Breaking a dogma: acute anti-inflammatory treatment alters both post-lesional functional recovery and endogenous adaptive plasticity mechanisms in a rodent model of acute peripheral vestibulopathy.

BACKGROUND: Due to their anti-inflammatory action, corticosteroids are the reference treatment for brain injuries and many inflammatory diseases. However, the benefits of acute corticotherapy are now being questioned, particularly in the case of acute peripheral vestibulopathies (APV), characterized by a vestibular syndrome composed of sustained spinning vertigo, spontaneous ocular nystagmus and oscillopsia, perceptual-cognitive, posturo-locomotor, and vegetative disorders. We assessed the effectiveness of acute corticotherapy, and the functional role of acute inflammation observed after sudde

Transneuronal tracing of neural pathways controlling an abdominal muscle, rectus abdominis, in the ferret.

Abdominal muscles participate in generating a large number of behaviors and reflex responses, including expiration, coughing, sneezing, vomiting, postural control, production of speech, straining, facilitation of venous return to the heart, and reaction to vestibular stimulation. However, the only premotor neurons that have been conclusively shown to influence abdominal motoneurons are located in nucleus retroambiguus, the expiratory region of the caudal ventral respiratory group. In the present study, the neural circuitry controlling the activity of one abdominal muscle, rectus abdominis, was

Behavioral and neural responses to high-strength magnetic fields are reduced in otolith mutant mice.

Static high magnetic fields (MFs) interact with the vestibular system of humans and rodents. In rats and mice, exposure to MFs causes perturbations such as head movements, circular locomotion, suppressed rearing, nystagmus, and conditioned taste aversion acquisition. To test the role of otoconia, two mutant mouse models were examined, head-tilt Nox3het (het) and tilted Otop1 (tlt), with mutations, respectively, in Nox3, encoding the NADPH oxidase 3 enzyme, and Otop1, encoding the otopetrin 1 proton channel, which are normally expressed in the otolith organs, and are critical for otoconia forma

[The adult brain produces new neurons to restore balance after vestibular loss].

Following partial or total loss of peripheral vestibular inputs, a phenomenon called central vestibular compensation takes place in the hours and days following the injury. This neuroplasticity process involves a mosaic of profound rearrangements within the brain stem vestibular nuclei. Among them, the setting of a new neuronal network is maybe the most original and unexpected, as it involves an adult reactive neurogenesis in a brain area not reported as neurogenic so far. Both the survival and functionality of this newly generated neuronal network will depend on its integration to pre-existin

[The significance of the interaural latency difference of VEMP].

OBJECTIVE: To investigate the significance of the interaural latency (IAL) difference of the latency of VEMP and to raise the sensitivity of the test. METHOD: Vestibular evoked myogenic potentials (VEMP) were tested in 20 healthy subjects; 13 patients with acoustic neuromaor cerebellopontile angle occupying lesions and 1 patient with multiple sclerosis. IAL differences of the wave p13,n23 and p13-n23 (abbreviatd as /delta p13/, /delta n23/ and /delta p13-n23/, respectively) were analysed to determine the normal range and the upper limit of the norm data. Four illustrative cases with the abnor

Distribution of androgen and estrogen receptor mRNA-containing cells in the rat brain: an in situ hybridization study.

The distribution of cells that express mRNA encoding the androgen (AR) and estrogen (ER) receptors was examined in adult male and female rats by using in situ hybridization. Specific labeling appeared to be largely, if not entirely, localized to neurons. AR and ER mRNA-containing neurons were widely distributed in the rat brain, with the greatest densities of cells in the hypothalamus, and in regions of the telencephalon that provide strong inputs in the medial preoptic and ventromedial nuclei, each of which is thought to play a key role in mediating the hormonal control of copulatory behavior

Projection neurons of the vestibulo-sympathetic reflex pathway.

