Ulcerative colitis (UC), a major form of inflammatory bowel disease (IBD), is characterized by chronic inflammation and ulceration of the colonic mucosa. Its etiology is multifactorial, involving genetic, environmental, and immune factors. Recent evidence highlights the crucial role of ferroptosis, an iron-dependent regulated cell death pathway, in UC pathogenesis. Ferroptosis is marked by excessive accumulation of lipid peroxides, reactive oxygen species, and iron overload, all contributing to epithelial injury. The 2,4,6-trinitrobenzene sulfonic acid (TNBS)-induced colitis model closely mimi

BACKGROUND: Inflammatory bowel disease (IBD) is characterized by chronic gastrointestinal inflammation and comprises ulcerative colitis (UC) and Crohn's disease (CD). Several cohort studies have shown that cigarette smoking (CS) is a risk factor for CD, but it has protective potential for UC. Although CS has been shown to cause different effects on UC and CD, the underlying mechanisms have not been fully determined.
METHODS: Colitis was induced in CS-exposed mice using two different chemical-induced models: dextran sodium sulfate (DSS) and 2,4,6-trinitrobenzene sulfonic acid (TNBS). We examin

BACKGROUND: Mesenchymal stem cells (MSCs) possess immunoregulatory properties and have been shown to alleviate symptoms of Crohn's disease (CD). While ferroptosis has been implicated in the onset of CD, the precise connection between MSCs, CD, and ferroptosis remains unclear. This study sought to explore whether MSCs could alleviate CD by inhibiting ferroptosis.
MATERIALS AND METHODS: We screened the hub genes involved in both ferroptosis and CD and validated them in clinical colon biopsies by immunohistochemistry (IHC). MSCs were administered intraperitoneally to mice with colitis induced by

PURPOSE: Ulcerative colitis (UC) pathogenesis involves neutrophil extracellular trap (NET) formation and intestinal barrier collapse. High mobility group box 1 (HMGB1) acts as a critical "alarmin," triggering neutrophil activation via the Toll-like receptor 4 (TLR4) axis. We investigated whether Astragalus polysaccharides (APS) could disrupt this "HMGB1-NETs" vicious cycle to treat UC.
METHODS: Severe colitis was induced in SD rats using 2,4,6-trinitrobenzene sulfonic acid (TNBS). Rats received APS (200, 400, 800 mg/kg), DNase I (to degrade NETs), or "APS + exogenous HMGB1" (functional rescue

BACKGROUND: Our earlier studies identified that non-SMC condensin I complex subunit D2 (NCAPD2) induces inflammation through the IKK/NF-κB pathway in ulcerative colitis. However, its role in the development of Crohn's disease (CD) and the specific molecular mechanism still need to be further studied.
METHODS: NCAPD2 expression in clinical ileal CD mucosa vs normal mucosa was examined, alongside its correlation with CD patients' clinical characteristics via their medical records. The biological function and molecular mechanism of NCAPD2 in CD were explored using a 2,4,6-trinitrobenzene sulfoni

OBJECTIVE: To study and analyze the varieties across long non-coding RNAs (lncRNAs), microRNAs (miRNAs), and mRNAs in spinal cord, construct the competing endogenous RNAs (ceRNAs) network, and discuss the possible central mechanism in the development of chronic inflammatory visceral pain (CIVP).
METHODS: The colitis-associated CIVP rat model was prepared using 2,4,6-trinitrobenzene sulfonic acid (TNBS) enema. The abdominal withdrawal reflex (AWR), mechanical withdrawal threshold (MWT), and thermal withdrawal latency (TWL) were used to evaluate the rat's pain behaviors. The whole transcriptome

1,3,5-trinamino-2,4,6-trinitrobenzene (TATB) as an important insensitive high explosive has excellent safety performance due to strong hydrogen bonds. Ionic impurities including sulfate ions (SO42-), nitrate ions (NO3-) and chloride ions (Cl-) formed during the preparation of TATB have negative effects on TATB-based explosives. However, strong hydrogen bonds result in extremely low solubility of TATB in traditional solvents, which poses a huge obstacle to extract and detect the impurities in TATB for quality control. In this study, a green extraction method based deep eutectic solvent (CS-1) w

Background: Baicalin is considered to be able to alleviate the progression of ulcerative colitis (UC), but the underlying molecular mechanism needs to be further elucidated. Methods: TNF-α-induced human normal colorectal mucosa cells (FHC) were used to mimic UC models in vitro , and trinitrobenzene sulfonic acid (TNBS)-injected rats were used to construct UC models in vivo . Cell proliferation and apoptosis were determined by CCK8 assay, EdU assay and flow cytometry. Inflammation factors were examined by ELISA, and ferroptosis-related markers were detected by corresponding kit. The mRNA and pr

PURPOSE: A ketogenic diet (KD), high in fat and low in carbohydrates, induces ketosis characterized by elevated circulating ketone bodies. While both KD and ketone bodies have demonstrated therapeutic potential in various pathophysiological conditions, their effect on inflammatory bowel diseases remains controversial. This study aimed to investigate the impact of a KD and ketone ester (KE), an ingestible form of ketone bodies, on intestinal inflammation.

