In vitro activity of pineapple extracts (Ananas comosus, Bromeliaceae) on Rhipicephalus (Boophilus) microplus (Acari: Ixodidae).

Measures to control the cattle tick, Rhipicephalus (Boophilus) microplus, based only on chemical products are becoming unsustainable, mainly because of the development of resistance. The objective of this study was to test the effect of the aqueous extract of pineapple skin (AEPS) and bromelain extracted from the stem (Sigma-Aldrich®, B4882) on engorged females and larvae of R. (B.) microplus in vitro. These substances were diluted in water and evaluated at eight concentrations. Engorged females were collected and distributed in groups of 10, with three repetitions for each treatment. After im

Innate immunity in wooless lamb to larvae of Amblyomma cajennense tick (Fabricius, 1787) (Acari: Ixodidae).

The Cayenne tick Amblyomma cajennense infests preferably horses in its adult form but other mammal species in its immature stages and is the main vector of Rickettsia rickettsii, the Brazilian spotted fever pathogen. As wooless lambs are often raised on pasture together with horses, an experiment was performed to investigate their possible acquisition of resistance to A. cajennense after experimental infestations. Seven naïve wooless lambs were infested thrice at 60 days interval with immature and adult instars of A. cajennense from a laboratory colony, the tick biotic potential being determin

Possible acquired resistance of dogs successively infested by Amblyomma cajennense (Fabricius, 1787) (Acari: Ixodidae) nymphs.

The present study aimed to evaluate the occurrence of immune resistance in dogs successively infested with Amblyomma cajennense nymphs. Five animals were submitted to four consecutive infestations with A. cajennense nymphs, at fourteen-day intervals. For each infestation, 50 nymphs were used per animal and data on the parasitic and non-parasitic periods were recorded. The average recovering rate of engorged nymphs in the successive infestations were 52.0, 29.2, 9.6 and 12.8%, respectively, with a significant reduction (p<0.05) of this parameter from the second infestation onwards. The modal dr

Babesia microti-like rodent parasites isolated from Ixodes persulcatus (Acari: Ixodidae) in Heilongjiang Province, China.

A Babesia microti-like rodent parasite was isolated from the tick, Ixodes persulcatus, collected from the northern forest area of Heilongjiang province, China. The collected I. persulcatus were allowed to feed on specific pathogen-free SCID mice and red blood cells from the mice were used to isolate Babesia spp. with the microareophilous stationary-phase culture technique. Paired and tetrad forms of merozoites were observed by light microscope in red blood cells of SCID mice. In vitro growth of the parasites was also achieved in mice erythrocytes, which indicated the presence of Babesia spp. i

Defining plasmids required by Borrelia burgdorferi for colonization of tick vector Ixodes scapularis (Acari: Ixodidae).

Maintenance in nature of Borrelia burgdorferi, the pathogenic bacterium that causes Lyme disease, requires transmission through an infectious cycle that includes a tick vector and a mammalian host. The genetic requirements for persistence in these disparate environments have not been well defined. B. burgdorferi has a complex genome composed of a chromosome and >20 plasmids. Previous work has demonstrated that B. burgdorferi requires two plasmids, lp25 and lp28-1, in the mammalian host. To investigate the requirement for these same two plasmids during tick infection, we experimentally infected

A new member of the trombiculid mite family Neotrombicula nagayoi (Acari: Trombiculidae) induces human dermatitis.

We present the first definitive evidence that the mite Neotrombicula nagayoi bites humans under natural conditions in Japan. Initially, bites resulted in mild pruritus without pain. However, skin reactions increased gradually year by year with severe pruritus with pain being reported by the victim after being bitten repeatedly. Six species of trombiculid mites comprising three genera were isolated from soil samples collected from August to October in both 2001 and 2002 at a study site where a man was bitten by N. nagayoi. The dominant species was L. intermedium (72.4%) followed by L. pallidum

Larval Gulf Coast ticks (Amblyomma maculatum) [Acari: Ixodidae] as host for Hepatozoon americanum [Apicomplexa: Adeleorina].

Laboratory-reared larval Gulf Coast ticks (GCTs) (Amblyomma maculatum) were exposed experimentally and found to acquire Hepatozoon americanum infection while feeding on parasitemic dogs. These ticks supported gamogonic and sporogonic development of the apicomplexan, and oocysts from newly molted nymphs were infectious for a dog. Other nymphs from this cohort that were allowed to feed on a blood-parasite naive sheep molted normally; the resulting adult ticks contained oocysts that were infectious for another dog. Merogonic development of H. americanum in the dogs and the resulting lesions/disea

Vertical transmission of Orientia tsutsugamushi in two lines of naturally infected Leptotrombidium deliense (Acari: Trombiculidae).

Vertical transmission of Orientia tsutsugamushi (Hayashi), the etiologic agent for scrub typhus, was studied in two lines of naturally infected Leptotrombidium deliense Walch. In one line of mites originating from a single adult (V3M), the rate of filial transmission was 100% for the first two laboratory generations, but declined to 86.6% in the third laboratory generation. The vertical infection rate in this line of mites was 100% for the parental generation, but declined to 95.6% for the F1 generation and 88.6% for F2. Impact of microclimate on immature tick-rodent host interactions (Acari: Ixodidae): implications for parasite transmission.

Rodents play a significant role in enzootic cycles of tick-borne pathogens, notably, in the northern hemisphere, tick-borne encephalitis virus and Lyme borreliosis spirochaetes. The relative numbers of nymphal and larval ticks feeding on rodents are crucial variables in determining the probability of rodent infection and the degree of amplification of infection prevalence in the tick population. Manipulation of the microclimate within quasinatural experimental arenas revealed that under increasingly dry conditions the numbers of unfed nymphal Ixodes ricinus L. questing in upper layers of the h

Ectoparasitic mites (Acari) of sympatric Brazilian free-tailed bats and big brown bats in Alabama.

