Methylation of OPRL1 mediates the effect of psychosocial stress on binge drinking in adolescents.

BACKGROUND: Nociceptin is a key regulator linking environmental stress and alcohol drinking. In a genome-wide methylation analysis, we recently identified an association of a methylated region in the OPRL1 gene with alcohol-use disorders. METHODS: Here, we investigate the biological basis of this observation by analysing psychosocial stressors, methylation of the OPRL1 gene, brain response during reward anticipation and alcohol drinking in 660 fourteen-year-old adolescents of the IMAGEN study. We validate our findings in marchigian sardinian (msP) alcohol-preferring rats that are genetically

A prospective study of genetic factors, human laboratory phenotypes, and heavy drinking in late adolescence.

Subjective responses to alcohol are considered candidate endophenotypes for alcohol use disorder and appear to anticipate future consumption. However, prospective studies have been rare, and laboratory research has typically examined subjective responses absent measures of self-administration. This study examined the association of subjective responses with subsequent laboratory self-administration, also evaluating laboratory phenotypes in relation to putative genetic risk factors [family history (FH) of alcohol dependence and OPRM1 genotype] and subsequent heavy drinking. Participants (N = 61

The Protocol for: Do peers enhance alcohol use outcomes in group motivational interviewing? A translational clinical trial investigating brain synchrony in underage emerging adults during a fMRI hyperscanning task and its association with 12 month alcohol use reductions.

BACKGROUND AND AIMS: Alcohol is the top substance used by US emerging adults under legal drinking age (underage emerging adults; U-EA; ages 18-19). U-EA are unlikely to seek or receive indicated alcohol interventions. It is imperative to find brief, effective interventions to reduce U-EA drinking. Developmental neuroscience reflects that peers hold higher neural salience during adolescence, as evidenced by differential neural response with real and/or simulated peers, even when those peers were not friends, and particularly in the context of alcohol. Peers also activate positive (prosocial) ne

Characterizing the co-occurrence of alcohol experimentation and suicidal thoughts and behaviors in early adolescence.

This study aims to investigate the roles of decision-making processes and genetics in the co-occurrence of alcohol use and suicidal thoughts/behaviors (STB) in adolescence. We used data from the ABCD® study (abcdstudy.org) and included behavioral (computerized tasks, self-report questionnaires) and genetic (polygenic scores [PGS]) measures related to cognitive (executive functions) and affective (delay-discounting, risk-taking, impulsivity) processes involved in decision-making. First, we evaluated the latent structure of decision-making in the full sample (N = 11,868) using a split-half explo

Personality, Social Factors, Brain Functioning, Familial Risk, and Trajectories of Alcohol Misuse in Adolescence.

IMPORTANCE: The development of an alcohol use disorder in adolescence is associated with increased risk of future alcohol dependence. The differential associations of risk factors with alcohol use over the course of 8 years are important for preventive measures. OBJECTIVE: To determine the differential associations of risk-taking aspects of personality, social factors, brain functioning, and familial risk with hazardous alcohol use in adolescents over the course of 8 years. DESIGN, SETTING, AND PARTICIPANTS: The IMAGEN multicenter longitudinal cohort study included adolescents recruited from

Assessment of the influential mechanisms of adolescent risk behaviors and protective and risk factors among high school students in Finnmark, Arctic Norway.

Adolescent risk behaviors, including alcohol use and antisocial behaviors, remain a public health concern in Finnmark, Arctic Norway. However, the mechanisms linking risk and protective factors to these behaviors remain underexplored. This study examines the influence of individual and environmental factors-such as family, peers, school, and local contexts-on adolescent alcohol use and antisocial behaviors. Using data from the 2021 Ungdata survey (N = 2,129 high school students), Partial Least Squares Structural Equation Modeling was applied to identify key associations. The results indicate t

The experience of physiological and psychosocial alcohol-related harms across adolescence and its association with alcohol use disorder in early adulthood: A prospective cohort study.

BACKGROUND: Different forms of alcohol-related harm (e.g., hangovers, fighting) may confer differential risk of clinically relevant alcohol problems. We examine: (i) patterns of transition in experiencing alcohol-related harms across adolescence; (ii) whether factors in early adolescence predict transition patterns; and (iii) whether transition patterns predict later alcohol use disorder (AUD) symptoms. METHODS: We used a longitudinal Australian cohort (n = 1828) to model latent class transition patterns of alcohol-related harms across three timepoints (Mage  = 13.9, 16.8, 18.8 years). Regres

Effects of long-term adolescent alcohol consumption on white matter integrity and their correlations with metabolic alterations.

