Coordinated Transport of Nitrate, Potassium, and Sodium.

Potassium (K+) and nitrogen (N) are essential nutrients, and their absorption and distribution within the plant must be coordinated for optimal growth and development. Potassium is involved in charge balance of inorganic and organic anions and macromolecules, control of membrane electrical potential, pH homeostasis and the regulation of cell osmotic pressure, whereas nitrogen is an essential component of amino acids, proteins, and nucleic acids. Nitrate (NO3 -) is often the primary nitrogen source, but it also serves as a signaling molecule to the plant. Nitrate regulates root architecture, st

Effects of iron oxide nanoparticles on Fe and heavy metal accumulation in castor (Ricinus communis L.) plants and the soil aggregate.

In this study, the effects of iron (Fe) nanoparticles (NPs) on Fe and heavy metal accumulations by castor (Ricinus communis L.) plants were investigated. The castor cultivar was planted in the soil contaminated with Pb and Zn for 48 days with Fe2O3 NPs treatments. The Fe and heavy metal concentrations in the plant tissues, the plant tissues' ultrastructures, and the Fe and heavy metal distributions in the soil aggregate were analyzed. The results of this study indicate that there is a mutual promotion relationship between Fe and heavy metals (Pb and Zn). The scanning electron microscopy (SEM)

Complexity and Conservation of Thermospermine-Responsive uORFs of SAC51 Family Genes in Angiosperms.

ACAULIS5 (ACL5) encodes thermospermine synthase in Arabidopsis and its loss-of-function mutant acl5 shows excess xylem differentiation and severe dwarfism. SAC51 encodes a basic helix-loop-helix (bHLH) protein and was identified from sac51-d, a dominant suppressor mutant of acl5, which restores the wild-type phenotype without thermospermine. The 5' leader of the SAC51 mRNA contains multiple upstream open-reading frames (uORFs) and sac51-d has a premature stop codon in the fourth uORF. This uORF is conserved among SAC51 family genes in vascular plants. According to the GUS reporter assay, the S

Silicon nutrition improves growth of salt-stressed wheat by modulating flows and partitioning of Na+, Cl- and mineral ions.

Silicon (Si) is reported to improve salt stress tolerance of cereals, but little is known about the effects of Si on flows and partitioning of sodium (Na+), chloride (Cl-), and essential mineral ions at the tissue and cellular level. Wheat (Triticum aestivum L.) was exposed to 200 mM NaCl for 30 d in hydroponics, with or without 2 mM Si. X-ray microanalysis coupled with scanning electron microscopy (SEM) was used to quantify the cell-specific ion profiles across root and leaf cells, paralleled by measurements of wheat growth and physiological responses. Under salt stress, higher Na+ and Cl- co

Interactions of Sulfate with Other Nutrients As Revealed by H2S Fumigation of Chinese Cabbage.

Sulfur deficiency in plants has severe impacts on both growth and nutrient composition. Fumigation with sub-lethal concentrations of H2S facilitates the supply of reduced sulfur via the leaves while sulfate is depleted from the roots. This restores growth while sulfate levels in the plant tissue remain low. In the present study this system was used to reveal interactions of sulfur with other nutrients in the plant and to ascertain whether these changes are due to the absence or presence of sulfate or rather to changes in growth and organic sulfur. There was a complex reaction of the mineral co

Nitrogen Use Efficiency Is Mediated by Vacuolar Nitrate Sequestration Capacity in Roots of Brassica napus.

Enhancing nitrogen use efficiency (NUE) in crop plants is an important breeding target to reduce excessive use of chemical fertilizers, with substantial benefits to farmers and the environment. In Arabidopsis (Arabidopsis thaliana), allocation of more NO3 (-) to shoots was associated with higher NUE; however, the commonality of this process across plant species have not been sufficiently studied. Two Brassica napus genotypes were identified with high and low NUE. We found that activities of V-ATPase and V-PPase, the two tonoplast proton-pumps, were significantly lower in roots of the high-NUE

Comparative uptake and translocation of pharmaceutical and personal care products (PPCPs) by common vegetables.

