Comprehensive behavioral phenotyping of male Sulf1-deficient mice reveals reduced locomotor activity, altered forelimb gait, and reduced prepulse inhibition.

Sulfatase 1 (Sulf1) is an extracellular sulfatase that regulates multiple signaling pathways by removing 6-O-sulfate from heparan sulfate. Although Sulf1 has established roles in development and cancer, its functions in the adult brain remain incompletely characterized. Recently we reported that Sulf1 is expressed in the nucleus accumbens, overlapping with dopamine D1 and D2 receptors, and that Sulf1 knockout (KO) mice showed altered reward and aversion learning. However, given that Sulf1 is expressed in many other brain regions, including the prefrontal cortex, midline thalamic nuclei, etc.,

Unilateral tactile agnosia as an onset symptom of corticobasal syndrome.

Tactile agnosia is the inability to recognize objects via haptic exploration, in the absence of an elementary sensory deficit. Traditionally, it has been described as a disturbance in extracting information about the physical properties of objects ("apperceptive agnosia") or in associating object representation with its semantic meaning ("associative agnosia"). However, tactile agnosia is a rare and difficult-to-diagnose condition, due to the frequent co-occurrence of sensorimotor symptoms and the lack of consensus on the terminology and assessment methods. Among tactile agnosia classification

Functional network disorganization and cognitive decline following fractionated whole-brain radiation in mice.

Cognitive dysfunction following radiotherapy (RT) is one of the most common complications associated with RT delivered to the brain, but the precise mechanisms behind this dysfunction are not well understood, and to date, there are no preventative measures or effective treatments. To improve patient outcomes, a better understanding of the effects of radiation on the brain's functional systems is required. Functional magnetic resonance imaging (fMRI) has shown promise in this regard, however, compared to neural activity, hemodynamic measures of brain function are slow and indirect. Understandin

Hindlimb motor responses to unilateral brain injury: spinal cord encoding and left-right asymmetry.

Mechanisms of motor deficits (e.g. hemiparesis and hemiplegia) secondary to stroke and traumatic brain injury remain poorly understood. In early animal studies, a unilateral lesion to the cerebellum produced postural asymmetry with ipsilateral hindlimb flexion that was retained after complete spinal cord transection. Here we demonstrate that hindlimb postural asymmetry in rats is induced by a unilateral injury of the hindlimb sensorimotor cortex, and characterize this phenomenon as a model of spinal neuroplasticity underlying asymmetric motor deficits. After cortical lesion, the asymmetry was

Longitudinal characterization of cognitive and motor deficits in an excitotoxic lesion model of striatal dysfunction in non-human primates.

As research progresses in the understanding of the molecular and cellular mechanisms underlying neurodegenerative diseases like Huntington's disease (HD) and expands towards preclinical work for the development of new therapies, highly relevant animal models are increasingly needed to test new hypotheses and to validate new therapeutic approaches. In this light, we characterized an excitotoxic lesion model of striatal dysfunction in non-human primates (NHPs) using cognitive and motor behaviour assessment as well as functional imaging and post-mortem anatomical analyses. NHPs received intra-str

Acoustic evaluation of short-term effects of repetitive transcranial magnetic stimulation on motor aspects of speech in Parkinson's disease.

Hypokinetic dysarthria in Parkinson's disease (PD) can be characterized by monotony of pitch and loudness, reduced stress, variable rate, imprecise consonants, and a breathy and harsh voice. Using acoustic analysis, we studied the effects of high-frequency repetitive transcranial magnetic stimulation (rTMS) applied over the primary orofacial sensorimotor area (SM1) and the left dorsolateral prefrontal cortex (DLPFC) on motor aspects of voiced speech in PD. Twelve non-depressed and non-demented men with PD (mean age 64.58 ± 8.04 years, mean PD duration 10.75 ± 7.48 years) and 21 healthy age-mat

The biochemical basis of the neurobehavioral abnormalities in the Lesch-Nyhan syndrome: a hypothesis.

