O-GlcNAc modification in diabetes and Alzheimer's disease.

Similar to phosphorylation, O-GlcNAcylation (or simply GlcNAcylation) is an abundant, dynamic, and inducible post-translational modification. In some cases, GlcNAcylation and phosphorylation occur at the same or adjacent sites, modulating each other. GlcNAcylated proteins are crucial in regulating virtually all cellular processes, including signaling, cell cycle, and transcription, among others. GlcNAcylation affects protein-protein interactions, activity, stability, and expression. Several GlcNAcylated proteins are involved in diabetes and Alzheimer's disease. Hyperglycemia increases GlcNAcyl

[Measurement of bile acid N-acetylglucosaminides in serum and urine of patients with chronic liver diseases during ursodeoxycholic acid treatment].

Bile acid N-acetylglucosaminide (GlcNAc) was investigated in serum and urine of patients with chronic liver diseases during ursodeoxycholic acid (UDCA) treatment by HPLC. Bile acid N-acetylglucosaminide was not detected other than primary biliary cirrhosis in serum and urine. All of these N-acetyglucosaminide was ursodeoxycholic acid N-acetylglucosaminide (GlcNAc-UDCA). GlcNAc-UDCA excretion was 17.3 +/- 5.7 mg/day in PBC stage I, 12.1 +/- 5.4 mg/day in stage II, 39.1 +/- 20.8 mg/day in stage III. Among the GlcNAc fraction GlcNAc-UDCA-gly occupied greatest part followed by GlcNAc-UDCA-tau and

Pretibial myxoedema: stimulation of mucopolysaccharide production of fibroblasts by serum.

We have shown that sera from normal individuals and from patients with pretibial myxoedema contain a factor which simulates mucopolysaccharide biosynthesis in normal human skin fibroblasts cultured in vitro. This factor was present in larger amounts in sera of patients with pretibial myxoedema. The role of growth stimulating factors in serum is reviewed and a hypothesis is put forward that the fibroblast stimulating factor is somatomedin and that its presence in increased amounts in thyroid disease may lead to pretibial myxoedema.

Lectin-histochemical study of O-linked glycoconjugates in dysplastic retina of Norrie disease.

The carbohydrate chains of O-linked glycoconjugates in dysplastic retina from a Japanese female infant with Norrie disease were examined by lectin histochemistry. The avidin-biotinylated peroxidase method was used. The retina was highly dysplastic and composed of undifferentiated embryonic tissues containing a number of rosettes of varying sizes. The lumina of the rosettes were stained by peanut agglutinin, which recognizes the Gal beta 1,3GalNAc sequence of O-linked glycans. However, the lumina were not labeled by wheat germ agglutinin, which reacts with sialic acid and/or N-acetylglucosamine

Characterization of the subunits and sugar moiety of human placental and leukemic beta-glucuronidase.

beta-Glucuronidase purified from human placenta and chronic myelogenous leukemic cells was composed of three components of 18, 64 and 80 kDa, though the relative contents of the components were different between the sources. Analysis of their N-terminal amino-acid sequences showed that the 18-kDa and 64-kDa components were derived from the 80-kDa component by cleavage between Val159 and Gly160. Furthermore, the enzyme was found to be glycosylated at Asn173 and Asn420 with high mannose-type oligosaccharides, based on the electrophoretic mobility of the components as well as the endopeptidic pep

beta-N-acetylhexosaminidases in the spleen of a patient with hairy-cell leukaemia.

The spleen from a patient with hairy-cell leukaemia had beta-N-acetylhexosaminidase activity that could be resolved into three isoenzymes by chromatography on phenyl boronate agarose. Two of these were the major forms, A and B, found in normal tissues but, in addition, there was an 'extra' form that accounted for 15% of total activity. The 'extra' form hydrolysed the synthetic substrate 4-methylumbelliferyl-beta-N-acetylglucosamine 6-sulphate, indicating the presence of alpha-subunits. It was more acidic than A, was less heat-stable and showed no generation of B on denaturation under a variety

Hypomorphic homozygous mutations in phosphoglucomutase 3 (PGM3) impair immunity and increase serum IgE levels.

