Conserved IL-2Rγc Signaling Mediates Lymphopoiesis in Zebrafish.

The IL-2 receptor γ common (IL-2Rγc) chain is the shared subunit of the receptors for the IL-2 family of cytokines, which mediate signaling through JAK3 and various downstream pathways to regulate lymphopoiesis. Inactivating mutations in human IL-2Rγc result in SCID, a primary immunodeficiency characterized by greatly reduced numbers of lymphocytes. This study used bioinformatics, expression analysis, gene ablation, and specific pharmacologic inhibitors to investigate the function of two putative zebrafish IL-2Rγc paralogs, il-2rγc.a and il-2rγc.b, and downstream signaling components during ea

Low temperature mitigates cardia bifida in zebrafish embryos.

The coordinated migration of bilateral cardiomyocytes and the formation of the cardiac cone are essential for heart tube formation. We investigated gene regulatory mechanisms involved in myocardial migration, and regulation of the timing of cardiac cone formation in zebrafish embryos. Through screening of zebrafish treated with ethylnitrosourea, we isolated a mutant with a hypomorphic allele of mil (s1pr2)/edg5, called s1pr2(as10) (as10). Mutant embryos with this allele expressed less mil/edg5 mRNA and exhibited cardia bifida prior to 28 hours post-fertilization. Although the bilateral hearts

Cyp1a reporter zebrafish reveals target tissues for dioxin.

2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) is the unintentional byproduct of various industrial processes, is classified as human carcinogen and could disrupt reproductive, developmental and endocrine systems. Induction of cyp1a1 is used as an indicator of TCDD exposure. We sought to determine tissues that are vulnerable to TCDD toxicity using a transgenic zebrafish (Danio rerio) model. We inserted a nuclear enhanced green fluorescent protein gene (EGFP) into the start codon of a zebrafish cyp1a gene in a fosmid clone using DNA recombineering. The resulting recombineered fosmid was then used t

Forkhead transcription factor foxe1 regulates chondrogenesis in zebrafish.

Forkhead transcription factor (Fox) e1 is a causative gene for Bamforth-Lazarus syndrome, which is characterized by hypothyroidism and cleft palate. Applying degenerate polymerase chain reaction using primers specific for the conserved forkhead domain, we identified zebrafish foxe1 (foxe1). Foxe1 is expressed in the thyroid, pharynx, and pharyngeal skeleton during development; strongly expressed in the gill and weakly expressed in the brain, eye, and heart in adult zebrafish. A loss of function of foxe1 by morpholino antisense oligo (MO) exhibited abnormal craniofacial development, shortening

Probing pineal-specific gene expression with transgenic zebrafish.

The pineal gland of zebrafish (Danio rerio) contains light-sensitive photoreceptor cells and plays an important role in the neuroendocrine system. The zebrafish exorhodopsin gene encodes a pineal-specific photoreceptive protein, whose promoter region harbors a cis-acting element, pineal expression-promoting element (PIPE), directing pineal-specific gene expression. For in vivo genetic studies on PIPE-binding proteins and their regulatory mechanisms, we generated a transgenic zebrafish line, Tg(P(20)-rh/P:gfp), that expresses green fluorescent protein (GFP) under the control of the zebrafish rh

Myc-induced T cell leukemia in transgenic zebrafish.

The zebrafish is an attractive model organism for studying cancer development because of its genetic accessibility. Here we describe the induction of clonally derived T cell acute lymphoblastic leukemia in transgenic zebrafish expressing mouse c-myc under control of the zebrafish Rag2 promoter. Visualization of leukemic cells expressing a chimeric transgene encoding Myc fused to green fluorescent protein (GFP) revealed that leukemias arose in the thymus, spread locally into gill arches and retro-orbital soft tissue, and then disseminated into skeletal muscle and abdominal organs. Leukemic cell

Production of triploid transgenic zebrafish, Brachydanio rerio (Hamilton).