Lesch-Nyhan syndrome (LNS) is a rare X-recessive disorder that leads to virtually complete deficiency of the purine salvage enzyme hypoxanthine-guanine phosphoribosyltransferase (HPRT). Partial HPRT deficiency results in uric acid overproduction with subsequent hyperuricemia, nephrolithiasis, renal failure and gouty arthritis. In contrast, at complete HPRT deficiency, besides overproduction of uric acid neurological problems appear including spasticity, choreoathetosis, mental retardation, and compulsive self-mutilation. The cause for the uric acid overproduction has been clarified, but the co

The effect of nitric oxide synthase inhibition on acute platelet accumulation and hemodynamic depression in a rat model of thromboembolic stroke.

The relative importance of hemodynamic factors in the pathogenesis of thrombotic or embolic stroke is unclear. Of particular therapeutic interest are those substances that facilitate vasodilation and the clearance of platelet aggregates in the compromised microvasculature. A likely contributor to these functions is nitric oxide because it is known to inhibit platelet aggregability and promote vascular relaxation. To investigate the involvement of nitric oxide in the hemodynamic changes after experimental ischemia, photochemically induced nonocclusive common carotid artery thrombosis (CCAT) was

Neural correlates of Parkinson's improvements after combined digital-levodopa therapy: a pilot study.

Parkinson's disease is characterized by progressive degeneration of dopaminergic neurons within the nigrostriatal pathway, leading to motor and non-motor deficits that become less responsive to chronic dopaminergic pharmacotherapy. Here, we examined whether a multimodal mobile digital intervention, DopApp™, delivering adaptive daily training across sensorimotor, psychological and rehabilitation domains, can induce functional reorganization of thalamocortical motor and limbic networks and augment dopaminergic treatment effects in Parkinson's disease. To test this hypothesis, we conducted a pilo

Functional brain alterations associated with acupuncture for chronic pain: a scoping review of fMRI studies.

BACKGROUND: Chronic pain (CP) is a public health challenge recognized as involving large-scale functional brain dysregulation. Acupuncture is widely used as a non-pharmacological intervention for CP, yet its central mechanisms remain incompletely understood. fMRI provides an approach for investigating acupuncture-related brain alterations in CP. METHODS: Eight databases were searched from inception to March 27, 2025 for fMRI studies investigating acupuncture's central effects in CP. Eligible studies included randomized controlled trials and observational studies involving migraine, knee osteo

Motor-related reorganization in left inferior frontal gyrus after subcortical stroke: neurochemical basis and response to targeted neuromodulation.

BACKGROUND: The left inferior frontal gyrus (IFG) is implicated in both language and motor processes. Its functional reorganization post-stroke, particularly in motor dysfunction without aphasia, remains poorly understood. We investigated effective connectivity (EC) alterations of the left IFG and their neurochemical basis, and responsiveness to neuromodulation in subcortical stroke. METHODS: Cross-sectional analysis included 32 left (LSS) and 27 right (RSS) stroke patients and 40 healthy controls (HCs). Seed-based EC of the left IFG was derived from resting-state fMRI and was compared betwee

Inhibition of Cortical Evoked Responses to Sound Pulses by Preceding Silent Gaps.

PURPOSE: The basic principle of sensorimotor gating (SMG) relies on the ability of a weak lead stimulus (such as a pre-pulse) to inhibit a startling effect of a following, more intense, abrupt stimulus-the so-called pre-pulse inhibition (PPI) paradigm. PPI has been used for near half a century as a means to investigate psychiatric disorders in which its disruption is a surrogate for altered SMG in schizophrenia. However, the blinking response is very variable, making it a poor outcome measure at the individual level. Unlike PPI, which is regulated in the lateral globus pallidus from the basal

Literature Review on Lesion-Symptom Mapping and Deep Brain Stimulation for Poststroke Spasticity: Restoring a Dysfunctional Network?

INTRODUCTION: The present study aimed to define a structural network of stroke-induced and spasticity-related lesions and to relate this network to target sites and reported effects of deep brain stimulation (DBS) to treat poststroke spasticity. METHODS: The Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines were followed (online suppl. Table 2). We performed two separate systematic literature reviews, collecting data from previously published voxel-based lesion-symptom mapping (VLSM) studies for poststroke spasticity patients searching the Medline database

Transdiagnostic Network Localization of Social, Language, and Motor Symptoms in Patients With Frontotemporal Lobar Degeneration.