Reuse of treated wastewater to irrigate agricultural crops is increasing in many arid and semi-arid areas around the world. The presence of numerous pharmaceutical and personal care products (PPCPs) in treated wastewater and their potential transfer into food produce such as vegetables poses an unknown human health risk. The goal of this study was to identify PPCPs that have a comparatively high potential for plant uptake and translocation. A total of 20 frequently-occurring PPCPs were compared for their accumulation into four staple vegetables (lettuce, spinach, cucumber, and pepper) grown in

Retention sites for Xylella fastidiosa in four sharpshooter vectors (Hemiptera: Cicadellidae) analyzed by scanning electron microscopy.

Xylella fastidiosa is a xylem-limited bacterium that causes citrus variegated chlorosis (CVC), Pierce's disease of grapevine, and leaf scald of coffee and plum and many other plant species. This pathogen is vectored by sharpshooter leafhoppers (Hemiptera: Cicadellidae: Cicadellinae) and resides in the insect foregut. Scanning electron microscopy was used to determine the retention sites of X. fastidiosa for the most common vector species in Brazilian citrus groves, Acrogonia citrina, Bucephalogonia xanthophis, Dilobopterus costalimai, and Oncometopia facialis. The Arabidopsis cinnamoyl CoA reductase irx4 mutant has a delayed but coherent (normal) program of lignification.

Previous studies have indicated that the Arabidopsis thalianairregular xylem 4 (irx4) mutant is severely lignin-deficient, forming abnormal lignin from aberrant monomers. Studies of lignin structure in dwarfed cinnamoyl CoA reductase (CCR)-downregulated tobacco were also previously reported to incorporate feruloyl tyramine derivatives. The lignin in the Arabidopsis irx4 mutant was re-investigated at 6 weeks and at maturation (9 weeks). Systemic response to aphid infestation by Myzus persicae in the phloem of Apium graveolens.

Little is known about the molecular processes involved in the phloem response to aphid feeding. We investigated molecular responses to aphid feeding on celery (Apium graveolenscv. Dulce) plants infested with the aphid Myzus persicae, as a means of identifying changes in phloem function. We used celery as our model species as it is easy to separate the phloem from the surrounding tissues in the petioles of mature leaves of this species. We generated a total of 1187 expressed sequence tags (ESTs), corresponding to 891 non-redundant genes. We analysed these ESTs in silico after cDNA macroarray hy

Loss of Arabidopsis ferroxidases LPR1 and LPR2 enhances plant immunity by promoting leaf iron deposition.

Iron (Fe) is a critical nutrient for both plants and their pathogens, and its distribution and homeostasis critically influence the outcome of plant-pathogen interactions. Low Phosphate Root 1 (LPR1) and LPR2 are ferroxidases that regulate Fe translocation in the xylem, but their role in plant immunity remains unknown. Here, we report that loss of LPR1 and LPR2 in Arabidopsis thaliana confers enhanced resistance to both virulent (Pseudomonas syringae pv. Tomato DC3000, Pst) and avirulent (Pst-avrRpt2) bacterial strains. This enhanced resistance is associated with excessive Fe deposition in the

Fraxinus mandshurica SPL2 enhances tolerance to drought stress by activating the phenylpropanoid pathway.

Drought is a major abiotic stressor that severely threatens forest tree growth and survival worldwide. The SQUAMOSA promoter binding protein-like (SPL) family is a group of plant-specific transcription factors that play important roles in plant growth, development, and environmental adaptation. Our study showed that overexpression of Fraxinus mandshurica SPL2 (FmSPL2) promotes lignin biosynthesis and enhances drought tolerance. Phenotypic analysis indicated that the stomatal aperture of FmSPL2-OE1 and FmSPL2-OE2 was reduced by 12.05% and 19.23%, respectively, compared to the wild type (WT). Ad

Magnaporthiopsis maydis reduces growth in maize plantlets without affecting root resistance to water flow.

Late wilt is a disease caused by the soil-borne fungus Magnaporthiopsis maydis that severely limits the production of maize in countries with warm to hot climates. A particular characteristic of this disease is that, although the pathogen penetrates the roots in the first weeks after sowing, the symptoms appear suddenly around flowering. Additionally, even with air temperatures and relative humidity that are optimal for disease development during the crop cycle, adequate soil water content results in undetected disease. Overall, physiological alterations caused by M. maydis in maize are poorly

Low-drift microneedle ion-selective electrodes for in situ monitoring of Mg2+ in plants.