BACKGROUND: Recurrent bacterial and fungal infections, eczema, and increased serum IgE levels characterize patients with the hyper-IgE syndrome (HIES). Known genetic causes for HIES are mutations in signal transducer and activator of transcription 3 (STAT3) and dedicator of cytokinesis 8 (DOCK8), which are involved in signal transduction pathways. However, glycosylation defects have not been described in patients with HIES. One crucial enzyme in the glycosylation pathway is phosphoglucomutase 3 (PGM3), which catalyzes a key step in the synthesis of uridine diphosphate N-acetylglucosamine, whic

Crystal structure of a heterodimer of phospholipase A2 from Naja naja sagittifera at 2.3 A resolution reveals the presence of a new PLA2-like protein with a novel cys 32-Cys 49 disulphide bridge with a bound sugar at the substrate-binding site.

The crystal structure of the phospholipase A2 (PLA2) heterodimer from Naja naja sagittifera reveals the presence of a new PLA2-like protein with eight disulphide bridges. The heterodimer is formed between a commonly observed group I PLA2 having seven characteristic disulfide bonds and a novel PLA2-like protein (Cys-PLA2) containing two extra cysteines at two highly conserved sites (positions 32 and 49) of structural and functional importance. The crystals of the heterodimer belong to tetragonal space group P41212 with cell dimensions, a = b = 77.7 A and c = 68.4 A corresponding to a solvent co

Biosynthesis in vitro of Caenorhabditis elegans phosphorylcholine oligosaccharides.

The biosynthesis in vitro of phosphorylcholine oligosaccharides in Caenorhabditis elegans has been investigated. Here we show that extracts of C. elegans' microsomes transfer phosphorylcholine from L-alpha-dipalmitoyl phosphatidylcholine to hybrid and complex type N-linked oligosaccharides containing mannose residues disubstituted with N-acetylglucosamine. The reaction products are consistent with structures reported for C. elegans as well those found in the filarial nematodes Acanthocheilonema viteae, Onchocerca volvulus, and Brugia malayi, strongly supporting the concept that the phosphorylc

MK-8719, a Novel and Selective O-GlcNAcase Inhibitor That Reduces the Formation of Pathological Tau and Ameliorates Neurodegeneration in a Mouse Model of Tauopathy.

Deposition of hyperphosphorylated and aggregated tau protein in the central nervous system is characteristic of Alzheimer disease and other tauopathies. Tau is subject to O-linked N-acetylglucosamine (O-GlcNAc) modification, and O-GlcNAcylation of tau has been shown to influence tau phosphorylation and aggregation. Inhibition of O-GlcNAcase (OGA), the enzyme that removes O-GlcNAc moieties, is a novel strategy to attenuate the formation of pathologic tau. Here we described the in vitro and in vivo pharmacological properties of a novel and selective OGA inhibitor, MK-8719. In vitro, this compoun

Lectin binding sites on the luminal surface of ependymal cells of the rat spinal cord: implications for neuropathological investigation.

Twenty fluorescein isothiocyanate-labeled lectins were applied to formalin-fixed, paraffin-embedded sections of rat spinal cord to delineate the specific lectin binding sites on the ependymal cells. Only two lectins, Solanum tuberosum agglutinin (STA) and wheat germ agglutinin (WGA), were found to bind intensely to the ciliated surface of rat ependymal cells. Inhibition studies indicated that terminal N-acetylglucosamine and sialic acid were abundantly present on the ciliated surface of ependymal cells and accounted for the binding of these two lectins. Experimentally induced hydromyelia was a

Bisecting GlcNAc in N-glycan regulates synaptic maturation.