Changes in head position and posture are detected by the vestibular system and are normally followed by rapid modifications in blood pressure. These compensatory adjustments, which allow humans to stand up without fainting, are mediated by integration of vestibular system pathways with blood pressure control centers in the ventrolateral medulla. Orthostatic hypotension can reflect altered activity of this neural circuitry. Vestibular sensory input to the vestibulo-sympathetic pathway terminates on cells in the vestibular nuclear complex, which in turn project to brainstem sites involved in the

Vestibular influences on autonomic cardiovascular control in humans.

There is substantial evidence that anatomical connections exist between vestibular and autonomic nuclei. Animal studies have shown functional interactions between the vestibular and autonomic systems. The nature of these interactions, however, is complex and has not been fully defined. Vestibular stimulation has been consistently found to reduce blood pressure in animals. Given the potential interaction between vestibular and autonomic pathways this finding could be explained by a reduction in sympathetic activity. However, rather than sympathetic inhibition, vestibular stimulation has consist

Vestibular primary afferent projection to the cerebellum of the rabbit.

The vestibular primary afferent projection to the cerebellum of the rabbit was studied with retrograde and orthograde tracers. We injected individual lobules of the cerebellum with horseradish peroxidase (HRP) or wheat germ agglutinin-HRP (WGA-HRP). Following these injections the numbers of labeled and unlabeled cells in Scarpa's ganglion were counted. Approximately 64-89% of the cells in Scarpa's ganglion were labeled retrogradely following uvula-nodular injections. About 2% of the cells in the ipsilateral Scarpa's ganglion were labeled after injections of the flocculus. Virtually no cells we

Vestibular compensation after ganglionectomy: ultrastructural study of the tangential vestibular nucleus and behavioral study of the hatchling chick.

The tangential nucleus is a major part of the avian vestibular nuclear complex, and its principal cells are structurally distinctive neurons participating in the vestibuloocular and vestibulocollic reflexes. After unilateral peripheral vestibular lesion, a behavioral recovery of function defined as vestibular compensation is observed. Because sprouting and hypertrophy of synapses have been reported in other regions of immature animals after central nervous system injury, we investigated whether this also occurs in the vestibular nuclei during compensation. To test this hypothesis, unilateral v

Morphology of single vestibulospinal collaterals in the upper cervical spinal cord of the cat: III collaterals originating from axons in the ventral funiculus ipsilateral to their cells of origin.

Some vestibulospinal pathways are composed of a homogeneous collection of axons with similar intraspinal collaterals. Other pathways contain axons whose collaterals vary in terms of shape, distribution, and complexity. The purpose of the present study was to extend the study of homogeneity versus heterogeneity of vestibulospinal axons to vestibulospinal axons that travel in the ventral funiculus ipsilateral to their cells of origin. Collaterals of these axons were stained following extracellular injections of Phaseolus vulgaris-leucoagglutinin in rostral parts of the medial and descending vest

Predictable and pseudo random saccades in patients with acoustic neuroma.

We studied predictive and pseudo random saccades in patients with cerebellopontine angle tumor. Target for the saccades was a laser beam which was reflected on the wall in front of the subject, and the stimulus was controlled by a computer. The fixed target was spaced at a distance of +/- 30 to each side of the subject. The random saccades jumped 20 degrees/40 degrees/60 degrees in an unpredictable manner. The saccade analysis program was based on pattern recognition. In the random saccade test none of the mean values of saccadic reaction time (SRT), saccadic accuracy (SA) and saccadic peak ve

Bilirubin-induced neurotoxic and ototoxic effects in rat cochlear and vestibular organotypic cultures.

Exposure to high levels of bilirubin in hyperbilirubinemia patients and animal models can result in sensorineural deafness. However, the mechanisms underlying bilirubin-induced damage to the inner ear, including the cochlear and vestibular organs, remain unknown. The present analyses of cochlear and vestibular organotypic cultures obtained from postnatal day 3 rats exposed to bilirubin at varying concentrations (0, 10, 50, 100, or 250 μM) for 24 h revealed that auditory nerve fibers (ANFs) and vestibular nerve endings were destroyed even at low doses (10 and 50 μM). Additionally, as the biliru

Ca2+ -activated K+ channels of the BK-type in the mouse brain.

