Treponema pallidum and Helicobacter pylori recovered in a case of chronic active gastritis.

A 49-year-old man complaining of epigastric pain underwent endoscopy, during which thickened stomach folds below the fundus were observed. Microscopic examination of gastric tissue biopsy specimens revealed chronic active gastritis. Dieterle stain revealed overwhelming numbers of "corkscrew-like" spirochetes. These were proved to be consistent with Treponema pallidum. A comprehensive study of the tissue revealed the added presence of Helicobacter pylori. This appears to be the first case report describing the involvement of H. pylori and T. pallidum together in a case of chronic active gastrit

Effects of cefetamet (Ro 15-8074) on Treponema pallidum and experimental syphilis.

Cefetamet pivoxil (Ro 15-8075) is a newly developed, expanded-spectrum cephalosporin that is orally active. In vitro, the active form, cefetamet (Ro 15-8074), at a concentration of 0.05 micrograms/ml killed and lysed Treponema pallidum. Rabbit serum did not diminish its effectiveness. The antibiotic rapidly entered the circulation following intramuscular injection into rabbits, attaining its highest levels of 24 to 37 micrograms/ml within 10 to 30 min. Animals were infected intradermally with T. pallidum and then treated with different doses of cefetamet. Accelerated healing was detected follo

A major antigen on the outer envelope of a human oral spirochete, Treponema denticola.

Human oral spirochetes are prominent inhabitants of subgingival plaque in patients with periodontal disease. By immunoelectron microscopy using protein A-gold complexes and either polyclonal mouse antiserum against the 53-kDa antigen or 53-kDa-antigen-specific monoclonal antibody, a major polypeptide antigen, with a molecular weight of 53,000 (molecular size, 53 kilodaltons [kDa]), of a human oral spirochete, Treponema denticola ATCC 33520, was found to localize on the surface of the outer envelope.

Comparative in vitro susceptibility of Treponema pallidum to ceftizoxime, ceftriaxone and penicillin G.

A procedure dating back to the early penicillin era is adapted in order to determine the activity of the new cephalosporins ceftizoxime and ceftriaxone against Treponema pallidum in vitro. While the well-known activity of penicillin G is confirmed for the virulent Nichols strain (0.002 micrograms/ml lead to 50% immobilisation) the new cephalosporins turn out to be almost as efficacious. The concentration of ceftizoxime leading to 50% inhibition amounts to 0.004 micrograms/ml. The corresponding figure for ceftriaxone is 0.01 micrograms/ml. The potential importance of these findings for the trea

Detection of Treponema pallidum in lesion exudate with a pathogen-specific monoclonal antibody.

The diagnosis of early syphilis currently requires dark-field microscopic or serologic demonstration of Treponema pallidum infection. Dark-field microscopy is not widely available and is complicated by the numerous saprophytic spirochetes which are present at oral and rectal mucosal surfaces. Serologic tests are positive in only 70 to 90% of patients with primary syphilis, and several days may be required for results to become available. We used a pathogen-specific, fluorescein-conjugated monoclonal antibody to examine lesion exudates from 61 patients for the presence of T. pallidum and compar

Detection of nonspecific resistance to Listeria monocytogenes in rabbits infected with Treponema pallidum.

Several lines of evidence suggest that cell-mediated immunity (CMI) is suppressed in the early stages of infection caused by Treponema pallidum and becomes activated at the time that latency is induced. In the studies reported in this paper, rabbits were infected intravenously with T. pallidum and subsequently challenged with Listeria monocytogenes. Enhanced ability to suppress the growth of Listeria was detected in their livers between 3 and 5 weeks after infection with T. pallidum, corresponding to the onset and regression of the generalized syphilitic eruption. A second infection of T. pall

Rapid visual detection of Treponema pallidum using the RPA-CRISPR/Cas12a system.

Syphilis, caused by Treponema pallidum, is a sexually transmitted infection that has re-emerged globally over the past decade, posing significant public health challenges. Conventional diagnostic methods are limited by lengthy processing times, operational complexity, and moderate sensitivity, highlighting the urgent need for rapid, sensitive, and user-friendly detection strategies. In this study, we developed a visual detection platform for T. pallidum DNA by integrating recombinase polymerase amplification (RPA) with CRISPR/Cas12a technology. The assay can be completed within one hour, with

A 5500-year-old Treponema pallidum genome from Sabana de Bogotá, Colombia.