Bisecting GlcNAc, a central branch in N-glycans synthesized by the N-acetylglucosaminyltransferase III (MGAT3; also known as GnT-III), is highly expressed in neurons. Although pathological roles of MGAT3 and bisecting GlcNAc have been demonstrated in Alzheimer's disease, their physiological functions in neurons remain unclear. To identify their physiological functions, here we examine the morphology of neurons in knockout (Mgat3-/-) mouse brains and investigate the expression and localization of bisecting GlcNAc-bearing glycoproteins involved in neuronal activity and morphogenesis. We found im

1H NMR studies of molecular interaction of d-glucosamine and N-acetyl-D-glucosamine with curcumin and caffeic acid phenethyl ester in DMSO.

Chitosan (Cs)-based polymers have been explored as potential drug-delivery systems that could enhance the practical applications of bioactive phenolic (Ph) substances, such as curcumin (CUR) and caffeic acid phenethyl ester (CAPE). In this study, we focused on designing CS-based drug carriers by examining the intermolecular interactions between the polymer components, d-glucosamine (Gn) and N-acetyl-d-glucosamine (AGn), and the target substances CUR and CAPE through 1H NMR titration in dimethyl sulfoxide (DMSO‑d6). The observed changes in chemical shifts indicated that Gn cation (GnH+) forms m

Increased protein O-GlcNAcYLATION in human placentas from hypertensive pregnancies.

INTRODUCTION: Key proteins involved in placentation are susceptible to O-linked β-N-acetylglucosamine modification (O-GlcNAcylation). Anomalies in protein O-GlcNAcylation have been associated with pathological conditions, including hypertension, and it is known that arterial hypertension negatively impacts placental function. However, the precise impact of protein O-GlcNAcylation on placental function and fetal growth remains unclear in humans. Therefore, the current study aimed to investigate O-GlcNAcylation expression, and its catalytic enzyme O-GlcNAc transferase (OGT), in the placenta of p

Synthesis and antiviral activity of 1,2,3-triazolyl nucleoside analogues with N-acetyl-d-glucosamine residue.

A series of 1,2,3-triazolyl nucleoside analogues bearing N-acetyl-D-glucosamine residue was synthesized by the copper-catalyzed alkyne-azide cycloaddition (CuAAC) reaction of N1-ω-alkynyl derivatives of uracil, 6-methyluracil, thymine and 3,4,6-tri-O-acetyl-2-deoxy-2-acetamido-β-D-glucopyranosyl azide. Antiviral assays revealed the lead compound 3f which showed both the same activity against the influenza virus A H1N1 (IC50=70.7 µM) as the antiviral drug Rimantadine in control (IC50=77 µM) and good activity against Coxsackievirus B3 (IC50=13.9 µM) which was one and a half times higher than the

N-acetylglucosaminylation of serine-aspartate repeat proteins promotes Staphylococcus aureus bloodstream infection.

Staphylococcus aureus secretes products that convert host fibrinogen to fibrin and promote its agglutination with fibrin fibrils, thereby shielding bacteria from immune defenses. The agglutination reaction involves ClfA (clumping factor A), a surface protein with serine-aspartate (SD) repeats that captures fibrin fibrils and fibrinogen. Pathogenic staphylococci express several different SD proteins that are modified by two glycosyltransferases, SdgA and SdgB. Here, we characterized three genes of S. aureus, aggA, aggB (sdgA), and aggC (sdgB), and show that aggA and aggC contribute to staphyloc

Biosynthesis of enterobacterial common antigen in Escherichia coli. Biochemical characterization of Tn10 insertion mutants defective in enterobacterial common antigen synthesis.

Twelve independent Tn10 insertion mutants of Escherichia coli K12 were isolated that were defective in the synthesis of enterobacterial common antigen (ECA). The mutants were identified by screening a random pool of Tn10 insertion mutants for their ECA phenotype using a colony-immunoblot assay. All 12 of the Tn10 insertion mutants were found to be located in the chromosomal region of the rff-rfe genes. Four of the Tn10 insertions were in rfe genes while the remaining eight Tn10 insertions were in rff genes. All of the rfe::Tn10 insertion mutants were defective in the synthesis of GlcNAc-pyroph