An antibody against the 442 carboxy-terminal amino acids of the BK channel alpha-subunit detects high immunoreactivity within the telencephalon in cerebral cortices, olfactory bulb, basal ganglia and hippocampus, while lower levels are found in basal forebrain regions and amygdala. Within the diencephalon, high density was found in nuclei of the ventral and dorsal thalamus and the medial habenular nucleus, and low density in the hypothalamus. The fasciculus retroflexus and its termination in the mesencephalic interpeduncular nucleus are prominently stained. Other mesencephalic expression sites

Presence of S-100 beta in cholinergic neurones of the rat hindbrain.

The double staining of S-100 beta and choline acetyltransferase (ChAT) revealed that S-100 beta immunoreactivity was localized in most, but not in all, cholinergic neurones in the somatomotor nuclei of the cranial nerves and in the ambiguus nucleus. S-100 beta was present in almost all cholinergic neurones in the brain stem reticular, red, vestibular (excluding medial), mesencephalic trigeminal and cerebellar nuclei. However, S-100 beta immunoreactivity was lacking in cholinergic neurones in the parabrachial complex, the dorsal motor nucleus of the vagal nerve and most sensory nuclei. No S-100

Connections of Purkinje cell axons of lobule X with vestibulocerebellar neurons projecting to lobule X or IX in the rat.

Connections of Purkinje cell axons of lobule X (nodulus) with vestibulocerebellar neurons projecting to lobule X or IX (uvula) were revealed in the rat. Purkinje cell axons were anterogradely labeled with biotinylated dextran (BD) injected into sublobule Xa while vestibular neurons were retrogradely labeled with cholera toxin subunit B (CTB) injected into sublobule Xa or IXc. Labeled terminals of Purkinje cell axons of lobule X were numerous in the superior vestibular nucleus (SV), medial parts of the parvocellular (MVpc) and the caudal part (MVc) of the medial vestibular nucleus (MV), and gro

[The role of glucocorticoids in the post-vibration changes in inhibition mediation in the brain structures].

Low-frequency vertical vibration for 2 hours together with three-fold increase of glucocorticoids is accompanied by the disruption of typical relationships of neuromediator processes (norepinephrine- and serotoninergic mediation) as well as the reduction of the intensity of choline trapping with glucocorticoid perception. Glucocorticoid-related conformation changes in plasma membranes and a drastic increase of the cAMP level provide plasticity of both nonspecific (hypothalamus, raphe bluish sites and nuclei) and specific (Deiters' nuclei) structures. The relationships of neuromediator processe

Progression of Vestibular Deficits Following Moderate-Intensity Blast Overpressure in Rats.

PURPOSE: Military personnel and veterans exposed to blast overpressure waves frequently report vestibular symptoms, including dizziness, vertigo, and imbalance. Although blast exposure is known to disrupt vestibular function, how vestibular deficits develop over time, particularly at the level of vestibular afferent signaling, remains unclear. METHODS: In this study, we used an ear-blast model to investigate the early injury responses and progression of vestibular deficits following a single, moderate-intensity blast exposure (20 PSI) in male and female Long-Evans rats. Vestibular function wa

Intranasal gentamicin irrigation impacts number and morphology of auditory brainstem neurons.

Gentamicin is an aminoglycoside antibiotic that broadly targets Gram-negative bacteria. Gentamicin exhibits ototoxic effects in both human subjects and animal models over several different routes of administration. While gentamicin is primarily vestibulotoxic, it causes sensorineural hearing loss attributed to hair cell damage at the base of the cochlea. Gentamicin can also be administered through intranasal irrigation to treat sinusitis in humans. While this route of delivery is believed to minimize ototoxic effects, we have shown gait ataxia, longer latency cervical vestibular-evoked myogeni