Treponematosis, a bacterial infection caused by Treponema pallidum subspecies and T. carateum (yaws, bejel, syphilis, pinta), has afflicted humans for millennia. Despite paleopathological evidence and emerging genomic data, little is known about the evolutionary history of these pathogens. We report a 5500-year-old Treponema genome (TE1-3) from Middle Holocene hunter-gatherer contexts of the rock shelter Tequendama I in Colombia. Our analyses place TE1-3 as a sister lineage to all known T. pallidum subspecies, positioning this pathogen in the Americas millennia before European contact and befo

Evaluation of Artificial Intelligence-Generated DNA Aptamers Against Treponema pallidum Surface Proteins.

DNA aptamer sequences were selected in silico with assistance from artificial intelligence (AI) using the new Xelari.com platform against three known immunogenic surface proteins of Treponema pallidum designated Tpp17, Tpp47 and Tp0751. The in silico aptamers were synthesized with 5' Alexa Fluor 647 labels and shown to bind live Treponema pallidum by fluorescence microscopy and spectrofluorometry. While AI-predicted Kd values differed somewhat from measured Kd values, it is clear that the aptamers bound the T. pallidum surface which is consistent with the molecular docking models for each apta

Treponema pallidum's neural invasion: from blood-brain barrier breach to immune sabotage.

Neurosyphilis is an infectious disease of the nervous system caused by Treponema pallidum. With the resurgence of syphilis worldwide, neurosyphilis has become prevalent again, but research on its pathogenesis remains challenging. T. pallidum exhibits remarkable invasive potential and immune evasion properties, which enable it to rapidly penetrate the blood-brain barrier (BBB) and infiltrate the central nervous system. Meanwhile, the immune response induced by this pathogen may cause tissue damage and accelerate disease progression. Additionally, host factors and the genotypes of T. pallidum st

A new typing scheme demonstrates high discriminatory power for Treponema pallidum subspecies.

The global resurgence of treponematoses, particularly syphilis, poses a growing public health challenge. Despite recent advances in sequencing technologies, obtaining complete Treponema pallidum genome sequences for epidemiological studies remains time-consuming and challenging due to the difficulty related to procuring clinical samples with sufficient treponemal burden to fulfil the sequencing requirements. There is an urgent need for rapid, cost-effective and accessible typing methods suitable for laboratories with Sanger sequencing resources. Based on the analysis of 121 T. pallidum genomes

Proteome Analysis of Seven Treponema pallidum subsp. pallidum Strains Grown In Vitro.

Treponema pallidum subsp. pallidum (T. pallidum), the fastidious causative agent of syphilis, has become more accessible for research with its recently developed in vitro cultivation method. In this work, the proteomes of seven T. pallidum strains (Nichols-like: DAL-1, Haiti B, and Madras; SS14-like: SS14, Mexico A, Philadelphia 1, and Grady), cultivated in vitro, were analyzed in biological triplicates by liquid chromatography-tandem mass spectrometry (LC-MS/MS). The MS/MS data were processed against their corresponding genomes using various annotation algorithms (DFAST, PGAP, Prodigal, Prokk

Proinflammatory CD14highCD16low monocytes/macrophages prevail in Treponema phagedenis-associated bovine digital dermatitis.

Digital dermatitis (DD) is a skin disease in cattle characterized by painful inflammatory ulcerative lesions in the feet, mostly associated with local colonization by Treponema spp., including Treponema phagedenis. The reason why most DD lesions remain actively inflamed and progress to chronic conditions despite antibiotic treatment remains unknown. Herein, we show an abundant infiltration of proinflammatory (CD14highCD16low) monocytes/macrophages in active DD lesions, a skin response that was not mitigated by topical treatment with oxytetracycline. The associated bacterium, T. phagedenis, iso

The growth-stimulating factor of Treponema phagedenis from bovine digital dermatitis lesions.

Bovine digital dermatitis (BDD) is an infectious skin disease of the hoof characterized by painful ulcerations that cause lameness in dairy cattle. Cell-free supernatants (CFS) of Falsiporphyromonas endometrii predominantly isolated from BDD lesions had the highest growth-stimulating effect on Treponema phagedenis among BDD-associated bacteria. Butyric acid was detected at a concentration of 45.4 mM in CFS of F. endometrii, and the growth of T. phagedenis was significantly promoted by butyric acid supplementation.

A defined syphilis vaccine candidate inhibits dissemination of Treponema pallidum subspecies pallidum.

Syphilis is a prominent disease in low- and middle-income countries, and a re-emerging public health threat in high-income countries. Syphilis elimination will require development of an effective vaccine that has thus far remained elusive. Here we assess the vaccine potential of Tp0751, a vascular adhesin from the causative agent of syphilis, Treponema pallidum subsp. pallidum. Tp0751-immunized animals exhibit a significantly reduced bacterial organ burden upon T. pallidum challenge compared with unimmunized animals. Introduction of lymph nodes from Tp0751-immunized, T. pallidum-challenged ani