BACKGROUND AND OBJECTIVES: Frontotemporal lobar degeneration (FTLD) includes different clinical syndromes with distinct patterns of symptoms and neuroanatomical locations of neurodegeneration. However, FTLD is clinically heterogeneous (with overlapping symptoms across several domains) and neuroanatomically heterogeneous (with brain atrophy in different locations in different patients). Traditional methods struggle to fully account for this heterogeneity. In this study, we use a relatively new neuroimaging approach, atrophy network mapping, to localize clinical symptoms in patients with FTLD to

A Multimodal Meta-Analytical Evidence of Functional and Structural Brain Abnormalities Across Alzheimer's Disease Spectrum.

BACKGROUND: Numerous neuroimaging studies have reported that Alzheimer's disease (AD) spectrum have been linked to alterations in intrinsic functional activity and cortical thickness (CT) of some brain areas. However, the findings have been inconsistent and the correlation with the transcriptional profile and neurotransmitter systems remain largely unknown. METHODS: We conducted a meta-analysis to identify multimodal differences in the amplitude of low-frequency fluctuation (ALFF)/fractional ALFF (fALFF) and CT in patients with AD and preclinical AD compared to healthy controls (HCs), using t

Connectivity by the Frontal Aslant Tract (FAT) Explains Local Functional Specialization of the Superior and Inferior Frontal Gyri in Humans When Choosing Predictive over Reactive Strategies: A Tractography-Guided TMS Study.

Predictive and reactive behaviors represent two mutually exclusive strategies in a sensorimotor task. Predictive behavior consists in internally estimating timing and features of a target stimulus and relies on a cortical medial frontal system [superior frontal gyrus (SFG)]. Reactive behavior consists in waiting for actual perception of the target stimulus and relies on the lateral frontal cortex [inferior frontal gyrus (IFG)]. We investigated whether SFG-IFG connections by the frontal aslant tract (FAT) can mediate predictive/reactive interactions. In 19 healthy human volunteers, we applied o

Distinct Functional and Structural Connectivity of the Human Hand-Knob Supported by Intraoperative Findings.

Fine motor skills rely on the control of hand muscles exerted by a region of primary motor cortex (M1) that has been extensively investigated in monkeys. Although neuroimaging enables the exploration of this system also in humans, indirect measurements of brain activity prevent causal definitions of hand motor representations, which can be achieved using data obtained during brain mapping in tumor patients. High-frequency direct electrical stimulation delivered at rest (HF-DES-Rest) on the hand-knob region of the precentral gyrus has identified two sectors showing differences in cortical excit

Rhythmic neural activity is comodulated with short-term gait modifications during first-time use of a dummy prosthesis: a pilot study.

BACKGROUND: After transfemoral amputation, many hours of practice are needed to re-learn walking with a prosthesis. The long adaptation process that consolidates a novel gait pattern seems to depend on cerebellar function for reinforcement of specific gait modifications, but the precise, step-by-step gait modifications (e.g., foot placement) most likely rely on top-down commands from the brainstem and cerebral cortex. The aim of this study was to identify, in able-bodied individuals, the specific modulations of cortical rhythms that accompany short-term gait modifications during first-time use

Altered intra- and inter-network functional connectivity in patients with persistent postural-perceptual dizziness.

BACKGROUND: Persistent postural-perceptual dizziness (PPPD) is a functional vestibular disorder characterized by persistent dizziness, unsteadiness, and non-spinning vertigo. It is the most common cause of chronic vestibular syndrome, but its pathogenesis is currently unclear. Recent studies have indicated that sensory integration may be altered in PPPD patients. OBJECTIVE: Using independent component analysis (ICA) combined with seed-based functional connectivity analysis, we aimed to analyze changes in brain network functional connectivity (FC) in PPPD patients during the resting state and

G-Causality Brain Connectivity Differences of Finger Movements between Motor Execution and Motor Imagery.

Motor imagery is one of the classical paradigms which have been used in brain-computer interface and motor function recovery. Finger movement-based motor execution is a complex biomechanical architecture and a crucial task for establishing most complicated and natural activities in daily life. Some patients may suffer from alternating hemiplegia after brain stroke and lose their ability of motor execution. Fortunately, the ability of motor imagery might be preserved independently and worked as a backdoor for motor function recovery. The efficacy of motor imagery for achieving significant recov