Eggs of zebrafish Brachydanio rerio were heat-shocked just after fertilisation to induce triploidy and subsequently subjected to microinjection with the linear recombinant plasmid containing the Rous Sarcoma Virus-Long Terminal Repeat (RSV-LTR) promoter linked to the rainbow trout (Oncorhynchus mykiss) growth hormone (GH)cDNA(pRSVrtGH). Slot and Southern analyses showed the extra chromosomal transfer of pRSVrtGH. Expression of pRSVrtGH was detected in the triploid transformants by RNA slot, Northern and Western analyses.

Zebrafish embryos are robust to local mechanical perturbations.

The investigation of morphogenesis during organism development has massively benefited from the interaction between developmental biology and biophysics, which gave new quantitative insights into nature's physical working principles. The process of zebrafish epiboly provides a beautiful model system as cells exhibit collective migration, meanwhile requiring symmetry breaking for gastrulation and the subsequent tissue differentiation. These elegantly robust processes are facilitated by both biochemical and mechanical cues. Here we test the hypothesis that the future body axis is robust against

Metabolism-Associated Hepatotoxicity of Gatifloxacin in Zebrafish Larvae.

Gatifloxacin (GTFX), a fourth-generation fluoroquinolone, causes metabolic disturbances in mammals, but its hepatotoxic mechanisms in aquatic vertebrates remain unclear. This study investigated whether GTFX induces liver injury in zebrafish larvae through oxidative stress or alternative pathways. Larvae at 3 days post-fertilization were exposed to 0.2-2.3 mg/mL GTFX for 48 h. Liver morphology, histopathology, intracellular reactive oxygen species (ROS), and expression of lipid metabolism (pparg) and xenobiotic biotransformation genes (cyp1a, cyp1b1) were assessed. GTFX exposure caused concentr

In Vivo Imaging of Zebrafish Larvae Heart Regeneration.

Cardiovascular disease is the leading cause of death globally, so learning about regeneration in the heart is a critical research topic. In the past, cardiomyocytes were thought to be terminally differentiated, and thus, the heart as an organ was thought to lack significant repair capacity. But newer data contradict it, depicting the MI and repair processes in a more active manner. Although histopathology can provide crucial structural information, it is limited to static snapshots of tissue structure; in vivo imaging, on the other hand, allows direct follow-up of regeneration kinetics. Synchronization of behavioral and cardiac dynamics in larval zebrafish.

Animals reprioritize behavioral goals in response to internal physiological states. Using larval zebrafish, we investigated whether engagement with a visuomotor task, the optomotor response (OMR), is coupled to cardiac dynamics. We discovered that threats lead to tachycardia that is synchronized with behavioral suppression. The change in heart rate is represented in the activity of specific neuronal populations. Severing the input to the sympathetic ganglia or ablating the vagus nerve revealed that the threat-related changes to behavioral state do not require interoceptive pathways. Direct tac

Balancing Discovery and Caution: Regeneration in zebrafish toxicology.

Zebrafish (Danio rerio) have emerged as a cornerstone of modern toxicology, providing unique advantages including strong genetic similarity to humans, experimental accessibility, and well-characterized behavioral assays. A defining feature of zebrafish biology is their remarkable regenerative capacity, which enables repair of fins, retina, spinal cord, liver, heart, and even brain regions. This regenerative ability supports important discoveries in developmental and regenerative biology, yet it also presents a limitation for toxicology. Tissue repair and functional recovery may mask persistent

Zebrafish as a translational filter for natural sleep modulators.

The escalating global burden of sleep disorders and their tight association with cognitive decline, metabolic abnormalities, and psychiatric comorbidities constitute a major public health challenge. Long term use of current chemically synthesized sedative hypnotics is constrained by dependence, tolerance, and cognitive side effects, creating an urgent demand for safer, mechanism traceable nutritional intervention strategies. Plant derived natural products, owing to their structural diversity, broad spectrum target potential, and favorable safety profiles, represent a critical reservoir of next

The novel role of foxi3 in zebrafish mandibular development.