BACKGROUND & AIMS: Colorectal cancer is a leading cause of cancer death, and a major risk factor is chronic inflammation. Despite the link between colitis and cancer, the mechanism by which inflammation leads to colorectal cancer is not well understood.
METHODS: To investigate whether different forms of inflammation pose the same risk of cancer, we compared several murine models of colitis (dextran sodium sulfate [DSS], 2,4,6-trinitrobenzene sulfonic acid, 4-ethoxylmethylene-2-phenyloxazol-5-one, Citrobacter rodentium, Fusobacterium nucleatum, and doxorubicin) with respect to their ability to

BACKGROUND: 5-hydroxytryptamine 4 receptors (5-HT4 Rs) are expressed in the colonic epithelium, and previous studies have demonstrated that luminal administration of agonists enhances motility, suppresses nociception, and is protective in models of inflammation. We investigated whether stimulation with a luminally acting 5-HT4 R agonist is comparable to previously tested absorbable compounds.
METHODS: The dextran sodium sulfate (DSS), trinitrobenzene sulfonic acid (TNBS), and interleukin 10 knockout (IL-10KO) models of colitis were used to test the protective effects of the luminally acting 5

BACKGROUND: Brain-gut axis dysregulation is observed in inflammatory bowel disease. However, the effect of altered gut flora on neuro- immunomodulation and its role in the pathogenesis of inflammatory bowel disease are unknown. The aims of this study are to determine (i) whether colitis modifies the expression of c-fos, a marker of general neuronal activation in the brain and (ii) whether this activation could be modulated by probiotic bacteria.
METHODS: In this study, 28 Sprague-Dawley rats were divided into 4 groups: colitis-probiotic group, non-colitis-fed-control group receiv- ing probiot

BACKGROUND: Cross-sensitization of pelvic organs is one theory for why symptoms of gut sickness and interstitial cystitis/bladder pain syndrome overlap. Experimental colitis has been shown to trigger bladder hyperactivity and hyperalgesia in rats. The chemokine receptor CXCR4 plays a key role in bladder function and central sensitization. We aim to study the role of CXCR4 and its inhibitor AMD3100 in colon-bladder cross-organ sensitization.
METHODS: The colitis model was established by rectal infusion of trinitrobenzene sulfonic acid. Western blot and immunofluorescence were used to assess th

Recent studies have implicated mitochondrial dysfunction as a trigger of inflammatory bowel diseases, including Crohn's disease (CD) and ulcerative colitis (UC). We have investigated the role of the mitochondria gate-keeper protein, the voltage-dependent-anion channel 1 (VDAC1), in gastrointestinal inflammation and tested the effects of the newly developed VDAC1-interacting molecules, VBIT-4 and VBIT-12, on UC induced by dextran sulfate sodium (DSS) or trinitrobenzene sulphonic acid (TNBS) in mice. VDAC1, which controls metabolism, lipids transport, apoptosis, and inflammasome activation, is o

BACKGROUND: Intestinal fibrosis is a common complication of Crohn's disease (CD). Adiponectin reportedly exerts anti-inflammatory effects in various disease models, including colitis models.
AIMS: In this study, we aimed to determine the effects of adiponectin on intestinal fibrosis and the underlying mechanisms.
METHODS: A murine model of intestinal fibrosis was established by administering increasing doses of 2,4,6-trinitrobenzene sulfonic acid to Balb/c mice via enema for 7 weeks. Primary human fibroblasts were isolated from the colon tissues of patients with CD. The fibroblasts were incu

The prevalence of autoimmune diseases is on the rise globally. Currently, autoimmunity presents in over 100 different forms and affects around 9% of the world's population. Current treatments available for autoimmune diseases are inadequate, expensive, and tend to focus on symptom management rather than cure. Clinical trials have shown that live helminthic therapy can decrease chronic inflammation associated with inflammatory bowel disease and other gastrointestinal autoimmune inflammatory conditions. As an alternative and better controlled approach to live infection, we have identified and ch

BACKGROUND: The accumulation of eosinophils is mediated by the chemokine receptor-3 (CCR3)-eotaxin axis. Increased expression of eotaxin and its receptor is associated with inflammatory bowel disease (IBD). Activation of eosinophils causes the release of cationic proteins that are neurotoxic such as eosinophil-derived neurotoxin (EDN). Damage to enteric neurons alters neurally controlled functions of the gut correlated with intestinal inflammation. We hypothesized that inhibition of the CCR3-eotaxin axis will prevent inflammation-induced functional changes to the gastrointestinal tract.

BACKGROUND: Irritable bowel disease (IBS) is viewed upon as a functional disorder of subclinical inflammatory changes in recent years, and there is no reliable biomarker. Triggering receptor expressed on myeloid cells 1 (TREM-1), also produced in a soluble form (sTREM-1), is involved in the activation of inflammatory cascades of intracellular events and may play a role in pathogenesis of IBS.
AIM: To investigate whether serum sTREM-1 level can be used as a marker of subclinical inflammation in D-IBS.
METHODS: Abdominal pain was quantified by a validated questionnaire. Expression level of TRE

Oenothera rosea L´Hér. ex Ait is a species traditionally used in the treatment of inflammation, headache, stomach pain, infections, among others. The aim of this study was evaluating the acute anti-inflammatory activity of the aqueous extract of O. rosea by 2,4,6-trinitrobenzene sulfonic acid (TNBS)-induced colitis. Rats were randomized into six groups: (I) Sham; (II) EtOH; (III) TNBS; and (IV-VI) 250, 500 and 750 mg/Kg, respectively. The colonic injury was induced (groups III-VI) by intrarectal instillation of 0.25 mL of TNBS (10 mg) in 50% ethanol. Groups I and II received an enema (0.25 mL)

The high-energy compound 3,4,5-trinitropyrazole (TNP) was developed as an alternative to other less energetic and more sensitive explosive materials, in particular 1-methyl-2,4,6-trinitrobenzene (TNT). However, the level of toxicity of TNP remains understudied. Here using an in vivo CD1 mouse model, we mimicked an acute exposure (24 h) to TNP, given either orally or intravenously, and determined the maximum administrable doses (190 mg/kg and 11 mg/kg, respectively), as well as the lethal dose for 50% (LD50) of female or male mice (390 mg/kg for both) treated intravenously with TNP alone.