Seven species of mites were recovered from 133 Brazilian free-tailed bats, Tadarida brasiliensis, and 94 big brown bats, Eptesicus fuscus, from February through November 1990 in colonies that shared roosting space in east-central Alabama. The macronyssid Chiroptonyssus robustipes (Ewing) was the most common mite on T. brasiliensis (964 mites, 87% of bats infested) and on E. fuscus (109 mites, 29% of bats infested). However, C. rubustipes normally is a specific parasite of T. brasiliensis. The macronyssids Steatonyssus ceratognathus (Ewing) and S. occidentalis (Ewing) were recovered from both s

Efficacy of three pyrethroids against Leptotrombidium fletcheri (Acari: Trombiculidae) infected and noninfected with scrub typhus.

Toxicities of three pyrethroids, d-phenothrin, decamethrin, and permethrin, were evaluated in the laboratory against Leptotrombidium fletcheri (Womersley & Heaslip). The susceptibilities between populations of the species infected and noninfected with scrub typhus were investigated. The three pesticides exhibited different toxicities to the chiggers. D- phenothrin was the most toxic, followed by decamethrin, then permethrin. There were no significant differences between susceptibilities of the infected and noninfected populations. Log-probit regression lines indicated that the species was most

Transmission of Demodex flagellurus (Acari: Demodicidae) in the house mouse, Mus musculus, under laboratory conditions.

Transmission of Demodex flagellurus Bukva, 1985 inhabiting the preputial and clitoral glands of the house mouse, Mus musculus L., was studied in mating experiments using pairs of infested, feral X mite-free, white laboratory mice. Spontaneous transfer of mites between sexually active hosts of the opposite sex, presumably through copulation, is reported for the first time for the hair-follicle mites under controlled conditions. Transfer of mites from host mother to her progeny was also proven. A comparatively low transmission effectiveness was observed in both modes of transmission, being 9.5%

Computer simulation of Rocky Mountain spotted fever transmission by the American dog tick (Acari: Ixodidae).

A computer model was developed for simulation of the transmission of Rickettsia rickettsii, the causative agent of Rocky Mountain spotted fever (RMSF), by the American dog tick, Dermacentor variabilis (Say). The model of RMSF was combined with a model for population dynamics of the American dog tick and included simulation of infection and transmission of rickettsiae between ticks and host mammals and transmission of RMSF to humans. The model simulated the effects of biotic and environmental variables such as weather, host density, habitat, transovarial transmission, fecundity of infected tick

Comparison of cryofixation methods for skin biopsies from sheep infested with Psoroptes ovis (Acari: Psoroptidae).

Two methods of cryofixing skin samples from sheep infested with Psoroptes ovis were compared. In one method the skin biopsy was placed in a capped vial before immersion, while the other allowed direct contact with liquid nitrogen. No statistically significant differences were found between the cryofixation methods with regard to location of P. ovis sections or engaged mouthparts. Direct contact with liquid nitrogen is recommended as the preferred method for cryofixing epidermal feeding ectoparasites on skin samples.

Mating behavior and preliminary evidence of putative odor-mediated interactions in Raoiella indica (Acari: Tenuipalpidae).

Odour and contact chemical traces play central roles in the sexual communication of mites, mediating partner encounter and recognition of the female reproductive status. We investigated the mating behavior of Raoiella indica Hirst and the role of female-associated chemical cues in male attraction. Behavioral observations were conducted under controlled laboratory conditions, and three complementary bioassays were performed: (i) detailed recording of courtship and copulation sequences, (ii) two-choice tests to evaluate male preference for arenas associated with females at different reproductive

Haemaphysalis longicornis (Acari: Ixodidae) failed to acquire and consequently transmit Babesia bigemina to naïve cattle.

Since its introduction into the United States, the Asian longhorned tick (Haemaphysalis longicornis) has spread to several states and poses a growing threat to livestock due to its broad host range and rapid reproductive capacity. Since H. longicornis can transmit livestock pathogens such as Theileria orientalis Ikeda, it is a high priority to determine the vector competence of a United States population of H. longicornis for other livestock pathogens that circulate or may re-emerge domestically, such as Babesia bigemina, the causative agent of bovine babesiosis. This study evaluated the vecto

Genome report: whole-genome assembly of the relapsing fever tick Ornithodoros turicata Dugès (Acari: Argasidae).

The soft tick family Argasidae contains vectors of medical and veterinary importance, but few molecular resources are available compared to hard ticks (Ixodidae). One example is Ornithodoros turicata, a recognized vector of Borrelia turicatae, causal agent of human relapsing fever, and a putative vector of African swine fever virus. To address the current lack of molecular resources for the Argasidae, we generated a chromosome-level genome assembly for O. turicata using PacBio sequencing in conjunction with an Illumina Hi-C library. The resulting reference genome has a total of 1.1 Gb in lengt

Artificial infestation of white-tailed deer with ticks (Acari: Ixodidae) to study tick-host interactions.

White-tailed deer (Odocoileus virginianus) are a main host for the adult life stages of tick species of medical and veterinary importance. Since white-tailed deer play a vital role in tick ecology, research has been conducted to understand this tick-host relationship. To date, research involving captive white-tailed deer and artificial infestation of these animals with ticks has focused on host suitability, the role of white-tailed deer in tick-borne diseases, and anti-tick vaccine research. The methodology reported for these studies was at times not descriptive and inconsistent regarding how