Recent studies have identified pathogenic variants in the FOXI3 gene associated with craniofacial microsomia pedigrees. In zebrafish, the foxi1 gene is considered a functional homolog of the mouse Foxi3. However, research on foxi3a and foxi3b, which display homologous genes in the naming of FOXI3 in zebrafish, has predominantly focused on their roles in epidermal ionocyte function. Our study reveals that disruption of foxi3a or foxi3b results in a reduced number of cranial neural crest cells (CNCCs) and hypoplastic mandibular cartilage in zebrafish. These findings introduce a new perspective o

Tssk6 is essential for the regulation of male zebrafish fertility.

Protein kinases are critical regulators of cellular functions and play a pivotal role in male reproduction. In this study, we investigated the role and regulatory mechanisms of the testis-specific serine/threonine kinase 6 (tssk6) in zebrafish. We found that tssk6 gene is highly expressed in the testis of adult male zebrafish, with its protein sequence showing high homology to those of mice and humans, highlighting its evolutionary conservation across vertebrates and suggesting an ancient, critical role in male reproduction. By constructing tssk6 mutants, we demonstrated that deletion of this

Cd248a regulates pericyte development and viability in zebrafish.

CD248 is a pericyte marker during embryonic and tumor neovascularization. Although its expression pattern and function in mammalian pericytes have been extensively studied, its role in zebrafish pericytes remains largely unexplored. In this study, we identify that among the two zebrafish orthologs of human CD248, cd248a, rather than cd248b, is predominantly expressed in pericytes during embryonic development. We generate cd248a and cd248b mutant zebrafish (cd248acc11/cc11 and cd248bcc12/cc12) and observe a significant reduction in pericyte numbers in cd248acc11/cc11 mutants, accompanied by dis

Development of Zebrafish model for Iron Induced Neuroinflammation.

Zebrafish models have been used to research Alzheimer's disease and other neurodegenerative disorders because of their similarities to the human genetic composition and behavior. Researchers have detected iron accumulation in the post-mortem brain sections of neurodegenerative disorder patients. Therefore, the development an animal model to simulate these clinical pathological findings is important. Iron is an important metal for maintaining homeostasis in the brain, depletion and accumulation of iron hamper neuronal development. Given the importance of iron overload in cognition impairment, t

Assessing the toxicity of bongkrekic acid in zebrafish embryos.

Bongkrekic acid (BA) is a potent bacterial toxin found in certain fermented foods, which poses a serious risk to human health through rapid multi-organ failure. Its toxic effects on the early stage of vertebrates remain poorly understood. In this study, we addressed this gap using zebrafish embryos as a model to evaluate BA's developmental toxicity. Following 96-h exposure to BA at concentrations of 0.01, 0.05, 0.1, and 1 mg/L, mortality rates increased sharply between 0.05 mg/L (25 %) and 0.1 mg/L (79 %). Morphological assessments at BA concentrations ≥0.1 mg/L revealed significant reductions

Fold-and-fuse neurulation in zebrafish requires vangl2.

Shaping of the future brain and spinal cord during neurulation is an essential component of early vertebrate development. In amniote embryos, primary neurulation occurs through a "fold-and-fuse" mechanism by which the edges of the neural plate fuse into the hollow neural tube. Failure of neural fold fusion results in neural tube defects (NTDs), which are among the most devastating and common congenital anomalies worldwide. Unlike amniotes, the zebrafish neural tube develops largely via formation of a solid neural keel that later cavitates to form a midline lumen. Although many aspects of prima

polr2i is Required for Zebrafish Early Cardiac Development.

BACKGROUND: Congenital heart disease (CHD) is characterized by structural and functional anomalies of the heart and major blood vessels present at birth. It is recognized as the most common congenital defect. Epidemiological studies highlight the substantial contribution of genetic factors to CHD pathogenesis. In our previous study, RNA polymerase II subunit I (POLR2I protein) was identified as a candidate genetic contributor to CHD. However, its functional role remains largely unexplored. METHODS: First, we performed bioinformatics analyses to evaluate the evolutionary conservation of the PO