Real-time in situ monitoring of magnesium ions (Mg2+) concentration in plants is essential for understanding ion transport and advancing precision agriculture. However, real-time in situ monitoring using all-solid-state ion-selective electrodes (ASS-ISEs) remains challenging because a water-layer tends to form at the interface between the sensing membrane and the metallic substrate, which leads to substantial potential drift. Here, we present an interfacial-engineering strategy for the microneedle electrode fabrication process, along with a method that enables accurate and stable in situ monit

A brassinosteroid signaling protein acts as an integrative regulator of K+ and Na+ transport in rice response to salt stress.

Maintaining the cytosolic K+/Na+ homeostasis is crucial for plants' salt tolerance. Therefore, elucidating the regulatory mechanism governing K+/Na+ transport is pivotal to engineering salt-tolerant crops. Although many regulators have been shown specifically to modulate either Na+ or K+ transporters, little is known about integrative regulators that orchestrate the transport of both cations. In this study, we reveal that a rice basic helix-loop-helix (bHLH) transcription factor, BES1-INTERACTING MYC-LIKE1 (OsBIM1), negatively regulates K+/Na+ homeostasis under salt stress. Loss of OsBIM1 func

Ethylene response factor 113 regulates the growth and development of poplar through the tryptophan-dependent auxin metabolism.

ERF transcription factors play a pivotal role in orchestrating plant growth and development processes. In this study, PagERF113 was isolated from the 84 K poplar (Populus alba × P. glandulosa) and its function was detected using molecular and genetic methods. Subcellular localization and trans-activation assays indicated that PagERF113 lacks transcriptional activation capability, residing in both the cell membrane and nucleus. Phenotypic analyses revealed that transgenic poplar lines overexpressing PagERF113 (OE) exhibited weedy growth compared to wild-type (WT), characterized by small leaves,

Fungicidal activity of curcumol against Phytophthora capsici via inhibiting phosphatidylcholine biosynthesis and its systemic translocation in plants.

Phytophthora capsici is an infamously soil-borne pathogen that poses a serious threat to agricultural production. Curcumol is a natural plant-derived sesquiterpene lactone, whose antimicrobial effect against plant pathogens remains unclear. In this study, curcumol exhibited pronounced antifungal activity against a diverse range of plant pathogens, particularly against plant pathogenic oomycetes, which including P. capsici, Phytophthora infestans, Phytophthora parasitica, and Phytophthora sojae. The median effective concentration values of curcumol against P. capsici for spore germination and m

Vessel- and ray-specific monolignol biosynthesis as an approach to engineer fiber-hypolignification and enhanced saccharification in poplar.

Lignin is one of the main factors determining recalcitrance to processing of lignocellulosic biomass towards bio-based materials and fuels. Consequently, wood of plants engineered for low lignin content is typically more amenable to processing. However, lignin-modified plants often exhibit collapsed vessels and associated growth defects. Vessel-specific reintroduction of lignin biosynthesis in dwarfed low-lignin cinnamoyl-CoA reductase1 (ccr1) Arabidopsis mutants using the ProSNBE:AtCCR1 construct overcame the yield penalty while maintaining high saccharification yields, and showed that monoli

Direct and Bicarbonate-Induced Iron Deficiency Differently Affect Iron Translocation in Kiwifruit Roots.

Bicarbonate-induced iron (Fe) deficiency (+Bic) is frequently observed in kiwifruit orchards, but more research attention has been paid to direct Fe deficiency (-Fe) in plants, including kiwifruit. Here we compared the differences of kiwifruit plants between -Fe and +Bic in: (1) the traits of 57Fe uptake and translocation within plants, (2) Fe forms in roots, and (3) some acidic ions and metabolites in roots. The concentration of 57Fe derived from nutrient solution (57Fedfs) in roots was less reduced in +Bic than -Fe treatment, despite similar decrease in shoots of both treatments. +Bic treatm
