extended-spectrum beta lactamases CTX-M confer resistance to several beta-lactams, are usually embedded into plasmids and thought to be mainly disseminated by conjugation. However, an increasing number of isolates carry these enzyme-encoding genes in the chromosome, suggesting that they can spread by other means of HGT. In this st

[Mechanisms of leucemogenesis].

The genetic origins of the development of malignant haematological disorders have been established at the beginning of the 80ies. Systematic characterization of chromosomal structural abnormalities and, more recently by DNA microarray approaches and sequencing of tumour genomes have allowed the identification of a large number of genes that are mutated during malignant transformation in humans. Functional studies of these human oncogenes have shown that most of them were not able to transform a haematologic progenitor when acting alone and that cooperation with other oncogenic events was requi

Enhancing the chemical transformation of Candida parapsilosis.

Candida parapsilosis is a leading cause of invasive mycoses and the major cause of nosocomial fungaemia amongst low and very low birth weight neonates. However, the molecular and physiological characteristics of this fungus remain understudied. To advance our knowledge about the pathobiology of this pathogen, we sought to develop and validate an effective method for chemical transformation of C. parapsilosis. Chemical transformation is the primary procedure for introducing foreign DNA into Candida yeast as it requires no special equipment, although its performance efficacy drops rapidly when t

BCL-6 mRNA expression in higher grade transformation of follicle center lymphoma: correlation with somatic mutations in the 5' regulatory region of the BCL-6 gene.

Follicle center lymphoma (FCL) is an indolent low-grade B cell non-Hodgkin's lymphoma (NHL) that frequently transforms to aggressive diffuse large B cell lymphoma (DLBCL). Histological transformation of FCL is commonly associated with accumulation of secondary genetic alterations. The BCL-6 gene is commonly implicated in the pathogenesis of DLBCL and its expression may be altered by clonal rearrangements and somatic point mutations in its 5' non-translated regulatory region. Recently, somatic mutations of the BCL-6 gene were associated with the transformation process. Here, we examined BCL-6 m

Hepatocellular Carcinoma Arising From Well-Differentiated Hepatocellular Neoplasm of Uncertain Malignant Potential in a Patient With Abernethy Malformation.

Abernethy malformation is a rare congenital extrahepatic mesocaval shunt (CEMS) in which mesenteric venous blood bypasses the liver completely (type I) or partially (type II) and drains directly into the vena cava. In complete CEMS (type I), the intrahepatic portal vein is absent. The resulting lack of portal flow is associated with the development of hepatic nodules, such as focal nodular hyperplasia (FNH), hepatocellular adenoma (HCA), and hepatocellular carcinoma (HCC). These lesions have a higher risk of malignant transformation than conventional adenomas. The term hepatocellular neoplasm

Agrobacterium tumefaciens-mediated transformation of Valsa mali: an efficient tool for random insertion mutagenesis.

Valsa mali is a causal agent of apple and pear trees canker disease, which is a destructive disease that causes serious economic losses in eastern Asia, especially in China. The lack of an efficient transformation system for Valsa mali retards its investigation, which poses difficulties to control the disease. In this research, a transformation system for this pathogen was established for the first time using A. tumefaciens-mediated transformation (ATMT), with the optimal transformation conditions as follows: 10(6)/mL conidia suspension, cocultivation temperature 22°C, cocultivation time 72 ho

Improved cotton transformation protocol mediated by Agrobacterium and biolistic combined-methods.

The combined Agrobacterium- and biolistic-mediated methods of cotton transformation provide a straightforward and highly efficient protocol for obtaining transgenic cotton. Cotton (Gossypium spp.) is the most important crop for natural textile fiber production worldwide. Nonetheless, one of the main challenges in cotton production are the losses resulting from insect pests, pathogens, and abiotic stresses. One effective way to solve these issues is to use genetically modified (GM) varieties. Herein, we describe an improved protocol for straightforward and cost-effective genetic transformation

[Hereditary colorectal carcinogenesis].

Hereditary cancer is characterized by the development of certain cancer types in combination with pathogenic germline mutations in genes known to predispose to these cancer types. Familial cancer differs from hereditary cancer in that no predisposing germline mutation is detected in affected families. However, familial cancer may have a genetic background of as yet unknown origin. Colorectal cancer is unique among human tumors since almost all cancers derive from macroscopically visible benign polypoid precursors. Molecular mechanisms of precursor development differ from that of malignant tran