Alcohol-related white matter (WM) microstructural changes have not been fully elucidated in adolescents. We aimed to investigate influences of subclinical alcohol use during adolescence on WM microstructure and to characterize those with serum metabolic alterations. 35 moderate-to-heavy drinkers (15 males, 20 females) and 27 controls (12 males, 15 females) were selected based on their ten-year Alcohol Use Disorders Identification Test scores measured at three time points. Magnetic resonance imaging was conducted at endpoint time. Whole brain analysis of fractional anisotropy (FA) was performed

Mild or borderline intellectual disability as a risk for alcohol consumption in adolescents - A matched-pair study.

BACKGROUND: Studies that investigate the association between mild or borderline intellectual disability (MBID) and alcohol use in adolescents have not examined whether MBID is an independent risk factor for drinking. AIM: It is important to examine whether MBID is a risk factor for alcohol consumption by controlling concomitant factors in a matched-pair design. METHOD: Overall, 329 students from two schools for children with MBID self-reported their drinking behavior via questionnaires, and 329 students from regular schools were matched to this group by gender, age, family composition, and p

Substance use and sexual orientation among adolescents: Differences by age group and sex in the 2023 National Survey of Drug Use and Health.

BACKGROUND AND OBJECTIVES: Adolescent substance use is a critical public health concern with significant long-term consequences, yet research on disparities by sexual orientation remains limited. Despite the unique stressors faced by sexual minority youth-particularly bisexual individuals-that increase their risk for substance use, most studies investigate disparities among older adolescents and adults. Using recent national data, this study aims to fill this gap and inform targeted prevention and intervention efforts for sexual minority youth. METHODS: We leveraged data from the 2023 Nationa

Historical variation in young adult binge drinking trajectories and its link to historical variation in social roles and minimum legal drinking age.

This study examines historical variation in age 18 to 26 binge drinking trajectories, focusing on differences in both levels of use and rates of change (growth) across cohorts of young adults over 3 decades. As part of the national Monitoring the Future Study, over 64,000 youths from the high school classes of 1976 to 2004 were surveyed at biennial intervals between ages 18 and 26. We found that, relative to past cohorts, recent cohorts both enter the 18 to 26 age band engaging in lower levels and exit the 18 to 26 age band engaging in higher levels of binge drinking. The reason for this rever

Associations between childhood risk factors and alcohol treatment outcomes in adolescence.

BACKGROUND: Early onset alcohol use, mental health problems, familial alcohol use, and adverse childhood experiences (ACEs) increase the likelihood of persistent alcohol-use disorders later in life. This study aimed to determine the relative influence of such risk factors when predicting adolescent alcohol treatment outcomes, an area overlooked in prior research. METHODS: Retrospective cross-sectional analysis using the National Drug Treatment Monitoring System, incorporating data from all publicly funded community alcohol services in England. The study included adolescents (aged 11-17) whose

The role of parental maladaptive emotion socialization in the risk process for negative urgency and drinking behavior in adolescence.

INTRODUCTION: Negative urgency (the tendency to act rashly when experiencing negative emotions) is a robust risk factor for a number of problem behaviors, including early adolescent drinking. Little is known about the factors that precede the development of negative urgency, and hence the full etiology of this component of risk. The current study aimed to investigate the possibility that facets of childhood maladaptive emotion socialization (the tendency for children's expressions of emotions to be met with punishment, minimized, or invoke a reaction of distress from their parents/caretakers)

Alcohol Use Trajectories Before and After Pregnancy Among Adolescent and Young Adult Mothers.

BACKGROUND: Moderate to heavy alcohol use during pregnancy (≥3 drinks/occasion) is linked to a range of adverse offspring outcomes. Prior studies suggest that adolescent and young adult mothers may be particularly vulnerable to these risky drinking behaviors during and after pregnancy. This study used latent class growth analysis (LCGA) to examine changes in risky drinking from prepregnancy to postpregnancy and identified prepregnancy predictors representing cognitive and social factors to inform prevention. METHODS: Participants included 432 adolescent and young adult mothers (aged 13 to 24;
